A Cross-Sectional Study on Intestinal Parasitic Infections in Children in Suburban Public Primary Schools, Saraburi, the Central Region of Thailand.

Intestinal parasitic infection rate among school-aged children in Thailand has been decreasing. However, certain intestinal parasites remain problematic in some regions. This cross-sectional study was conducted between February and September 2016 in three suburban government primary schools (KK, BR, and HK), Saraburi, Thailand. Stool was collected from 263 asymptomatic subjects (4-15 years old), using simple direct smear, formalin-ether concentration, Boeck and Drbohlav's Locke-Egg-Serum (LES) medium culture, and agar plate culture. A self-administered questionnaire was used to collect data ab

A retrospective study comparing agar plate culture, indirect immunofluorescence and real-time PCR for the diagnosis of Strongyloides stercoralis infection.

Strongyloides stercoralis is a parasite that can cause death in immunocompromised people. A proper diagnosis is hence essential. The real-time polymerase-chain reaction (RT-PCR) is a novel, promising diagnostic method, that detects the DNA of the parasite in stool samples. In this retrospective study, we compared the sensitivity of agar plate coproculture (APC), an in-house immunofluorescence test (IFAT) and an in-house RT-PCR for the diagnosis of S. stercoralis infection. The study sample was composed by 223 samples. Samples resulting positive to APC, IFAT and RT-PCR were 20, 140 and 25, resp

Albendazole and ivermectin for the control of soil-transmitted helminths in an area with high prevalence of Strongyloides stercoralis and hookworm in northwestern Argentina: A community-based pragmatic study.

BACKGROUND: Recommendations for soil-transmitted helminth (STH) control give a key role to deworming of school and pre-school age children with albendazole or mebendazole; which might be insufficient to achieve adequate control, particularly against Strongyloides stercoralis. The impact of preventive chemotherapy (PC) against STH morbidity is still incompletely understood. The aim of this study was to assess the effectiveness of a community-based program with albendazole and ivermectin in a high transmission setting for S. stercoralis and hookworm. METHODOLOGY: Community-based pragmatic trial

Enteric pathogens of food sellers in rural Gambia with incidental finding of Myxobolus species (Protozoa: Myxozoa).

BACKGROUND: Ongoing surveillance of enteric pathogens of public health significance among casual food sellers is undertaken in many resource-limited countries. We report the results of a survey in Kiang West province, The Gambia, and provide an exemplar methodology for such surveys in resource-limited laboratories. METHODS: Unpreserved, unrefrigerated stool samples were subjected to Salmonella, Shigella and agar plate culture for rhabditoid nematodes. Direct microscopy, formalin-ethyl acetate concentration and iron-hematoxylin staining was performed later, following preservation. RESULTS: Of

CD4+ T cell subsets and Tax expression in HTLV-1 associated diseases.

Human T lymphotropic virus type 1 (HTLV-1) infection displays variable clinical manifestations. These include inflammatory diseases such as HTLV-1 associated myelopathy (HAM) or immunosuppressive conditions such as Strongyloides stercoralis hyperinfection. The viral protein, Tax causes activation and proliferation of T cells. We hypothesize that the expression of Tax in T cell subsets characterizes the clinical manifestations of HTLV-1. To test this hypothesis, we measured T helper 1 effector cells and regulatory T cells (Tregs) among Tax expressing lymphocytes from peripheral blood mononuclea

Changing patterns of gastrointestinal parasite infections in Cambodian children: 2006-2011.

We studied gastrointestinal parasites in symptomatic Cambodian children attending a provincial hospital in Siem Reap, Cambodia between 2006 and 2011. A total of 16 372 faecal samples were examined by direct microscopy. Parasites were detected in 3121 (19.1%) samples and most common were Giardia lamblia (8.0% of samples; 47.6% disease episodes), hookworm (5.1%; 30.3%) and Strongyloides stercoralis (2.6%; 15.6%). The proportion of infected children increased, and the number of disease episodes effectively treated with a single dose of mebendazole decreased, over the 5-year period.

[Selective vs. mass treatment with antihelminthic drugs: experience in two hyperendemic communities].

OBJECTIVE: To compare the prevalence and intensity of soil-transmitted nematode infection among children from two hyperendemic communities who were treated with antihelminthic drugs under two different regimens: selective or individual treatment was administered to members of one of the communities, and repeated mass treatment was given to the control community. METHODS: The study population was comprised of 909 children of both sexes between the ages of 2 and 13 from two peri-urban communities: Las Lomas and El Abastos, both in Santa Fe, Argentina. A prospective, longitudinal, quasi-experime

Selective loss of gastrointestinal mast cells and impaired immunity in PI3K-deficient mice.

