The antioxidants vitamin E and beta-carotene protect against ethanol-induced neurotoxicity in embryonic rat hippocampal cultures.

Fetal alcohol syndrome is characterized by numerous nervous system anomalies with the developing hippocampus being highly vulnerable. Other conditions can result from maternal ethanol consumption including oxidative stress. Critical antioxidants, such as vitamin E, can be decreased and antioxidative defenses altered. Gestational day 18 rat hippocampal cultures were exposed to ethanol ranging from 400 to 2400 mg/dl (16 h). MTT assays assessed neurotoxicity. Viability was decreased dose dependently. Supplementation with vitamin E or beta-carotene afforded neuroprotection against all ethanol conc

Effect of exogenous thyroxine on the development of the Purkinje cell in fetal alcohol effects in the rat.

The amelioration of fetal alcohol effects on the postnatal development of the Purkinje cell by exogenous L-thyroxine was investigated in the neonatal rat. Time-pregnant rats were divided into three groups. Group A (n = 6) received 35% liquid ethanol diet; Group B (n = 6) was fed a liquid diet in which maltose dextrins replaced alcohol isocalorically, constituting the pair-fed group; Group C (n = 6) received the 35% liquid ethanol diet and, in addition, received exogenous thyroxine (5 microg/kg/day) subcutaneously. After the pups were born, the mothers were removed and the pups of each were sur

Commentary: mal-intentioned illiteracy, willful ignorance, and fetal protection laws: is there a lexicologist in the house?

Author argues that fetal protection laws result from the antiabortion agenda, not from concern for child health. Such laws derive from rhetorical dissembling, not careful crafting of public health policy.

Effect of fetal alcohol exposure on postnatal pituitary adenosine 3', 5'-cyclic phosphate content and growth hormone release.

This study examines the influence of fetal ethanol (ETOH) exposure and pair-feeding dams on postnatal, releasing factor-induced pituitary growth hormone (GH) release and adenosine 3',5'-cyclic phosphate (cAMP) accumulation. Fetuses were exposed to ETOH in utero by feeding dams a 36% (calories derived from ETOH: 6.6% v/v) ETOH liquid diet. Postnatal body weights were measured at sacrifice to evaluate the influence of ETOH on growth. Pituitary weight and protein content were measured to determine if changes in GH secretion or cAMP are proportional to the overall effect of ETOH on the pituitary.

Magnetic resonance imaging and spectroscopy in fetal ethanol exposed Macaca nemestrina.

Magnetic resonance imaging (MRI) and proton magnetic resonance spectroscopy (1H-MRS) offer noninvasive ways to observe structural and biochemical changes which might serve as valuable diagnostic markers for detecting brain damage from prenatal ethanol teratogenesis. Cranial MR imaging and spectroscopy were performed on 20 nonhuman primates (Macaca nemestrina) with known prenatal ethanol exposures and well-documented cognitive and behavioral levels of performance. The choline: creatine ratio detected by 1H-MRS in the brain increased significantly with increasing duration of in utero ethanol exp

Sex differences in vulnerability to developmental spatial learning deficits induced by limited binge alcohol exposure in neonatal rats.

The two main objectives of this study were (1) to replicate previous findings that 6 days of binge-like exposure to alcohol during the neonatal brain growth spurt induces significant place learning deficits in juvenile rats and (2) to determine whether more limited (3-day) binge-like exposure during the neonatal period induces place learning deficits and whether the effects depend on the developmental timing of the exposure. Using artificial rearing methods and a split-litter experimental design, groups of male and female neonatal rats were given binge-like exposure to 4.5 g/kg/day of ethanol

[A cytogenetic study of the functions of the variable regions in human C heterochromatin. III. The relationship between the amount of C heterochromatin and the occurrence of the fetal alcohol syndrome].

It has been shown elsewhere that in children with growth retardation and gene syndromes (Russell-Silver, Dubowitz etc.), and in healthy children with inherited short stature the total amount of C heterochromatin in chromosomes 1, 9, and 16 was reduced as compared to the control. A correlation between the C heterochromatin reduction and the intrauterine growth retardation was suggested. A study of C heterochromatin in patients with fetal alcohol syndrome has shown that the chromosome sets of all the 11 children examined are identical to those of the above-mentioned patients. This fact may indic

Postnatal high-peak blood ethanol concentration and external cue-based discrimination learning and reversal in the preweanling rat: comparison with memory-based discrimination learning.

