Gene therapy in children with AIPL1-associated severe retinal dystrophy: an open-label, first-in-human interventional study.

BACKGROUND: Retinal dystrophy caused by genetic deficiency of AIPL1 causes severe and rapidly progressive impairment of sight from birth. We sought to evaluate whether early intervention by gene supplementation therapy was safe and could improve outcomes in children with this condition. METHODS: This non-randomised, single-arm, clinical study conducted in the UK involved four children aged 1·0-2·8 years with severe retinal dystrophy associated with biallelic disease-causing sequence variants in AIPL1. We designed a recombinant adeno-associated viral vector comprising the human AIPL1 coding se

Effect of binasal occlusion in children with esotropia.

BACKGROUND: Esotropia is a common pediatric ophthalmologic disorder that, if left untreated, can have a significant impact on the binocular visual function and appearance of the child. Binasal occlusion (BNO) is a non-surgical measure to alleviate the impact of esotropia. But there is no consistent theoretical basis for BNO and its therapeutic efficacy has been controversial. This study aimed to investigate the effect of BNO in the treatment of children with esotropia. METHODS: We performed a retrospective review of children with multiple types of esotropia who still had symptoms of obvious e

Profile of Amblyopia in School going (5-15 years) Children at State Level Referral Hospital in Uttarakhand.

INTRODUCTION: Amblyopia is one of the common causes of childhood visual impairment. The prevalence of amblyopia usually varies according to the age group of the studied population and the factors prevailing in that region. The upper limit of the critical time when amblyopia can develop is around eight years of age and it can be permanent if corrective measures are not taken in time. AIM: The purpose of study was to know the profile and pattern of amblyopia in children aged 5-15 years with refractive error in Uttarakhand and to compare it with national and regional (South Asian) studies. Paradoxical improvement of visual acuity in macular disease.

PURPOSE: Improvement in visual acuity is often considered the best indicator of the effectiveness of a treatment in age-related maculopathy. However, during the course of the disease, the location of the patients' preferred retinal locus of fixation may change. This can lead to an unexpected functional improvement, unrelated to treatment. METHODS: From a running database of 1,369 retina patients, we identified 116 patients over 60 years of age when age-related maculopathy was diagnosed based on the following inclusion criteria: one study eye with an initial acuity of the logarithm of the mini

Association between progressive retinal nerve fiber layer loss and longitudinal change in quality of life in glaucoma.

IMPORTANCE: Evaluation of structural optic nerve damage is a fundamental part of diagnosis and management of glaucoma. However, the relationship between structural measurements and disability associated with the disease is not well characterized. Quantification of this relationship may help validate structural measurements as markers directly relevant to quality of life. OBJECTIVE: To evaluate the relationship between rates of retinal nerve fiber layer (RNFL) loss and longitudinal changes in quality of life in glaucoma. DESIGN, SETTING, AND PARTICIPANTS: Observational cohort study including
