Drinks like a fish: using zebrafish to understand alcoholic liver disease.

Steatosis is the most common consequence of acute alcohol abuse, such as occurs during a drinking binge. Acute alcohol induced steatosis may predispose to more severe hepatic disease. We have developed a model of alcoholic liver disease (ALD) in zebrafish larvae to provide a system in which the genes and pathways that contribute to steatosis can be rapidly identified. Zebrafish larvae represent an attractive vertebrate model for studying acute ALD because they possess the pathways to metabolize alcohol, the liver is mature by 4 days post-fertilization (dpf), and alcohol can be simply added to

[And wine maketh glad the heart of man: is it so?--allergy to alcoholic drinks].

Adverse reactions to alcoholic drinks are common, especially in asthmatic patients. Since these drinks are the end product of a long process consisting of multiple ingredients, many factors may induce such reactions. This is a case study of a woman who has suffered from recurrent episodes of cutaneous eruption and dyspnea. Meticulous inquiry eventually revealed that drinking white wine was responsible for those symptoms. Grapes, barley, various fruits, alcohol, preserving substances like sulfite, biogenic amines like histamine and other ingredients can all be potential triggers of the adverse

Does an energy drink modify the effects of alcohol in a maximal effort test?

BACKGROUND: There are popular reports on the combined use of alcohol and energy drinks (such as Red Bull and similar beverages, which contain caffeine, taurine, carbohydrates, etc.) to reduce the depressant effects of alcohol on central nervous system, but no controlled studies have been performed. The main purpose of this study was to verify the effects of alcohol, and alcohol combined with energy drink, on the performance of volunteers in a maximal effort test (cycle ergometer) and also on physiological indicators (oxygen uptake, ventilatory threshold, respiratory exchange rate, heart rate,

Comparison of the effect of smoking and alcohol drinking between oral and pharyngeal cancer.

To compare the separate and combined effects of alcohol drinking and smoking between the 2 sites, we evaluated 274 men with oral cancer, 364 with pharyngeal cancer and 1,254 controls, frequency-matched for age and area of residence, from Italy and Switzerland. Extremely elevated risk increases for oral cancer (odds ratio, OR = 228) and pharyngeal cancer (OR = 100) were found for the highest joint level of drinking (>/=77 drinks/week) and smoking (>/=25 cigarettes/day). Ratios of ORs between oral cancer and pharyngeal cancer vs. controls, obtained by polytomous logistic regression, suggested th…

[Epidemiological study on diet, smoking and alcohol drinking in the relationship to prostatic weight].

BACKGROUND: There have been no epidemiological studies on the relationship between the prostatic weight and blood examination data. This study was carried out to evaluate this relationship. SUBJECTS AND METHODS: Blood examination (beta-carotene, GOT, GPT, total cholesterol, etc.) and measurement of the prostatic weight were performed in 432 subjects aged 50 years or more of a town in Japan, and the possible relationship between blood analysis data and the prostatic weight was evaluated. The prostates were examined by transrectal ultrasonography (TRUS) and classified into the normal prostate a

Problem drinking in the Naval Service: a study of personnel identified as alcohol abusers.

A group of naval personnel referred for alcohol problems was studied for a number of characteristics including sex, service status, age group, Branch, presenting factors, where serving, referral source, marital status, length of service and family history of alcohol abuse. They were compared with a control group of other naval personnel. The alcohol referral group displayed a number of statistically significant differences from the control group. Junior rates, those aged below 20, communications branch personnel and those serving at sea were over represented. The alcohol referrals group were a

Alcoholism, politics, and bureaucracy: the consensus against controlled-drinking therapy in America.

Controlled drinking has long been rejected as a therapeutic goal in traditional American alcoholism treatment. More striking has been the adoption of this position by behaviorists who once fostered controlled-drinking (CD) therapy, such as Peter Nathan, former director of the Rutgers Center of Alcohol Studies. This shift has occurred even while the evidence for rejecting the value of moderation training is unclear. Rather than being justified empirically, this rejection must instead be understood as a strategy behaviorists have used to jettison a politically explosive element in the behavioral

[Dysontogenesis of the brain of the progeny born to mothers drinking alcohol during pregnancy].

A study was made of the development of the brain from 44 embryos (5-12 weeks) and 1 fetus (14 weeks) obtained from mothers who used alcohol during pregnancy. 16 cases made up the control group. In 34 cases out of 45 (75.5%), deviations in the development of the brain were established and distributed into the three degrees of gravity: mild, moderate and severe. 11 variants of brain pathology are described. Deviations in the formation of brain walls and disorders of the development of the cortical lamina occurred most frequently (70.6 and 57.9%, respectively). In alcoholic mothers, brain patholo

Alcohol drinking in the rat after destruction of serotonergic and catecholaminergic neurons in the brain.

Rats were offered solutions of ethyl alcohol that were increased in strength from 3-30% concentration over a 12 day interval. Then, 50 mug 5,6-dihydroxytryptamine (5,6-DHT), 200 mug 6-hydroxydopamine (6-OHDA), or 100 mug 5, 7-dihydroxytryptamine (5,7-DHT) were injected acutely into the lateral cerebral ventricle in a 20 mul volume. The selection of ethyl alcohol was enhanced following the lesioning of serotonergic neurons by 5,6-DHT in terms of preference-aversion curves as well as the gm/kg intakes. Conversely, a similar destruction of dopamine and norepinephrine containing neurons by 6-OHDA

Lifetime Drinking Patterns Shape Awareness of Alcohol-Related Chronic Disease Risks in U.S. Adults: Findings From the National Alcohol Survey.

