Dopamine transporter-mediated cytotoxicity of beta-carbolinium derivatives related to Parkinson's disease: relationship to transporter-dependent uptake.

Endogenous or exogenous beta-carboline (betaC) derivatives structurally related to the selective dopaminergic neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and its active metabolite 1-methyl-4-phenylpyridinium (MPP(+)) may contribute to dopaminergic neurodegeneration in Parkinson's disease (PD). We addressed the importance of the dopamine transporter (DAT) for selective dopaminergic toxicity by testing the differential cytotoxicity and cellular uptake of 12 betaCs in human embryonic kidney HEK-293 cells ectopically expressing the DAT gene. Cell death was measured using [4,5-Di

Developmental expression profile of the optic atrophy gene product: OPA1 is not localized exclusively in the mammalian retinal ganglion cell layer.

PURPOSE: Autosomal dominant optic atrophy (ADOA) is characterized by primary degeneration of retinal ganglion cells and atrophy of the optic nerve. The OPA1 gene encodes a 960-amino-acid protein. In the current study the temporal and spatial localization of OPA1 were examined in developing and adult murine ocular tissues and the adult human eye. Because the Bst/+ mouse has been postulated as a model of ADOA, the mOPA1 expression in the Bst/+ retina was also examined. METHODS: A polyclonal antibody generated against a C-terminal peptide of OPA1 was used to assess by immunohistochemistry the ex

Antipsychotic drugs antagonize human serotonin type 3 receptor currents in a noncompetitive manner.

The serotonin type 3 (5-HT(3)) receptor is the only ligand-gated ion channel receptor for serotonin (5-HT). 5-HT(3) receptors play an important role in modulating the inhibitory action of dopamine in mesocorticolimbic brain regions. Neuroleptic drugs are commonly thought to exert their psychopharmacological action mainly through dopamine and serotonin type 2 (5-HT(2)) receptors. Except for clozapine, a direct pharmacological interaction of neuroleptics with 5-HT(3) receptors has not yet been described. Using the concentration-clamp technique, we investigated the effects of flupentixol, various

FoxO6, a novel member of the FoxO class of transcription factors with distinct shuttling dynamics.

Forkhead transcription factors of the FoxO-group are associated with cellular processes like cell cycle progression and DNA-repair. FoxO function is regulated by protein kinase B (PKB) via the phosphatidylinositol 3-kinase/PKB survival pathway. Phosphorylation of serine and threonine residues in specific PKB phosphorylation motifs leads to exclusion of FoxO-proteins from the nucleus, which excludes them from exerting transactivating activity. Members of the FoxO-group have three highly conserved regions containing a PKB phosphorylation motif. This study describes the cloning and characterizati

Developmental changes in the structure of the rat fetal lung, with special reference to the airway smooth muscle and vasculature.

Structural changes in the developing rat lung were studied by a combined use of light microscopy including immunohistochemistry for a-smooth muscle actin (alpha-SMA) and scanning electron microscopy (SEM) using the KOH-collagenase digestion method. In the embryonic stage (E11-E13), the lung bud appeared as an outgrowth from the ventral wall of the foregut which grew caudally into the splanchnic mesoderm to form a pair of bronchial buds at the end. At E13, the airway smooth muscle cells first appeared around the bifurcation of the trachea. These smooth muscle cells were restricted to the dorsal

Comparison of two murine models of familial hypertrophic cardiomyopathy.

Although sarcomere protein gene mutations cause familial hypertrophic cardiomyopathy (FHC), individuals bearing a mutant cardiac myosin binding protein C (MyBP-C) gene usually have a better prognosis than individuals bearing beta-cardiac myosin heavy chain (MHC) gene mutations. Heterozygous mice bearing a cardiac MHC missense mutation (alphaMHC(403/+) or a cardiac MyBP-C mutation (MyBP-C(t/+)) were constructed as murine FHC models using homologous recombination in embryonic stem cells. We have compared cardiac structure and function of these mouse strains by several methods to further define m

Periodontal tissue regeneration by combined applications of recombinant human osteogenic protein-1 and bone morphogenetic protein-2. A pilot study in Chacma baboons (Papio ursinus).

