Biosynthesized silver and gold nanoparticles are potent antimycotics against opportunistic pathogenic yeasts and dermatophytes.

BACKGROUND: Epidemiologic observations indicate that the number of systemic fungal infections has increased significantly during the past decades, however in human mycosis, mainly cutaneous infections predominate, generating major public health concerns and providing much of the impetus for current attempts to develop novel and efficient agents against cutaneous mycosis causing species. Innovative, environmentally benign and economic nanotechnology-based approaches have recently emerged utilizing principally biological sources to produce nano-sized structures with unique antimicrobial properti

Fundamental niche prediction of the pathogenic yeasts Cryptococcus neoformans and Cryptococcus gattii in Europe.

Fundamental niche prediction of Cryptococcus neoformans and Cryptococcus gattii in Europe is an important tool to understand where these pathogenic yeasts have a high probability to survive in the environment and therefore to identify the areas with high risk of infection. In this study, occurrence data for C. neoformans and C. gattii were compared by MaxEnt software with several bioclimatic conditions as well as with soil characteristics and land use. The results showed that C. gattii distribution can be predicted with high probability along the Mediterranean coast. The analysis of variables …

Quantitative Microplate-Based Growth Assay for Determination of Antifungal Susceptibility of Histoplasma capsulatum Yeasts.

Standardized methodologies for determining the antifungal susceptibility of fungal pathogens is central to the clinical management of invasive fungal disease. Yeast-form fungi can be tested using broth macrodilution and microdilution assays. Reference procedures exist for Candida species and Cryptococcus yeasts; however, no standardized methods have been developed for testing the antifungal susceptibility of yeast forms of the dimorphic systemic fungal pathogens. For the dimorphic fungal pathogen Histoplasma capsulatum, susceptibility to echinocandins differs for the yeast and the filamentous …

Screening of plant extracts for antimicrobial activity against bacteria and yeasts with dermatological relevance.

There is cumulative resistance against antibiotics of many bacteria. Therefore, the development of new antiseptics and antimicrobial agents for the treatment of skin infections is of increasing interest. We have screened six plant extracts and isolated compounds for antimicrobial effects on bacteria and yeasts with dermatological relevance. The following plant extracts have been tested: Gentiana lutea, Harpagophytum procumbens, Boswellia serrata (dry extracts), Usnea barbata, Rosmarinus officinalis and Salvia officinalis (supercritical carbon dioxide [CO2] extracts). Additionally, the followin…

[Rapid identification and susceptibility to killer toxins of yeasts isolated from non-systemic mycoses].

Rapid identification and susceptibility to killer toxins of yeasts isolated from non-systemic mycoses. The use of quick and reliable yeast identification methods, as well as the development of new antifungal agents with more specific targets, will enable a more efficient treatment of mycoses. In the present work, a total of 53 clinical isolates obtained from non-systemic infections in Neuquén Hospitals and an ophthalmologic clinic in Buenos Aires during 2005, were identified by means of a rapid molecular method (ITS1-5.8S ADNr-ITS2 PCR-RFLP). Additionally, the killer susceptibility of the isol…

[Yeasts of the genus Malassezia: taxonomic classification and significance in (veterinary and) clinical medicine].

The historical development of the taxonomic classification of Malassezia yeasts until today yielded the description of seven different species based upon molecularbiological, morphological and biochemical parameters (M. furfur, M. pachydermatis, M. sympodialis, M. globosa, M. obtusa, M. restricta, M. slooffiae). Tests like Cremophor EL assimilation, esculin degradation, catalase test, pigment production and determination of polidocanol sensitivity appear to be suitable procedures for routine identification of the different species. Their colonization of clinically healthy humans (in particular…

The effect of UV-light on pityrosporum yeasts: ultrastructural changes and inhibition of growth.

The effect of UV-light on Pityrosporum yeasts (P.) was studied: P. yeasts cultured from the skin were spread on Dixon plates and irradiated with different UVB-and UVA-light dosages and read after three days, controls were not irradiated. Also P. yeasts, immediately after irradiation, were isolated from the plates and studied with an electron microscope. A significant growth inhibition or no growth at all was seen after 25, 50, 75 J/cm2 UVA and 900 mJ/cm2 UVB, a moderate inhibition after irradiation with 250 mJ/cm2 UVB. The growth inhibition was paralleled by ultrastructural degenerative altera…

The 'tape-method': a new and simple method for quantitative culture of Pityrosporum yeasts.

A new method for quantitative culture of Pityrosporum yeasts, the 'tape-method', is presented. Samples for culture were taken from the skin by stripping with 1 cm2 of tape, whereafter the tapes were placed over a drop of sterile olive oil which was pipetted on a Sabouraud medium. Plates were incubated at 37 degrees C, and after 7 days the numbers of Pityrosporum colonies growing under the tapes were counted. With this method a difference in numbers of Pityrosporum colonies between seborrheic dermatitis and normal skin could be discerned. This difference was significant. It appeared that two su…

Evolution of yeasts and lactic Acid bacteria during fermentation and storage of bordeaux wines.

The levels of yeasts and lactic acid bacteria that naturally developed during the vinification of two red and two white Bordeaux wines were quantitatively examined. Yeasts of the genera Rhodotorula, Pichia, Candida, and Metschnikowia occurred at low levels in freshly extracted grape musts but died off as soon as fermentation commenced. Kloeckera apiculata (Hanseniaspora uvarum), Torulopsis stellata, and Saccharomyces cerevisiae, the dominant yeasts in musts, proliferated to conduct alcoholic fermentation. K. apiculata and eventually T. stellata died off as fermentation progressed, leaving S. c…

Two budding yeast RAD4 homologs in fission yeast play different roles in the repair of UV-induced DNA damage.

We have identified two fission yeast homologs of budding yeast Rad4 and human xeroderma pigmentosum complementation group C (XP-C) correcting protein, designated Rhp4A and Rhp4B. Here we show that the rhp4 genes encode NER factors that are required for UV-induced DNA damage repair in fission yeast. The rhp4A-deficient cells but not the rhp4B-deficient cells are sensitive to UV irradiation. However, the disruption of both rhp4A and rhp4B resulted in UV sensitivity that was greater than that of the rhp4A-deficient cells, revealing that Rhp4B plays a role in DNA repair on its own. Fission yeast h

Integrated metabolite, volatile, and genome annotation profiling reveals distinct functional phenotypes in cocoa-derived yeasts.

Cocoa fermentation shapes the precursor chemistry underlying chocolate flavour, but the isolate-level contributions of cocoa-associated yeasts remain incompletely resolved. This study compared six cocoa-derived yeasts with a reference Saccharomyces cerevisiae strain by profiling sugar utilisation, organic-acid turnover, volatile formation, and targeted genome annotation under controlled cocoa-relevant conditions. In cocoa pulp extract-based medium, Kluyveromyces marxianus Y1A, Meyerozyma guilliermondii Y1, and Ogataea polymorpha Y3A showed the strongest sugar depletion and organic-acid transfo…
