The retinal nerve fiber layer mean thickness in patients with early Parkinson's disease reflects striatal dopamine function.

BACKGROUND: The purpose of this study was to investigate the relationship between the retinal nerve fiber layer (RNFL) mean thickness and striatal dopamine (DOPA) uptake in patients with early Parkinson's disease (PD) and to evaluate the potential value of the RNFL mean thickness in the diagnosis of early PD. METHODS: Plasma levels of α-synuclein (α-syn) were measured by enzyme-linked immunosorbent assay (ELISA). Motor and nonmotor symptoms were assessed with related scales. DOPA uptake was measured with DOPA positron emission tomography (PET)/ computed tomography (CT). The RNFL mean thicknes

A53T α-Synuclein Expression is Associated with Altered Dopaminergic-Like Differentiation and Reduced DNA Topoisomerase IIβ Levels in an In Vitro Model of Parkinson's Disease.

The study explores the interaction between topoisomerase IIβ (topo IIβ) and α-synuclein aggregation in Parkinson's disease (PD) using an in vitro model overexpressing A53T mutant α-synuclein, revealing key pathological features. The overexpression of A53T mutant α-synuclein was associated with impaired neuronal differentiation, accompanied by delayed neurite outgrowth and reduced levels of MAP2 and NF-L. Transcriptomic study during differentiation revealed substantial gene expression changes, with 57 of 84 PD-associated genes exhibiting differential regulation on days 8 and 12. Dopaminergic ma

Pharmacological inhibition of O-GlcNAcase reduces pS129-α-synuclein positive aggregates in the substantia nigra of mThy1-hSNCA mice.

BackgroundThe aggregation and spread of α-synuclein within brain are associated with the loss of dopaminergic neurons and the formation of Lewy bodies as seen in Parkinson's disease. Blocking the initiation of α-synuclein aggregation, or the spread of such aggregates, may offer disease-modifying approaches to slow disease progression. Previous studies have demonstrated that modification of aggregation prone proteins, including α-synuclein, with O-linked β-N-acetylglucosamine (O-GlcNAc) reduces their aggregation. Small molecule inhibitors of the enzyme O-GlcNAcase (OGA), which removes O-GlcNAc

Diagnostic Performance of the α-Synuclein Seed Amplification Assay for Dementia With Lewy Bodies: A Comparison Across 4 Laboratories.

BACKGROUND AND OBJECTIVES: The α-synuclein (α-syn) seed amplification assay (SAA) has shown promising results for diagnosing dementia with Lewy bodies (DLB) using CSF samples. A barrier to implementing α-syn SAA clinically is the use of different protocols for the assay. It is unknown how different protocols perform in comparison with each other. We compared the performance of α-syn SAA across 4 laboratories using CSF samples of patients with DLB. METHODS: This was a retrospective cross-sectional study that included data from 4 different European laboratories. We included probable patients wi

Loss-of-Function (G603R) Lrp10 Fails to Downregulate mRNA of Pathologic α-Synuclein and Causes Neurodegeneration of Substantia Nigra Dopaminergic Cells in Parkinson's Disease Knockin Mice.

Heterozygous (G603R) LRP10 mutation causes autosomal dominant Parkinson's disease (PD). Heterozygous Lrp10G603R/+ mice were prepared to unravel pathomechanisms underlying (G603R) Lrp10-induced death of substantia nigra (SN) dopaminergic neurons. Lrp10G603R/+ mouse exhibited PD movement deficits, neurodegeneration of SN dopaminergic cells and existence of SN phospho-α-synuclein-containing aggregates. Lrp10 was expressed in mouse SN dopaminergic neurons, and WT LRP10 exerted neuroprotection function on dopaminergic cells by repressing α-synuclein gene transcription and downregulating α-synuclein

Crude proline modulates the microbiota-gut-brain axis by attenuating curli-associated α-synuclein accumulation and regulating Wnt/β-catenin signaling in Parkinson's disease.

This study investigated whether crude proline (CP), a metabolite fraction derived from heat-killed Enterococcus faecium (E. faecium) FBL1 isolated from makgeolli, is associated with the modulation of microbiota-host interactions within the microbiota-gut-brain axis and attenuation of PD-related pathology. CP was purified using fast protein liquid chromatography and liquid chromatography-mass spectrometry. Its biological effects were evaluated in SH-SY5Y and Caco-2 cell models and in an MPTP-induced mouse model of PD, followed by behavioral, microbiota, and molecular analyses. CP treatment sign

A Pilot Study Evaluating the Effect of Artesunate on Glymphatic-Related Fluid Transport in Parkinson's Disease Mouse Models In Vivo and Ex Vivo.

