Preformed Stainless-Steel Crowns: Their Effects on Oral Hygiene Maintenance and Gingival Health - A Prospective Original Research.

The stainless-steel crown (SSC) is a durable restoration and has several indications for use in primary teeth such as following a pulpotomy/pulpectomy, fractured teeth, teeth with developmental defects, or large multi-surface caries lesions where amalgam is likely to fail. Due to its durability and a lifespan like the primary tooth, it could well be the gold standard in restorative care. SSCs protect the crown from fracture, reduce the possibility for leakage, and ensure a biological seal. However, the placement of the SSC should follow a meticulous technique. There are some clinical situation

Continual integration of single-cell multimodal data with MIRACLE.

Single-cell sequencing has transformed our understanding of cellular heterogeneity, enabling the construction of multi-omics atlases through data integration. However, conventional atlas updates require full reintegration of all datasets, creating scalability challenges that limit the timeliness and adaptability of biomedical research. Here we present multimodal integration with continual learning (MIRACLE), an online learning framework for scalable multimodal integration. Using dynamic architecture adaptation and data rehearsal, MIRACLE continually integrates diverse datasets while preserving

Adaptive chaos: mild disorder may help contain major disease.

We have qualitatively analyzed different cases of human disorders and displacement activities of animals and hypothesize that some of them are examples of low-dimensional dynamical chaos in biological organisms. We also considered a biological organism in the framework of the control system theory and found that chaotic regime in one subsystem may be compensating for the loss of chaos in another subsystem for the sake of stability of the whole system. According to the hypothesis chaotic behavior of different organs sets in a human body as an alternative to serious diseases or even death. The p

Transcriptional profile of Paracoccidioides spp. in response to itraconazole.

BACKGROUND: Itraconazole is currently used to treat paracoccidioidomycosis. The mechanism of action of azoles has been elucidated in some fungi, although little is known regarding its mechanism of action in Paracoccidioides spp. The present work focused on identification of regulated transcripts using representational difference analysis of Paracoccidioides spp. yeast cells treated with itraconazole for 1 and 2 h. RESULTS: Paracoccidioides Pb01 genes up-regulated by itraconazole included genes involved in cellular transport, metabolism/energy, transcription, cell rescue, defense and virulence

The road not taken: Evolution of tetrodotoxin resistance in the Sierra garter snake (Thamnophis couchii) by a path less travelled.

The repeated evolution of tetrodotoxin (TTX) resistance provides a model for testing hypotheses about the mechanisms of convergent evolution. This poison is broadly employed as a potent antipredator defence, blocking voltage-gated sodium channels (Nav ) in muscles and nerves, paralysing and sometimes killing predators. Resistance in taxa bearing this neurotoxin and a few predators appears to come from convergent replacements in specific Nav residues that interact with TTX. This stereotyped genetic response suggests molecular and phenotypic evolution may be constrained and predictable. Here, we

Transcriptome analysis of pika heart tissue reveals mechanisms underlying the adaptation of a keystone species on the roof of the world.

High-altitude environments impose intense stresses on living organisms and drive striking phenotypic and genetic adaptations, such as hypoxia resistance, cold tolerance, and increases in metabolic capacity and body mass. As one of the most successful and dominant mammals on the Qinghai-Tibetan Plateau (QHTP), the plateau pika (Ochotona curzoniae) has adapted to the extreme environments of the highest altitudes of this region and exhibits tolerance to cold and hypoxia, in contrast to closely related species that inhabit the peripheral alpine bush or forests. To explore the potential genetic mec

Investigating the genetic and epigenetic basis of big biological questions with the parthenogenetic marbled crayfish: A review and perspectives.

In the last 15 years, considerable attempts have been undertaken to develop the obligately parthenogenetic marbled crayfish Procambarus virginalis as a new model in biology. Its main advantage is the production of large numbers of offspring that are genetically identical to the mother, making this crustacean particularly suitable for research in epigenetics. Now, a draft genome, transcriptome and genome-wide methylome are available opening new windows for research. In this article, I summarize the biological advantages and genomic and epigenetic features of marbled crayfish and, based on first

[Trends of physical development and adaptation to the study load and health-improving sports activities in elementary schoolchildren of Moscow].

UNLABELLED: The inclusion of 7-10-year-old children in the school system occurs in the context of the interaction of two processes: activation of ontogenesis at this stage of age development and stimulation of biological and psychological adaptation of younger schoolchildren to the learning activity. The main factors negatively affecting the health of elementary school students are the following: hypokinesia, mental and psycho-emotional stress related to computer and digital learning technologies, prolonged stay in closed rooms of the educational institution rather than in the natural environm

Capsid-Targeting Biologic Achieves Broad HIV-1 Neutralization with a High Barrier to Resistance.

Antiretroviral therapy (ART) has proven effective in suppressing HIV-1 replication, but further development of HIV-1 inhibitors is continually driven by the challenge of drug resistance and viral adaptation. The HIV-1 capsid is a promising target for treatment due to its high sequence conservation as well as its crucial role in the viral life cycle. Recently, we have developed a novel capsid-targeting biologic that prevents HIV-1 replication by efficient degradation of newly synthesized capsid. Here, we have investigated the sensitivity to viral escape as well as the breadth of this biologic a

Pneumococcal Colonization and Virulence Factors Identified Via Experimental Evolution in Infection Models.

