Multi-level intervention of Tangluoning on mitochondrial thioredoxin deficiency in Schwann cells ameliorates diabetic peripheral neuropathy.

BACKGROUND: Diabetic peripheral neuropathy (DPN), the most prevalent complication of diabetes, currently lacks disease-modifying therapies. While Tangluoning recipe (TLN)-a traditional Chinese herbal medicine derived from Huangqi Guizhi Wuwu decoction-demonstrates therapeutic efficacy against DPN symptoms, its mechanism remained elusive. This study pioneers two transformative advances: Uncovering a previously unrecognized pathogenic axis linking mitochondrial thioredoxin (Trx2) deficiency to DPN demyelination; Establishing a novel dual-target therapeutic strategy that concurrently rescues mito

Cepharanthine-mediated endoplasmic reticulum stress inhibits Notch1 via binding GRP78 for suppressing hepatocellular carcinoma metastasis.

BACKGROUND: The metastasis of hepatocellular carcinoma (HCC) leads to a poor prognosis, wherein the activation of Notch1 is an essential contributor. Cepharanthine (Cep) has been identified for its effective antiviral function and versatile intracellular targets. Our previous study has only reported the anti-cancer efficacy of Cep in lung cancer, without an in-depth exploration. Herein, the present study aims to investigate the anti-metastasis effect in HCC, the target involved, and the molecular mechanism of Cep. METHODS: Stable over-expression of Notch1-N1ICD yielded C5WN1 cells compared wi

Experimental acute thrombotic stroke in baboons.

To study the effects of antithrombotic therapy in experimental stroke, we have characterized a baboon model of acute cerebrovascular thrombosis. In this model an inflatable silastic balloon cuff has been implanted by transorbital approach around the right middle cerebral artery (MCA), proximal to the take-off of the lenticulostriate arteries (LSA). Inflation of the balloon for 3 hours in six animals produced a stereotypic sustained stroke syndrome characterized by contralateral hemiparesis. An infarction volume of 3.2 +/- 1.5 cm3 in the ipsilateral corpus striatum was documented by computerize

Synthesis and Characterization of Bisphosphonate-Functionalized Gadolinium Oxide Nanoparticles as Nonionizing Contrast Agents to Detect Bone Turnover.

We developed a one-pot polyol synthetic strategy for generating bisphosphonate-conjugated citric acid gadolinium oxide nanoparticles (BP-GdOx-NPs) for use as a medical imaging contrast agent to detect bone turnover. This agent, akin to the current nuclear medicine 99mTechnetium MDP bone scan, utilizes bisphosphonates for bone-seeking properties while substituting the former radioisotope with GdOx-NPs, enabling detection via computed tomography (CT) and magnetic resonance imaging (MRI). Our synthetic approach ensures covalent linkage between citric acid-coated NPs and one of the R-groups on the

Highly sensitive and label-free detection of influenza H5N1 viral proteins using affinity peptide and porous BSA/MXene nanocomposite electrode.

Influenza viruses are known to cause pandemic flu through inter-human and animal-to-human transmissions. Neuraminidase (NA), which is a surface glycoprotein of both influenza A and B viruses, is a minor immunogenic determinant; however, it has been proposed as an ideal candidate for a real testing. We successfully identified an affinity peptide which is specific to the influenza H5N1 virus NA via phage display technique and observed initially its binding affinities using enzyme-linked immunosorbent assay (ELISA). In addition, four synthetic peptides were chemically synthesized to develop an af

A mandibular advancement device attenuates the abnormal morphology and function of mitochondria from the genioglossus in obstructive sleep apnea-hypopnea syndrome rabbits.

BACKGROUND: Obstructive sleep apnea hypopnea syndrome (OSAHS) is a serious and potentially life-threatening disease. Mandibular advancement device (MAD) has the characteristics of non-invasive, comfortable, portable and low-cost, making it the preferred treatment for mild-to-moderate OSAHS. Our previous studies found that abnormal contractility and fibre type distribution of the genioglossus could be caused by OSAHS. However, whether the mitochondria participate in these tissue changes is unclear. The effect of MAD treatment on the mitochondria of the genioglossus in OSAHS is also uncertain.

VDR activation attenuate cisplatin induced AKI by inhibiting ferroptosis.

Our preliminary work has revealed that vitamin D receptor (VDR) activation is protective against cisplatin induced acute kidney injury (AKI). Ferroptosis was recently reported to be involved in AKI. Here in this study, we investigated the internal relation between ferroptosis and the protective effect of VDR in cisplatin induced AKI. By using ferroptosis inhibitor ferrostatin-1 and measurement of ferroptotic cell death phenotype in both in vivo and in vitro cisplatin induced AKI model, we observed the decreased blood urea nitrogen, creatinine, and tissue injury by ferrostatin-1, hence validate

Interleukin-1-mediated release of interleukin-8 by asbestos-stimulated human pleural mesothelial cells.

The pleuropulmonary response to inhaled asbestos frequently involves inflammation and release of various cytokines from lung cells. Among these, interleukin-8 (IL-8) released from the mesothelium could augment inflammation of the pleura by attracting neutrophils to the pleural space. We used cultures of human pleural mesothelial cells (HPMC) to examine the mechanism of IL-8 production by asbestos and cytokines. Suspensions of amosite, chrysotile, or crocidolite asbestos in concentrations as low as 5 micrograms/ml enhanced release of IL-8 from HPMC during 6 h of incubation at 37 degrees C. Elec

Cell responses to Hydron by a new in-vitro method.

An in-vitro biotoxicity test system, suitable for the assessment of endodontic filling materials, has been developed and used to test cell responses to Hydron, AH26 and Tubliseal. A robust, well-characterized and stable cell line (L-cells) which was grown as uniform cultures on Millipore filters, has been used as indicator cells. As they approached confluence they were exposed to test substances for 24 h and biosynthetic activities were measured. The test system is a modification of that described by Wennberg et al. (1979). By inverting the cultures on organ-culture rafts, cells were separated

Structural analysis of the venom glands of the armed spider Phoneutria nigriventer (Keyserling, 1891): microanatomy, fine structure and confocal observations.

Spiders belonging to the genus Phoneutria (Perty, 1833), most commonly known as 'armed' spiders, are among the most dangerous species in Brazil due to high toxicity of their venom, associated with their habit of invading domestic or specific areas such as banana plantations. The venom of Phoneutria spiders is secreted by a pair of venom glands located inside their cephalothoraxes and connected to the chelicerae by two independent ducts. In the present study, the microanatomy and histological structure of the venom glands of Phoneutria nigriventer (Keyserling, 1891) were examined in detail by h

Amphiphilic Lipid-Polymer Triblock Architectures as Nanocarriers for Therapeutic Delivery.

This study investigates the formation and characterization of nanocarriers derived from the self-assembly of amphiphilic lipid-polymer hybrids (LPHs) engineered with a symmetric ABA block copolymer architecture. In this design, the terminal A-blocks comprise branched hydrophobic fatty acid (FA) moieties, while the central B-block consists of a hydrophilic polymer segment, either polyethylene glycol (PEG) or a polypeptide-based glycine-lysine (GK) construct. By systematically varying the degree of branching within the hydrophobic blocks and the chemical identity of the hydrophilic segments, thi