OBJECTIVE: Understanding the relationship between people's lifetime alcohol use and their awareness of the risk of common chronic diseases from alcohol use is important for informing health messaging efforts. This article aimed to (a) assess the level of awareness that alcohol use increases the risk of cancer, diabetes, and hypertension, and (b) examine the association between lifetime drinking patterns and levels of awareness. METHOD: We used data from the 2019-2020 National Alcohol Survey that included awareness that cancer, hypertension, and diabetes could result from drinking too much alc

It Pays to Ask Twice: Alcohol Consumption Amongst Aboriginal and Torres Strait Islander Peoples Who Self-Reported Not Drinking Alcohol.

INTRODUCTION: It is important that Aboriginal and Torres Strait Islander peoples who experience worries from alcohol use are linked with support. Screening is important to help link people to support. However, individuals may not freely report consumption, especially if they perceive discrimination or drink less than others. Here we report on the use of a 'check question' asking about alcohol consumption on special occasions. METHODS: We analysed data from an urban and remote community in South Australia, collected between July and October 2019. Participants who self-reported not drinking alc

Evaluating sex and line differences in successive negative contrast and ethanol consumption using alcohol preferring and high alcohol drinking rats.

BACKGROUND: The loss of a job or relationship are a couple of examples of unexpected reward loss. Life events, such as these can induce negative emotional reactions (e.g., anxiety and stress), which have been associated with increased alcohol consumption and in turn, an increased risk of developing an alcohol use disorder (AUD). The present study analyzed consummatory successive negative contrast (SNC) for the first time in alcohol preferring (P) and high alcohol drinking (HAD) rats that have been selectively bred to consume high amounts of ethanol. Following reward loss, animals were given fr…

Effects of a Peripherally Restricted Hybrid Inhibitor of CB1 Receptors and iNOS on Alcohol Drinking Behavior and Alcohol-Induced Endotoxemia.

Alcohol consumption is associated with gut dysbiosis, increased intestinal permeability, endotoxemia, and a cascade that leads to persistent systemic inflammation, alcoholic liver disease, and other ailments. Craving for alcohol and its consequences depends, among other things, on the endocannabinoid system. We have analyzed the relative role of central vs. peripheral cannabinoid CB1 receptors (CB1R) using a "two-bottle" as well as a "drinking in the dark" paradigm in mice. The globally acting CB1R antagonist rimonabant and the non-brain penetrant CB1R antagonist JD5037 inhibited voluntary alc

Pharmacological characterization of the 20% alcohol intermittent access model in Sardinian alcohol-preferring rats: a model of binge-like drinking.

BACKGROUND: Binge drinking is defined as a pattern of alcohol drinking that brings blood alcohol levels to 80 mg/dl or above. In this study, we pharmacologically characterized the intermittent access to 20% ethanol (EtOH) model (Wise, Psychopharmacologia 1973;29:203) in Sardinian alcohol-preferring (sP) rats to determine to which of the compounds known to reduce drinking in specific animal models this binge-like drinking was sensitive to. METHODS: Adult male sP rats were divided into 2 groups and allowed to drink either 20% v/v alcohol or water for 24 hours on alternate days (Monday, Wednesda…

The Role of Identity in Positive Alcohol Use Attitudes, Risky Drinking, and Alcohol-Related Problems Among Racially/Ethnically Diverse Adolescents.

PURPOSE: Positive attitudes toward alcohol use, risky drinking, and the likelihood of experiencing alcohol-related problems tend to increase during adolescence. Although prior studies suggest that identity coherence protects against, and identity confusion increases, vulnerability to negative health-related outcomes, less is known about how identity coherence and confusion uniquely predict trajectories of positive alcohol-use attitudes, risky drinking, as well as the onset of alcohol-related problems across adolescence-particularly among racially/ethnically diverse youth. In the present study,

Grouping motivational interviewing for alcohol craving in individuals with alcohol use disorder: the influence of drinking history on its efficacy.

BACKGROUND: Motivational interviewing (MI) is a psychological intervention for patients with alcohol use disorder (AUD). This study examined the role of the history of drinking alcohol in the efficacy of group MI (GMI) on alcohol craving in AUD. The sample (N = 108) consisted of patients with AUD. METHODS: A total of 108 patients with AUD received a 6-week GMI or treat-as-usual (TAU) intervention and were interviewed at baseline, weeks 2, 4, and 6. The severity of alcohol craving was evaluated by the Penn Alcohol Craving Scale (PACS), and the depressive symptoms were assessed by the Hamilton

Chronic Voluntary Alcohol Drinking Causes Anxiety-like Behavior, Thiamine Deficiency, and Brain Damage of Female Crossed High Alcohol Preferring Mice.

The central nervous system is vulnerable to chronic alcohol abuse, and alcohol dependence is a chronically relapsing disorder which causes a variety of physical and mental disorders. Appropriate animal models are important for investigating the underlying cellular and molecular mechanisms. The crossed High Alcohol Preferring mice prefer alcohol to water when given free access. In the present study, we used female cHAP mice as a model of chronic voluntary drinking to evaluate the effects of alcohol on neurobehavioral and neuropathological changes. The female cHAP mice had free-choice access to

Baclofen and naltrexone, but not N-acetylcysteine, affect voluntary alcohol drinking in rats regardless of individual levels of alcohol intake.

In humans, there is profound individual variation in the risk of alcohol use disorder (AUD). Because GABA, opioid and glutamate neurotransmission have been implicated in AUD, functional differences in these neural systems may underlie the individual vulnerability to AUD. We therefore determined the effects of drugs affecting GABA, opioid and glutamatergic neurotransmission on alcohol consumption in rats that differed in baseline alcohol intake. Subgroups of low-, medium- and high-alcohol-drinking rats were selected on the basis of alcohol consumption using an intermittent alcohol access proced
