Solriamfetol enhances wakefulness and improves cognition and anxiety in a murine model of OSA.

BACKGROUND: Obstructive sleep apnea (OSA) is a chronic condition characterized by intermittent hypoxia (IH). Excessive daytime sleepiness (EDS) is a common consequence of OSA and is associated with cognitive deficits and anxiety. Modafinil (MOD) and Solriamfetol (SOL) are potent wake-promoting agents clinically used to improve wakefulness in OSA patients with EDS. METHODS: Male C57Bl/6J mice were exposed to either IH or room air (RA) controls during the light phase for 16 weeks. Both groups were then randomly assigned to receive once-daily intraperitoneal injections of SOL (200 mg/kg), MOD (2…

The use of amantadine in patients with unresponsive wakefulness syndrome after severe cerebral hemorrhage.

BACKGROUND: Amantadine is currently recommended for use in patients of post-traumatic brain injury with unresponsive wakefulness syndrome (UWS). However, the application of amantadine in UWS after cerebral hemorrhage has only been rarely reported. This allows for a further exploration of the role of amantadine in the treatment of UWS resulting from a severe cerebral hemorrhage. METHODS: We observed the changes of seven patients with UWS of intracerebral hemorrhage after taking amantadine. We also carried out a detailed neurological examination of the patient with disorders of consciousness to…

Using tracheal breathing sounds and anthropometric information for screening obstructive sleep apnoea during wakefulness.

Obstructive sleep apnoea (OSA) is a common yet underdiagnosed disorder. Undiagnosed OSA significantly increases perioperative morbidity and mortality for OSA patients undergoing surgery, requiring full anaesthesia. Tracheal breathing sounds characteristics during wakefulness have shown a high correlation with the apnoea-hypopnea index (AHI), while they are also affected by the anthropometric parameters, e.g., sex, age, etc. This study investigates the effects of the anthropometric parameters on our new quick objective OSA screening tool during wakefulness. Breathing sounds of 122 individuals (…

Maintenance of Wakefulness Test scores and driving performance in sleep disorder patients and controls.

OBJECTIVE: Sleepiness at the wheel is a risk factor for traffic accidents. Past studies have demonstrated the validity of the Maintenance of Wakefulness Test (MWT) scores as a predictor of driving impairment in untreated patients with obstructive sleep apnea syndrome (OSAS), but there is limited information on the validity of the maintenance of wakefulness test by MWT in predicting driving impairment in patients with hypersomnias of central origin (narcolepsy or idiopathic hypersomnia). The aim of this study was to compare the MWT scores with driving performance in sleep disorder patients and …

[Effect of apomorphine on the wakefulness--sleep cycle of the common frog Rana temporaria].

This work considers effects of introduction into spinal lymphatic sac of dopamine agonist--apomorphine-(APO) at doses of 0.1, 1.0, 2.0 and 4.0 mg/kg body weight on the common frog wakefulness-sleep cycle (WSC). Usually the frog WSC is represented by wakefulness and three types of passive-protective behavior: by immobility states of the type of catalepsy, catatonia, and cataplexy that are characterized by high thresholds of arousal and by different (corresponding to the name) skeletal musculature tones. These immoboloty forms are considered as homologues of mammalian stress-reaction, hibernatio…

The dual role of the orexin/hypocretin system in modulating wakefulness and respiratory drive.

PURPOSE OF REVIEW: Today, numerous studies show that orexin peptides act as regulators of many functions including the control of sleep-wake … othalamic orexinergic neurons is subjected to maturation and is mandatory to maintain long bouts of wakefulness in adults. At wake onset, this activity progressively builds up as a result of synaptic interactions and reinforces the awake state. Orexin deficiency attenuates the hypercapnic reflex only during wakefulness and is correlated with an increase in sl …

Effect of uvulopalatopharyngoplasty on work of breathing during wakefulness in obstructive sleep apnea syndrome.

OBJECTIVES: We evaluated the effects of uvulopalatopharyngoplasty (UPPP) on the work of breathing (WOB) in obstructive sleep apnea syndrome (OSAS). METHODS: Fifteen healthy subjects and 30 subjects with OSAS who desired UPPP were prospectively enrolled. All underwent measurement of WOB while awake as well as in a sleep study. These studies were repeated 3 months after UPPP in the patients with OSAS. RESULTS: In OSAS before UPPP, the WOB while supine was increased above that of normal subjects. After UPPP, the WOB while supine remained elevated in those whose OSAS did not respond to surgery,

[Effect of swinging on EEG of rats of juvenile age in the wakefulness state].

