BACKGROUND: Tuberculosis (TB), caused by the intracellular bacterium Mycobacterium tuberculosis complex (Mtbc), remains a significant global health challenge, with Mtbc once again being the leading infectious killer worldwide. Despite over a century of research, the disease continues to pose a major threat, with an estimated one-fourth of the global population latently infected. According to the World Health Organization (WHO), approximately 1.3 million deaths were attributed to TB in 2024 alone. The emergence of multidrug-resistant (MDR) strains, resistant to isoniazid and rifampicin, and ext

Tuberculous meningitis (TBM) is the most severe form of Mycobacterium tuberculosis (Mtb) infection in humans and is a public health concern worldwide. We evaluated the performance of GeneXpert MTB/RIF (GeneXpert) for the diagnosis of TBM. In addition, genetic diversity and drug susceptibility profiling of Mtb strains isolated from TBM patients were also investigated. A total of 293 TBM suspected cerebrospinal fluid (CSF) samples were collected and subjected to GeneXpert and Mycobacterial Growth Indicator Tube (MGIT 960) culture, respectively. Sensitivity and specificity of GeneXpert was 72.7%

Bacteriophage can be used as an alternative or complementary therapy to antibiotics for treating multidrug-resistant bacterial infections. However, the rapid emergence of resistant host variants during phage treatment has limited its therapeutic applications. In this study, a potential phage-resistant mechanism of Klebsiella pneumoniae was revealed. After phage GH-K3 treatment, a smooth-type colony, named K7RB, was obtained from the K. pneumoniae K7 culture. Treatment with IO4- and/or proteinase K indicated that polysaccharides of K7 played an important role in phage recruitment, and protein r

Peripheral T-cell lymphomas (PTCL) are usually characterized by aggressive clinical behaviour and poor clinical outcome, but their biological background has not been extensively investigated to date, due to their low incidence, about 10% of all non-Hodgkin's lymphoma cases in Western countries, and also to the paucity of specific molecular-genetic abnormalities. Neverthless, there is increasing biological and clinical evidence that primary nodal PTCL should be considered separately from extra-nodal cases, but little is known about biological factors of possible clinical and prognostic impact.

UNLABELLED: Multidrug-resistant (MDR) Pseudomonas aeruginosa infections pose a major challenge to effective treatment. Understanding genomic adaptations during antimicrobial therapy in patients infected with this pathogen is crucial for preventing therapeutic failure. Here, we investigated the population diversity and evolution of P. aeruginosa collected longitudinally from six patients who evolved multidrug-resistant infections. Serial clinical P. aeruginosa single-colony isolates (n = 63) and culture-enriched metagenomic population samples (n = 39) were collected and subjected to whole-genom

Multidrug-resistant (MDR) Gram-negative bacteria-particularly carbapenem-resistant Enterobacterales, Pseudomonas aeruginosa and Acinetobacter baumannii-remain among the hardest infections to treat, and Resistance-Nodulation-Division (RND) efflux pumps are central to that resistance, making them a recurring target for antibiotic-adjuvant strategies. Natural products offer chemically diverse starting points for efflux-modulating adjuvants, but a fall in MIC is hard to attribute to pump inhibition: membrane permeabilisation, proton-motive-force (PMF) dissipation, efflux-gene downregulation, intri

BACKGROUND: Antimicrobial resistance (AMR) represents a pressing global health challenge, influencing human, animal, and environmental health within the interconnected One Health framework. ESBL- and carbapenemase-producing Enterobacteriaceae constitute a major global public health threat. In Lebanon, their presence is particularly concerning, as they are detected across diverse environments, including hospitals, surface water, wastewater, poultry, and livestock. Despite close contact between pets and their owners, the role of companion animals, particularly cats, in spreading AMR determinants

Salmonella enterica are important foodborne bacterial pathogens globally associated with poultry. Exposure to Salmonella-contaminated eggs and egg-related products is a major risk for human salmonellosis. Presently, there is a huge data gap regarding the prevalence and circulating serovars of Salmonella in chicken eggs sold in Ghana. In this study, 2,304 eggs (pools of six per sample unit) collected from informal markets in Accra, Kumasi and Tamale, representing the three ecological belts across Ghana, were tested for Salmonella. Antimicrobial susceptibility testing and Whole Genome Sequencing

The World Health Organization recommends diagnosing Multidrug-Resistant Tuberculosis (MDR-TB) in high burden countries by detection of mutations in Rifampin (RIF) Resistance Determining Region of Mycobacterium tuberculosis rpoB gene with rapid molecular tests GeneXpert MTB/RIF and Hain MTBDRplus. Such mutations are found in >95% of Mycobacterium tuberculosis strains resistant to RIF by conventional culture-based drug susceptibility testing (DST). However routine diagnostic screening with molecular tests uncovered specific "low level" rpoB mutations conferring resistance to RIF below the critic

