Antineutrophil Cytoplasmic Antibodies: Integrating History, Pathogenesis, and Modern Diagnostics.

Antineutrophil cytoplasmic antibodies (ANCA) define a distinct group of small-vessel vasculitides collectively termed ANCA-associated vasculitis (AAV). Since their initial recognition as immunofluorescence patterns, ANCA have evolved into well-characterized pathogenic autoantibodies directed primarily against proteinase 3 (PR3) and myeloperoxidase (MPO). This review integrates the historical evolution of ANCA discovery with current understanding of disease pathogenesis and contemporary diagnostic practice. We summarize key biological mechanisms underlying AAV, including genetic susceptibility,

Endoplasmic Reticulum Stress, Redox Signaling, and Neutrophil Effector Functions in Chronic Lung Diseases: Evidence and Research Priorities.

Endoplasmic reticulum (ER) stress and unfolded protein response (UPR) signaling intersect with inflammatory and redox pathways. Experiments in primary neutrophils link selected UPR pathways to mitochondrial reactive oxygen species, neutrophil extracellular trap (NET) formation, antimicrobial activity, and immune regulation under defined conditions. This review organizes these findings by cell type, experimental model, and the relationship tested, distinguishing them from cell-line experiments, other-cell mechanisms, clinical associations, and database connections. We examine calcium signaling,

Fecal microbiota transplantation in ulcerative colitis: mucosal immune mechanisms and precision microbiota therapy.

Fecal microbiota transplantation (FMT) has emerged as an investigational microbiota-targeted strategy for ulcerative colitis (UC), aiming to restore dysbiotic gut ecosystems and microbiota-host homeostasis. Mechanistic studies suggest that FMT may reshape microbial community structure, enrich short-chain fatty acid-producing bacteria, remodel bile acid and tryptophan-aryl hydrocarbon signaling, enhance epithelial barrier integrity, and regulate mucosal immunity, including Th17/Treg balance, IgA-associated responses, macrophage reprogramming, IL-10/IL-22 signaling, and neutrophil extracellular

From Physiology to Clinical Practice in Pancreatic Cancer-Related Thromboembolism-A Review.

Pancreatic cancer (PC) is a highly lethal malignancy linked to the highest rate of thromboembolic complications (TEC) among all solid tumors. TECs occur in approximately 5-40% of PC patients. The most common type of TEC in PC is venous thromboembolism (VTE). The mechanisms leading to frequent TEC in PC are complex and involve interactions between tumor-derived procoagulant factors and the prothrombotic tumor microenvironment (TME). Secretion of tissue factor and proinflammatory cytokines by tumor cells and the TME, overexpression of heparanase and podoplanin, impaired fibrynolysis and increase

RGS16 Aggravates Hepatic Ischemia-Reperfusion Injury via Hepatocyte-Intrinsic Apoptosis/Inflammation & Neutrophil Recruitment/NETosis.

Hepatic ischemia-reperfusion injury (HIRI) is a frequent and severe complication after liver transplantation and hepatectomy, driven by sterile inflammation and lacking effective therapies. Regulator of G-protein signaling 16 (RGS16) is an inflammatory modulator, but its role in HIRI remains unclear. We found that RGS16 expression was markedly increased in the livers of post-hepatectomy patients and positively correlated with neutrophil extracellular trap formation (NETosis). Using hepatocyte-specific Rgs16 knockout and transgenic mouse models, we demonstrated that Rgs16 deficiency attenuated

Multi-Omics Reveals Gut Microbiota Shifts and Hepatic Metabolic-Immune Alterations in "Short-Leg" Malformed Frog (Pelophylax nigromaculatus).

Amphibian malformation syndromes significantly impact both conservation efforts and aquaculture, yet their underlying systemic pathophysiological mechanisms remain poorly characterized. This study comprehensively examines the multi-level pathological processes associated with the "short-leg" malformation syndrome in the black-spotted frog (Pelophylax nigromaculatus) using an integrated methodology, encompassing morphological, histopathological, gut microbiome, and hepatic transcriptomic analyses. Affected frogs demonstrated shortened limbs, impaired motor function, and a distinctive metabolic

Antiphospholipid Syndrome and Cardiovascular Disease.

