Molecular switches of SQSTM1: the impact of post-translational modifications on autophagy and neurodegeneration.

SQSTM1/p62 (sequestosome 1) is an important receptor protein involved in many cellular signaling processes, including macroautophagy/autophagy. It is a molecular hub for cellular homeostasis and cellular responses. Within autophagy, SQSTM1 targets ubiquitinated cargo for degradation, maintaining cellular proteostasis. Structurally, SQSTM1 consists of several domains that facilitate its binding to ubiquitinated cargo, the formation of SQSTM1 aggregate inclusions, interactions with MAP1LC3/LC3, and the mediation of clearance via the autophagy pathway. Beyond its structure, post-translational mod

Tuberous sclerosis as an underlying basis for infantile spasm.

The study of the molecular pathogenesis of epilepsy in tuberous sclerosis has taken on a new dimension with the identification of the TSC1 and TSC2 genes. While the development of seizures is ultimately related to mutations in one of the two genes, the mechanism underlying the genotype-phenotype relationship remains a puzzle. This chapter, presented arguments in favor of the hypothesis that abnormal cortical excitability originates in and around focal areas of structural malformations (i.e., cortical tubers and dysplasia) and that these "lesions" are the biologic consequences of tuberin and/or

The RXRalpha ligand-dependent activation function 2 (AF-2) is important for mouse development.

We have engineered a mouse mutation that specifically deletes the C-terminal 18 amino acid sequence of the RXRalpha protein. This deletion corresponds to the last helical alpha structure (H12) of the ligand-binding domain (LBD), and includes the core of the Activating Domain of the Activation Function 2 (AF-2 AD core) that is thought to be crucial in mediating ligand-dependent transactivation by RXRalpha. The homozygous mutants (RXRalpha af2(o)), which die during the late fetal period or at birth, exhibit a subset of the abnormalities previously observed in RXRalpha -/- mutants, often with inc

N-cadherin identifies human endometrial epithelial progenitor cells by in vitro stem cell assays.

STUDY QUESTION: Is there a specific surface marker that identifies human endometrial epithelial progenitor cells with adult stem cell activity using in vitro assays? SUMMARY ANSWER: N-cadherin isolates clonogenic, self-renewing human endometrial epithelial progenitor cells with high proliferative potential that differentiate into cytokeratin+ gland-like structures in vitro and identifies their location in some cells of gland profiles predominantly in basalis endometrium adjacent to the myometrium. WHAT IS KNOWN ALREADY: Human endometrium contains a small population of clonogenic, self-renewi

Cyclin proteolysis as a retinoid cancer prevention mechanism.

The retinoids, natural and synthetic derivatives of vitamin A, are active in cancer therapy and prevention. Their biological effects are mediated through ligand-dependent interactions with retinoid receptors that associate with specific co-regulators. A better understanding of retinoid chemopreventive mechanisms is needed. Our prior work revealed that all-trans-retinoic acid (RA) prevented tobacco-specific carcinogenic transformation of cultured human bronchial epithelial cells. RA signaled G1 arrest that permitted repair of genomic DNA damage caused by these carcinogens. RA triggered G1 arres

Fra-1 replaces c-Fos-dependent functions in mice.

Structure-function analysis as well as studies with knock-out and transgenic mice have assigned distinct functions to c-Fos and Fra-1, two components of the transcription factor AP-1 (activator protein-1). To test whether Fra-1 could substitute for c-Fos, we generated knock-in mice that express Fra-1 in place of c-Fos. Fra-1 rescues c-Fos-dependent functions such as bone development and light-induced photoreceptor apoptosis. Importantly, rescue of bone cell differentiation, but not photoreceptor apoptosis, is gene-dosage dependent. Moreover, Fra-1 fails to substitute for c-Fos in inducing expr

A novel bacterial symbiont in the seaweed Gracilaria tenuistipitata and its promising anti-inflammatory and antiproliferative potential in relation to special secreted metabolites.

