Advances in vaccine development through the controlled human infection models for hookworm and schistosomiasis.

Controlled human infection models can play an important role in vaccine development, particularly for neglected tropical diseases such as helminth infections. Currently, controlled infection models have been established for schistosomiasis and hookworm. This review highlights the developments in the controlled human schistosomiasis infection model (CHI-S) and the controlled human hookworm infection model (CHHI) and their contributions to vaccine development. In general, both models are considered safe and well-tolerated. Measures to decrease risk of potential adverse events were taken when dev

Widespread Soil-Transmitted Parasitic Contamination in Raw Vegetables at Fresh Markets in the Inner Zone of Bangkok: Findings from a Cross-Sectional Study.

Bangkok, a globally popular travel destination, hosts numerous fresh markets and street food vendors catering to locals and visitors. Although raw vegetables are integral to Thai cuisine, their consumption may pose risks of soil-transmitted parasitic infections. This cross-sectional study assessed the prevalence of parasitic contamination in raw vegetables from Bangkok's inner zones, focusing on five districts: Huai Khwang, Khlong Toei, Pathum Wan, Chatuchak, and Phra Nakhon. A total of 200 samples across eight vegetable types were randomly collected from these areas and prepared for analysis

Modeling the effect of different drugs and treatment regimen for hookworm on cure and egg reduction rates taking into account diagnostic error.

BACKGROUND: Hookworm infections, caused by Ancylostoma duodenale and Necator americanus, are of considerable public health importance. The World Health Organization recommends preventive chemotherapy as the key strategy for morbidity control. Meta-analyses have been conducted to estimate treatment efficacy of available drugs and drug combinations. However, in most studies, the relation between the diagnostic error and infection intensity have not been considered, resulting in an overestimation of cure rates (CRs). METHODOLOGY: A Bayesian model was developed to compare the 'true' CR and egg re

Multiple drug resistance in hookworms infecting greyhound dogs in the USA.

Ancylostoma caninum is the most prevalent nematode parasite of dogs. We confirmed multiple-drug resistance (MDR) in several A. caninum isolates to all anthelmintic drug classes approved for the treatment of hookworms in dogs in the USA. Cases of MDR hookworms appear to be highly overrepresented in greyhounds. The aims of this study were to evaluate the drug-resistant phenotypes and genotypes of the A. caninum infecting greyhounds. Fecal samples from greyhounds of the USA were acquired from two greyhound adoption kennels, one active greyhound racing kennel, and three veterinary practices. Fecal

Environmental characteristics around the household and their association with hookworm infection in rural communities from Bahir Dar, Amhara Region, Ethiopia.

Soil-Transmitted Helminths (STH) are highly prevalent Neglected Tropical Disease in Ethiopia, an estimated 26 million are infected. Geographic Information Systems and Remote Sensing (RS) technologies assist data mapping and analysis, and the prediction of the spatial distribution of infection in relation to environmental variables. The influence of socioeconomic, environmental and soil characteristics on hookworm infection at the individual and household level is explored in order to identify spatial patterns of infection in rural villages from Zenzelema (Amhara region). Inhabitants greater th

Immunopathology and modulation induced by hookworms: From understanding to intervention.

Hookworm infection is considered the most prevalent human soil-transmitted helminth infection affecting approximately 500 million people and accounting for 3.2 million disability-adjusted life years lost annually. As with many other neglected tropical diseases, no international surveillance mechanisms that show accurate data on the prevalence of hookworm infection are in place, thus hindering strategies to control parasite transmission. In this review, we unravel the current knowledge in immunopathology and immunoregulation of hookworm infection and present discoveries in drug therapies based

Soil contamination of a public park by human and canine mastadenovirus, as well as hookworms and Toxocara spp eggs.

Public parks are leisure environments widely used by both, adults and children, often accompained by their pets. Soil contamination of these environments by enteric viruses and intestinal parasites occurs through these animals feces. The aim of this work was to detect Carnivore protoparvovirus 1 (CPV-1) and different species of Mastadenovirus in soils samples from a park located in a medium-sized city in Brazil and evaluate the presence of helminth eggs and larvae in 18 points of a public park soil samples, as well as feces found on this site during six months. Parasitological analyzes were co

[Investigation of soil - transmitted nematode infections in Xiding Township, Menghai County, Yunnan Province].

OBJECTIVE: To understand the status of soil-transmitted nematode infections in Xiding Township, Menghai County, Yunnan Province, so as to provide the reference for formulating the strategy of soil-transmitted nematodosis control. METHODS: Soil-transmitted nematode eggs in feces were detected by the Kato-Katz method, and the eggs of Enterobius vermicularis were detected by the cellophane tape method in children. The soil samples were collected from vegetable, fruit and other crop fields of 15 residents randomly to detect hookworm. RESULTS: The stool samples from 1 002 residents were examined

Geohelminths: public health significance.

The worldwide prevalence of geohelminths and their unique place in evolutionary biology have attracted research focus. These major soil-transmitted intestinal nematodes that cause human diseases are the nematode roundworm (Ascaris lumbricoides), the whipworm (Trichuris trichiura) and the two hookworms (Ancylostoma duodenale and Necator americanus), often collectively referred as geohelminths. Studies of geohelminthiasis in poorly nourished children in developing regions report that geohelminths contribute to stunted growth and cognitive impairment. Insights into immunology have shed light on t

Quantitative interpretation of coprocultures in a population infected with Oesophagostomum bifurcum.

Coproculture is used in northern Togo and Ghana in the research on Oesophagostomum bifurcum, a common parasite of man in these regions. Prior to a follow-up study to investigate patterns of reinfection in Oesophagostomum and hookworm, it was attempted to evaluate the relevance of counting larvae for the assessment of the intensity of infection at the population level. Of 102 samples, one egg count (Kato-smear) and three coprocultures were carried out. Frequency distributions of counts of larvae of Oesophagostomum and of hookworm isolated in three coprocultures, showed log-normality. There was

Detection of hookworm and hookworm-like larvae in human fecocultures in Suriname.

Estimation of the prevalence of hookworm infection is usually based on finding eggs of the parasite in the stool. Eggs of the different hookworm species are difficult to distinguish and can be confused with those of hookworm-like species. Proper identification can be made after hatching the eggs and cultivating larvae to the infective stage. We have used the Harada-Mori method to study the stools of Bushnegroes in Suriname and have identified the emerged larvae. Of the 804 individuals examined, 53.6% had positive fecal cultures. The identification of emerged larvae revealed that 75% of the pos

Use of the enzyme-linked immunosorbent assay in the diagnosis of Dictyocaulus filaria infection.

The enzyme-linked immunosorbent assay (ELISA) was used to detect and quantify antibodies to whole worm extract antigens of Dictyocaulus filaria. The results provided evidence that the test was highly sensitive and could be used as a tool for epidemiological studies. No cross reactions were observed with sera from lambs infected with the hookworm Gaigeria pachyscelis.
