Effects of prenatal ethanol and long-chain n-3 fatty acid supplementation on development in mice. 1. Body and brain growth, sensorimotor development, and water T-maze reversal learning.

Pregnant mice were fed equivalent daily amounts of a liquid diet containing 25% (kcal) ethanol, or with maltose dextrin substituted isocalorically for ethanol. In addition, the diet contained 20% oil; this was either of two mixtures, one comprised of predominantly n-6 (18:2n-6) fatty acids, and the other containing an equivalent amount of n-6, but supplemented with a source of long chain n-3 (20:5n-3, 22:6n-3) fatty acids. An additional control group was fed lab chow ad libitum. The treatment was implemented from day 7 to 17 of gestation, whereafter all groups were fed lab chow. Ethanol decrea

[Effects of prenatal exposure to alcohol on the binding of H3-diazepam to the synaptic membranes of the cerebral hemisphere cortex at various stages of ontogenesis in the rat offspring].

Exposure to ethanol during pregnancy results in the alteration of (3H)-diazepam binding to synaptosomal neocortical membranes from the rat offspring. In male rats, 14 days of age, binding level diminished to 11%. In two-month-old control rats Scatchard plot was biphasic. It has been shown that prenatal exposure to ethanol leads to changes in the nature of binding in two-month-old experimental animals, as compared with the control ones. Possible relationship is discussed between the brain benzodiazepine system disorders and behaviour.

Ethanol-induced cytoskeletal dysgenesis with dietary protein manipulations.

Ethanol-induced cytoskeletal abnormalities occur in many cell types, affecting structure and function of the respective organs. When affecting the developing myocyte, damage is responsible for the hypotonicity and congestive heart failure of Fetal Alcohol Syndrome (FAS) infants. To test the hypothesis that ethanol damages myocytes in utero, and determine that damage is not affected by dietary protein manipulations, female Sprague-Dawley rats were fed Leiber-DeCarli diets containing either 10% or 25% protein-derived calories, isocalorically balanced with or without 36% of the total calories con

A model for later-life effects of perinatal drug exposure: maternal hormone mediation.

In the female rat chronic ethanol intake increased plasma levels of corticosterone; acute stress in the pregnant or lactating rat increased plasma levels of corticosterone in the fetuses or sucklings. In the lactating rat, corticosterone rapidly equilibrated between plasma and milk, so that variations in the mother's adrenocortical secretion could be promptly reflected in the suckling's body. The role of maternal adrenocortical hormone with regard to the development of pituitary-adrenocortical and behavioral activities was indicated by endocrine (plasma corticosterone), neurochemical (hippocam

Glycoproteins and proteins in an axolemma-enriched fraction and myelin from developing rats: effect of maternal ethanol consumption.

The present study examined proteins and glycoproteins from an axolemma-enriched fraction from the developing offspring of female rats that were pair-fed control or 6.6% (50 g/liter) ethanol liquid diets on a chronic basis prior to parturition. In addition, this study examined the synthesis of the major CNS myelin-associated glycoprotein (MAG) as an index of myelin maturation. The results of the latter study demonstrated normal MAG maturation in ethanol-treated rats. However, a significant decrease in the proportion of radioactivity associated with MAG was found in the developing offspring of e

Acute alcohol exposure during gestational day 8 in the rat: effects upon physical and behavioral parameters.

Pregnant rats on the eighth day of gestation (GD 8) received 2 intraperitoneal injections (0.015 ml/g body weight) spaced by an interval of 4 hours, of either 0, 6, 12 18, or 24% (v/v) alcohol solution. Brains of pups sacrificed 24 hours after delivery revealed a dose-related impairment in hemispheric and cerebellar length, width and weight. Statistically significant differences were also observed in body weights of culled pups during Post-partum Day 1. An open field test performed when offspring were 40-45 days old demonstrated a dose-dependent increase in ambulation and rearing scores for bo

Functional connectivity of cognition-related brain networks in adults with fetal alcohol syndrome.

BACKGROUND: Fetal alcohol syndrome (FAS) can result in cognitive dysfunction. Cognitive functions affected are subserved by few functional brain networks. Functional connectivity (FC) in these networks can be assessed with resting-state functional MRI (rs-fMRI). Alterations of FC have been reported in children and adolescents prenatally exposed to alcohol. Previous reports varied substantially regarding the exact nature of findings. The purpose of this study was to assess FC of cognition-related networks in young adults with FAS. METHODS: Cross-sectional rs-fMRI study in participants with FAS

Differential effects of maternal diets on birth outcomes and metabolic parameters in rats after ethanol consumption during pregnancy.

Maternal nutrition status plays an important role in the development of fetal alcohol spectrum disorders (FASD), but its direct evidence is lacking. This study compared a standard chow with a semi-purified energy-dense (E-dense) diet on birth and metabolic outcomes in rats after ethanol (EtOH) consumption during pregnancy. Pregnant Sprague-Dawley rats were randomised into four groups: chow (n 6), chow + EtOH (20 %, v/v) (n 7), E-dense (n 6) and E-dense + EtOH (n 8). Birth outcomes including litter size, body and organ weights were collected. Metabolic parameters were measured in dams and pups …

Role of zinc insufficiency in fetal alveolar macrophage dysfunction and RSV exacerbation associated with fetal ethanol exposure.