BACKGROUND: Patients with peripheral T-cell lymphomas (PTCLs) have inferior progression-free survival (PFS) and overall survival (OS) compared with patients who have aggressive B-cell non-Hodgkin lymphoma. Because PTCLs over express multidrug resistance gene 1/P-glycoprotein (MDR-1/P-gp), we devised platinum, etoposide, gemcitabine, and methylprednisolone (PEGS) with agents that are not substrates of the efflux pump. Gemcitabine was included because of its excellent single-agent activity in PTCL.
METHODS: Patients who had PTCL with stage II bulky disease, stage III or IV disease with extra-no

Clinical findings indicate that co-administration of the isoxazolyl-penicillin flucloxacillin with cyclosporine may reduce the plasma concentrations of cyclosporine. We have explored in the present study if induction of cytochrome P450 3A4 or P-glycoprotein may offer a mechanistic explanation of the observed effects. Flucloxacillin is neither an inhibitor nor a substrate of drug metabolizing cytochrome P450 isoenzymes (CYP3A4, 1A2, 2C9, 2C19 and 2D6) or P-glycoprotein as shown by an in vitro assay for CYP inhibition, a fluorescent indicator assay for P-glycoprotein inhibition and a functional

Nature-based wastewater treatment systems are increasingly implemented to support water reuse in arid regions, yet their effectiveness in removing viable culturable opportunistic pathogens remains insufficiently characterized. In this exploratory, culture-based study, we assessed bacterial population dynamics across a five-stage natural wastewater treatment system (Wadi Hanifa, Riyadh, Saudi Arabia), tracking culturable bacteria from secondary-treated influent to sand-filtered effluent. Water samples were collected at five sequential treatment stages and analyzed for physicochemical parameters

BACKGROUND: Mycobacterium tuberculosis resistance to anti-tuberculosis drugs is a major threat to global public health. Whole genome sequencing (WGS) is rapidly gaining traction as a diagnostic tool for clinical tuberculosis settings. To support this informatically, previous work led to the development of the widely used TBProfiler webtool, which predicts resistance to 14 drugs from WGS data. However, for accurate and rapid high throughput of samples in clinical or epidemiological settings, there is a need for a stand-alone tool and the ability to analyse data across multiple WGS platforms, in

Non-typhoidal Salmonella (NTS) is a major foodborne pathogen causing significant morbidity and mortality worldwide, particularly affecting children under 5 years of age. Despite its public health importance, systematic studies on NTS infection in children in Hainan Province, China, are limited. We performed a comprehensive retrospective analysis of 77 NTS isolates collected from children at the Hainan Women and Children's Medical Center between April 2023 and May 2024. The study integrated antimicrobial susceptibility testing, whole-genome sequencing, and phylogenetic reconstruction to elucida

The banning of colistin as a feed additive for food-producing animals in mainland China in 2017 caused the decline in the prevalence of Escherichia coli-mobilized colistin resistance (mcr-1) in China. Salmonella Typhimurium and its monophasic 1,4,[5],12:i:- variants are also the main species associated with the spread of mcr-1; however, the evidence of the prevalence and transmission of mcr-1 among Salmonella is lacking. Herein, the 5,354 Salmonella isolates recovered from fecal samples of diarrheal patients in Guangdong, Southern China, from 2009 to 2019 were screened for colistin resistance

Backgrounds: Autocrine motility factor (AMF) represents a paradoxical protein with dual roles in cancer progression and therapy. This study investigated AMF-derived tetradecapeptides as novel chemosensitizing agents to overcome multidrug resistance (MDR) in hematological malignancies (HMs). Methods: Seven AMF variants were screened for anticancer activity across 14 human cancer cell lines using MTT and Cell Counting Kit-8 (CCK-8) assays. Four tetradecapeptides (AMF-derived peptides AAP, HGP, HTP, and SKP) corresponding to the AMF206-219 region were designed and evaluated in three HM cell lines

BACKGROUND: Fluoroquinolone (FQ) resistance in Mycobacterium tuberculosis (MTB) is a major cause of treatment failure in multidrug-resistant tuberculosis (MDR-TB). This resistance primarily results from mutations within the quinolone resistance-determining region (QRDR) of the gyrA gene encoding DNA gyrase. Conventional phenotypic drug susceptibility testing (DST) is labor-intensive and time-consuming, making it unsuitable for rapid clinical decision-making. Therefore, developing a rapid, sensitive, and point-of-care testing (POCT) assay is of great importance.
METHODS: A cartridge-based POCT
