Analysis of inter-leg coordination mechanisms in cricket locomotion: insights from thoracic Ganglion network transection.

Adaptive insect locomotion depends on interactions between the nervous system and body dynamics, yet how these components contribute to inter-leg coordination remains unclear. We investigated the role of direct neural coupling by unilaterally transecting the posterior intermediate connective linking the mesothoracic and metathoracic ganglia in the cricketGryllus bimaculatus. We analyzed kinematic changes and compared them with predictions from a purely neural phase oscillator network model featuring hierarchical asymmetric coupling. The experiments showed a clear dissociation: transection disr

West Nile Virus and Other Nationally Notifiable Arboviral Diseases - United States, 2024.

In the United States, arthropodborne viruses (arboviruses) are primarily transmitted by infected mosquitoes or ticks; rarely, they can be transmitted through blood transfusion or organ transplantation. Whereas a majority of arboviral infections are asymptomatic, symptomatic infections range from mild febrile illness to severe neuroinvasive disease. This report summarizes data for nationally notifiable domestic arboviral diseases with illness onset in 2024. Forty-eight states and the District of Columbia reported 1,955 arboviral disease cases, including 189 (10%) deaths. Although the number of

Disrupting vector competence: exploring radiofrequency exposure as a novel approach to mosquito-borne disease prevention in a changing climate.

The global expansion of mosquito-borne diseases such as dengue, chikungunya, Zika, and West Nile virus is a major public health concern, intensified by climate change and environmental alterations. Aedes aegypti, Aedes albopictus, and Culex pipiens are among the most important vectors for these pathogens, contributing to their transmission across increasingly broad geographic areas. In Europe, the expanding distribution and vectorial competence of Ae. albopictus and Cx. pipiens highlight the need for innovative control strategies beyond traditional chemical and mechanical interventions, which

Multi-sample long-read nanopore sequencing of Agabus bipustulatus (Coleoptera: Dytiscidae: Agabinae) mitogenome produces effectively reliable data for downstream analyses.

Mitochondrial genomes are a rich source of data for various downstream analyses such as population genetics, phylogeny, and systematics. Today it is possible to assemble rapidly large numbers of mitogenomes, mainly employing next-generation sequencing and third-generation sequencing. However, verification of the correctness of the generated sequences is often lacking, especially for noncoding, length-variable parts. Here we have assembled the mitochondrial genome (mitogenome) from four specimens of Agabus bipustulatus (L.) using long-read nanopore sequence data. The use of the latest nanopore

Persistence and recovery of dinotefuran in eastern hemlock (Tsuga canadensis) foliage and twigs by UPLC-MS/MS and ELISA.

BACKGROUND: Dinotefuran, a systemic neonicotinoid insecticide, is approved for control of hemlock woolly adelgid (HWA) (Adelges tsugae Annand), an invasive sap-feeding insect that can kill eastern hemlocks (Tsuga canadensis). Dinotefuran is highly water soluble, facilitating more rapid translocation and HWA control than other neonicotinoids, but its persistence is not well-known. Samples of needles and twigs were collected in spring 2021 from 50 hemlocks treated with a dinotefuran basal trunk spray in 2018 or 2019 (131-145 weeks and 85-93 weeks before sampling, respectively). Processed samples

Comparative multi-tissue analyses identify testis-specific serine/threonine protein kinase (TSSK) genes involved in male fertility in the melon fly Zeugodacus cucurbitae.

BACKGROUND: Zeugodacus cucurbitae is an agricultural pest species with robust reproductive capabilities capable of causing extensive damage. The advent of novel male fertility-related pest control strategies has been an area of active entomological research focused on the sterile insect technique (SIT) strategy. RESULTS: RNA-sequencing analyses were conducted using 16 tissue samples from adult male Z. cucurbitae, leading to the identification of 5338 genes that were differentially expressed between the testes and three other analyzed tissue types. Of these genes, 808 exhibited high levels of

Functional characterization of the kinin receptor in the Chagas disease vector Rhodnius prolixus; activity of native kinins and potent biostable Aib-containing insect kinin analogs.

The causative agent for Chagas disease, Trypanosoma cruzi, is transmitted to a human host in the urine/feces of the kissing bug, Rhodnius prolixus, following blood feeding. Kinins are important chemical messengers in the overall control of blood feeding physiology in R. prolixus, including hindgut contractions and excretion. Thus, disruption in kinin signaling would have damaging consequences to the insect but also interfere with the transmission of Chagas Disease. Here, a heterologous functional receptor assay was used to confirm the validity of the previously cloned putative kinin G-protein-

Gamma-radiation of Glossina palpalis gambiensis revisited: effect on fertility and mating competitiveness.

African animal trypanosomoses are vector-borne diseases that cause enormous livestock losses in sub-Saharan Africa, with drastic socio-economic impacts. Vector control in the context of an area-wide integrated pest management program with a sterile insect technique component requires the production of high-quality sterile male tsetse flies. In our study, we evaluated the effect of irradiation on the fecundity of Glossina palpalis gambiensis to identify the optimal dose that will induce maximum sterility while maintaining biological performance as much as possible. In addition, male mating perf

[Research progress on pesticide residues of Panax notoginseng].

Panax notoginseng is a perennial Chinese medicinal plant, which has serious continuous cropping obstacles and is prone to a variety of diseases and insect pests during the growth process. At present, the prevention and control of pests and diseases is mainly carried out through chemical pesticides, and the consequent pesticide residues of P. notoginseng have attracted much attention. This study reviewed the types and detection methods of pesticide residues in P. notoginseng from 1981 to 2021, and compared the limits of pesticide residues in P. notoginseng in China and abroad to provide a refer

Inheritance and fitness costs of Spodoptera frugiperda (Lepidoptera: Noctuidae) resistance to spinosad in Brazil.

