Among Oriental anopheline mosquitoes (Diptera: Culicidae), several major vectors of forest malaria belong to the group of Anopheles (Cellia) leucosphyrus Dönitz. We have morphologically examined representative material (> 8000 specimens from seven countries) for taxonomic revision of the Leucosphyrus Group. Six new species are here described from adult, pupal and larval stages (with illustrations of immature stages) and formally named as follows: An. latens n. sp. (= An. leucosphyrus species A of Baimai et al., 1988b), An. cracens n. sp., An. scanloni n. sp., An. baimaii n. sp. (formerly An. d…

Anopheles (Cellia) sundaicus (Rodenwaldt) is an important malaria vector in the Andaman and Nicobar islands of India where it breeds in freshwater as well as in brackish water. To establish the molecular identity of An. sundaicus on these islands we analyzed samples from four geographically isolated areas-Teressa, Nancowry, Car Nicobar and Katchal islands. PCR-amplification and nucleotide sequence analysis were performed for internal transcribed spacer 2 (ITS2) and domain-3 (D3) of 28S rRNA. The ITS2 region of An. sundaicus from all four islands was identical but different from An. sundaicus A…

We measured the seasonal abundance and bloodfeeding behavior of species A and C of the mosquito Anopheles minimus Theobald 1901 in an endemic malarious area of western Thailand. An. minimus s.l. is a major vector of human malarial and filarial parasites in Southeast Asia. Mosquitos were collected once a month for one year using four collection methods: human-baiting indoors, human-baiting outdoors, human-baiting in the forest, and cow-baiting. We found that both species A and C tend to feed from cows rather than humans; we did not find any preference for indoor, outdoor or forest-biting in eit…

The population genetic structure of the Anopheles gambiae in western Kenya was studied using length variation at five microsatellite loci and sequence variation in a 648-nt mtDNA fragment. Mosquitoes were collected from houses in villages spanning up to 50 km distance. The following questions were answered. (i) Are mosquitoes in a house more related genetically to each other than mosquitoes between houses? (ii) What degree of genetic differentiation occurs on these geographical scales? (iii) How consistent are the results obtained with both types of genetic markers? At the house level, no diff…

A study was made on the distribution of anophelines in Suriname with special emphasis on the principal malaria vector Anopheles darlingi and on the occurrence of other possible vector species. Peridomestic human bait collections of adult mosquitoes and collections of larvae were made in many localities with a recent history of malaria transmission. Stable populations of An. darlingi were only found in the interior, south of the limit of tidal influence, due to year-round availability of breeding habitats in quiet partly sunlit places in flooded forest areas and along river banks. In the area w…

A nonradiometric method has been developed for distinguishing between the sibling species Anopheles gambiae Giles and An. arabiensis Patton, two important Afrotropical vectors of malaria. DNA fragments of species diagnostic length are amplified by polymerase chain reaction (PCR) from a small amount of unknown DNA and three different PCR primers. All three PCR primers are based on ribosomal DNA (rDNA) sequences. A universal plus-strand primer (A0) is derived from a conserved region at the 3' end of the 28S rDNA coding region. Two species-specific minus-strand primers (Aa0.5 and Ag1.3) are deriv…

Malaria vector mosquitoes belonging to the Anopheles gambiae complex were studied in four hamlets in The Gambia. All inhabitants were given bednets treated either with a placebo (milk) in two hamlets or with the pyrethroid insecticide permethrin (500 mg/m2) in two other hamlets. Malaria transmission occurred mainly during a few weeks of the rainy season, in September and October 1987. The indoor resting densities of mosquitoes in permethrin-treated hamlets were reduced, and we estimated over 90% reduction in biting on man by An. gambiae Giles sensu stricto in these hamlets. No mosquitoes were …

O'nyong-nyong virus (ONNV) is a mosquito-borne alphavirus responsible for large-scale epidemics in sub-Saharan Africa. As the closest evolutionary relative of Chikungunya virus (CHIKV), ONNV shares substantial genetic similarity and overlapping clinical manifestations with CHIKV. Mechanistic understanding of ONNV infection has therefore largely been extrapolated from CHIKV rather than directly established. However, ONNV exhibits distinct biological features, including predominant transmission by Anopheles mosquitoes and a clinical presentation characterized by prominent lymphadenopathy with li…

BACKGROUND OBJECTIVES: India has made significant progress in combating malaria and reducing malaria-related mortality and morbidity. Yet, local transmission and malaria hotspots are important hurdles as India looks to achieve malaria elimination by 2030. Gadchiroli district is a highly malaria-endemic area in Maharashtra state of India. It accounts for more than 50% of the reported malaria cases in the state. Anopheles culicifacies and An. fluviatilis are the primary malaria vectors in the area. However, the status of vector sibling species is unknown in the district. In this study, we aimed …

