The results of the laboratory assays of the Yugoslav typhoid vaccines used in the field trials at Osijek are disappointing when compared with the results of the field trials. The tests carried out at the Lister Institute in England, the Central Institute of Hygiene in Zagreb, Yugoslavia, and the Walter Reed Army Institute of Research in the USA indicate that, except for the agglutination tests for H antigen, there is little or no demonstrable difference in the potency of the two Yugoslav vaccines in either active immunization tests or passive protection tests.Although only a relatively small nu

Enteric fever, a systematic bacterial infection caused by Salmonella Typhi and Paratyphi, continues to impose a significant public health burden in low and middle-income countries, yet our understanding of the serum antibody dynamics following infection remains incomplete. Although previous work has characterized the longitudinal seroresponses following acute typhoid infection, gaps persist in deciphering how repeated exposures influence antibody decay and protection. In our longitudinal cohort study of blood culture-confirmed enteric fever cases enrolled in Bangladesh, Nepal, and Pakistan, we

Cross-reactivity in a delayed-type hypersensitivity (DH) response was studied in mice immunised with live Salmonella typhi, S. paratyphi A and S. paratyphi B. Extensive cross-reactions outside the serogroup limits were observed. The ability of DH cross-reacting and non-cross-reacting sonicates to generate activated macrophages was studied in mice immunised 3 months earlier with S. paratyphi B. Whereas DH cross-reacting S. poona sonicate generated activated macrophages the non-cross-reacting S. typhi sonicate did not. To determine whether infections due to diarrhoea-causing salmonellae generate

The purpose of our study was to analyze the use of febrile agglutinin (FA) serologic tests in the diagnosis of Salmonella (typhi and paratyphi), Rickettsia, Brucella, and Francisella infections. Data were collected in a retrospective chart study of 202 titer requests covering a 42-month period at our teaching hospital. We found that no diagnoses were made by utilizing complete six-antigen FA panels. Three cases of infection were diagnosed serologically, all three with more discriminately selected single-antigen titers. These three cases each had a positive epidemiological history suggestive of

The in vivo growth of Salmonella paratyphi A, S. paratyphi B, S. paratyphi C, and S. typhi, as well as of an S. typhi-typhimurium hybrid, was studied in three different strains of mice. S. paratyphi A and B and S. typhi demonstrated very little growth potential in any of the intravenously infected mice, even after as many as 20 serial mouse passages. It was noted, however, that small numbers of viable S. paratyphi B and S. typhi persisted in the spleens of infected mice for up to 28 days. Salmonella paratyphi C and the S. typhi-typhimurium hybrid gave rise to progressive systemic infections be

Annually, there are 5.57 to 9.23 million typhoid fever cases and 46,200 to 163,000 associated deaths, mainly in low- and middle-income countries. High incidence is estimated in the Southeast Asian, African, and Eastern Mediterranean (EMR) regions of the WHO. We reviewed EMR typhoid surveillance and vaccination recommendations to inform regional prevention and control efforts. Separate surveys were sent to surveillance and immunization officers in all 21 EMR countries and the occupied Palestinian territory, with 17 surveys (77%) and 13 surveys (59%) completed, respectively. Fifteen countries ro

Licensed typhoid vaccines contain Vi polysaccharide (ViPS), a capsular antigen of Salmonella. Typhi (S.typhi), either unconjugated or conjugated to carrier proteins like tetanus toxoid (TT), diphtheria toxoid (DT), and Cross-reactive material 197 (CRM-197). Quantitation of ViPS during vaccine development, is challenging, as ViPS demonstrates limited adsorption to solid phases, resulting in poor binding and reduced sensitivity in conventional ELISA formats. We developed a novel competitive enzyme-linked immunosorbent assay (c-ELISA) using biotinylated ViPS as a detector probe for ViPS quantitat