Mice that lack the p85alpha regulatory subunit of phosphatidylinositol-3 kinase (PI3K) are deficient in gastrointestinal and peritoneal mast cells but have dermal mast cells. Accordingly, these mice show impaired bacterial clearance in response to acute septic peritonitis and are highly susceptible to infection by the intestinal nematode Strongyloides venezuelensis. Systemic anaphylactic shock responses, however, are intact. We found that although reconstitution of PI3Kminus sign/minus sign mice with bone marrow--derived mast cells (BMMCs) restored anti-bacterial immunity, only T helper type 2

Role of IL-5 in innate and adaptive immunity to larval Strongyloides stercoralis in mice.

Protective immunity to Strongyloides stercoralis infective larvae in mice has been shown to be dependent on IL-5 based on mAb depletion studies. The goal of this study was to determine the functional role of IL-5 during the innate and adaptive immune response to larval S. stercoralis in mice. In these studies, three strains of mice were used: wild-type C57BL/6J (WT), IL-5 knockout (KO), and IL-5 transgenic (TG). Innate responses to the larvae indicated that there was enhanced survival in the KO animals and decreased survival in the TG animals compared with WT. Furthermore, killing of larvae in

Analysis of T cell populations and IL-3 mRNA expression in mesenteric lymph node cells and intestinal intraepithelial lymphocytes in Strongyloides ratti-infected mice.

T cell populations and IL-3 mRNA expression were analysed in mesenteric lymph node cells and intestinal intraepithelial lymphocytes (IEL) in Strongyloides ratti-infected mice. On days 7 and 12 post-infection, 2.6 times as many mesenteric lymph node cells were present in S. ratti-infected mice compared with uninfected mice. Although the percentages of CD3+, CD4+ and CD8+ cells decreased during infection, the absolute numbers of these cell types increased on day 7 due to an overall increase in the mesenteric lymph node cell number. The CD4/CD8 ratio in IEL was increased on day 5, whereas no sign

Intestinal immune cells in Strongyloides stercoralis infection.

BACKGROUND: Strongyloides stercoralis can cause a wide spectrum of disease in man, ranging from a chronic asymptomatic infection to a hyperinfective, often fatal syndrome. In rodents, spontaneous expulsion of Strongyloides spp occurs after experimental infection. Mast cells, goblet cells, and eosinophils have been identified as possible effectors of this expulsion. AIMS: To investigate intestinal histopathology and mucosal immunity in immunocompetent patients with chronic S stercoralis infection. METHODS: Jejunal biopsies were performed in 19 immunocompetent patients with a positive stool ex

Massive experimental infection with Strongyloides venezuelensis in rats and absence of sudden death.

Ten rats were divided into five groups, A - E, to determine the larval dose-effect of infection with Strongyloides venezuelensis (SVZ). The rat groups were exposed to SVZ infective larvae as follow: (A) 100, (B) 1,000, (C) 10,000, (D) 100,000 and (E) 1,000,000. The eight rats in Groups A - D survived the infection. Rats exposed to the higher doses of larvae had the higher egg per gram counts (EPG), but more rapid reduction in EPG counts. The four rats in Groups C and D had EPG counts greater than 10,000 EPG during days 7 - 21 after infection. Maximum EPG values in Group C were 85,400 and 106,6

Intestinal opportunistic parasites among different groups of immunocompromised hosts.

The present study was designed to find out the prevalence of different intestinal opportunistic parasites in different groups of immunocompromised hosts; patients suffering from malignancy with or without immuno-suppressive drugs, or with chronic renal failure, or diabetes mellitus, or subjects under cortisone therapy. Examination of stool samples collected from 427 immunocompromised hosts revealed the detection of intestinal opportunistic parasites in 98 samples, with a prevalence of 23%. Infection with opportunistic parasites was higher in males than in females (32.6% versus 12.9%). There wa

A comparative study of gastrointestinal nematode egg output in N'Dama, Zebu and N'Dama x Zebu crossbred cattle.

Strongyle faecal egg output was estimated in N'Dama, Zebu and N'Dama x Zebu crossbred (F1) cattle. N'Dama cattle showed a significantly lower prevalence of strongyle infection, as measured by faecal egg output, than F1 (P < 0.01) and Zebu (P < 0.001) cattle. In strongyle-infected animals, mean egg output was also significantly lower in N'Damas (P < 0.03) than in Zebus. A previous trypanosomiasis infection did not affect the results. The presence of a natural resistance trait to strongyle infection in N'Dama cattle is postulated.

Transmission of intestinal helminths in Danish sow herds.

Faecal samples from sows and their litters, all untreated with anthelmintics, were examined in eight sow herds, ranging from very intensive to very traditional management. Four helminth genera were recorded, namely Oesophagostomum sp., Ascaris suum, Trichuris suis and Strongyloides ransomi. The herds with the most intensive management were only infected with A. suum, while the more traditionally managed herds were infected with three or four species. In one herd, the sows showed a small relative increase in the excretion of Oesophagostomum eggs during lactation, but in general there was no con