Reliable estimates of the age structure of malaria mosquitoes would aid evaluation of the efficacy of vector control and inform models of malaria transmission. We have previously shown that rapid evaporative ionisation mass spectrometry (REIMS) could determine the age and species of laboratory reared mosquitoes and culicine mosquitoes from a river estuary in the United Kingdom. Here we investigated the robustness of this methodology by introducing additional environmental, genetic and physiological diversity in experiments using laboratory reared and field collected Anopheles gambiae mosquitoe…

BACKGROUND: The malaria control programmes are facing a challenge due to a rapidly increasing resistance to currently available synthetic in … idal and adulticidal activity against late 3rd to early 4th instar larvae and 3 to 5 days adults of Anopheles gambiae, respectively. For both assays, the plants were incubated with the parasites for 48 h at concentrations of 25, 50, 100, 200, and 400 ppm. Percent mean mortality rate was compared using one-way ANOVA, and LC50 and LC90 values were calculated …

BACKGROUND: Mosquitoes are important drivers of infectious diseases transmission, with Anopheles mosquitoes being responsible of malaria transmission. In Cambodia, where malaria is prevalent in forested regions, understanding the ecology of these vectors is crucial. This study aimed to investigate the abundance, distribution, seasonal patterns, biting behaviour of Anopheles mosquitoes, and prevalence of Plasmodium, in Mondulkiri province, Northeastern Cambodia.
METHODS: Conducted in 9 sites, seven in forested and two in neighbouring villages, over one year, the collection of Anopheles mosquit…

BACKGROUND: In Colombia, mining is one of the main economic activities in four eco-epidemiological areas that contributes approximately 80% of annual malaria cases. Mining activity generates changes in the environment and creates newly available breeding sites to be colonized by malaria vector mosquitoes, which increases the risk of malaria transmission. The study aimed to identify the presence of Anopheles species and their role in malaria transmission in five malaria-endemic localities with gold mining extraction in El Bagre, Antioquia, Colombia.
METHODS: A cross-sectional study was conduct…

BACKGROUND: The mosquito family Culicidae diverged into the subfamilies Anophelinae and Culicinae approximately 179 million years ago. Most female mosquitoes are anautogenous and must blood-feed on a vertebrate to produce eggs. Regulation of egg-producing gonotrophic cycles is best understood in the culicine Aedes aegypti. Anopheline mosquitoes encode all of the hormones that regulate gonotrophic cycles in Ae. aegypti, but the processes regulating egg formation may not be fully similar. In this study, we conducted experiments that compared egg formation in Anopheles stephensi to prior findings…

Anopheles darlingi is the primary malaria vector in Central and South America and is responsible for most malaria transmission in the Amazon region. In this study, we perform whole-genome analysis of individual An. darlingi mosquitoes to explore genomic diversity, signatures of selection, and insecticide resistance markers. We analysed wild-caught (n = 20) and colony-maintained (n = 8) mosquitoes from the State of Rondônia, Brazil. In total, 1.54 million high-quality single-nucleotide polymorphisms (SNPs) were identified. Population genomic analysis revealed genetic differentiation between the…

A major insecticide resistance mechanism in insect pests is knock-down resistance (kdr) caused by mutations in the voltage-gated sodium channel (Vgsc) gene. Despite being common in most malaria Anopheles vector species, kdr mutations have never been observed in Anopheles funestus, the principal malaria vector in Eastern and Southern Africa, with resistance mainly being conferred by detoxification enzymes. In a parallel study, we monitored 10 populations of An. funestus in Tanzania for insecticide resistance unexpectedly identified resistance to a banned insecticide, DDT, in the Morogoro region…

BACKGROUND: The high burden of malaria in Africa is largely due to the presence of competent and adapted Anopheles vector species. With invasive Anopheles stephensi implicated in malaria outbreaks in Africa, understanding the genomic basis of vector-parasite compatibility is essential for assessing the risk of future outbreaks due to this mosquito. Vector compatibility with P. falciparum arises from ancient coevolution and involves genes like Pfs47 in P. falciparum and P47Rec in Anopheles. Questions remain about whether sub-continental vector variation is a selective pressure on current Plasmo…

Females from many mosquito species feed on blood to acquire nutrients for egg development. The oogenetic cycle has been characterized in the … wever limited in this and other mosquito species. Here, we demonstrate that in the malaria mosquito Anopheles gambiae, Lp and Vg are reciprocally regulated in a timely manner to optimize egg development and ensure fertility. Defective lipid transport via Lp knockdown triggers abortive ovarian follicle development, leading to misregulation of Vg and aberrant …
