Cryptic Production of trans-3-Hydroxyproline in Echinocandin B Biosynthesis.

Echinocandins are antifungal nonribosomal hexapeptides produced by fungi. Two of the amino acids are hydroxy-l-prolines: trans-4-hydroxy-l-proline and, in most echinocandin structures, (trans-2,3)-3-hydroxy-(trans-2,4)-4-methyl-l-proline. In the case of echinocandin biosynthesis by Glarea lozoyensis, both amino acids are found in pneumocandin A0, while in pneumocandin B0 the latter residue is replaced by trans-3-hydroxy-l-proline (3-Hyp). We have recently reported that all three amino acids are generated by the 2-oxoglutarate-dependent proline hydroxylase GloF. In echinocandin B biosynthesis b

From synthesis to function via iterative assembly of N-methyliminodiacetic acid boronate building blocks.

The study and optimization of small molecule function is often impeded by the time-intensive and specialist-dependent process that is typically used to make such compounds. In contrast, general and automated platforms have been developed for making peptides, oligonucleotides, and increasingly oligosaccharides, where synthesis is simplified to iterative applications of the same reactions. Inspired by the way natural products are biosynthesized via the iterative assembly of a defined set of building blocks, we developed a platform for small molecule synthesis involving the iterative coupling of

Exploring genetic diversity, population structure, and phylogeography in Paracoccidioides species using AFLP markers.

Paracoccidioidomycosis (PCM) is a life-threatening systemic fungal infection acquired after inhalation of Paracoccidioides propagules from the environment. The main agents include members of the P. brasiliensis complex (phylogenetically-defined species S1, PS2, PS3, and PS4) and P. lutzii. DNA-sequencing of protein-coding loci (e.g., GP43, ARF, and TUB1) is the reference method for recognizing Paracoccidioides species due to a lack of robust phenotypic markers. Thus, developing new molecular markers that are informative and cost-effective is key to providing quality information to explore gene

Profile of Invasive Fungal Infections in a Paediatric Haemato-Oncology Unit: A Five-Year Retrospective Cohort Study From a Tertiary Care Centre in South India.

BACKGROUND: Invasive fungal infections (IFIs) are a leading cause of morbidity and mortality in paediatric haemato-oncology (PHO) patients. Empirical antifungal therapy in high-risk children must be informed by local epidemiological, species-level and antifungal-susceptibility data, yet contemporary Indian paediatric cohort data remain limited. OBJECTIVE: To describe the clinical, microbiological, therapeutic and outcome profile of probable and proven IFI in a tertiary care South Indian PHO unit over five years, and to benchmark the findings against contemporary international literature. Umami-active components in edible Fungi: Distribution, structure-function relationships, molecular mechanisms, computational insights and industrial applications.

Edible mushrooms are a biodiverse yet underutilized source of umami compounds providing clean-label alternatives to synthetic flavourings such as monosodium glutamate (MSG). The characteristic savory taste arises from interactions among free amino acids (glutamate, aspartate), 5'-ribonucleotides (5'-GMP, 5'-IMP), organic acids (succinic, citric), polyols (trehalose, mannitol), and bioactive peptides, whose accumulation is influenced by genetic composition, growth substrate, developmental stage, and postharvest management. This review systematically examines the distribution of umami compounds

Host and Environmental Factors Outweigh Pathogen in Shaping Skin Microbiome of a Threatened Andean Amphibian Community.

The fungal pathogen Batrachochytrium dendrobatidis (Bd) has driven severe amphibian declines across the Neotropics, yet some species persist under enzootic pathogen conditions following epizootics. We investigated whether skin bacterial communities and host mucosome activity contributed to the persistence of seven amphibian species in a once megadiverse Chocó-Andean community that experienced drastic Bd-associated declines. Across dry and wet seasons, we characterized skin bacterial communities using 16S rRNA metabarcoding and assessed mucosome-mediated inhibition of a local Bd lineage using i

Confrontations between Aspergillus nidulans and microbial biocontrol agents cause differential regulation of secondary metabolism and synthesis of chemicals toxic to human kidney and colon cells.

Microbial interactions in agricultural ecosystems are chemically dynamic, with significant implications for ecological balance and crop protection. As synthetic fungicides face increasing regulatory and resistance challenges, applying microbial biological control agents (MBCAs) presents a potential alternative crop-protection strategy. However, the chemical and toxicological consequences of such applications remain poorly understood. Using the model ascomycete Aspergillus nidulans, the response to confrontations with twelve microbial partners, including bacterial and fungal MBCAs, plant pathog

Upcycling rice husk ash into antiseptic-encapsulated ordered mesoporous silica materials for antimicrobial applications.

Rice husk ash (RHA), an abundant agricultural and industrial waste, was upcycled into functional mesostructured silica encapsulating cetylpyridinium chloride (CPC), an FDA-approved antiseptic. Sodium silicate was efficiently extracted from RHA and leveraged as a biosourced silica precursor for the aqueous sol-gel synthesis of mesostructured hybrid silica at 30 °C. Micelles of CPC simultaneously acted as structure-directing agents and as integrated antimicrobial cargo, enabling the direct formation of ordered hybrids in one-pot. The condensation pH influenced the synthesis yield, the bonding co

Exometabolome variation in the fungal pathogen of humans Candida albicans reveals specificities at the genetic clade and strain levels.

