Xeroderma pigmentosum: squamous cell carcinoma of the tongue.

A case report is presented of a 9-year-old boy with xeroderma pigmentosum complementation group C, who died from a squamous cell carcinoma arising from the tip of the tongue. Treatment with etretinate and in a later phase with indomethacin and prednisolone was not effective, except for a limited initial response of the tumour to etretinate.

Susceptibility of Xeroderma pigmentosum cells to transformation with oncogenes.

Oncogenes capable of transforming 3T3-Vill cells were not detected in 'normal' Xeroderma pigmentosum (XP) fibroblasts but were detected in two out of six XP epitheliomas. Preliminary results concerning the transfection of 'normal' XP fibroblasts with activated ras genes seem to indicate that these cells are as resistant as the healthy controls to the transforming action of the group II oncogenes. However, after transfection with v-myb oncogene in XP fibroblasts several cellular clones have been isolated showing some new phenotypic characteristics.

Hypersensitivity of xeroderma pigmentosum cells to dietary carcinogens.

Xeroderma pigmentosum patients, in addition to ultraviolet-induced skin cancers, have an increased prevalence of neoplasms occurring in sites shielded from ultraviolet radiation. We postulated that these internal neoplasms might be related to ingestion of dietary carcinogens. As model dietary carcinogens, we studied the tryptophan pyrolysis products, 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indole (Trp-P-1) and 3-amino-1-methyl-5H-pyrido[4,3-b]indole (Trp-P-2). These dietary compounds bind to DNA and are highly mutagenic and carcinogenic. Cytotoxicity of these compounds was examined in cultured ly…

Multiple melanomas occurring in a patient with xeroderma pigmentosum.

Xeroderma pigmentosum (XP) is associated with an increased frequency of cutaneous malignancies including malignant melanoma. We report a patient who developed eleven primary melanomas and numerous highly atypical nevi in addition to the basal cell carcinomas and squamous cell carcinomas more commonly seen in patients with XP.

Abnormal response of xeroderma pigmentosum cells to bleomycin.

The repair of bleomycin-damaged DNA was examined in human fibroblasts isolated from patients having the disease xeroderma pigmentosum (XP). In normal fibroblasts, the appearance of low-molecular-weight DNA was observed in the presence of increasing amounts of the drug. The studies in XP fibroblasts produced results which differed from those obtained in normal cells in two ways. (a) Prelabeled XP cells from most complementation groups contained more low-molecular-weight DNA than observed in the other human fibroblasts examined. (b) When XP cells were exposed to low doses of bleomycin, the low-m…
