Effects of ultraviolet light on thymidine incorporation and DNA chain elongation in photoreactivable insect cells.

An insect cell line, IAL-PID2, was exposed to UV and analyzed for its ability to incorporate [3H]-thymidine and to elongate replicon-sized DNA fragments. After exposure to 5 or 10 J/m2 UV, the cells exhibited a rapid and prolonged depression in the rate of thymidine incorporation. Photoreactivation reduced this depression but did not entirely reverse it. For exposures of 5 J/m2 or above, full recovery did not occur until 18 h after exposure. The blockage of fork progression after UV exposure was fluence-dependent, with replication segments after exposure to 20 J/m2 being shorter than those obs

Synthesis of a gene coding for an insect-specific scorpion neurotoxin and attempts to express it using baculovirus vectors.

We have explored the possibility of improving baculovirus pesticides by incorporating an insect-specific neurotoxin gene into a baculovirus genome. A 112-bp gene (BeIt) encoding insectotoxin-1 of the scorpion Buthus eupeus was synthesized and cloned in Escherichia coli. For expression, BeIt was transferred to the DNA genome of Autographa californica nuclear polyhedrosis virus (AcMNPV). Three different recombinant AcMNPVs, carrying BeIt under the control of the strong AcMNPV polyhedrin promoter, were constructed and expression of BeIt was monitored upon infection of Spodoptera frugiperda (Sf) c

Phenylalanine-driven metabolic induced resistance activates phenylpropanoid defenses in tomato against Tetranychus urticae.

Tetranychus urticae (Koch), the two-spotted spider mite (TSSM), is a highly polyphagous agricultural pest, infesting more than 1,100 plant species. Chemical control of TSSM is limited by the rapid development of resistant mite populations. Previous studies have shown that exogenous application of phenylalanine to plants is efficiently absorbed and metabolized into phenylpropanoids, thereby enhancing plant resistance to fungal and insect pests. Here, we demonstrate that phenylalanine treatment also increases plant resistance to TSSM. In choice assays, mites preferentially colonized non-treated

From invasion to infection: The worldwide expansion and arboviral transmission potential of Aedes albopictus.

The Asian tiger mosquito, Aedes albopictus, has emerged as a globally invasive vector of major public health concern. This review synthesises current evidence on its invasion biology, vector competence, ecological plasticity, and expanding epidemiological significance, with particular emphasis on arboviral transmission dynamics. A comprehensive review of experimental, field-based, and modelling studies was conducted to identify determinants of vector competence and transmission risk, including Wolbachia-mediated viral interference, temperature-driven changes in extrinsic incubation period, ver

Plant-mediated RNAi via Panonychus citri-derived miRNAs disrupts adult emergence and confers mite resistance.

MicroRNAs (miRNAs), small non-coding RNAs that trigger RNA interference (RNAi), are promising candidates for pest control, yet their efficacy against non-insect pests remains poorly explored. This study evaluates endogenous mite-derived miRNAs as transgenes for crop resistance against Panonychus citri, a major pest with rapid acaricide resistance that severely damages citrus, soybean, and other cash crops. Through developmental miRNA libraries of P. citri, we identify miR-750 and miR-200a-5p as key regulators during the deutonymph-to-adult transition, whose overexpression significantly inhibit

Temperature and Pollinators Drive Evolutionary and Plastic Variation in Leaf and Petal Epicuticular Waxes.

Plants rely on epicuticular waxes to withstand stress and mediate interactions with other organisms, yet floral waxes remain largely unexplored despite their potential key role in reproduction. We examined how temperature and pollinators shaped leaf and petal waxes in Brassica rapa after six generations of experimental evolution under hot and ambient conditions with bumblebees (Bombus terrestris), Pieris rapae butterflies, acting as "pollinating herbivores" and also involving larval herbivory, or hand-pollination as a control. Leaves contained more wax but shorter aliphatic chains than petals.

Bactrocera dorsalis and Its Gut Microbiota: An Emerging Insect Model.

The gut microbiota influences host health, development, nutrition, and behavior, positioning it as a frontier research area in life sciences. Bactrocera dorsalis is a major agricultural pest, with a short life cycle, ease of laboratory rearing, and the availability of germ-free larvae. The gut microbiota of B. dorsalis is complex and relatively insensitive to environmental influences. Due to these advantages, B. dorsalis has emerged as a promising model organism for gut microbiota research. This review synthesizes the advantages of B. dorsalis as a model organism, detailing its gut structure a

Phytoplasma-plant interactions: effector-mediated host reprogramming, hormonal crosstalk, metabolic alterations and plant-mediated vector manipulation.

Phytoplasmas are wall-less, phloem-restricted bacterial pathogens that infect over 1,000 plant species, causing substantial losses in agriculture, horticulture, and forestry worldwide. Despite their reduced genomes and limited metabolic autonomy, these obligate parasites colonize diverse hosts through secreted effector proteins that extensively reprogram plant development, metabolism, immune signalling, and vector interactions. Advances in genomics, transcriptomics, proteomics, metabolomics, and functional studies have substantially clarified the molecular basis of phytoplasma pathogenicity an

Whole testicular organ and seminal cryopreservation in the malarial mosquito Anopheles gambiae (Diptera: Culicidae).

