Adenosine induced atrial fibrillation precipitating polymorphic ventricular tachycardia.

An 86-year-old female developed supraventricular tachycardia 36 hours after a myocardial infarction (MI). She developed atrial fibrillation and polymorphic ventricular tachycardia (PVT) following administration of 12 mg of adenosine. The PVT caused hemodynamic instability with no response to cardioversion, but termination with procainamide. The heart is vulnerable to hemodynamically unstable, possibly lethal, PVT early after MI under some circumstances. This vulnerability may be exposed following administration of adenosine. Extra caution is warranted when using adenosine in the post-MI period…

Molecular biology of adenosine triphosphate-sensitive potassium channels.

KATP channels are a newly defined class of potassium channels based on the physical association of an ABC protein, the sulfonylurea receptor, and a K+ inward rectifier subunit. The beta-cell KATP channel is composed of SUR1, the high-affinity sulfonylurea receptor with multiple TMDs and two NBFs, and KIR6.2, a weak inward rectifier, in a 1:1 stoichiometry. The pore of the channel is formed by KIR6.2 in a tetrameric arrangement; the overall stoichiometry of active channels is (SUR1/KIR6.2)4. The two subunits form a tightly integrated whole. KIR6.2 can be expressed in the plasma membrane either

Exogenous adenosine potentiates hypnosis induced by intravenous anaesthetics.

We investigated the effect of adenosine on hypnosis induced by thiopentone, propofol and midazolam in mice. The onset and duration of hypnosis were determined by the loss of righting reflex. Adenosine and 2-chloroadenosine caused a significant shortening of onset of sleep-time and prolongation of duration of sleep-time in all groups (p < 0.05). Dipyridamole administration before combined intravenous anaesthetic-adenosine or intravenous anaesthetic-2-chloroadenosine administration produced similar effects to adenosine (p < 0.05). The adenosine antagonist theophylline, given before intravenous a…

Adenosine receptor activation modulates intraocular pressure in rabbits.

The relatively selective adenosine A1 agonists N6-cyclohexy-ladenosine (CHA), R(-)-N6-(2-phenylisopropyl)adenosine (R-PIA) and S(+)-N6-(2-phenylisopropyl)adenosine (S-PIA); the A2a agonist 2-p-carboxyethyl)phenethyl-amino-5'-N-ethylcarbox-amidoadenosin e (CGS-21680) and the nonselective A2 agonist 2-phenylaminoadenosine (CV-1808) were evaluated in vivo for their effects on aqueous humor dynamics and in vitro for their action on 3H-norepinephrine release and cAMP accumulation in the isolated iris/ciliary body. These studies demonstrated that adenosine agonists can modulate intraocular pressure …

Diagnostic value of cerebrospinal fluid adenosine deaminase determination.

We measured the activity of adenosine deaminase (ADA) in the cerebrospinal fluid of 3 patients with tuberculous meningitis, 38 with viral meningitis, 15 with bacterial meningitis, 5 with malignant lymphoma, 11 with cerebrovascular diseases and 13 with miscellaneous neurological disorders. The highest ADA activities were observed in patients with tuberculous meningitis (median 21.3 U/l, range 20.0-23.0) and lymphoma (13.0 U/l, range 4.0-25.0). The sensitivity of the test for diagnosing tuberculous meningitis was 100% and the specificity 99% when a cut-off value of 20.0 U/l was used. We conclude…

Adenosine analogs inhibit fighting in isolated male mice.

The potent adenosine analogs N-ethylcarboxamide adenosine (NECA) and phenylisopropyladenosine (PIA) inhibit fighting and associated agonistic behaviors in isolated male mice. These effects are reversed by methylxanthines; moderate doses of NECA which inhibit fighting have minimal effects on spontaneous locomotor activity. At very low doses, both NECA and PIA increase fighting in parallel with previously reported increases of motor activity. Brain levels of [3H]-NECA and [3H]-PIA achieved at behaviorally effective doses suggest an involvement of adenosine receptors. The biochemical mechanism of…

Adenosine release may mediate spinal analgesia by morphine.

Spinal analgesia produced by morphine is blocked by methylxanthine adenosine receptor antagonists. In biochemical studies, morphine releases adenosine from spinal cord synaptosomes prepared from the dorsal spinal cord, as well as from the intact spinal cord in vivo. Adenosine release is reduced by intrathecal and neonatal pretreatment with capsaicin but not by intrathecal pretreatment with 6-hydroxydopamine or 5,7-dihydroxytryptamine, indicating that adenosine originates from small-diameter primary afferent neurons but not descending monoaminergic pathways. In this Viewpoint Jana Sawynok and c…

Increased erythrocyte adenosine deaminase activity without haemolytic anaemia.

