Extracellular vesicles (EVs) contribute to the damage caused by traumatic brain injury (TBI) and can cross the blood-brain barrier (BBB). We analyzed plasma-derived EVs from human TBI patients to identify factors potentially contributing to TBI pathology. EVs were isolated using membrane affinity (ExoEasy) and size exclusion chromatography (iZone), both yielding CD9(+) and CD63(+) EVs with minimal contamination by serum albumin and apolipoprotein. Immunoblotting detected GFAP in TBI but not control EVs, indicating astrocyte-derived EVs crossing the BBB. Proteomic analysis and immunoblotting of

Drought stress exacerbates non-stomatal water loss, which hinders agricultural growth and global food security. The cuticle, a layer of cutin, waxes, and other polymers, protects plants from the negative impact of drought. One crucial adaptation that has enabled terrestrial plants to survive and flourish in drought-prone regions is the development of cuticle structures. In this review, we (1) provide an overview of the molecular and enzymatic pathways involved in the biosynthesis of cutin and wax; (2) analyze important regulatory networks that control the formation of cuticles, such as abscisi

Obesity-associated cognitive decline represents a growing public health concern, yet the mechanisms linking high-fat diet (HFD) to neurological impairment remain incompletely understood. Xanthohumol (XN) and its non-estrogenic derivatives, tetrahydroxanthohumol (TXN) and α,β-dihydro-xanthohumol (DXN), improve metabolic dysfunction and cognitive impairment associated with diet-induced obesity. The mechanisms underlying these cognitive benefits remain poorly defined, but all three compounds improve glucose tolerance, spatial learning and memory in obese C57BL/6J mice. We hypothesized that the gu

Background: The SARS-CoV-2 3CLpro is essential for viral replication and an attractive target for antiviral intervention. While most strategies target the catalytic site, recent studies suggest that the dimerization interface and cryptic allosteric pockets offer alternative mechanisms for inhibition. Objective: This study investigated lipid metabolites from the marine sediment-derived Streptomyces sp. DSD454T as potential multi-site 3CLpro inhibitors. Methods: Metabolites were extracted from cultured biomass and characterized using LCMS-QTOF, MS/MS (LCMS-TQ), and 1H NMR, with identities confir

PURPOSE: The general population is at risk of exposure to ionizing radiation due to nuclear warfare, terrorism, or radiological accidents. Such exposures can lead to acute radiation syndrome (ARS), a set of debilitating and often fatal health effects that start within the first week after irradiation. ARS is largely defined by injuries to the bone marrow and intestinal tract, two organ systems that are highly sensitive to the deleterious effects of ionizing radiation. There is currently no FDA-approved, minimally invasive assay to identify absorbed radiation dose in subjects suspected to have

Bisphenol A (BPA) and alcohol (ALC) are recognized hepatotoxicants to which humans are chronically exposed through environmental sources, dietary habits, and preferences for alcoholic beverages. However, there is currently a lack of conclusive evidence concerning the synergistic hepatotoxic effects associated with co-exposure. This study provides mechanistic insights into BPA and ALC co-exposure-induced subcellular changes mimicking hepatotoxicity using acute and chronic 2D models and 3D HepG2/LX-2 spheroids. Results demonstrated that co-exposure amplified oxidative stress, DNA damage, and apo

OBJECTIVE: The incidence of metabolic-associated steatotic liver disease (MASLD) remains persistently high, and there is a lack of specific therapeutic drugs in clinical practice. Elucidating the mechanistic pathways of therapeutic drugs for MASLD and exploring biomarkers with high specificity and sensitivity are of great significance for the precise diagnosis and treatment of the disease. The present study aims to review the research progress in this relevant field.
METHODS: We systematically collated recent domestic and international literature on the mechanistic pathways of therapeutic dru

BACKGROUND: Aberrations in neuro-immune, metabolic, and oxidative stress (NIMETOX) pathways are implicated in major depressive disorder (MDD). First-episode simple dysmood disorder (FE-SDMD) without metabolic syndrome offers a unique model to investigate early lipid alterations using a systems biology approach.
METHODS: Plasma samples were collected from 88 university students (44 FE-SDMD, 44 healthy controls). Participants underwent psychiatric assessments, including adverse childhood experiences (ACEs), negative life events (NLEs), depression, anxiety, suicidal behaviors, and insomnia. Unta

Epidemiological evidence suggests fetal hyperglycemia predisposes the offspring of mothers with gestational diabetes (GDM) to insulin resistance. While breastfeeding is known to improve metabolic outcomes in offspring from GDM mothers, the breast-milk (BM) composition may be significantly altered. How the BM in GDM-induced changes influence insulin signaling and contribute to-or interfere with-these clinical benefits remains unknown. We hypothesized that impaired maternal glucose tolerance (IGT) during gestation alters BM-composition. Using a high-calorie nutritional rodent GDM-model, we inves

Human breast milk composition is inextricably linked to maternal physiology, yet the impact of thyroid dysfunction on this biological matrix remains undercharacterized. Given that lactation serves as the primary metabolic conduit for the neonate, alterations in the milk metabolic profile could have profound developmental consequences. This study investigates metabolomic and proteomic differentiations in breast milk associated with maternal thyroid abnormalities to evaluate their potential implications for infant growth and neurodevelopment. A systematic search was conducted across PubMed, Emba