BACKGROUND: We previously reported that maternal alcohol use significantly increases the risk of sepsis in premature and term newborns. In the mouse, fetal ethanol exposure results in an immunosuppressed phenotype for the alveolar macrophage (AM) and decreases bacterial phagocytosis. In pregnant mice, ethanol decreased AM zinc homeostasis, which contributed to immunosuppression and impaired AM phagocytosis. In this study, we explored whether ethanol-induced zinc insufficiency extended to the pup AMs and contributed to immunosuppression and exacerbated viral lung infections. METHODS: C57BL/6 f

Regional dysregulation of taurine and related amino acids in the fetal rat brain following gestational alcohol exposure.

The fetal brain exhibits exquisite alcohol-induced regional neuronal vulnerability. A candidate mechanism for alcohol-mediated brain deficits is disruption of amino acid (AA) bioavailability. AAs are vitally important for proper neurodevelopment, as they comprise the most abundant neurotransmitters in the brain and act as neurotransmitter precursors, nitric oxide donors, antioxidants, and neurotrophic factors, which induce synaptogenesis, neuronal proliferation, and migration. We hypothesized that gestational alcohol alters brain AA concentrations, disrupts AAs associated with neuropathogenesi

Exploring the feasibility of using electronic health records in the surveillance of fetal alcohol syndrome.

BACKGROUND: Explore the use of electronic health records (EHRs) in fetal alcohol syndrome (FAS) surveillance systems. METHODS: Using EHRs we identified diagnoses and anthropometric measurements related to the FAS criteria developed by the Fetal Alcohol Syndrome Surveillance Network (FASSNet) among children aged 0 to 12 years. RESULTS: There were 143,393 distinct children aged between 0 and 12 years enrolled in Kaiser Permanente, Georgia, during the study period. Based on diagnoses and anthropometric measurements, 20,101 children met at least one criterion of interest, and when grouped into c

Transient exposure to ethanol during zebrafish embryogenesis results in defects in neuronal differentiation: an alternative model system to study FASD.

BACKGROUND: The exposure of the human embryo to ethanol results in a spectrum of disorders involving multiple organ systems, including the impairment of the development of the central nervous system (CNS). In spite of the importance for human health, the molecular basis of prenatal ethanol exposure remains poorly understood, mainly to the difficulty of sample collection. Zebrafish is now emerging as a powerful organism for the modeling and the study of human diseases. In this work, we have assessed the sensitivity of specific subsets of neurons to ethanol exposure during embryogenesis and we h

Feasibility and promise of a remote-delivered preconception motivational interviewing intervention to reduce risk for alcohol-exposed pregnancy.

BACKGROUND: Alcohol-exposed pregnancy (AEP) is a leading cause of birth defects. Effective face-to-face preconception interventions based on motivational interviewing (MI) exist and should be translated into remote formats for maximum public health impact. This study investigated the feasibility and promise of a one-session, remote-delivered, preconception, MI-based AEP intervention (EARLY Remote) for non-treatment-seeking community women. SUBJECTS AND METHODS: This was a single-arm, prospective pilot intervention study. All participants received the intervention via telephone and mail. Feasi

Neuroprotective peptide ADNF-9 in fetal brain of C57BL/6 mice exposed prenatally to alcohol.

BACKGROUND: A derived peptide from activity-dependent neurotrophic factor (ADNF-9) has been shown to be neuroprotective in the fetal alcohol exposure model. We investigated the neuroprotective effects of ADNF-9 against alcohol-induced apoptosis using TUNEL staining. We further characterize in this study the proteomic architecture underlying the role of ADNF-9 against ethanol teratogenesis during early fetal brain development using liquid chromatography in conjunction with tandem mass spectrometry (LC-MS/MS). METHODS: Pregnant C57BL/6 mice were exposed from embryonic days 7-13 (E7-E13) to a 25

Normal distribution of palpebral fissure lengths in Canadian school age children.

BACKGROUND: Fetal alcohol syndrome (FAS) includes the facial dysmorphic feature of short palpebral fissures (PFs) and short PFs are a key physical marker for identifying children with FAS and some other rarer conditions. There is concern that normative data on PFs now available may not reflect all racial/ethnic groups and might be inaccurate in general. OBJECTIVES: To accomplish a large population based study that would accurately determine normative PF values across the full diversity of the Canadian school age population. METHODS: A normative sample of school age children was identified in

Persistent impairment of hippocampal neurogenesis in young adult rats following early postnatal alcohol exposure.

BACKGROUND: Prenatal alcohol exposure can cause damage to the developing fetus with outcomes including growth deficiency, facial dysmorphology, brain damage, and cognitive and behavioral deficits. Smaller brains in children with FASD have been linked both with reduced cell proliferation in the developing CNS and with apoptotic cell loss of postmitotic neurons. Prenatal alcohol exposure in rodents during the period of brain development comparable to that of the first and second trimesters of human pregnancy persistently alters adult neurogenesis. Long-term effects of alcohol exposure during the

Fetoprotectivity of the flavanolignan compound siliphos against ethanol-induced toxicity.

Of the three flavanolignans that are found in silymarin (Silybum marianum [L.] Gaertn.), silybin is thought to be the primary therapeutic constituent. To test the capacity of silybin to protect the rat fetus from toxic effects of maternally ingested EtOH we did the following: Adult female rats were assigned to one of four groups; EtOH, EtOH/silybin, pair-fed control, and chow fed control. Silybin was orally administered as Siliphos(R), which is one part silybin to two parts phosphatidylcholine. All groups except the chow-fed control were maintained on a liquid diet throughout pregnancy. On day