A 4-fold increase of red blood cell adenosine deaminase (ADA) activity was found in a patient without haemolytic anaemia, but with mild anisopoikilocytosis. High-performance liquid chromatography showed a 40% reduction of adenosine-5'-triphosphate (ATP) while all the other nucleotides were in normal ranges. The patient's parents (first cousins) and a brother displayed the same enzyme activities as the controls. This observation suggests that mild increases of ADA activity is neither a marker for congenital hypoplastic anaemia as previously reported nor associated with haemolytic anaemia.

Intracoronary adenosine deaminase reduces canine myocardial reactive hyperemia.

We employed intracoronary infusions of calf intestine adenosine deaminase (ADA) to test the hypothesis that adenosine regulates coronary blood flow during myocardial reactive hyperemia (RH). Infusions of 4.5 U ADA/min per kg body weight into the left circumflex coronary artery of 10 open-chest dogs reversibly reduced repayment of flow debt by 30-39% (P less than 0.05) following 5-, 10-, 15-, 20-, and 30-second coronary occlusions, the percentage reduction being independent of occlusion length. ADA reduced peak RH flow rate (17%, P less than 0.05) only after 5-second occlusions. Intracoronary i…

Severe combined immunodeficiency disease with adenosine deaminase deficiency.

Severe combined immunodeficiency disease with adenosine deaminase deficiency was reported of a 1-year-2-month-old boy, who showed clinical picture of pulmonary candidiasis. The serum immunoglobulin level and absolute lymphocyte count were markedly diminished. But immunological study revealed both T- and B-cell surface markers on the peripheral lymphocytes, in spite of complete deficiency of both systems.

Adenosine signaling: the convergent hub in antidepressant action.

A groundbreaking study by Yue et al. reveals adenosine signaling as the convergent mechanism underlying the rapid antidepressant effects of both ketamine and electroconvulsive therapy. This discovery not only resolves a longstanding mechanistic puzzle but also opens new therapeutic avenues, including novel ketamine derivatives and nonpharmacological interventions that harness adenosine's antidepressant potential.

Adenosine receptors in blood cells: Biomarkers and beyond.

The identification of reliable, non-invasive biomarkers that reflect molecular alterations in deep and inaccessible tissues remains a major challenge in translational medicine. In this context, peripheral blood cells have emerged as valuable surrogates of tissue-level pathophysiology. Among molecular systems expressed in circulating cells, adenosine receptors (ARs) represent particularly attractive candidates due to their central role in regulating inflammation, immune responses, vascular function, and tissue homeostasis. This review focuses on the expression and functional modulation of AR su…

Extracellular Adenosine in Gastric Cancer: The Role of GCSCs.

Gastric cancer (GC) is among the most common and deadliest types of cancer, with a poor prognosis primarily due to late-stage detection and the presence of cancer stem cells (CSCs). This study investigates the mechanisms regulating extracellular adenosine levels in gastric cancer stem-like cells (GCSCs) derived from the MKN-74 cell line. Our results show that GCSCs release more ATP into the extracellular medium and exhibit higher levels of CD39 expression, which enables them to hydrolyze a greater amount of ATP. Furthermore, we also found that GCSCs possess a greater capacity to hydrolyze AMP,…

SAM cycle and adenosine at the heart of immune conversations.

Recent work by Nedbalova et al. reframes the SAM transmethylation pathway as a biosensor and signaling hub, linking immune activation to systemic metabolic changes via adenosine export. Their compelling findings reveal a key mechanism by which immune cells influence organism-wide physiology beyond traditional methylation roles.

Detection of Adenosine Phosphates in Blood by nh-PHIP.

Hyperpolarization techniques are rapidly advancing and beginning to make an impact on chemical analysis by NMR spectroscopy, bringing notice … anol-chloroform mixture are essential steps to yield extracts compatible with nh-PHIP. Detection of adenosine phosphates in the extracts requires the removal of interfering Mg2+ cations from the ATP-Mg complex. Addition of the Et3N/Et3N-HCl organic buffer helped to maintain the protonation/deprotonation state of the analytes in the NMR sample. This work pre …

The Immune Regulatory Role of Adenosine in the Tumor Microenvironment.

Adenosine, an immunosuppressive metabolite, is produced by adenosine triphosphate (ATP) released from dying or stressed cells and is found at high levels in the tumor microenvironment of most solid tumors. It mediates pro-tumor activities by inducing tumor cell proliferation, migration or invasion, tumor tissue angiogenesis, and chemoresistance. In addition, adenosine plays an important role in regulating anti-tumor immune responses and facilitating tumor immune escape. Adenosine receptors are broadly expressed by tumor-infiltrated immune cells, including suppressive tumor-associated macrophag…
