Cancer-associated fibroblasts from cervical cancer modulate protumoral neutrophil functions: evidence from a pilot exploratory study.

Cancer-associated fibroblasts (CAFs) are key regulators of the tumor microenvironment; however, their role in modulating neutrophil function during cervical cancer (CC) progression remains poorly understood. In this study, we evaluated whether CAF supernatants (CSN) derived from early (FIGO I-II) and late stages (FIGO III-IV) of CC differentially regulate neutrophil activity. Peripheral blood neutrophils from healthy donors were stimulated with CSN, and their functional responses were assessed, including cytokine production, metabolic activity (MTT), reactive oxygen species (ROS) generation, a

A microwave-conditioned MoS₂-tomato extracellular vesicle hydrogel drives photothermal-photoelectric reprogramming of chronic wounds.

Chronic diabetic wounds remain difficult to treat because the wound bed is trapped in a hostile microenvironment marked by oxidative stress, persistent inflammation, and impaired tissue remodeling. Here, we developed a microwave-conditioned gellan gum (GG) hydrogel incorporating molybdenum disulfide (MoS₂) and tomato-derived extracellular vesicle-enriched fraction (tEVs) as an externally activatable platform for diabetic wound repair. The isolated tEVs showed nanoscale size uniformity, colloidal stability, and concentration-dependent radical-scavenging activity, supporting their role as a bioa

PDHA1 Hyperactivation-Mediated Glucose Metabolism Reprogramming Fuels Neutrophil NETosis and Promotes Acute Lung Injury.

Neutrophil extracellular trap (NET) formation, termed NETosis, is a key driver of lung tissue injury in acute lung injury/acute respiratory distress syndrome (ALI/ARDS). However, the precise mechanisms that trigger neutrophil NETosis remain incompletely understood. Here, we observed that neutrophils from both ALI mice and ARDS patients exhibited NETosis alongside a pronounced selective metabolic shift, characterized by enhanced oxidative phosphorylation (OXPHOS) and attenuated glycolytic activity. Phosphoproteomic profiling revealed that this shift was driven by aberrant activation of pyruvate

Bidirectional Crosstalk Between Intestinal Epithelium and Immune Microenvironment in Inflammatory Bowel Disease: Mechanisms and Therapeutic Implications.

Inflammatory bowel disease (IBD), encompassing Crohn's disease and ulcerative colitis, is characterized by chronic mucosal inflammation driven by dysregulated interactions between intestinal epithelial cells (IECs) and immune components. This review systematically explores the dynamic interplay between epithelial barrier integrity and immune-microenvironmental regulation in IBD pathogenesis. We highlight the dual roles of innate immunity (neutrophils, macrophages, dendritic cells, and innate lymphoid cells) and adaptive immunity (Th1, Th17, and Treg cells) in orchestrating inflammatory cascade

SomaScan proteomics reveals novel biomarkers in the progression of liver cirrhosis to hepatocellular carcinoma.

BACKGROUND: Hepatocellular carcinoma (HCC) typically develops from liver cirrhosis (LC), however early diagnosis is difficult due to a lack of reliable biomarkers. The goal of this study was to use SomaScan proteomics technology to find plasma protein profiles that differentiated LC and HCC from healthy controls in order to develop novel biomarkers for HCC early detection and targeted therapy. METHODS: We used SomaScan technology to evaluate 10,893 plasma proteins from LC, HCC, and normal populations. Differentially expressed proteins (DEPs) were discovered and functionally annotated using HP

Forsythiae Fructus polysaccharides mitigate DSS colitis by modulating gut microbiota-derived palmitic acid and PI3K/AKT pathway.

Inflammatory bowel disease (IBD) is a chronic inflammatory disorder with limited therapeutic options, and the contribution of microbiota-associated metabolites to disease progression remains incompletely understood. Here, we demonstrate that Forsythia polysaccharides (FPs) exert protective effects against dextran sulfate sodium (DSS)-induced colitis through coordinated regulation of gut microbiota, microbial metabolism, and inflammatory signaling. FPs treatment markedly alleviated colonic injury and inflammation, restored intestinal barrier integrity, and remodeled the gut microbial community.

Complement in acute kidney injury: a convergent pathogenic pathway in multifactorial renal damage.