Antiphospholipid syndrome (APS) is an acquired systemic autoimmune thrombo-inflammatory disorder characterized by venous, arterial, and microvascular thrombosis and/or pregnancy morbidity in the setting of persistent antiphospholipid antibodies. Cardiovascular disease in APS extends well beyond classic thrombosis and includes ischemic stroke, myocardial infarction, venous thromboembolism, valvular heart disease, pulmonary embolism, accelerated atherosclerosis, and microvascular ischemic injury. Mechanistically, APS is increasingly understood as a disorder of immunothrombosis, in which antiphos

ANCA vasculitis: what is in the pipeline.

PURPOSE OF REVIEW: Anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV) is now recognized as a complex immune-mediated inflammatory disorder rather than a purely pauci-immune small-vessel vasculitis. Advances in understanding ANCA specificity, complement activation, neutrophil extracellular trap formation, genetic susceptibility, and environmental triggers have reshaped concepts of disease classification, prognosis, and therapy. A focused review of these developments is timely because they are directly informing current treatment strategies, disease stratification, and futur

3D nanoscale imaging of amyloid-β oligomer interactions with extracellular vesicles by cryo-ET.

Central to Alzheimer's disease pathology are prefibrillar oligomer assemblies of amyloid-β (Aβ) peptide. A widely discussed hypothesis proposes that amyloid-β oligomers insert into neuronal lipid membranes, disrupting their integrity and causing a loss of cellular homeostasis in Alzheimer's disease. This membrane disruption is believed to be a major source of Aβ-induced neurotoxicity. Cryo electron tomography (cryo-ET) has facilitated 3D nanoscale imaging of Aβ-membrane interactions under near-native conditions. Analyses of small extracellular vesicles (sEVs) reveals that Aβ oligomers includin

Mitochondrial transfer via ADSC-EVs reprograms glucose/glutamine metabolism to restore TCA cycle-driven M2 polarization in diabetic wounds.

Chronic diabetic wounds are trapped in a persistent inflammatory state, largely due to macrophage failure to transition from pro-inflammatory (M1) to pro-reparative (M2) phenotypes. Here, we show that adipose-derived stem cell extracellular vesicles (ADSC-EVs) deliver functional mitochondria into diabetic wound macrophages, thereby restoring tricarboxylic acid (TCA) cycle-driven M2 polarization. Mechanistically, ADSC-EV-mediated mitochondrial transfer reactivates pyruvate dehydrogenase (PDH) and pyruvate carboxylase (PC), increases TCA cycle flux, and suggests enhanced glutamine anaplerosis, a

Psoralen ameliorates sepsis-induced acute lung injury by regulating microbiota-dependent tryptophan metabolism.

BACKGROUND: Sepsis frequently leads to acute lung injury (ALI). Despite the documented anti-inflammatory effects of Psoralen (PSO), the exact mechanism by which it confers protection against septic ALI remains poorly understood. PURPOSE: This research explored the protective efficacy and mechanistic aspects of PSO in sepsis-induced ALI. METHODS: Lipopolysaccharide (LPS) was employed to create a murine ALI model. Pathological and inflammatory responses were evaluated. The dependency on gut microbiota was assessed through intestinal flora ablation and fecal microbiota transplantation. The aryl

Source-renewing ectopic ApoB-TRL lipolysis in non-resolving ARDS: a testable spatial hypothesis.

Acute respiratory distress syndrome (ARDS) may remain regionally permeable and poorly reparative after the initiating insult has weakened. We propose that, in a biologically enriched subset of non-resolving ARDS, repeated entry of circulating apolipoprotein B-containing triglyceride-rich lipoproteins (ApoB-TRLs) into an injured perivascular or interstitial compartment provides a renewable substrate for local injury. Catalytically active lipoprotein lipase (LPL) converts particle triglyceride (TG) to free fatty acids (FFAs). When FFA generation exceeds albumin buffering, cellular disposal, and

Galectin-10 and Charcot-Leyden Crystals in Chronic Rhinosinusitis with Nasal Polyps: Pathogenetic Insights and Clinical Implications.

Chronic rhinosinusitis with nasal polyps (CRSwNP) is a heterogeneous inflammatory disease in which type 2 inflammation predominates in most patients in Western populations. Galectin-10 (Gal-10) and Charcot-Leyden crystals (CLCs), linked to eosinophil activation, cytolysis and eosinophil extracellular trap cell death (EETosis), have emerged as potential biomarkers and mediators of eosinophilic inflammation. This narrative review summarises current evidence on their biological role and clinical relevance in CRSwNP. The literature was identified through searches of PubMed/MEDLINE, Web of Science

ROS-responsive nanogels enable inhaled CD24 immunotherapy for selective hyperinflammation suppression in severe pneumonia.