Simultaneous recording of the EEG activity of superficial cortical and deep (caudate nucleus, dorsal hippocampus, anterior hypothalamus) brain parts has been performed for the first time after a 2-h swinging of frequency of 0.2 Hz in Wistar rats of juvenile age. Swinging was produced on a 4-bar parallel swing. Using a Neuron-Spectr electroencephalograph and a Diana program, normalized power spectra of wave EEG components, synchronization coefficients, and coefficients of cross-correlation between bioelectrical potentials of various brain structures were determined. After a 2-h swinging, the me

Impact of motivation on Multiple Sleep Latency Test and Maintenance of Wakefulness Test measurements.

BACKGROUND: The Multiple Sleep Latency Test (MSLT) and Maintenance of Wakefulness Test (MWT) are standard clinical tests used to measure sleep tendency in clinical populations in which test results may lead to significant life change for patients. Loss of a driving license or drug seeking may provide significant motivation to patients to obtain needed results on these tests. In the current study, the effect of motivation on the ability to fall asleep or stay awake was examined. METHODS: Twelve subjects spent 3 nights and the following days in the laboratory. On the day following the first lab…

Hemispheric lateralization of the EEG during wakefulness and REM sleep in young healthy adults.

EEG recordings confirm hemispheric lateralization of brain activity during cognitive tasks. The aim of the present study was to investigate spontaneous EEG lateralization under two conditions, waking and REM sleep. Bilateral monopolar EEG was recorded in eight participants using a 12-electrode montage, before the night (5 min eyes closed) and during REM sleep. Spectral analysis (0.75-19.75 Hz) revealed left prefrontal lateralization on total spectrum amplitude power and right occipital lateralization in Delta activity during waking. In contrast, during REM sleep, right frontal lateralization i

Respiratory function during wakefulness and sleep among survivors of respiratory and non-respiratory poliomyelitis.

The purpose of this study was to determine whether there is a difference in respiratory mechanics and gas exchange between polio survivors and healthy, age-matched controls during wakefulness and sleep. Polio survivors were divided into four groups. The first group included those who had evidence of respiratory muscle involvement originally (PRM) and the second group included those who had bulbar muscle involvement originally (PBM). The third and fourth groups had only limb involvement originally but were separated by absence (PSL) or presence of a scoliosis (PSS) at the time of their evaluati…

[Somatosensory evoked potentials in the sleep-wakefulness cycle in healthy subjects and narcolepsy patients].

Studies of somatosensory evoked potentials (SSEPs) were conducted in various functional states of sleep-alertness cycle (relaxed alertness, 2-nd and delta-stages of slow sleep, rapid sleep) in 7 healthy subjects and 8 patients with polysymptom narcolepsy. Integrated amplitude (IA) was calculated in poststimulus intervals, accordingly to SSEPs division into groups of early (20-80 ms), mean (80-200 ms) and late (200-400 ms) components. It has been shown that in patients with polysymptom narcolepsy IA of all SSEPs components in alertness was lower than in healthy subjects; during sleep higher IA

Abnormally abrupt transitions from sleep-to-wake in Huntington's disease sheep (Ovis aries) are revealed by automated analysis of sleep/wake transition dynamics.

Sleep disturbance is a common and disruptive symptom of neurodegenerative diseases such as Alzheimer's and Huntington's disease (HD). In HD patients, sleep fragmentation appears at an early stage of disease, although features of the earliest sleep abnormalities in presymptomatic HD are not fully established. Here we used novel automated analysis of quantitative electroencephalography to study transitions between wake and non-rapid eye movement sleep in a sheep model of presymptomatic HD. We found that while the number of transitions between sleep and wake were similar in normal and HD sheep, t

Why Does Sleep Slow-Wave Activity Increase After Extended Wake? Assessing the Effects of Increased Cortical Firing During Wake and Sleep.

UNLABELLED: During non-rapid eye movement (NREM) sleep, cortical neurons alternate between ON periods of firing and OFF periods of silence. This bi-stability, which is largely synchronous across neurons, is reflected in the EEG as slow waves. Slow-wave activity (SWA) increases with wake duration and declines homeostatically during sleep, but the underlying mechanisms remain unclear. One possibility is neuronal "fatigue": high, sustained firing in wake would force neurons to recover with more frequent and longer OFF periods during sleep. Another possibility is net synaptic potentiation during w

Fluctuations between sleep and wakefulness: wake-like features indicated by increased EEG alpha power during different sleep stages in nightmare disorder.

Although a growing body of research indicates that frequent nightmares are related to impaired sleep regulation, the pathophysiology of nightmare disorder is far from being fully understood. We examined the relative spectral power values for NREM and REM sleep separately in 19 individuals with nightmare disorder and 21 healthy controls, based on polysomnographic recordings of the second nights' laboratory sleep. Nightmare subjects compared to controls exhibited increased relative high alpha (10-14.5Hz) and fronto-central increases in high delta (3-4Hz) power during REM sleep, and a trend of in