BACKGROUND: Spodoptera frugiperda is a pest of economically important crops in South America. In Brazil, this species is considered the most destructive pest of maize. Use of spinosyn insecticides in insect resistance management (IRM) has been one strategy to control this pest. In this study, we selected a strain of S. frugiperda resistant to spinosad and evaluated the inheritance and fitness costs of the resistance. RESULTS: Estimated LC50 (concentration required to kill 50% of larvae) values were 0.011 and 9.80 µg cm-2 for the spinosad-susceptible (Sus) and -resistant (Spin-res) strains, re

Critical phases of viral production processes monitored by capacitance.

Over the last decade industrial manufacturing of viral vaccines and viral vectors for prophylactic and therapeutic applications is experiencing a remarkable growth. Currently, the quality attributes of viral derived products are assessed only at the end-point of the production process, essentially because in-process monitoring tools are not available or not implemented at industrial scale. However, to demonstrate process reproducibility and robustness, manufacturers are strongly advised by regulatory agencies to adopt more on-line process monitoring and control. Dielectric spectroscopy has bee

Delivery of Nucleic Acids through Embryo Microinjection in the Worldwide Agricultural Pest Insect, Ceratitis capitata.

The Mediterranean fruit fly (medfly) Ceratitis capitata (Wiedemann) (Diptera: Tephritidae) is a pest species with extremely high agricultural relevance. This is due to its reproductive behavior: females damage the external surface of fruits and vegetables when they lay eggs and the hatched larvae feed on their pulp. Wild C. capitata populations are traditionally controlled through insecticide spraying and/or eco-friendly approaches, the most successful being the Sterile Insect Technique (SIT). The SIT relies on mass-rearing, radiation-based sterilization and field release of males that retain

Blood meal-derived heme decreases ROS levels in the midgut of Aedes aegypti and allows proliferation of intestinal microbiota.

The presence of bacteria in the midgut of mosquitoes antagonizes infectious agents, such as Dengue and Plasmodium, acting as a negative factor in the vectorial competence of the mosquito. Therefore, knowledge of the molecular mechanisms involved in the control of midgut microbiota could help in the development of new tools to reduce transmission. We hypothesized that toxic reactive oxygen species (ROS) generated by epithelial cells control bacterial growth in the midgut of Aedes aegypti, the vector of Yellow fever and Dengue viruses. We show that ROS are continuously present in the midgut of s

Characterization of the immune response of domestic fowl following immunization with proteins extracted from Dermanyssus gallinae.

Dermanyssus gallinae is the most significant ectoparasite of European poultry egg laying production systems due to high costs of control and associated production losses as well as adverse effects on bird welfare. In this study, soluble proteins were extracted from unfed D. gallinae (DGE) using a urea-based detergent and ultra-filtration, passed through a 0.22 microm filter and blended aseptically with adjuvant. One group of laying hens was immunized with DGE and adjuvant (Montanide ISA 50 V) whilst another group (Control) received physiological saline and adjuvant. All birds were immunized on

Phytotoxicity of tolylfluanid in tomatoes.

The use of Euparen Multi (tolylfluanid) for controlling Botrytis cinerea in tomatoes has been decreased the last decade for several reasons. Because of the lack of different fungicides with a good efficacy it is important that growers can use different fungicides to prevent development of resistance of Botrytis cinerea against many fungicides. Tolylfluanid has negative side effects on some insect populations that are used for biological control. It is known that Euparen Multi and Euparen can have a negative effect on some predatory mites (Schmidt and Zeller, 1998) such as Phytoseiulus persimil

Equilibrium properties of a coupled contagion model of mosquito-borne disease and mosquito preventive behaviors.

Although different strategies for mosquito-borne disease prevention can vary significantly in their efficacy and scale of implementation, they all require that individuals comply with their use. Despite this, human behavior is rarely considered in mathematical models of mosquito-borne diseases. Here, we sought to address that gap by establishing general expectations for how different behavioral stimuli and forms of mosquito prevention shape the equilibrium prevalence of disease. To accomplish this, we developed a coupled contagion model tailored to the epidemiology of dengue and preventive beh

Chemically-mediated colonization of black cherry by the peach bark beetle, Phloeotribus liminaris.

The peach bark beetle (Phloeotribus liminaris Harris, PBB) affects the health, quality, and value of black cherry (Prunus serotina Ehrh.) within the Central Hardwoods Forest Region of North America. When colonized by adult beetles, black cherry trees produce a defensive exudate, or 'gum', staining the wood and decreasing its value up to 90%. Current management tactics are inadequate to avoid extensive damage to most veneer-sized black cherry in the region. We test the hypothesis that PBB colonization behavior is chemically-mediated and determine the extent to which PBB is attracted to compound

Use of temperature to improve West Nile virus forecasts.

Ecological and laboratory studies have demonstrated that temperature modulates West Nile virus (WNV) transmission dynamics and spillover infection to humans. Here we explore whether inclusion of temperature forcing in a model depicting WNV transmission improves WNV forecast accuracy relative to a baseline model depicting WNV transmission without temperature forcing. Both models are optimized using a data assimilation method and two observed data streams: mosquito infection rates and reported human WNV cases. Each coupled model-inference framework is then used to generate retrospective ensemble