Breast cancer (BC) is estimated to be around 2.3 million new cases and 670 000 deaths in 2020 resulting it into the most frequent cancer globally and ranks fifth among mortalities. Numerous studies have established the link between BC and microRNAs (miRNAs), which are endogenous non-protein-coding master regulators that modulate genetic expressions and influences many physiological and pathological cellular processes. In addition, hypoxia, which also consequently leads to cancer, is a pivotal driver of biological behaviour and malignant traits of cancer cells and eventually tumour cell surviva

INTRODUCTION: Maternal obesity contributes to programming of offspring obesity. A causal pathway for placental inflammatory changes in utero with obesity remains to be established. We examined the role of the toll-like receptor (TLR) pathway in mediating maternal obesity associated changes in placental gene expression using trophoblast-specific deletion of the TLR adapter protein, MyD88.
METHODS: Female MyD88 conditional knockouts (MyD88-CKO) at 5 weeks of age were provided control (17% fat calories, CON) or high-fat diets (HFD, 45% fat calories) for 12 weeks and mated with MyD88 (flox-contro

Dilated cardiomyopathy (DCM) was the most prevalent cardiomyopathy worldwide. Although ferroptosis has been implicated in cardiac pathogenesis, its regulatory mechanism in DCM remained poorly defined. In this study, we found that GIPC1 (GAIP/RGS19-interacting protein), a scaffolding protein, was significantly downregulated in cardiac tissues from DCM patients and doxorubicin (DOX)-induced DCM models. Integrated proteomic and lipidomic analysis revealed that cardiac-specific knockout of GIPC1 disrupted mitochondrial fatty acid metabolism, increased the abundance of polyunsaturated fatty acid-co

INTRODUCTION: It is uncertain whether hyperoxia contributes to adverse intensive care unit outcomes. Potential harm is likely mediated by oxidative stress driving pulmonary and systemic inflammation.
METHODS: We conducted a randomised, double-blind, placebo-controlled trial to test the hypothesis that hyperoxia modifies pulmonary and systemic inflammation following inhaled lipopolysaccharide (LPS) challenge. Healthy adult volunteers were randomised (1:1) to receive hyperoxia administered via high-flow nasal oxygen (FiO2 1.0, 60 L/min) or synthetic medical air for 6 hours. Bronchoalveolar lava

BACKGROUND & AIMS: Fatty liver diseases are highly prevalent worldwide, driven by hepatocyte metabolic dysfunction and alcohol consumption. Both cell-autonomous and non-cell-autonomous mechanisms contribute to hepatic lipid accumulation. Here, we investigated the role of liver endothelial cell-hepatocyte communication in regulating lipid metabolism.
METHODS: The in vivo function of Heg1 in liver endothelial cells was investigated using 2 endothelial-specific knockout models, conditional (Lyve1-Cre;Hegfl/fl) and inducible (Cdh5-CreERT2;Hegfl/fl), in the context of 3 liver disease models: metab

Age is a critical determinant of COVID-19 severity, yet the underlying metabolic mechanisms driving disease progression remain poorly understood. Using a well-characterized Syrian hamster model, we investigated how aging shapes intestinal and systemic sterol metabolism during SARS-CoV-2 infection. Young (7-10 weeks) and aged (12 months) hamsters were intranasally infected, and clinical outcomes were integrated with virological, histopathological, and lipidomic profiles across intestinal tissues, lungs, and serum. Despite comparable pulmonary pathology, aged hamsters exhibited significantly hig

Pericytes are critical for maintaining blood-brain barrier (BBB) integrity and have emerged as key contributors to Alzheimer's disease (AD) pathogenesis. Although the apolipoprotein E2 (APOE2) allele is associated with reduced AD risk and increased longevity, its impact on pericyte function is unclear. We measured pericyte density in the brains of humanized APOE2, APOE3, and APOE4 knock-in mice and found that APOE2 mice revealed increased pericyte markers and enhanced BBB integrity. To uncover the underlying mechanisms, we used CRISPR/Cas9 editing to generate isogenic human iPSC-derived pericy

This study investigated whether a diet rich in lauric acid, based on Hermetia illucens (HI) larvae meal, would induce measurable changes in lipid-related metabolic pathways in the liver and in the fatty acid composition of tissues. Twenty-four weaned piglets were assigned to three dietary groups: control (0% HI), 2.5% HI, and 5% HI. Multilevel analyses, including histology, biochemical assays, fatty acid profiling, and gene expression, were performed to assess liver function and metabolic adaptations of animal organisms. Dietary HI markedly reshaped the tissue lipid composition, increasing the

Background: Parkinson's disease (PD) is a progressive neurodegenerative disorder with a well- documented prodromal phase during which non-motor symptoms appear years before motor onset. Seborrheic dermatitis (SD) is a chronic inflammatory skin disease that is significantly more prevalent in PD patients than in the general population. The biological mechanisms underlying this association have not been integrated into a single framework. Objectives: This narrative review aims to synthesise the available evidence on the molecular, genetic, and pathophysiological mechanisms linking SD and PD. Meth