Postnatal exposure to ethanol that produces high-peak blood ethanol concentrations (HP-BEC) in artificially reared infant rats affects hippocampal neuroanatomy and discrimination learning based on memorial cues from a patterned (single) alternation (PA) schedule in preweanling rats (P. L. Greene, J. L. Diaz-Granados, & A. Amsel, 1992). In the present experiments, discrimination by preweanling rats exposed to ethanol in the same way was tested with nonmemorial, external cues. In this external cue-based discrimination and in its reversal, ethanol-exposed rats were not different from normal or ar

Ethanol-induced disturbances of gliogenesis and neuronogenesis in the developing murine brain: an in vitro and in vivo immunohistochemical and ultrastructural study.

The effects of ethanol on murine brain development were studied in a model of whole embryo culture and in vivo. Ethanol locally enhances cell death in the primitive neuroepithelium. During neuronal migration, ethanol induces a premature transformation of the radial glial guides into astrocytes. Ethanol also inhibits the late gliogenesis. The resulting postmigratory neocortex displays an abnormal neuronal pattern almost completely deprived of vertical columnization. These glial-neuronal disturbances can explain neuropathological and clinical features of the fetal alcohol syndrome. Our study pro

Effects of prenatal stress and ethanol on cerebellar fiber tract maturation in B6D2F2 mice: an image analysis study.

Two studies were carried out to evaluate the role of prenatal stress in the outcome of prenatal ethanol exposure on central nervous system maturation in offspring. B6D2F1 mice were paid-fed liquid diets containing either 25% ethanol-derived calories or an isocaloric amount of maltose-dextrin on days 12 to 17 of gestation. During this period, half of the dams in each dietary condition also underwent two daily one-hour periods of restraint stress. A fifth group, provided with lab chow and water ad lib, was left undisturbed throughout pregnancy. At birth, all pups were fostered to untreated dams.

Ultrastructural changes in the mouse fetal neocortex following chronic maternal alcoholization.

Female mice (RAP strain) were alcoholized for 30-50 days before mating and during pregnancy until killing, with a 20% solution of ethanol administered instead of drinking water. From foetuses of 16, 18, 20 days and from newborn puppies on day 1 parietal neocortex fragments were excised and examined electronmicroscopically. Chronic maternal alcoholization induces in the neocortex of mouse foetuses and newborn puppies various ultrastructural changes: swelling of mitochondria with the disappearance of cristae and vacuolation, both in the capillary endothelium and in the cells of the neural tissue

Hyperresponsiveness to stress: differential effects of prenatal ethanol on males and females.

In this study we investigated the hypothesis that pituitary-adrenal response inhibition is compromised in animals prenatally exposed to ethanol. In the first experiment, we examined whether opportunity to perform a consummatory response reduces the adrenocortical response to a novel test cage. Animals were water deprived for 24 hr and tested in one of three conditions: (a) removed from home cage, blood sample obtained immediately; (b) placed into empty novel cage, blood sample obtained 30 min later; (c) placed into novel cage with water available, blood sample obtained 30 min later. All animal

CHOICES: an integrated behavioral intervention to prevent alcohol-exposed pregnancies among high-risk women in community settings.

CHOICES is an integrated behavioral intervention for prevention of prenatal alcohol exposure in women at high risk for alcohol-exposed pregnancies. The intervention uses motivational interviewing and cognitive-behavioral strategies, and targets adoption of effective contraception and reduction of alcohol use. The CHOICES intervention includes four manual-guided counseling sessions delivered by behavioral health counselors and one contraceptive session with a family planning clinician. CHOICES's efficacy has been established through a series of randomized controlled trials in settings including

History of physical abuse and periconceptional drinking in pregnant women.

This study examines the relationship between physical abuse and periconceptional drinking in women presenting to a mid-Atlantic, urban hospital-based OB/GYN clinic serving a largely indigent population between April 2003 and May 2004. During their first prenatal visit, 308 women completed a screening battery that included the Abuse Assessment Screen (AAS) and measures of alcohol use, including the CAGE, T-ACE, TWEAK, and the PRIME-MD Patient Health Questionnaire (PHQ). Bivariate analyses, including odds ratios (ORs) and 95% confidence intervals (CIs), revealed that women with a history of phys

Acute Ethanol Exposure during Synaptogenesis Rapidly Alters Medium Spiny Neuron Morphology and Synaptic Protein Expression in the Dorsal Striatum.

Fetal alcohol spectrum disorders are caused by the disruption of normal brain development in utero. The severity and range of symptoms is dictated by both the dosage and timing of ethanol administration, and the resulting developmental processes that are impacted. In order to investigate the effects of an acute, high-dose intoxication event on the development of medium spiny neurons (MSNs) in the striatum, mice were injected with ethanol on P6, and neuronal morphology was assessed after 24 h, or at 1 month or 5 months of age. Data indicate an immediate increase in MSN dendritic length and bran…
