A collaborative search for albumin genetic variants (alloalbumins) was undertaken by cellulose acetate and agarose electrophoresis at pH 8.6 of the sera of patients at two major medical centers in the United States and of nearly 20,000 blood donors in Japan. Seventeen instances of alloalbuminemia were ascertained, and seven different alloalbumin types were characterized by structural study. Two previously unreported alloalbumin types were identified. In one type, which was present in a Caucasian family and designated Iowa City-1, aspartic acid at position 365 was replaced by valine (365 Asp---…

The permeability of the microcirculation to native and glycosylated albumin was tested in 25 non-diabetic Syrian hamsters. The microvasculature of the cheek pouch was studied by a fluorescent video-microscopy technique after the animals had been injected with fluorescent native or glycosylated albumin or with both. Native albumin remained in the cheek pouch microvasculature, whereas glycosylated albumin leaked out of the microvascular bed along the postcapillary and collecting venules. The extravascular leakage of glycosylated albumin, probably due to its electrical or conformational change, m

Human serum albumin was incubated at 37 degrees in 0.01 M phosphate buffer (pH 7.4) under sterile conditions for up to 10 days with labeled [14C]glucose (1-25 mg/ml). Glucose incorporated into albumin was calculated following extensive dialysis of the incubation mixture. The results indicated that glucose reacted with albumin by a nonenzymatic process involving Schiff base formation and Amadori rearrangement to a stable ketoamine derivative. The degree of glucosylation was dependent on the reaction time, glucose concentration, and pH. Glucosylation was enhanced when albumin was fatty acid free

A method of large scale plasma fractionation where PEG precipitation combined with absorption is described. This method permits isolation of most globulins under mild conditions. The final purification of the albumin is achieved by heat treatment at 60 degrees C pH 4.8. The quality of albumin preparations for therapeutic use is discussed from two points of view, purity and nativity. The main problem of purity is the content of high molecular weight aggregates formed by the pasteurization (60 degrees C, 10 h.). The aggregates give a risk for immunological reactions. The nativity of albumin is c

The use of polyacrylamide gel electrophoresis for the quantitation of albumin dimer in commercial albumin samples typically results in an overestimation of the percentage of dimer, as a consequence of significant deviations from Beer's law. This report describes a polyacrylamide gel electrophoresis technique which employs an internal standard for quantitation of albumin dimer. A constant amount of beta-2-glycoprotein I is added to the sample prior to electrophoresis, and the amount of albumin dimer is determined by comparison to that of beta 2-glycoprotein I. A number of commercial albumin sol

In plasma fractionation industry different methods are used for producing albumin for therapeutic use. We wanted to investigate whether the different ways of fractionating plasma have an effect on the nativity of the albumin molecules. Isoelectric focusing in 6M urea has shown the presence of variable amounts of oxidized albumin in different preparations. The presence of oxidized forms indicates that an initial denaturation has taken place. To evaluate if the alteration in the albumin molecules has any effects in vivo, metabolic studies in rabbits and humans with radioiodinated albumin were un

Albumin is the most effective oncotic agent and has wide clinical application. In the resuscitation of injured patients with hypovolemic shock (during war and peace) the use of supplemental albumin is purported to be more effective in restoring plasma volume, increasing cardiac output, preventing pulmonary oedema and maintaining organ perfusion than resuscitation without supplemental albumin. Most important in a study of albumin are criterias for appropriate use because it was sometimes unjustified. Therapeutic use of albumin should not be dictated by arbitrary or pragmatic restriction but rat

The influence of free fatty acids (FFA's) on the albumin binding of bilirubin was studied in vitro in the plasma of infants with neonatal hyperbilirubineamia and in solutions employing crystalline albumin to which bilirubin and FFA (oleic acid) were added. The bilirubin saturation index (SI) was utilized to distinguish between that fraction of bilirubin bound at the primary (high-affinity) site of albumin and bilirubin bound at secondary sites from which it is easily dissociated by salicylate. The relative saturation of albumin with bilirubin was also measured by addition of salicylate to whol

Examination of the records of 340 children admitted to hospital in Uganda with oedematous malnutrition (kwashiorkor) showed that the serum-albumin concentration at admission was closely related to mortality-rates. Furthermore, in this series, it was not possible to show a simple relation between mortality-rates and the degree of wasting present. The results further emphasise the need to include serum-albumin concentration in international classification systems for the assessment of protein-energy malnutrition, especially that of the kwashiorkor type.

Two clinical trials were designed to test the effect of albumin administration before phototherapy for non-haemolytic neonatal jaundice. The first with 18 hours of phototherapy showed that the albumin-treated group (23 infants) had higher and more prolonged jaundice than the group (27 infants) given phototherapy alone. In the second study on 29 infants there were two groups as before, but the duration of phototherapy was increased to at least 48 hours and a third untreated group was included. These results showed significant differences between all three groups, the cases receiving phototherap