Acute kidney injury (AKI) is a common clinical syndrome characterized by substantial etiologic heterogeneity, whose pathophysiology encompasses inflammatory amplification, microcirculatory dysfunction, tubular injury, and maladaptive repair. Evidence from preclinical models and clinical samples indicates that dysregulated complement activation represents a potential convergent mechanism of injury in multiple forms of AKI, although its relative contribution varies across etiologies. Experimental models support mechanistic and pathogenic roles for complement activation, whereas clinical evidence

Transcriptomic insights into epigenetic and immunomodulatory roles of nuclear histone genes in bladder cancer: A bioinformatics approach.

Bladder cancer (BC) poses a significant global health and economic burden due to its high recurrence rates, progression risks, and the need for lifelong surveillance. Despite advances in diagnosis and treatment, reliable molecular biomarkers for prognosis and therapeutic targeting remain limited. In this study, we used an integrative bioinformatics approach to identify key dysregulated nuclear genes in BC, focusing on histone variants because of their essential role in chromatin organization and gene regulation. Using RNA-seq data, we performed a quality assessment, aligned reads with STAR, an

Mitochondrial Metabolism in Neutrophils: Emerging Roles in Inflammatory Skin Diseases.

Neutrophils, the most abundant of the circulating leucocytes, play crucial roles in antimicrobial defence, tissue remodelling, and immune regulation. Traditionally regarded as predominantly glycolytic, relying on aerobic glycolysis (the Warburg effect) for rapid ATP generation, neutrophils are now recognised to possess broader metabolic flexibility. Emerging evidence has revealed their capacity for oxidative phosphorylation (OXPHOS), fatty acid oxidation (FAO), and glutaminolysis. As a result, mitochondrial metabolism is dynamically reprogrammed during differentiation and activation. These met

The C5a-C5aR1 axis facilitates blood-brain barrier disruption induced by EV-A71 infection through neutrophil activation.

Enterovirus A71 (EV-A71), a member of the genus Enterovirus within the family Picornaviridae, induces neuroinflammation; however, the underlying mechanisms remain incompletely understood. This study demonstrates that the C5a-C5aR1 axis plays a pivotal role in EV-A71-induced blood-brain barrier (BBB) disruption and neuroinflammation, primarily by regulating neutrophil migration. Using human brain specimens and a mouse model, we observed pronounced inflammatory cell infiltration in the brainstem and BBB disruption following EV-A71 infection. Immunofluorescence analysis revealed robust activation

Oral cascade-responsive nanomedicine to disrupt macrophage-neutrophil crosstalk for intestinal barrier repair.

Intestinal barrier dysfunction underlies inflammatory bowel disease (IBD) and is driven, in part, by pathological crosstalk between macrophages and neutrophils that sustains pro-inflammatory cytokine release and excessive formation of neutrophil extracellular trap (NET). In this study, an oral cascade-responsive nanomedicine, PLBD, was developed to co-deliver the prostaglandin D2 receptor agonist BW245C and the NET scavenger DNase I with precise spatiotemporal control and minimal off-target release. PLBD comprises mucus-penetrating, reactive oxygen species (ROS)-sensitive liposomes (LBD) formu

Immune-endothelial-coagulation crosstalk as a driver of multi-organ dysfunction in severe viral pneumonia.

Viral burden or pathogen identity alone cannot adequately explain the progression of severe viral pneumonia from a compartmentalized respiratory infection to acute respiratory distress syndrome, multi-organ failure, and death. Maladaptive immunity, endothelial damage, and coagulation dysregulation are all functionally integrated in a host-driven pathological mechanism that mediates disease escalation. Systemic microvascular damage and pulmonary inflammation are linked by immune-endothelial-coagulation interaction. This review investigates the ways in which immunothrombosis and microcirculatory

Microbiome-innate immune crosstalk in acute exacerbation of idiopathic pulmonary fibrosis: an amplification framework.