Acute pneumonia triggered by pathogen infection and lung injury persists as a critical global life-threatening issue. Cluster of differentiation 24 (CD24), by selectively inhibiting the inflammatory response associated with damage-associated molecular patterns (DAMPs) through the interaction with Siglec, has appeared as a unique paradigm to alleviate acute pneumonia, yet its effective pulmonary delivery remains challenging. Here, we report on reactive oxygen species (ROS)-responsive and mucus-penetrable nanogels (ROSμNG) for pulmonary CD24 delivery to rescue acute pneumonia by selective inhibi

Temporal proteomic characterization of SARS-CoV-2 infected mouse lungs.

Understanding of dynamic activations of pathophysiological processes in the infected lungs is important for effective treatment of SARS-CoV-2 infection. Time-course transcriptome analyses of infected lungs have been performed to address this issue. Since proteins actually execute the pathophysiological processes, however, the time-course transcriptome data provide limited information regarding their temporal transitions. Here, we present time-course proteomic profiling of lung tissues from K18-hACE2 transgenic mice using liquid chromatography-tandem mass spectrometry analysis at day 0, 1, 2, 5

Deguelin suppresses Nalm6 leukemia cell proliferation through lactate accumulation and ATP depletion.

BACKGROUND: Acute lymphoblastic leukemia (ALL) is characterized by metabolic reprogramming, including elevated glycolysis and lactate production. Monocarboxylate transporter 1 (MCT1/SLC16A1) is frequently overexpressed in ALL, sustaining glycolytic flux to promote cell proliferation. While the natural flavonoid deguelin is known to induce necroptosis at high concentrations, the mechanisms underlying its anti-leukemic effects at lower, potentially more therapeutically relevant doses remain unclear. METHODS AND RESULTS: Nalm6 cells were treated with deguelin (0.08-10 µM) for up to 72 h. Cell vi

Aortic aneurysm and dissection: complement and precision medicine in aortic disease.

Aortic disease encompasses life-threatening conditions such as aortic aneurysm and dissection, which are associated with high prevalence, morbidity, and mortality. The complement system, a key component of innate immunity, not only defends against pathogens but also maintains tissue homeostasis. Recent discoveries have expanded its role beyond immunity, linking complement dysregulation to numerous diseases and positioning it as a target for pharmacotherapy. Complement-based treatments for precision medicine are emerging, with several pharmaceuticals either already approved or under investigati

The Roles and Molecular Mechanisms of HIF-1α in Pulpitis.

Pulpitis is characterized by inflammation within dental pulp tissue, primarily triggered by bacterial infection. Hypoxia-inducible factor-1α (HIF-1α), a key transcriptional regulator, is stabilized under the hypoxic conditions associated with pulpitis. This review examines the roles and molecular mechanisms of HIF-1α in the pathogenesis and progression of pulpitis. Hypoxia in pulpitis prevents the degradation of HIF-1α, leading to its elevated expression. Furthermore, lipopolysaccharide from invading bacteria upregulates HIF-1α transcription through nuclear factor kappa B and mitogen-activated

Concomitant use of sivelestat sodium hydrate and antithrombotic drugs worsens the treatment outcome of patients with acute respiratory distress syndrome and suppression of fibrinolysis: a single-center, retrospective study.

OBJECTIVES: Sivelestat sodium hydrate (SSH) may be effective in the early stage of acute respiratory distress syndrome (ARDS) before the neutrophil extracellular trap scaffold structure is complete. Therefore, patients with suppression of fibrinolysis (SF) before the secondary fibrinolytic process might benefit from SSH administration. The primary aim of this study was to determine the effect of the SF state and combination therapy on the effect of SSH administration. METHODS: We retrospectively reviewed the data of patients diagnosed with ARDS at Fujita Health University Hospital between Jul

Prolonged exposure to low-dose traffic-derived PM2.5 causes fatty liver disorder in mice.

There is no safe level of exposure to air pollution, including particulate matter smaller than 2.5 µm (PM2.5), to human health. Whilst it is well known that exposure to heavily polluted air is associated with several liver disorders, it is unclear how long-term exposure to low-level traffic-derived PM2.5 affects liver health. BALB/c mice (5 weeks, male) were exposed to traffic-derived PM2.5 (10 µg/mouse/day, intranasally) daily for 4, 8 and 12 weeks. Markers of inflammation and fibrosis were measured at each time point. Changes in liver proteome and lipid profiles were measured using proteomic