Marine macroalgae host compositionally complex microbiomes that constitute an evolutionarily refined, yet largely unexplored source of structurally diverse natural products. Thus, the bioactivities of the algae may be related to those of their associated microorganisms. Here we describe the discovery of strain S.100, an endophytic bacterium isolated from the red alga Gracilaria tenuistipitata and displaying remarkable anti-inflammatory and antiproliferative activities, through a primary screening on ethyl acetate (EtAc), chloroform and n-hexane extracts of its culture. S.100 was taxonomically

Uncertainty-aware quantitative analysis of high-throughput live cell migration data.

Cell migration is a fundamental biological process essential for embryonal development, immune function, and cancer metastasis, with migration velocity representing a key parameter of this behaviour. Today, cell migration velocity can be measured in high-throughput assays that generate complex, hierarchically structured datasets with technical noise, batch effects, and biological variability, introducing significant uncertainty in velocity estimates. Current statistical approaches often fail to rigorously quantify this uncertainty, limiting reproducibility and comparisons across independent ex

Transcription Factor Binding and Individual Genetic Risk of Valproate Teratogenicity.

BACKGROUND AND OBJECTIVES: Valproate (VPA) use during pregnancy is associated with a wide range of structural birth defects, but not all exposed children are affected and there is evidence for a genetic predisposition. The development of a pharmacogenomic biomarker test that can be used for preconception counseling, allowing access to women unnecessarily denied VPA treatment because of concern about teratogenic risks if they were to become pregnant, is challenged by a poor understanding of how variation in maternal DNA could modify the risk. We hypothesized that genomic variants that affect th

Dose-Dependent and Non-Autonomous Signaling in CAKUT: A Lineage-Specific Framework from Conditional Knockout Studies.

BACKGROUND/OBJECTIVES: Congenital anomalies of the kidney and urinary tract (CAKUTs) represent the leading cause of pediatric chronic kidney disease, yet the molecular mechanisms underlying these malformations remain incompletely understood. While genetic studies have identified numerous CAKUT-associated genes, conventional knockout approaches often result in embryonic lethality or fail to reveal tissue-specific gene functions. This review aims to synthesize findings from conditional knockout mouse studies that have elucidated the spatiotemporal requirements of key signaling pathways during ki

Direct and indirect evidence for pyroptosis and immune dysregulation in IVF/ICSI-ET-related early pregnancy loss.

BACKGROUND: Early pregnancy loss(EPL) remains a major obstacle to successful in vitro fertilization/intracytoplasmic sperm injection-embryo transfer(IVF/ICSI-ET). Immune dysregulation at the maternal-fetal interface has been increasingly implicated in implantation failure and early miscarriage, while inflammasome-mediated pyroptosis has emerged as a potential contributor to inflammatory tissue injury. However, the direct evidence linking pyroptosis to IVF/ICSI-ET-related EPL remains limited. OBJECTIVE: To critically appraise direct and indirect evidence regarding immune dysregulation and the

Balancing efficacy and risk: A toxicological review and safety transformation of Berberis plants and their alkaloids.

BACKGROUND: Berberis plants are widely distributed medicinal herbs with a long history of traditional use worldwide. Their primary active constituents are isoquinoline alkaloids, including berberine, palmatine, jatrorrhizine, and berbamine, which exhibit promising pharmacological activities in anti-inflammation, antibacterial, hypoglycemic, lipid-regulating, and antitumor effects. However, under specific doses, routes of administration, or individual conditions, these constituents may also induce multi-system toxic reactions, making the safety profile of Berberis plants a critical concern that

Concordant epigenetic and discordant clinical PHP1B/iPPSD3 manifestations in two monozygotic adolescent twins.

BACKGROUND: Imprinting disorders are caused by epigenetic alterations leading to abnormal gene expression from imprinted loci. Pseudohypoparathyroidism type 1B is characterized by methylation defects at the GNAS locus, affecting hormone responsiveness. Monozygotic twins discordant for imprinting disorders provide a unique opportunity to investigate the timing, maintenance, and tissue-specific distribution of epimutations and their relation to clinical expression. Most previous studies focus on infants, where fetal blood chimerism may influence epigenetic profiles, but data in adolescents remai

Unlocking the regenerative properties of extraembryonic membrane-derived biomaterials in tissue engineering.