AIMS: Artesunate (ART) has shown neuroprotective potential, but its effects on glymphatic-related fluid transport in Parkinson's disease (PD) remain unclear. This pilot study aimed to explore whether ART affects glymphatic-related fluid transport in a PD mouse model and to assess the feasibility of evaluating these changes in vivo. METHODS: C57BL/6 WT and A53T mice (n = 6 each) were divided into four groups (WT, WT + ART, A53T, and A53T + ART) and treated with ART or DMSO for 7 days. Glymphatic-related fluid transport was assessed by dynamic MRI after intraventricular Gd-DTPA injection. Immun

iSCORE-PD: an isogenic stem cell collection to research Parkinson's disease.

Genome-edited human pluripotent stem cells (hPSCs) provide a powerful platform to study complex diseases such as Parkinson's disease (PD). Here, we describe iSCORE-PD, an isogenic collection of 65 genome-edited hPSC lines carrying disease-causing or high-risk variants in 11 PD-linked genes (SNCA, PRKN, PINK1, DJ1/PARK7, LRRK2, ATP13A2, FBXO7, DNAJC6, SYNJ1, VPS13C, and GBA1). All lines are derived from a well-characterized female hESC line and subjected to extensive quality control. Whole-genome sequencing reveals that genetic variation between lines, largely confined to non-coding regions, is

Attention-deficit/hyperactivity disorder-related psychomotor activity and altered neuronal activity in the medial prefrontal cortex and striatum in the A53T mouse model of Parkinson's disease and other synucleinopathies: Findings from an "endophenotype" approach.

Attention-Deficit/Hyperactivity Disorder (ADHD) is associated with an increased risk of Parkinson's disease (PD) and other synucleinopathies later in life. The severity of the ADHD phenotype may play a significant role in this association. There is no indication that any of the existing animal models can unify these disorders. Using the Open Field Test, amphetamine-challenge test, Western blot and immunohistochemical analysis of neuronal activity markers (c-Fos, FosB and ΔFosB) we performed a deliberate neurobehavioral characterization of 6-month-old hemizygous A53T carriers (A53T+) of the JAX

Neuroprotective efficacy of berberine and caffeine against rotenone-induced neuroinflammatory and oxidative disturbances associated with Parkinson's disease via inhibiting α-synuclein aggregation and boosting dopamine release.

Parkinson's disease (PD) is characterized by motor impairment, glial-mediated inflammation, redox imbalance, and α-synuclein (α-syn) aggregation. Conventional therapies relieve early PD symptoms, but they do not repair dopaminergic neurons. Berberine (BBR) and caffeine (CAF), both natural alkaloids, exhibited neuroprotective effects in many neurodegenerative disorders. Consequently, we hypothesized that the combination of BBR and CAF therapies would offer protection against PD-related impairments in the rotenone (ROT)-induced rat model when compared to the commercial drug, metformin (MTF). NHR-85 modulates mitochondrial and lipid homeostasis to protect against α-synuclein aggregation in C. elegans.

Peroxisome proliferator-activated receptors (PPARs), such as PPARδ, are transcription factors that play a pivotal role in energy and fat metabolism. PPARδ activates genes involved in lipid and glucose metabolism and is expressed in various human tissues, including all brain regions and especially neurons, where it regulates lipid homeostasis and contributes to neuroprotection. However, the precise molecular mechanisms underlying these protective effects remain poorly understood. Here, we identify the Caenorhabditis elegans nuclear hormone receptor NHR-85 as a putative orthologue of human PPARδ

Apigenin enhances Nrf2-induced chaperone-mediated autophagy and mitigates α-synuclein pathology: Implications for Parkinson's disease therapy.

BACKGROUND: The aggregation of α-synuclein (SNCA) in dopaminergic neurons of the substantia nigra is a key factor in the pathogenesis of Parkinson's disease (PD). Despite years of drug discovery efforts targeting SNCA aggregation, no disease-modifying drugs have been approved to date. The failure of numerous clinical trials can be attributed, at least in part, to the difficulty in identifying potent compounds during preclinical investigations. OBJECTIVE: Establish a screening approach based on molecular docking and autophagic flux detection to identify natural compounds from new perspectives

Progress in Disease-Modifying Therapies for Parkinson's Disease.

