High-resolution genomics uncovers region-specific evolution and virulence of ocular Streptococcus pneumoniae.

UNLABELLED: Streptococcus pneumoniae is the leading cause of sight-threatening ocular infection. However, comprehensive molecular epidemiological analyses that focus on ocular pneumococci are limited. We performed a high-resolution whole-genome analysis (WGS) of ocular pneumococcal isolates from the Zhejiang province of China and a global data set (n = 58). Using latex agglutination and the Quellung reaction, nine serotypes were identified, with PCV13 covering 54.5% of the Chinese isolates, and no non-typeable (NT) strains were found in conjunctivitis. Illumina short-read data were assembled d

Subconjunctival Administration of Adeno-associated Virus Vectors in Small Animal Models.

Ocular diseases include a wide range of inherited genetic and acquired disorders that are appealing targets for local drug delivery due to their relative ease of accessibility via multiple administration routes. Subconjunctival (SC) injections offer advantages over other intraocular administration routes as they are simple, safe, require only local anesthesia, and are usually performed in an outpatient setting. Although SC injections in small animals usually require the assistance of an operating microscope due to the size of the eye, they are widely utilized for drug delivery, including gene

IgA and IgG1 reactivities assessed by flow cytometry mirror clinical aspects of infants with ocular congenital toxoplasmosis.

This study intended to apply the flow cytometric analysis of IgA and IgG reactivity and intracytoplasmic cytokine analysis to understand and decode the clinical aspects of infants with ocular congenital toxoplasmosis. The Toxoplasma gondii-infected infants (TOXO) were subdivided according to their clinical aspects based on the absence (NRL), presence of active (ARL), active/cicatricial (ACRL) or cicatricial retinochoroidal lesions (CRL) and compared to non-infected controls (NI). The reactivity of anti-T. gondii IgG subclasses resembles the clinical aspects of ocular lesions. IgG and IgG1 disc

αB-crystallin elastin-like polypeptides for sustained ocular drug delivery.

The unfolded protein response (UPR) has been mechanistically implicated in multiple ocular disorders, including age-related macular degeneration (AMD). Therapeutic intervention using an αB-crystallin-derived 'mini-cry' chaperone peptide has potential to restore proteostasis; however, its low molecular weight results in rapid ocular clearance. To prolong mini-cry's ocular effects, our team linked it with elastin-like polypeptides (ELPs). ELPs are thermoresponsive polymers that phase-separate into microscale coacervates. Provided ELPs do not interfere with biological activity, cry-ELPs could pro

Self-assembling biomimetic peptide hydrogel regulates tissue homeostasis to promote repair of persistent corneal epithelial defects.

Persistent corneal epithelial defects (PCED) remain a significant clinical challenge due to rapid tear turnover, blinking-induced shear stress, and a protease-rich ocular surface that limit the efficacy of soluble bioactive factors. We report a dual-functional self-assembling peptide hydrogel, mIGF-Gel (Biotin-DFYIGSSSR), incorporating the adhesion motif YIGSR derived from laminin and the SSSR bioactive motif derived from insulin-like growth factor 1 (IGF-1) in a single design. The peptide forms a β-sheet-rich nanofibrous hydrogel with shear-thinning, viscoelastic, and self-recovering properti

Primary position upbeat nystagmus.

Primary position upbeat nystagmus is a rare clinical finding. We report a patient with clinically isolated syndrome suggestive of multiple sclerosis who presented with primary position upbeat nystagmus. MRI revealed a demyelinating lesion in the lower medulla, which affected the nucleus intercalatus; this type of lesion inhibits the flocculovestibular inhibitory pathway, thereby causing upbeat nystagmus. Nystagmus still persisted after pulsed corticosteroid therapy. This could be due to a loss of central adaptation of the vestibulo-ocular system in our patient, because of more diffuse brainste

Ocular surface immune cell diversity in dry eye disease.

Dry eye disease (DED) is a multifactorial chronic ocular surface inflammatory condition. Disease severity has been directly related to the immuno-inflammatory status of the ocular surface. Any perturbation in the orchestrated functional harmony between the ocular surface structural cells and immune cells, both resident and trafficking ones, can adversely affect ocular surface health. The diversity and contribution of ocular surface immune cells in DED have been of interest for over a couple of decades. As is true with any mucosal tissue, the ocular surface harbors a variety of immune cells of

Differential effect of ischemia/reperfusion on pigmented and albino rabbit retina.

