mosGILT antibodies interfere with Plasmodium sporogony in Anopheles gambiae.

Plasmodium, the causative agents of malaria, are obtained by mosquitoes from an infected human. Following Plasmodium acquisition by Anopheles gambiae, mosquito gamma-interferon-inducible lysosomal thiol reductase (mosGILT) plays a critical role in its subsequent sporogony in the mosquito. A critical location for this development is the midgut, a tissue we show expresses mosGILT. Using membrane-feeding and murine infection models, we demonstrate that antibodies against mosGILT reduce the number of P. falciparum and P. berghei oocysts in the midgut and the infection prevalence of both species in

Characterization of temporal expression of immune genes in female locust challenged by fungal pathogen, Aspergillus sp.

INTRODUCTION: The innate immune system provides defense against invading pathogens in insects and mammals. METHODS: We conducted transcriptomic analyses of the locust Locusta migratoria under Aspergillus oryzae infection to clarify temporal variation in its molecular immune response. RESULTS: We found that fat body cells and hemocytes play different roles in the immune response of locusts to Aspergillus infection at different time points after inoculation, and melanization was the main process underlying the immune response of female locusts. Most pattern recognition receptors (PRR) genes we

Ongoing insecticide resistance evolution in beet armyworm via changes in the utilization of pre-existing and new mutations.

Little has been known until now about the population genomic processes underpinning pest insect species' rapid and ongoing adaptation to chemical insecticides. This paper elucidates these processes for six mutations encoding resistances to four insecticide classes in Chinese Spodoptera exigua: F116V in CYP9A186 against avermectins, T929I and L1014F in VGSC against pyrethroids, V1848I in VGSC against indoxacarb and I4790M and I4790K in RyR against diamides. Whole genome sequencing of 139 individuals from 20 populations across China revealed little genetic differentiation among the populations a

Studies on the Design, Synthesis, and Insecticidal Activity Evaluation of New Caffeine-Acrylate Derivatives as Potential Ryanodine Receptor Activators.

On the basis of insecticidal leading structures of methylxanthine-acrylates I and II, a series of new caffeine-acrylate derivatives A-F were designed and synthesized via various structural optimization strategies. The structures of new compounds were confirmed through 1H NMR, 19F NMR, 13C NMR, and high-resolution mass spectrometry (HRMS). The bioassays showed that some of the new ester compounds possessed high insecticidal potentials against Plutella xylostellaL. and Mythimna separata Walker with improved insecticidal activity compared to the lead compounds. Notably, new compounds B8, B10, C1,

Physicochemical characteristics and anti-inflammatory properties of Zophobas morio (super mealworm) protein extracted by different methods.

In this study, Zophobas morio protein (ZMP) was extracted via combining alkaline extraction with ultrasound-assisted (AE-UAE) or microwave-assisted (AE-MAE) extraction in comparison with their respective single extraction methods and conventional method. AE-UAE and AE-MAE exhibited a higher extraction yield (40.68 % and 36.80 %, respectively) than single methods and conventional method (29.19 %-35.89 %). SDS-PAGE showed AE-UAE and AE-MAE induced new formation of smaller molecular weight proteins. Moreover, the hybrid methods decreased α-helix content, whereas increased β-sheet by unfolding the

MDR49 coding for both P-glycoprotein and TMOF transporter functions in ivermectin resistance, trypsin activity inhibition, and fertility in the yellow fever mosquito, Aedes aegypti.

This study investigated the function of the MDR49 gene in Aedes aegypti. MDR49 mutants were constructed using CRISPR/Cas9 technology; the mutation led to increased sensitivity to ivermectin (LC50: from 1.3090 mg L-1 to 0.5904 mg L-1), and a reduction in midgut trypsin activity. These findings suggest that the P-gp encoded by MDR49 confers resistance to ivermectin and impacts the reproductive function in Ae. aegypti. RNA interference technology showed that knockdown of MDR49 gene resulted in a significant decrease in the expression of VGA1 after a blood meal, as well as a decrease in the number

Cleavage activation and functional comparison of Manduca sexta serine protease homologs SPH1a, SPH1b, SPH4, and SPH101 in conjunction with SPH2.

Phenoloxidase (PO) is a crucial component of the insect immune response against microbial infection. In the tobacco hornworm Manduca sexta, PO is generated from its precursor proPO by prophenoloxidase activating proteases (PAPs) in the presence of two noncatalytic serine protease homologs (SPHs). cDNA cloning and genome analysis indicate that SPH1a (formerly known as SPH1), SPH1b, SPH4, SPH101, and SPH2 contain a clip domain, a linker, and a protease-like domain (PLD). The first 22 residues of the SPH1b, SPH4, and SPH101 PLDs are identical, and differ from SPH1a only at position 4, Thr154 subs

Exome-wide association study reveals largely distinct gene sets underlying specific resistance to dengue virus types 1 and 3 in Aedes aegypti.

Although specific interactions between host and pathogen genotypes have been well documented in invertebrates, the identification of host genes involved in discriminating pathogen genotypes remains a challenge. In the mosquito Aedes aegypti, the main dengue virus (DENV) vector worldwide, statistical associations between host genetic markers and DENV types or strains were previously detected, but the host genes underlying this genetic specificity have not been identified. In particular, it is unknown whether DENV type- or strain-specific resistance relies on allelic variants of the same genes o

Detrimental Effects of Induced Antibodies on Aedes aegypti Reproduction.

