Methaemoglobinaemia Associated With Mixed Cocaine and Amphetamine Overdose: A Case Report.

Methaemoglobinaemia is a rare disorder characterized by increased levels of methaemoglobin, a form of haemoglobin with oxidized iron that cannot efficiently bind oxygen. This leads to inadequate oxygen delivery to tissues with various clinical manifestations from asymptomatic to severe persistent hypoxia, CNS symptoms, and cardiovascular collapse. Acquired methaemoglobinaemia is typically a sudden condition, often resulting from poisoning by specific drugs and compounds, which can potentially have fatal consequences. We present a case of a patient who came with severe methaemoglobinaemia due t

GPR160 is not a receptor of anorexigenic cocaine- and amphetamine-regulated transcript peptide.

Cocaine- and amphetamine-regulated transcript peptide (CARTp) is an anorexigenic neuropeptide whose receptor is undisclosed. Previously, we reported the specific binding of CART(61-102) to pheochromocytoma PC12 cells, where CART(61-102) affinity and the number of binding sites per cell corresponded to ligand-receptor binding. Recently, Yosten et al. designated orphan GPR160 as the CARTp receptor, because the GPR160 antibody abolished neuropathic pain and anorexigenic effects induced by CART(55-102) and exogenous CART(55-102) coimmunoprecipitated with GPR160 in KATOIII cells. As no direct evide

Past, present and future of cocaine- and amphetamine-regulated transcript peptide.

The existence of the peptide encoded by the cocaine- and amphetamine-regulated transcript (Cartpt) has been recognized since 1981, but it was not until 1995, that the gene encoding CART peptide (CART) was identified. With the availability of the predicted protein sequence of CART investigators were able to identify sites of peptide localization, which then led to numerous approaches attempting to clarify CART's multiple pharmacologic effects and even provide evidence of potential physiologic relevance. Although not without controversy, a picture emerged of the importance of CART in ingestive b

Use of Mixed Amphetamine Salts in a Patient with Depersonalization/Derealization Disorder.

Depersonalization and derealization symptoms are common and often transient. Recurrent, persistent symptoms can result in a diagnosis of depersonalization/derealization disorder (DDD). This is a diagnosis with little evidence available regarding effective interventions, and there are currently no pharmacological treatments for DDD approved by the United States Food and Drug Administration (FDA). Here, we reported a case of an adult female whose presentation was consistent with DDD. Her DDD symptoms notably reduced after treatment with mixed amphetamine salts. We also reviewed the limited resea

Statistical comparison of mass spectra for identification of amphetamine-type stimulants.

A method for the statistical comparison of mass spectral data is demonstrated for applications in controlled substance analysis. The method uses an unequal variance t-test at each mass-to-charge ratio in the scan range to determine if two spectra are statistically associated or discriminated. If the two spectra are associated, a random-match probability is calculated to estimate the likelihood that the mass spectral fragmentation pattern in question occurs by random chance alone. If the two spectra are discriminated, the fragment ions responsible for the discrimination are determined. In this

Amphetamine sensitization alters reward processing in the human striatum and amygdala.

Dysregulation of mesolimbic dopamine transmission is implicated in a number of psychiatric illnesses characterised by disruption of reward processing and goal-directed behaviour, including schizophrenia, drug addiction and impulse control disorders associated with chronic use of dopamine agonists. Amphetamine sensitization (AS) has been proposed to model the development of this aberrant dopamine signalling and the subsequent dysregulation of incentive motivational processes. However, in humans the effects of AS on the dopamine-sensitive neural circuitry associated with reward processing remain

Amphetamines modulate prefrontal γ oscillations during attention processing.

Amphetamine-based medications robustly suppress symptoms of attention-deficit/hyperactivity disorder (ADHD), but their exact mechanisms remain poorly understood. Recent hemodynamic imaging studies have suggested that amphetamines may modulate the prefrontal and anterior cingulate brain regions, although few studies have been published and the results have not been entirely consistent. Meanwhile, several electrophysiological studies have shown that abnormal fast oscillations (in the γ range) may be closely linked to inattention and other cardinal symptoms of ADHD. In this study, we utilized mag

The cocaine- and amphetamine-regulated transcript (CART) immunoreactivity in the amygdala of the pig.

The distribution and morphology of neurons containing cocaine- and amphetamine-regulated transcript (CART) was investigated in the pig amygdala. CART- immunoreactive (CART-IR) cell bodies were rarely observed in the pig amygdala and most often they were present in the posterior (small-celled) parts of the basolateral and basomedial nuclei. In all other subdivisions only a small number of randomly scattered pericarya were present. In every region studied the CART-IR neurons formed a heterogeneous population consisting mostly of small, rounded or slightly elongated cell bodies, with a few poorly

Cardiac safety of methylphenidate versus amphetamine salts in the treatment of ADHD.

OBJECTIVES: Safety concerns about central nervous system stimulants for the treatment of attention-deficit/hyperactivity disorder (ADHD) include adverse cardiac effects. This study aimed to compare the risk for cardiac events in users of methylphenidate and amphetamine salts. METHODS: A retrospective cohort design using claims data from the Florida Medicaid fee-for-service program representing a total of 2131953 children and adolescents was used. The analysis included all beneficiaries who were between 3 and 20 years of age, enrolled between July 1994 and June 2004, had at least 1 physician d

Potential adverse effects of amphetamine treatment on brain and behavior: a review.