Acute exacerbation of idiopathic pulmonary fibrosis (AE-IPF) remains a major cause of short-term mortality, yet its biological drivers are incompletely understood. Increasing evidence indicates that the distal lung is not sterile and that microbial burden, ecological disruption, and host immune responses may interact within the fibrotic niche. Here, we summarize current evidence and propose a focused, hypothesis-generating framework for microbiome-immune crosstalk in AE-IPF. In this model, pulmonary dysbiosis and persistent microbial stimulation may converge with epithelial injury-derived dang

Gout Inflammation Time Programming: Molecular Clock from Crystal Triggering to Tissue Remodeling.

This review introduces and elaborates a novel temporal paradigm, the "Gout Inflammation Time Programming" model, conceptualized through the Gout-STAT™ framework. This model redefines gout inflammation as a dynamic continuum progressing through three precisely timed phases: an acute Perception phase (0-24 h) initiated by monosodium urate (MSU) crystal recognition, triggering the NOD-like receptor thermal protein domain associated protein 3 (NLRP3) inflammasome and neutrophil-driven burst; a critical Adaptation phase (24-72 h) where outcomes are determined by immunometabolic reprogramming of mac

Granulomonocytapheresis for Sepsis: Mechanistic Rationale, Therapeutic Potential, and Potential Concerns in Thromboinflammatory Critical Illness.

Granulomonocytapheresis (GMA) is an extracorporeal leukocyte adsorption therapy that selectively removes activated granulocytes and monocytes through interactions involving Fcγ receptors (FcγR) and complement receptor 3 (CR3). Initially developed for chronic inflammatory diseases such as ulcerative colitis, GMA has recently attracted attention as a potential adjunctive therapy for sepsis. Contemporary understanding of sepsis emphasizes thromboinflammation, immunothrombosis, endothelial injury, and inflammatory cell death as central mechanisms underlying organ dysfunction. Activated FcγR-positi

Systemic Inflammatory Response During Coronary Artery Bypass Grafting with Cardiopulmonary Bypass: Cellular and Molecular Mechanisms.

Coronary artery bypass grafting (CABG) utilizing cardiopulmonary bypass (CPB) remains a cornerstone of treatment for advanced multivessel coronary artery disease. During the ischemic period, myocardial cells become injured and increasingly depend on anaerobic metabolism. After reperfusion, exposure of blood components to artificial surfaces, combined with ischemia-reperfusion injury, surgical trauma, and gut translocation of endotoxins, triggers a complex systemic inflammatory response syndrome (SIRS). This inflammatory cascade involves a tightly regulated network of humoral cascades (compleme

Immune dysregulation and infection susceptibility in Anti-MDA5 dermatomyositis.

Antibodies targeting melanoma differentiation-associated gene 5 protein (MDA5) are associated with a distinct subtype of dermatomyositis characterised by cutaneous ulceration, systemic inflammation, and a high risk of rapidly progressive lung disease. Beyond these manifestations, patients experience disproportionately high rates of infection compared with other inflammatory myopathies, including viral, bacterial, and fungal complications. This review synthesises cohort data demonstrating an excess burden of infections, particularly Pneumocystis jirovecii pneumonia (PJP) and cytomegalovirus, at

Molecular basis of coronary artery disease-malignancy comorbidity: inflammation, immunometabolism, thrombosis, and cardio-oncology translation.

Coronary artery disease (CAD) and malignancy frequently coexist in aging populations and share multiple risk factors, including smoking, obesity, diabetes, dyslipidemia, chronic inflammation, and metabolic dysfunction. However, increasing evidence suggests that CAD-malignancy comorbidity cannot be fully explained by epidemiological coincidence alone, although its causal direction remains difficult to establish because of shared risk factors, surveillance bias, reverse causality, cancer stage, and treatment exposure. Instead, convergent inflammatory, metabolic, thrombotic, and immune mechanisms

How does the Immunological System Change during the SARS-COV-2 Attack? A Clue for the New Immunotherapy Discovery.

The COVID-19 pandemic caused by the Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-COV-2) is one of the biggest unsolved global problems of the 21st century for which there has been no definitive cure yet. Like other respiratory viruses, SARS-COV-2 triggers the host immunity dramatically, causing dysfunction in the immune system, both innate and adaptive, which is a common feature of COVID-19 patients. Evidence shows that in the early stages of COVID-19, the immune system is suppressed while it is overactive in severe patients characterized by excessive and prolonged inflammatory respon