Extraembryonic tissues play a crucial role in embryonic development and fetal growth and exhibit remarkable biological and biomechanical properties including, pro-angiogenic, anti-inflammatory, immunomodulatory, anti-fibrotic, antimicrobial, and viscoelastic. These characteristics have driven the development of innovative therapies for regenerative medicine applications. Unlike other biological tissues, the mammalian placenta is considered waste material, making it a highly accessible and easily obtainable resource that poses no additional risks to the health of the mother or fetus. Among the

Single-cell-pore-sized 3D printed scaffolds for retinal pigment epithelial cell therapy.

Cell therapy is one of the most promising methods to treat retinal degenerative diseases, and crucial to its success is optimizing biomaterials to facilitate the delivery of retinal pigment epithelial (RPE) cells. This study explores the application of single-cell-pore-sized 3D printed polycaprolactone (PCL) scaffolds for cultivating human embryonic stem cell-derived RPE cell sheets. It compares them with track-etched polyethylene terephthalate (PET) membranes, the commercial products used in clinical trials for RPE cell delivery. We engineered two types of scaffolds at the microscale to optim

Lack of ANKMY2 suppresses kidney cystogenesis in embryonic- and adult-onset polycystic kidney disease.

Autosomal dominant polycystic kidney disease (ADPKD) is characterized by progressive bilateral cyst formation. Multiple cellular pathways including second messenger cAMP signaling are dysregulated in ADPKD, but mechanisms initiating cysts are unknown. ADPKD is caused by mutations in PKD1/PKD2 genes encoding for polycystins that localize to primary cilia-nonmotile, microtubule-based dynamic compartments sensing extracellular chemical/mechanical signals. The compact cylindrical structure of cilia enables tunable signaling amplification regulatable by ciliary length. Severe cystogenesis from poly

Genomic Structural Equation Modelling Reveals the Shared Genetic Architecture for Oral Frailty.

PURPOSE: Oral frailty, an age-related decline in oral function and health linked to adverse geriatric outcomes, involves multiple phenotypes. Chronic periodontitis, a key inflammatory driver of tooth loss and systemic disease, is a cornerstone of this syndrome, yet the shared genetic architecture connecting it to other oral conditions remains uncharacterised. METHODS AND MATERIALS: We employed genomic structural equation modelling (genomic SEM) to integrate genome-wide association studies (GWAS) summary statistics from five oral frailty-related phenotypes, defining a common latent factor refl

Modulation of transferrin receptor by HIV-2.

BACKGROUND: Human immunodeficiency viruses (HIV-1 and HIV-2), are the causative agents of the acquired immunodeficiency syndrome (AIDS), that share substantial genomic and structural similarities, yet differ in replication dynamics and disease progression. While HIV-1 primarily enters host cells via CD4 and the chemokine co-receptors CCR5 and CXCR4, HIV-2 engages a broader range of chemokine receptors. The transferrin receptor (TFRC/CD71), a membrane protein essential for iron uptake and immune function, has recently been implicated in viral entry by other pathogens. In this study, we investig

Antagonism between wild-type and mutant β-catenin controls hepatoblastoma differentiation via fascin-1.

BACKGROUND & AIMS: β-catenin is a well-known effector of the Wnt pathway, and a key player in cadherin-mediated cell adhesion. Oncogenic mutations of β-catenin are very frequent in paediatric liver primary tumours. Those mutations are mostly heterozygous, which allows the co-expression of wild-type (WT) and mutated β-catenins in tumour cells. We investigated the interplay between WT and mutated β-catenins in liver tumour cells, and searched for new actors of the β-catenin pathway. METHODS: Using an RNAi strategy in β-catenin-mutated hepatoblastoma (HB) cells, we dissociated the structural and