The purpose of the study was to compare the retinal sensitivity of pigmented and albino rabbits to ischemia/reperfusion-induced electroretinogram (ERG) alterations and optic nerve morphological changes. High intraocular pressure (HIOP) was induced by applying a suction-cup on the eye and a depression with an ophthalmodynamometer. HIOP was maintained for lengths of time (30-75 min). Flash ERGs were recorded in dark-adapted animals for ischemia and 2 h reperfusion periods. Two weeks later, histological examination of the retinas and optic nerves was done. Albino rabbits submitted to 45 min HIOP

Perceptual adaptation to the correction of natural astigmatism.

BACKGROUND: The visual system adjusts to changes in the environment, as well as to changes within the observer, adapting continuously to maintain a match between visual coding and visual environment. We evaluated whether the perception of oriented blur is biased by the native astigmatism, and studied the time course of the after-effects following spectacle correction of astigmatism in habitually non-corrected astigmats. METHODS AND FINDINGS: We tested potential shifts of the perceptual judgments of blur orientation in 21 subjects. The psychophysical test consisted on a single interval orienta

ERG rod a-wave in Oguchi disease.

We analyzed the change in the ERG rod a-wave waveform during the course of dark adaptation in two patients with Oguchi disease. Two Japanese patients showed a homozygous arrestin 1147delA mutation. Scotopic flash ERGs were recorded after different periods of dark adaptation. ERG rod a-waves were obtained after subtraction of the cone ERG contribution. The rod a-waves were fitted with a model of the rod receptor signal. The parameters, Rm(p3) (maximum a-wave amplitude) and S (sensitivity) were calculated. Longer periods in the dark produced larger rod a-wave but only to the first flash presente

Effective medical examination of VDT workers using a new testing method of ocular alignment.

To investigate the relationship between asthenopia of VDT workers and the ocular alignment, we measured far and near alignment of 407 subjects (74 VDT workers with asthenopia, 137 VDT workers without asthenopia, and 196 non-VDT workers) using Total Vision tester (VT-500). There was no relationship between the asthenopia and the records of far alignment test. On the other hand, the incidence of exophoria and convergence insufficiency type in near alignment test was significantly high in the group of VDT workers with asthenopia. From these results, we speculate that cases who have exophoria or s

[Presbytia: are eyeglasses unavoidable?].

The power of eye accommodation to close vision decreases with age. This physiological process, called presbytia (or presbyopia), occurs in all people reaching maturity, irrespective of sex and race. This paper describes the evolution of our knowledge concerning this phenomenon and the various means of correction utilized nowadays to compensate for the loss of accommodation power.

Analysis of the electroretinogram in toxoplasma retinochorioiditis.

The decision to use therapy in toxoplasma retinochorioiditis depends on the location of the active lesion and the presence of vitreous activity. In eyes with dense vitreous clouding it can be difficult to see whether the macular region is involved or not. In theory the localisation of a lesion can be estimated on the basis of the flash ERG. The standard flash electroretinogram was recorded in 23 patients with inactive toxoplasma retinochorioiditis lesions in the retina. In 17 cases a lesion was present within the central 12 degrees of the visual field, 8 of these had a reduced photopic ERG. In

European vestibular experiments on the Spacelab-1 mission: 7. Ocular counterrolling measurements pre- and post-flight.

The static ocular counterrolling (OCR) of the four scientific crew members in the first Spacelab mission was measured during baseline-data-collection before and after the flight of SL-1. It was presumed that the modification of otolithic responses during spaceflight will be reflected in specific changes of the OCR-gain on the first days after recovery. The magnitude of OCR was determined analysing colour-transparencies of subjects right eyes that were produced in different positions of lateral body tilt. In general, one subject did not show any changes at all; three subjects exhibited a signif

A new cause for difficulty in seeing at night.

A syndrome is described in which patients complain of inability to see at night, although their rod and cone thresholds are normal. In such persons the rods, when dark-adapted, elevate cone flicker thresholds by 2 log units or more, and this condition is the basis of their complaint. Patients are often thought to be hysterics or to have congenital stationary night blindness.

Effects of pattern deprivation on visual cortical cells in the rabbit: a reevaluation.

The response properties of 217 cells recorded from the monocular segment of primary visual cortex in rabbits reared with lid suture of the contralateral eye (monocular deprivation, MD) were studied. These data were compared with 280 cells recorded from normal rabbits. There was no change in the percentage of orientation-selective cells, nonorientation-selective cells, or unmappable/unresponsive cells in MD animals compared with normals. Among orientation selective cells the orientation-tuning range of cells in MD animals was normal, and the predominance of cells with horizontal preferred orien
