Association of arthritis with a gene complex encoding C-type lectin-like receptors.

OBJECTIVE: To identify susceptibility genes in a rat model of rheumatoid arthritis (RA) and to determine whether the corresponding human genes are associated with RA. METHODS: Genes influencing oil-induced arthritis (OIA) were position mapped by comparing the susceptibility of inbred DA rats with that of DA rats carrying alleles derived from the arthritis-resistant PVG strain in chromosomal fragments overlapping the quantitative trait locus Oia2. Sequencing of gene complementary DNA (cDNA) and analysis of gene messenger RNA (mRNA) expression were performed to attempt to clone a causal gene. A

TRAF1-C5 as a risk locus for rheumatoid arthritis--a genomewide study.

BACKGROUND: Rheumatoid arthritis has a complex mode of inheritance. Although HLA-DRB1 and PTPN22 are well-established susceptibility loci, other genes that confer a modest level of risk have been identified recently. We carried out a genomewide association analysis to identify additional genetic loci associated with an increased risk of rheumatoid arthritis. METHODS: We genotyped 317,503 single-nucleotide polymorphisms (SNPs) in a combined case-control study of 1522 case subjects with rheumatoid arthritis and 1850 matched control subjects. The patients were seropositive for autoantibodies aga

The FCRL3 -169T>C polymorphism is associated with rheumatoid arthritis and shows suggestive evidence of involvement with juvenile idiopathic arthritis in a Scandinavian panel of autoimmune diseases.

BACKGROUND AND OBJECTIVES: The Fc receptor-like 3 (FCRL3) gene -169T>C single nucleotide polymorphism (SNP) has been reported to be associated with several autoimmune diseases (AIDs) in Japanese populations. However, association results in other populations have been conflicting. Therefore, we investigated this SNP in a Scandinavian panel of AIDs. METHODS: We genotyped patients with rheumatoid arthritis (RA; n = 708), juvenile idiopathic arthritis (JIA; n = 524), systemic lupus erythaematosus (SLE; n = 166), ulcerative colitis (UC; n = 335), primary sclerosing cholangitis (PSC; n = 365), Croh

Periodontal and hematological characteristics associated with aggressive periodontitis, juvenile idiopathic arthritis, and rheumatoid arthritis.

BACKGROUND: Periodontitis shares several clinical and pathogenic characteristics with chronic arthritis, and there is some degree of coexistence. The aims of this study were to elucidate whether patients with localized aggressive periodontitis (LAgP), generalized aggressive periodontitis (GAgP), juvenile idiopathic arthritis (JIA), and rheumatoid arthritis (RA) share periodontal and hematological characteristics distinguishing them from individuals free of diseases. METHODS: The study population consisted of white adults (<or=35 years old) with LAgP (N=18), GAgP (N=27), JIA (N=10), RA (N=23),

The phenotype of ostensibly healthy women who are carriers for ornithine transcarbamylase deficiency.

Ornithine transcarbamylase (OTC) deficiency is an X-linked disorder of urea synthesis. Among females who carry a mutant OTC allele, there is a wide range of phenotypic variability, ranging from apparent normality to a severe onset and the resulting profound neurologic impairment observed in hemizygous males. This study was designed to define the phenotypic variability of OTC deficiency in ostensibly healthy carrier females and to compare them to noncarrier females from their own and other families. One hundred seventy-five women from 89 families participated in this study. Each completed a mai

Neutrophil-derived superoxide anion induces lipid peroxidation and stimulates collagen synthesis in human hepatic stellate cells: role of nitric oxide.

Experimental evidence indicates that the lipid peroxidation of biological membranes is often associated with the development of liver fibrosis. We have studied the effect of neutrophil-derived reactive oxygen species (ROS) on collagen synthesis by human hepatic stellate cells (HSC), the major source of collagen in the liver, in a coculture system. Lipid peroxidation in the cocultures was evaluated in terms of either malondialdehyde (MDA) production or the formation of MDA/4-hydroxynonenal protein adducts. The expression of cellular messenger RNAs (mRNAs) was evaluated by either Northern blotti

Decreased pulmonary nitric oxide synthase activity in the rat model of congenital diaphragmatic hernia.