Anopheles gambiae (Giles) is a medically important mosquito species and a vector of the malaria Plasmodium parasite. There have been many advancements in vector control research that involves genetic manipulation of the mosquito. Malaria vector research requires rearing continuous mosquito colonies that are costly and carry risks to maintain. Therefore, we studied the feasibility of germplasm cryopreservation as a way to maintain genetic background as well as restore mosquito colonies. One part of germplasm cryopreservation is the cryopreservation of the whole germinal organ such as the testis

Landscape genetics of Cochliomyia hominivorax (Diptera: Calliphoridae): predicting dispersal pathways and expansion potential after reintroduction.

The New World screwworm fly, Cochliomyia hominivorax (Coquerel, 1858) (Diptera: Calliphoridae), is an obligate parasitic blowfly that causes traumatic myiasis and represents a major threat to livestock production in the Neotropics. Although eradication programs based on the Sterile Insect Technique (SIT) successfully eliminated the species from North and Central America, the risk of reinvasion remains a persistent concern. Understanding how landscape structure influences dispersal and connectivity is therefore essential for improving surveillance and control strategies. We evaluated the influe

Engineering resilient gene drives for sustainable malaria control by predicting, testing and overcoming target site resistance in Anopheles gambiae.

CRISPR-based gene drives are selfish genetic elements with the potential to spread through entire insect populations for sustainable vector control. Gene drives designed to disrupt the reproductive capacity of females can suppress laboratory populations of the malaria mosquito, Anopheles gambiae. However, any suppressive intervention will inevitably exert an evolutionary pressure for resistance, and the likelihood of resistance emerging at natural population scales remains poorly defined. Here, we present a pipeline to quantify the evolutionary space for resistance, enabling accelerated discov

Mark-Release-Recapture of Packed and Shipped Aedes aegypti with Wolbachia: Implications for Conducting Remote Incompatible Insect Technique Programs.

Male mosquitoes containing the endosymbiont Wolbachia (Wb+) can be used as a tool to suppress wild mosquito populations through a technique termed incompatible insect technique (IIT). IIT programs reduce wild mosquitoes via incompatible matings between released males and wild females to reduce the number of viable offspring produced in the next generation. Successful programs rely on regular release of incompatible males to outcompete wild males for female mates. Past IIT programs have relied on local production of Wb+ males to support regular releases of incompatible males. Here, we evaluated

Perinatal Exposure to the Neonicotinoid Thiacloprid Impacts Transcription of Neuroplasticity and Neuroendocrine Markers in Mice but Not in the Zebrafish Model.

Neonicotinoids are widely used insecticides in agriculture, aquaculture, pet care, and urban pest control. Initially developed to selectively target the insect cholinergic system, their extensive use has raised concerns about adverse effects on nontarget vertebrates. This study investigated the developmental neurotoxicity of the neonicotinoid thiacloprid using two vertebrate models: zebrafish and mice. Transgenic cyp19a1b-GFP zebrafish eleutheroembryos, which report estrogenic activity, were exposed to thiacloprid (10-6-10-8 M) for 4-5 days. No significant changes were observed in GFP expressi

Suppression of Aedes aegypti may not affect sympatric Aedes albopictus populations: findings from two years of entomological surveillance in Singapore.

Globally, multiple trials have successfully demonstrated the effectiveness of novel tools, such as the sterile and incompatible insect techniques, in suppressing Aedes aegypti populations. However, there is concern that Aedes albopictus, another arbovirus-competent vector, may occupy the niches vacated by Ae. aegypti in areas where these species occur in sympatry. Here we investigate these concerns within the context of a Wolbachia-based Ae. aegypti suppression programme in highly urban Singapore, where general mosquito management is sustained through environmental management. Using nationally

Immunogenicity of intraperitoneal and intranasal liposome adjuvanted VLP vaccines against SARS-CoV-2 infection.

Humans get SARS-CoV-2 infection mainly through inhalation; thus, vaccine that induces protective immunity at the virus entry site is important for early control of the infection. In this study, two anionic liposome (L)-adjuvanted VLP vaccines against SARS-CoV-2 were formulated. Baculovirus-Sf21 insect cell system was used for production of VLPs made of full-length S, M and E proteins. S protein of one vaccine (L-SME-VLPs) contained furin cleavage site at the S1/S2 junction, while that of another vaccine (L-S'ME-VLPs) did not. Both vaccines were innocuous and immunogenic when administered IP an

Effects of seed infection by Fusarium verticillioides on maize performance against Sesamia nonagrioides attack.

In maize (Zea mays L), the fungus Fusarium verticillioides can behave as a pathogen, but it is also able of asymptomatic colonization as an endophyte. Therefore, it would be of great value to identify metabolites and/or metabolic pathways implicated in mutualistic and pathogenic interactions. The objectives of the present study were: (i) to investigate the effect of seed colonization by F. verticillioides on maize growth in a group of inbreds with contrasting resistance to F. verticillioides; (ii) to know if maize priming by Fusarium seed infection affects maize response to other parasites and