Candida albicans is a fungal pathobiont that thrives in different host niches and successfully confronts the host immune defenses. The success of C. albicans as a pathogen relies on a highly plastic metabolism, with examples of variations across isolates; however, detailed characterization of how C. albicans genetic diversity impacts the content of the metabolome is still missing. Here, we have implemented an NMR (nuclear magnetic resonance)-based high-throughput workflow to probe exometabolome variation in in vitro cultures of 96 clinical isolates spread across several genetic clusters (clade

Early-stage intercropping modulates rhizosphere soil properties, microbiome composition, and photosynthetic physiology of Phellodendron chinense: a comparative analysis of five cultivation systems.

INTRODUCTION: Phellodendron chinense is an important medicinal plant in China. However, continuous monoculture has caused soil degradation and physiological constraints, which have become a key bottleneck limiting its sustainable production. Intercropping with companion plants is an effective approach to break this dilemma. METHODS: We selected five representative cultivation systems commonly practiced of Phellodendron chinense and systematically measured their rhizosphere soil properties, microbial community composition, and leaf gas-exchange traits. The overarching goal was twofold: (i) to

Divergent Association Pathways of Soil Organic Carbon Variation Under Grazing Exclusion Across Three Grassland Sites: Relationships with Microbial Network Structure and Community Assembly.

Grazing exclusion is a widely implemented restoration strategy for degraded grasslands, but its effects on soil organic carbon (SOC) remain highly variable, and the ecological pathways associated with these changes-particularly the roles of soil microorganisms-are still insufficiently understood. To investigate context-dependent SOC responses, we conducted a 12-year grazing exclusion study across three contrasting grassland sites representing an environmental gradient (temperate desert, temperate steppe, and mountain meadow) on the northern slope of the Tianshan Mountains. By integrating multi

A mycovirus-encoded homologue of plant viral movement proteins is not functional in movement complementation assays in plants.

Valsa mali negative strand RNA virus 1 (VmNSRV1), a fungal virus infecting Valsa mali and belonging to the Phenuiviridae family, was recently shown to be capable of naturally infecting plants. Its RNA2 encodes a homologue of a plant virus movement protein (MP) that can complement MP-deficient virus vectors and localizes to plasmodesmata, allowing natural cross-kingdom infection. To determine whether this property is a common feature among mycoviruses possessing distant MP homologues or an isolate-specific trait, we investigated Trichoderma gamsii cogu-like virus 1 (TgClV1), a Phenuiviridae mem

Laccase-peroxidase synergy modifies Kraft lignin structure and enhances the electrospinnability for bio-based materials.

Lignin is a promising renewable macromolecular feedstock for advanced carbon-based materials; however, its intrinsic molecular heterogeneity, relatively low molecular weight, and limited thermal stability often hinder direct fiber formation. This study demonstrates that a multienzymatic oxidative system, integrating laccase (O₂-driven, 6.8 × 10⁵ U L⁻¹), manganese peroxidase (Mn3+ shuttle, 1.5 × 10³ U L⁻¹), and lignin peroxidase (high redox potential, veratryl alcohol-free, 3.0 × 10⁴ U L⁻¹),effectively expands the electrospinnability window of Kraft lignin (KL). The redox-diverse enzymatic cock

Challenges in Diagnosis and Treatment of HIV-Negative Host Pulmonary Talaromyces marneffei in Non-Endemic Areas: A Case Report with a History of Pulmonary Tuberculosis.

Talaromyces marneffei is an important opportunistic fungal pathogen closely related to acquired immunodeficiency syndrome (AIDS), and its infection is relatively rare in human immunodeficiency virus (HIV)-negative populations. Although HIV-related immunosuppression remains the main risk factor, the history of treated tuberculosis and subsequent structural lung damage may constitute an underrecognized predisposing condition in non-epidemic areas, especially in elderly patients. This report described a 69-year-old HIV-negative male pulmonary infection case from a non-endemic area of T. marneffei

Test, Treat, Repopulate™ method reduces red complex bacteria and stabilizes the oral microbiome.

Despite advances in dental hygiene and clinical interventions, preventable conditions such as tooth decay and gum disease remain widespread public health concerns. Recent research implicates complex, polymicrobial interactions as central drivers of disease pathogenesis, which often limits the effectiveness of traditional therapeutic approaches. Here, researchers evaluate the effectiveness of the Test, Treat, Repopulate method, a multi-phase oral health protocol combining scaling and root planing, targeted antibiotic therapy, use of a pH-balanced, prebiotic containing toothpaste and rinse, and

Azole-resistant Aspergillus fumigatus and related species in Japan.

Azole-resistant Aspergillus, particularly azole-resistant Aspergillus fumigatus (ARAf), is an increasingly consequential problem in Japan because triazoles remain central to aspergillosis management and chronic pulmonary aspergillosis (CPA) often requires prolonged oral therapy. CPA provides a within-host evolutionary niche in which persistent fungal burden and long-term azole exposure select resistant subpopulations, contributing to non-response, relapse, and progressive lung destruction. This review synthesizes Japan-specific clinical and environmental epidemiology, resistance mechanisms, an