Because nitric oxide (NO) dilates vascular smooth muscle cells, a deficiency of endogenous pulmonary nitric oxide production by nitric oxide synthase (NOS) has been suggested to be involved in the pathophysiology of pulmonary hypertension in congenital diaphragmatic hernia (CDH). Our aim was to determine whether experimentally induced CDH in rats results in a decrease in the synthesis of NO in the lungs. Adult Sprague-Dawley rats were fed 300 mg/kg of nitrofen at 10.5 days' gestation. CDH, control, and sham (dosed with nitrofen, but without CDH) lungs were homogenized at full term (22 days' ge

Inhibition of CA V decreases glucose synthesis from pyruvate.

The carbonic anhydrase inhibitor acetazolamide reduces citrulline synthesis by intact guinea pig liver mitochondria and also inhibits mitochondrial carbonic anhydrase (CA V) and the more lipophilic carbonic anhydrase inhibitor ethoxzolamide reduces urea synthesis by intact guinea pig hepatocytes in parallel with its inhibition of total hepatocytic carbonic anhydrase activity. Intact hepatocytes from 48-h starved male guinea pig livers were incubated at 37 degrees C in Krebs-Henseleit with 95% O2/5% CO2 at pH 7.1 with 5 mM pyruvate, 5 mM lactate, 3 mM ornithine, 10 mM NH4Cl, 1 mM oleate; with t

Isolation of an avian erythrocyte protein possessing ADP-ribosyltransferase activity and capable of activating adenylate cyclase.

An ADP-ribosyltransferase was purified approximately 500-fold from the supernatant fraction of turkey erythrocytes. The enzyme hydrolyzed [carbonyl-(14)C]NAD to ADP-ribose and [carbonyl-(14)C]nicotinamide at a low rate. Nicotinamide formation from NAD was enhanced by arginine methyl ester > D-arginine approximately L-arginine > guanidine; lysine, histidine, and citrulline were ineffective. Incubation of [adenine-U-(14)C]NAD and arginine methyl ester or arginine with the purified enzyme resulted in the formation of new compounds that contained (14)C, reacted with ninhydrin, and quenched backgro

Arbuscular mycorrhizal fungi reshape carbon-nitrogen balance and metabolism in nitrogen-limited escarole (Cichorium endivia L.).

Nitrogen nutrition remains a major challenge for sustainable crop production, particularly under the current need to reduce fertilizer inputs. This study evaluated whether inoculation with arbuscular mycorrhizal fungi (AMF) improves the physiological, nutritional, and metabolic performance of escarole (Cichorium endivia L.) plants under nitrogen-limited conditions. Escarole plants were grown under two nitrogen regimes, full nitrogen supply and 25% of the optimal nitrogen concentration, either inoculated or non-inoculated with Rhizophagus irregularis. Plant growth, mineral composition, soluble

Urinary and Faecal Amino-Acids as Biomarkers for Colorectal Neoplasia in Lynch Syndrome-A Prospective Longitudinal Study.

In Lynch syndrome, colonoscopy surveillance for colorectal cancer (CRC) is burdensome and post-colonoscopy CRCs still occur. Unlike the faecal immunochemical test (FIT), gut metabolomic alterations including amino-acids have shown potential as non-invasive biomarkers for detecting both advanced and non-advanced sporadic colorectal neoplasia. We aimed to evaluate the potential of faecal and urinary amino-acids to refine colonoscopy surveillance in Lynch syndrome. Individuals with Lynch syndrome collected faeces and urine 3 months before and 3-6 months after surveillance colonoscopy. Individuals

Metabolic profiling of human follicular fluid to unravel the pathways involved in polycystic ovary syndrome - a systematic review.