Streptococcus pneumoniae is a commensal of the human nasopharynx and a major cause of respiratory and invasive disease. We examined adaptation and evolution of pneumococcus, within nasopharynx and lungs, in an experimental system where the selective pressures associated with transmission were removed. This was achieved by serial passage of pneumococci, separately, in mouse models of nasopharyngeal carriage or pneumonia. Passaged pneumococci became more effective colonizers of the respiratory tract and we observed several examples of potential parallel evolution. The cell wall-modifying glycosy

Whole-Genome Sequencing of African Dogs Provides Insights into Adaptations against Tropical Parasites.

Natural selection in domestic dogs is of great interest in evolutionary biology since dogs have migrated to every inhabited continent of the world alongside humans, and adapted to diverse environments. Here, we explored their demographic history and genetic basis of adaptation to the tropical African environment using whole genome analyses of 19 African indigenous dogs from Nigeria. Demographic analysis suggests that the ancestors of these dogs migrated into Africa from Eurasia 14,000 years ago and underwent a severe founder effect before population expansion. Admixture analysis further reveal

The roles of plasticity and evolutionary change in shaping gene expression variation in natural populations of extremophile fish.

The notorious plasticity of gene expression responses and the complexity of environmental gradients complicate the identification of adaptive differences in gene regulation among populations. We combined transcriptome analyses in nature with common-garden and exposure experiments to establish cause-effect relationships between the presence of a physiochemical stressor and expression differences, as well as to test how evolutionary change and plasticity interact to shape gene expression variation in natural systems. We studied two evolutionarily independent population pairs of an extremophile f

Genetics, evolution, and adaptive significance of the selfing syndrome in the genus Capsella.

The change from outbreeding to selfing is one of the most frequent evolutionary transitions in flowering plants. It is often accompanied by characteristic morphological and functional changes to the flowers (the selfing syndrome), including reduced flower size and opening. Little is known about the developmental and genetic basis of the selfing syndrome, as well as its adaptive significance. Here, we address these issues using the two closely related species Capsella grandiflora (the ancestral outbreeder) and red shepherd's purse (Capsella rubella, the derived selfer). In C. rubella, petal siz

Transient genital abnormalities in striped hyenas (Hyaena hyaena).

The highly masculinized genitalia of female spotted hyenas Crocuta crocuta is unique among mammals: Crocuta have no external vagina so urination, penile intromission and parturition take place through the clitoris, which mimics a fully erectile male penis. Among hyenids, virilization of external female genitalia has previously been observed only in Crocuta, so functional explanations of masculinization have focused on aspects of social ecology unique to the species. Here we first show that the striped hyena Hyaena hyaena exhibits both unusual similarity in male and female androgen concentratio

Rapid recovery and identification of anthrax bacteria from the environment.

Bacillus anthracis has been recognized as a highly likely biological warfare or terrorist agent. We have designed culture techniques to rapidly isolate and identify "live" anthrax from suspected environmental release. A special medium (3AT medium) allows for discrimination between closely related bacilli and non-pathogenic strains. Nitrate was found to be a primary factor influencing spore formation in Bacillus anthracis. Nitrate reduction in anthrax is not an adaptation to saprophytic environmental existence, but it is a signal to enhance environmental survival upon the death of the anthrax h

Toward an ecology of women's reproductive health.

Medical ecology has been one of the dominant theoretical strands in medical anthropology since the 1960s. It has come to be identified with a rather narrow range of research issues, concerning such matters as adaptation to climate and infectious disease, and has dealt with small-scale foraging societies. An ecological model has potential for dealing broadly with other health issues, in the case explored here, women's reproductive health, a topic that has frequently been approached from either a cultural or biological standpoint rather than the integrated one advocated here.

AP2/ERF transcription factors in plant stress adaptation: evolutionary expansion, functional diversification and hormone crosstalk.

APETALA2/ethylene response factor (AP2/ERF) transcription factors constitute a plant-specific regulatory family that coordinates plant development, environmental stress responses, and hormone signaling. However, the contribution of AP2/ERF family expansion and functional diversification to environmental adaptation has not been fully integrated across evolutionary and mechanistic scales. This review synthesizes classical findings and representative advances from the past five years in structural classification, duplication-driven expansion, functional diversification, stress adaptation, and hor

Biological Foundation Models for Complex Disease Research and Clinical Translation.

Complex diseases, including cancer, rare genetic disorders, neurodevelopmental and psychiatric conditions, and neurodegenerative diseases, arise from interactions among genetic variation, gene regulation, and cellular states that are difficult to capture using a single data type or biological scale. Biological foundation models address this challenge by treating nucleotides and genes as tokens and learning representations that can be transferred to downstream biomedical and clinical tasks. In this review, we examine two major model classes, genomic sequence foundation models and cell foundatio

Possible models for multiple chemical sensitivity: conceptual issues and role of the limbic system.

Conceivably, chemicals contacting olfactory nerve projections in the nose could either be transported into or relay electrical signals to the limbic region, leading to a vast array of symptoms. Likewise, thought processes and mood states may trigger or interrupt pre-existing limbic activity. At present, however, no evidence suggests that limbic activity triggered by environmental exposures can be entirely overcome by psychologic interventions. One important ramification of a limbic hypothesis, if true, is that no convenient biologic marker for multiple chemical sensitivity may exist at the pre

Climate-driven adaptive microevolution in wild yeasts.

Environmental variation imposes strong selection on microbial populations, yet linking ecological gradients to genomic and phenotypic diversification remains challenging due to limited sampling of wild microbes. Here, we leveraged the exceptional abundance of the cold-adapted yeast Saccharomyces eubayanus throughout Patagonian Nothofagus forests to investigate how climatic gradients shape natural microbial diversity. Using systematic sampling across ~1,000 km of Chilean Patagonia, spanning multiple seasons, altitudes, and thermal regimes, we integrated environmental, genomic, and phenotypic an