Native and recombinant human bone morphogenetic/osteogenic proteins (BMPs/ OPs) singly initiate bone induction in vivo. The finding of synchronous but spatially different BMPs/OPs expression during periodontal tissue morphogenesis suggests novel therapeutic approaches using morphogen combinations based on recapitulation of embryonic development. Twelve furcation defects prepared in the first and second mandibular molars of three adult baboons (Papio ursinus) were used to assess whether qualitative histological aspects of periodontal tissue regeneration could be enhanced and tissue morphogenesi

Avian olfactory receptors: differentiation of olfactory neurons under normal and experimental conditions.

The bird olfactory system has a simple structure and affords an attractive developmental model for the study of olfactory morphogenesis and differentiation. We have cloned and characterized several chick olfactory receptor (COR) genes belonging to the superfamily of seven-transmembrane domain proteins. In situ hybridization analysis of their spatiotemporal patterns of expression during development reveals several important characteristics. COR expression starts early in placodal cells (Embryonic Day 5, E5). Changes in their expression pattern then correlate with the onset of synaptogenesis (E8

Receptivity is a polarity dependent special function of hormonally regulated uterine epithelial cells.

Useful knowledge of the mechanisms which regulate ovoreceptivity and implantation remains elusive in spite of increasing efforts to apply the technology of biochemistry and to a lesser extent, cellular and molecular biology to the analysis of the problem. Existing models used to analyze interactions of the blastocyst and endometrial cells of the uterus have been unable to account for nongenotypic embryonic losses, particularly those following in vitro fertilization and embryo transfer. Separation of endometrial uterine epithelial (UE) and uterine stromal (US) cells was used to demonstrate that

Pathobiology of cortical neurons in metabolic and unclassified amentias.

Visualization of the neuron in its entirety through the use of the rapid Golgi method has permitted detection of several pathobiological features of neurons that are intimately associated with profound mental retardation in infants and children. In cases of unclassified mental retardation, dendrites and particularly dendritic spines exhibit severe developmental abnormalities. Dendritic spines, the postsynaptic components of axospinodendritic synapses, may be absent or abnormally long and thin in retardates. Evidence is presented that some cases of progressive neurobehavioral deterioration in i

Pharmacological characterization and functional divergence of tyramine receptors PxTAR2 and PxTAR1A in the diamondback moth, Plutella xylostella.

Tyramine signaling orchestrates essential physiological processes in insects, yet the functional diversification of tyramine receptors (TARs) and the evolutionary significance of receptor gene duplication remain poorly understood. While most lepidopterans harbor a single TAR1, the diamondback moth Plutella xylostella uniquely possesses two TAR1 paralogs (PxTAR1 and PxTAR1A) alongside PxTAR2, providing an exceptional model to investigate sub-functionalization. However, functional characterization of PxTARs has been limited to TAR1. Here, we uncover striking functional differentiation of TARs th

Incorporation of low-oxidation graphene oxide into electrospun poly(ε-caprolactone) /gelatin scaffolds for human embryonic stem cell-derived retinal pigment epithelium cell transplantation.

Scaffold-based retinal pigment epithelium (RPE) transplantation is a candidate approach to preserve or recover vision for macular degeneration. Electrospun poly(ε-caprolactone) (PCL)/Gelatin (Gel) scaffolds have demonstrated support for cellular proliferation and function, yet their inadequate mechanical performance compromises surgical handling and deliverability. In this study, we incorporated low-oxidation graphene oxide (GO) into electrospun PCL/Gel scaffolds and evaluated their performance for human embryonic stem cell (hESC)-derived RPE cell transplantation. We found that the incorporati

Pregnancy establishment and early embryonic survival in lactating Holstein cows are differentially constrained by parity, body reserves, and embryo transfer.

Pregnancy success in lactating dairy cows depends on both pregnancy establishment and maintenance during early gestation. This study quantified the independent and combined associations of parity, days in milk at service (DIM), body condition score (BCS), milk yield, service number, and reproductive technology, artificial insemination (AI) or embryo transfer (ET), with pregnancy at 28 days (PR28) and embryonic and early fetal loss between days 28 and 50 of gestation (EEFL) in lactating Holstein cows managed under commercial conditions. After exclusion of heifers and incomplete records, 9178 se

Life-Cycle Diphenyl Phosphate (DPhP) Exposure Reveals Sex-Differential Functional Alterations in the Zebrafish Gut Microbiota.