Amphetamine stimulants have been used medically since early in the twentieth century, but they have a high abuse potential and can be neurotoxic. Although they have long been used effectively to treat attention deficit hyperactivity disorder (ADHD) in children and adolescents, amphetamines are now being prescribed increasingly as maintenance therapy for ADHD and narcolepsy in adults, considerably extending the period of potential exposure. Effects of prolonged stimulant treatment have not been fully explored, and understanding such effects is a research priority. Because the pharmacokinetics o

Cocaine- and amphetamine-regulated transcript (CART) is expressed in the ovine pancreas.

Indirect immunohistochemistry was applied to demonstrate the presence of cocaine- and amphetamine-regulated transcript (CART) peptide expression in the pancreas of the sheep. Using double immunocytochemical staining, the co-incidence of vasoactive intestinal polypeptide (VIP), neuropeptide Y (NPY) or substance P (SP) in CART-immunoreactive (IR) nerve fibers and intrapancreatic neurons was analyzed. Immunoreactivity to CART was detected in endocrine cells predominantly localized at the islet periphery. The exocrine pancreas and blood vessels were intensively innervated by CART-IR nerve fibers.

Bimodal effect of amphetamine on motor behaviors in C57BL/6 mice.

Amphetamine-induced motor behaviors, i.e., locomotor and stereotypic activities, were simultaneously characterized in C57BL/6 mice, a strain commonly used for genetic studies. Our findings show relatively high levels of focused activities in drug-naive C57BL/6 mice, confirming the lively nature of this mouse strain. Acute amphetamine induced a dose-dependent, bimodal response: locomotion predominated at lower doses of amphetamine and was gradually displaced by stereotypic behavior as dose and time increased. The sum total of both behavioral activities increased with amphetamine dose, supportin

Sensitization to amphetamine, but not phencyclidine, disrupts prepulse inhibition and latent inhibition.

RATIONALE: Schizophrenia has been linked to dysregulation of dopamine and glutamate transmitter systems. Attempts to model aspects of schizophrenia in animals have made use of treatments that primarily affect dopaminergic (e.g., amphetamine, Amp) and glutamatergic (e.g., phencyclidine, PCP) function. In addition to exerting short-term acute effects, these agents also induce long-term effects, as seen, for example, in neurochemical and behavioural sensitization. OBJECTIVES: The goal of this work was to compare Amp- and PCP-sensitized states on two measures of information processing that are im

An activity indicator of acute withdrawal depends on amphetamine dose in rats.

A moderate dose of amphetamine (AMPH) produces hypoactivity around 20 h post-administration. This hypoactivity may be an indicator of an acute withdrawal state. The purpose was to see how AMPH doses affected the expression of this hypoactivity and, by inference, AMPH-induced acute withdrawal. Rats were housed in individual open fields, with free access to food and water. Light-dark cycles were scheduled such that drug-elicited patterns could be readily detected. Animals first received a series of eight control treatments, and then a series of 10 experimental treatments spaced at 33-h intervals

Risperidone attenuates the discriminative-stimulus effects of d-amphetamine in humans.

Studies conducted with nonhuman laboratory animals have consistently shown that atypical antipsychotics that are mixed dopamine and serotonin antagonists attenuate the discriminative-stimulus effects of amphetamine. In the present experiment, eight healthy humans learned to discriminate 15 mg of oral d-amphetamine. After acquiring the discrimination (i.e., > or = 80% correct responding on four consecutive days), the effects of a range of doses of d-amphetamine (0, 2.5, 5, 10, and 15 mg), alone and after pretreatment with risperidone (0 and 1 mg), a D2 dopamine and 5-hydroxytryptamine (5-HT)2 s

Motor effects of amphetamine in rats pretreated with either dizocilpine or phencyclidine.

The aim of the present study was to examine motor effects of amphetamine (AMPH) in rats pretreated with either dizocilpine (MK-801) or phencyclidine (PCP), and to estimate possible differences in these effects. Our results showed that AMPH increases the duration of motor effects of PCP, while it does not change motor effects of MK-801. These findings may reflect different mechanisms of action of MK-801 and PCP, as well as selective influence of AMPH on metabolism of these drugs.

Altered response to intravenous thiopental and succinylcholine in acute amphetamine abuse.

Substance abuse has become increasingly prevalent: illegal drugs have profound and varied physiologic effects which create a large potential for anesthetic problems and complications (1). Amphetamine is a strong sympathomimetic and may therefore influence the course of anesthesia. We report the case of a patient with acute amphetamine abuse presenting difficulties during anesthesia.

Requirement of endogenous basic fibroblast growth factor for sensitization to amphetamine.

Repeated exposure to amphetamine produces long-lasting increases in sensitivity to its effects. We reported previously that repeated amphetamine treatment results in increased astrocytic expression of basic fibroblast growth factor (bFGF) in the ventral tegmental area (VTA) and substantia nigra compacta (SNc) and that this effect is prevented by coadministration of a nonspecific glutamate receptor antagonist. Here we show that the development of sensitization to amphetamine is prevented when amphetamine injections are preceded by infusions of a neutralizing antibody to bFGF into the VTA. In ad

Susceptibility to amphetamine-induced locomotor sensitization is modulated by environmental stimuli.

We have previously reported that intravenous (i.v.) administrations of 0.5-1.0 mg/kg of amphetamine in the absence of any environmental stimuli predictive of drug administration failed to induce psychomotor sensitization whereas the same drug did produce robust sensitization when given in association with environmental novelty. These results were obtained by studying rotational behavior in animals with a unilateral 6-OHDA lesion of the mesostriatal dopamine system. The purpose of this study was to determine if environmental novelty has a similar effect on sensitization to the locomotor activat