Aedes aegypti (Linnaeus) (Diptera: Culicidae) is the main vector of viruses causing dengue, chikungunya, Zika, and yellow fever, worldwide. This report focuses on immuno-blocking four critical proteins in the female mosquito when fed on blood containing antibodies against ferritin, transferrin, one amino acid transporter (NAAT1), and acetylcholinesterase (AchE). Peptides from these proteins were selected, synthetized, conjugated to carrier proteins, and used as antigens to immunize New Zealand rabbits. After rabbits were immunized, a minimum of 20 female mosquitos were fed on each rabbit, per

Endocrine regulation of airway clearance in Drosophila.

Fluid clearance from the respiratory system during developmental transitions is critically important for achieving optimal gas exchange in animals. During insect development from embryo to adult, airway clearance occurs episodically each time the molt is completed by performance of the ecdysis sequence, coordinated by a peptide-signaling cascade initiated by ecdysis-triggering hormone (ETH). We find that the neuropeptide Kinin (also known as Drosokinin or Leukokinin) is required for normal respiratory fluid clearance or "tracheal air-filling" in Drosophila larvae. Disruption of Kinin signaling

Molecular cloning, spatiotemporal and functional expression of GABA receptor subunits RDL1 and RDL2 of the rice stem borer Chilo suppressalis.

Insect γ-aminobutyric acid (GABA) receptor (GABAR) is one of the major targets of insecticides. In the present study, cDNAs (CsRDL1A and CsRDL2S) encoding the two isoforms of RDL subunits were cloned from the rice stem borer Chilo suppressalis. Transcripts of both genes demonstrated similar expression patterns in different tissues and developmental stages, although CsRDL2S was ∼2-fold more abundant than CsRDL1A throughout all development stages. To investigate the function of channels formed by CsRDL subunits, both genes were expressed in Xenopus laevis oocytes singly or in combination in diff

Towards improving tsetse fly paratransgenesis: stable colonization of Glossina morsitans morsitans with genetically modified Sodalis.

BACKGROUND: Tsetse flies (Glossina sp.) refractory to trypanosome infection are currently being explored as potential tools to contribute in the control of human and animal African trypanosomiasis. One approach to disrupt trypanosome transmission by the tsetse fly vector involves the use of paratransgenesis, a technique that aims to reduce vector competence of disease vectors via genetic modification of their microbiota. An important prerequisite for developing paratransgenic tsetse flies is the stable repopulation of tsetse flies and their progeny with its genetically modified Sodalis symbion

Evidence for the presence of Ctenocephalides orientis in livestock dwellings in northwest Iran.

Fleas are important vectors of diseases such as murine typhus, tularaemia, hymenolepiasis and plague. The presence of active foci and history of human- and flea-transmitted plague in northwest Iran prompted the present group to collect and identify fleas from human and livestock dwellings across West Azerbaijan Province. Adult fleas were collected and identified using routine taxonomic keys. Species designation was confirmed by sequencing the cytochrome oxidase subunit I (COI). Of the collected specimens (n = 989), 104 were collected off-host (30 from human dwellings and 74 in light traps) and

Expression and function of spineless orthologs correlate with distal deutocerebral appendage morphology across Arthropoda.

The deutocerebral (second) head segment is putatively homologous across Arthropoda, in spite of remarkable disparity of form and function of deutocerebral appendages. In Mandibulata this segment bears a pair of sensory antennae, whereas in Chelicerata the same segment bears a pair of feeding appendages called chelicerae. Part of the evidence for the homology of deutocerebral appendages is the conserved function of homothorax (hth), which has been shown to specify antennal or cheliceral fate in the absence of Hox signaling, in both mandibulate and chelicerate exemplars. However, the genetic bas

Identification of glutathione S-transferase genes in Leptinotarsa decemlineata and their expression patterns under stress of three insecticides.

Glutathione S-transferases (GSTs) is a family of multifunctional enzymes that are involved in detoxification of poisonous compounds. In the present paper, the Leptinotarsa decemlineata genome and transcriptome dataset were mined and 30 GST genes were identified. These GSTs belonged to cytosolic (29 genes) and microsomal (1 gene) classes. Among them 3 GSTs (LdGSTe2, LdGSTs4, and LdGSTo3) possessed splice variants. Of the 29 cytosolic LdGSTs, 3, 10, 5, 4, 4, and 1 members were classified as delta, epsilon, omega, sigma, theta, and zeta subclasses respectively, along with 2 unclassified genes. Ph

Effective protection induced by three different versions of the porcine S3Pvac anticysticercosis vaccine against rabbit experimental Taenia pisiformis cysticercosis.

In an effort to develop an effective and affordable oral vaccine against porcine Taenia solium cysticercosis, the S3Pvac anti-cysticercosis vaccine was expressed in papaya calli. Taenia pisiformis experimental rabbit cysticercosis was used as a model to compare the efficacy of the oral vaccine vs. the injectable S3Pvac-synthetic and S3Pvac-phage versions. Oral S3Pvac-papaya significantly reduced the expected number of hepatic lesions and peritoneal cysticerci to a similar extent than the injectable vaccines. This study reports for the first time an effective oral vaccine against T. pisiformis

High-throughput approach to detection of knockdown resistance (kdr) mutation in mosquitoes, Culex quinquefasciatus, based on real-time PCR using single-labelled hybridisation probe/melting curve analysis.

BACKGROUND: Knockdown resistance (kdr) mutation (L1014F) is a well-defined mechanism of resistance to pyrethroids and DDT in many insect species. Sensitive detection of the mutations associated with resistance is a prerequisite for resistance management strategies. The authors have developed a new real-time molecular diagnostic assay based on SimpleProbe(®)/melting curve analysis for large-scale kdr genotyping in the wild population of Culex quinquefasciatus Say, the principal vector of bancroftian filariasis. Melting curve analysis is based on the thermal stability difference between matched