Parkinson's disease (PD) is a highly prevalent neurodegenerative disorder, and current therapeutic approaches fail to prevent the progressive loss of dopaminergic neurons. To date, no disease-modifying therapies (DMTs) have been approved for PD. Developing effective DMTs remains the foremost objective in PD research. Here, we review the rationale for α-synuclein, LRRK2, GBA1, the PINK1-Parkin axis, and GLP-1R as potential therapeutic targets for PD. Additionally, we summarize the functional alterations observed in cellular organelles, including mitochondria, lysosomes, and the endoplasmic reti

A Conserved MicroRNAs Motif Reveals TDP-43 Pathology and MAPK Pathway Activation in Parkinson's Disease.

Parkinson's disease (PD), the second most common neurodegenerative disorder, involves dopaminergic neuron loss and α-synuclein (α-syn) aggregation. Recent studies have shown that dysregulation of microRNAs (miRNAs) is associated with PD, but little is known about its pathological mechanism. Here, we identified a conserved 5-nucleotide motif (HBCCC; H = U, A, or C; B = U, C, or G) in 52 downregulated miRNAs from the salivary RT-QuIC (real-time quaking-induced conversion)-positive PD patients, accounting for 82.5% of these dysregulated miRNAs. The motif-containing miRNAs, including miRNA-6812-3p

CHCHD2 mutant mice link mitochondrial deficits to PD pathophysiology.

Mitochondrial dysfunction is a hallmark of Parkinson's disease (PD), but the mechanisms by which it drives autosomal dominant and idiopathic forms of PD remain unclear. To investigate this, we generated and performed a comprehensive phenotypic analysis of a knock-in mouse model carrying the T61I mutation in the mitochondrial protein CHCHD2 (coiled-coil-helix-coiled-coil-helix domain-containing 2), which causes late-onset symptoms indistinguishable from idiopathic PD. We observed pronounced mitochondrial disruption in substantia nigra dopaminergic neurons, including distorted ultrastructure and

Genome-wide epistasis analysis reveals significant epistatic signals associated with Parkinson's disease risk.

Genome-wide association studies have increased our understanding of Parkinson's disease genetics by identifying common disease-associated variants. However, much of the heritability remains unaccounted for, and we hypothesized that this could be explained, in part, by epistasis, the statistical interaction between two or more genetic variants. Here, we developed a genome-wide non-exhaustive epistasis screening pipeline called variant-variant interaction through variable thresholds (VARI3) and applied it to diverse Parkinson's disease genome-wide association study cohorts. We used 14 cohorts of

A Biomarker-Based Classification of Corticobasal Syndrome.

BACKGROUND: Corticobasal syndrome (CBS) is a clinically defined syndrome with progressive movement and cortical dysfunction, caused by various underlying pathologies, most commonly tau-predominant pathologies such as progressive supranuclear palsy and corticobasal degeneration, or Alzheimer's disease (AD). Lewy-type α-synucleinopathies (LTS), TDP-43 proteinopathies, and mixed pathologies may also underlie CBS. The clinical impact of these pathologies remains poorly understood. OBJECTIVES: To subclassify CBS patients in vivo using biomarkers for amyloid-β (Aβ), Tau, and α-synuclein (αSyn), and

Chenodeoxycholic Acid-Mediated neuroprotection via α-synuclein and BDNF Modulation in MPTP-Induced mouse model of Parkinson's disease.

Parkinson's disease (PD) remains a major challenge in the field of neurodegenerative diseases and requires innovative therapeutic approaches. In this study, we investigated the therapeutic potential of chenodeoxycholic acid (CDCA) in PD using a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced mouse model. CDCA, a naturally occurring bile acid, has previously shown promise in various neurological disorders by reducing neuronal degeneration and promoting neuronal health, however its utility in PD has not been studied. We divided mice into a control group, an MPTP-induced PD model and

Propagation of pathologic α-synuclein from kidney to brain may contribute to Parkinson's disease.

The pathogenesis of Lewy body diseases (LBDs), including Parkinson's disease (PD), involves α-synuclein (α-Syn) aggregation that originates in peripheral organs and spreads to the brain. PD incidence is increased in individuals with chronic renal failure, but the underlying mechanisms remain unknown. Here we observed α-Syn deposits in the kidneys of patients with LBDs and in the kidney and central nervous system of individuals with end-stage renal disease without documented LBDs. In male mice, we found that the kidney removes α-Syn from the blood, which is reduced in renal failure, causing α-S