Fish gut microbiota is essential for maintaining host physiological health but is constantly threatened by environmental chemicals. Diphenyl phosphate (DPhP), a major degradation product of organophosphate flame retardants and an environmental pollutant widely detected in aquatic ecosystems, is a chemical of emerging concern; however, few studies have addressed its effects on the gut microbiota. In this study, zebrafish were exposed to environmentally relevant concentrations of DPhP (0.8, 3.9, or 35.6 μg/L) from the embryonic stage to adulthood over 120 days. Using V3-V4 region 16S rRNA gene s

Effects of Maternal Tetramethyl Bisphenol F Exposure on Neurodevelopment and Behavior in Mouse Offspring.

Bisphenol A (BPA) has long been used in plastics, resins, and food packaging materials; however, extensive research has demonstrated its reproductive, developmental, and endocrine-disrupting effects. Consequently, BPA has been increasingly restricted and replaced with structural analogues. Among these, tetramethyl bisphenol F (TMBPF) has emerged as one of the most widely used substitutes, particularly in epoxy resins and food-can coatings. Although initially regarded as a safer alternative, accumulating evidence suggests that TMBPF may exert multiple toxicological effects, raising concerns abo

Mechanical compression causes lung hypoplasia in congenital diaphragmatic hernia with GATA4 genetic variants.

Congenital diaphragmatic hernia (CDH) is a common and severe structural malformation in which the high rate of morbidity and mortality is caused by lung hypoplasia and pulmonary hypertension. The severity of lung and pulmonary vascular defects in patients with CDH is heterogeneous with both intrinsic defects during development and mechanical compression playing important roles. Genetic variants have been identified in 30% of CDH patients and are associated with increased morbidity and mortality, but it is unclear how these variants impact lung and pulmonary vascular defect severity. Extracellular vesicle-mediated release of bis(monoacylglycerol)phosphate is regulated by LRRK2 and glucocerebrosidase activity.

The endolysosomal phospholipid bis(monoacylglycerol)phosphate (BMP) is aberrantly elevated in urine from Parkinson's patients carrying mutations in leucine-rich repeat kinase 2 (LRRK2) and glucocerebrosidase (GCase). Because BMP resides on, and regulates biogenesis of, endolysosomal intralumenal membranes that become extracellular vesicles (EVs) upon release, we hypothesized that increased urinary BMP reflects enhanced exocytosis of BMP-enriched EVs. We analyzed BMP metabolism and EV-associated BMP release in wild-type (WT) and R1441G LRRK2 mouse embryonic fibroblasts (MEFs). Immunofluorescenc

HABP4/VIM-mediated focal adhesion assembly regulates enteric neural crest cell migration via the FAK/SRC pathway.

BACKGROUND: The enteric nervous system (ENS) arises from enteric neural crest cells (ENCCs), and defects in this process can lead to Hirschsprung's disease (HSCR), a congenital disorder characterized by the absence of enteric neurons in the distal colon. Our previous work has identified HABP4 as a potential susceptibility gene for HSCR; however, the mechanisms by which it regulates ENS development remain unclear. METHODS: Single-cell RNA sequencing combined with immunofluorescence staining was used to characterize the spatiotemporal expression pattern of HABP4 in mouse intestinal tissues. HAB

Classification of different light colors applied during the incubation period based on small intestine morphology with XGBoost algorithm.

BACKGROUND: Light is a significant environmental stimulus affecting embryonic development during incubation, and it is known that different wavelengths can have distinctive effects on incubation performance and post-hatching physiological characteristics. However, the extent to which the traces left by different light colors applied during incubation on the small intestine morphology of chicks can be classified has not yet been sufficiently clarified. Therefore, the aim of this study is to evaluate the classifiability of different light colors (red, white, and green) applied during incubation

First LDLRAP1 and Recurrent LDLR Mutations in Tunisian Families With Familial Hypercholesterolemia.

Familial hypercholesterolemia (FH) is a genetic disorder characterised by elevated plasma LDL-cholesterol, predisposing to premature atherosclerotic cardiovascular disease. Most cases follow an autosomal dominant pattern (ADH) caused by pathogenic variants in LDLR, APOB or PCSK9. In contrast, the rare autosomal recessive form (ARH) results from biallelic mutations in LDLRAP1, leading to defective LDL receptor-mediated endocytosis. Despite the high rate of consanguinity in Tunisia, LDLRAP1 variants have not yet been reported in this population. In this study, Whole Exome Sequencing of two consa