Polycystic ovary syndrome (PCOS), an endocrine-metabolic disorder, affects women of reproductive age. Follicular fluid (FF) reflects the biochemical microenvironment surrounding the oocyte, providing valuable insight into molecular mechanisms associated with PCOS. Metabolomics enables comprehensive profiling of small-molecule metabolites allowing the identification of disease-specific metabolic signatures, dysregulated pathways, and potential diagnostic/therapeutic targets. This systematic review summarizes available evidence on the FF metabolomic profile of women with PCOS compared to normo-o

Metabolic Profiling of Circulating and Tumour-Infiltrating T Cells Uncovers Bioenergetic Checkpoints Linked With Immune and Clinical Skewing in Melanoma Patients.

Metabolism is critical for immune cell functions. Tumours shape their microenvironment to render it metabolically hostile for infiltrating immune cells. While targeting immunometabolism emerges as a promising way to reinvigorate anticancer immunity, a deeper understanding of the metabolic disturbances of immune cells is needed. Here, we explored how the metabolic status of T cells governs immune skewing from circulating and tumour-infiltrating CD4+ and CD8+ T cells of melanoma patients at a single-cell level using the SCENITH method and targeted metabolomics. Circulating and tumour-infiltratin

Isoliquiritin alleviates sepsis-induced intestinal barrier dysfunction in mice potentially by promoting autophagy.

Intestinal barrier function protection is essential to ameliorate sepsis. Isoliquiritin (ILQ) is a natural compound with broad biological activities. However, whether ILQ attenuates sepsis-induced intestinal damage remains unclear. An LPS-induced sepsis mouse model was established. ILQ inhibited body weight loss in sepsis mice. ILQ enhanced mouse intestinal function, as evidenced by decreased serum LDH levels and increased citrulline levels. Alcian blue staining showed that ILQ increased goblet cells in sepsis mouse ileum. ILQ increased the MUC2 RNA levels and decreased the claudin-2 protein e

IL-36 activates neutrophil extracellular traps and exacerbates LPS-induced ARDS in mice.

Sepsis-induced acute respiratory distress syndrome (ARDS) is characterized by uncontrolled pulmonary inflammation and neutrophil-driven pathology. Neutrophil extracellular traps (NETs) have been implicated in ARDS progression, but the role of interleukin-36 (IL-36) in this process remains unclear. This study aimed to investigate the regulatory mechanism of IL-36 via NETs and its impact on NF-κB activation in a murine model of LPS-induced ARDS. A lipopolysaccharide (LPS)-induced ARDS model was established in C57BL/6 mice. Animals were divided into Control, ARDS, ARDS + IL-36, and ARDS + IL-36Ra

NETosis in the Kidney, Liver, and Lung of Mice With Cecal Ligation and Puncture-Induced Sepsis and the Ameliorating Effects of Adipose-Derived Stem Cell Exosomes.

Sepsis-induced organ failure involves dysregulated neutrophil responses, including neutrophil extracellular trap (NET) formation, via NETosis. Exosomes generated from adipose-derived stem cells (ADSCs) have shown potential in sepsis treatment. This study characterized organ-specific NETosis in the kidney, liver, and lungs using a murine cecal ligation and puncture (CLP) model and assessed the modulatory effects of ADSC exosomes. Male C57BL/6 mice were grouped as follows: control, CLP, and CLP with ADSC exosome therapy (n = 6 per group). Plasma cell-free DNA (cfDNA) and tissue citrullinated his

Evaluation of the anti-inflammatory effect of ectoine-leflunomide combination in adjuvant-induced arthritis in rats, and its colonic protective action against leflunomide-induced gastrointestinal injury.

Rheumatoid arthritis (RA) is an autoimmune disease with an intriguing pathophysiological mechanism that primarily affects the joints and leads to extra-articular manifestations. Leflunomide is one of the most effective disease-modifying anti-rheumatic drugs (DMARDs), utilized in RA treatment, but its use comes with frequent gastrointestinal adverse effects, which might necessitate therapy discontinuation. The current work aimed to explore the antirheumatic and gastroprotective effects of ectoine, a compatible solute found in nature, on adjuvant-induced arthritis (AIA) in rats, and to compare i
