In vitro assessment of the activity of anthelmintic compounds on adults of Onchocerca volvulus.

The viability of adult Onchocerca volvulus and the effect of 12 known anthelmintic compounds on the parasites have been evaluated in an in vitro culture system. Three different parameters, a colorimetric assay, using NADH-dependent reduction of a tetrazolium salt to dark blue formazan by living adult worms, motility indices of male worms and lactate excretion of female worms were used to determine worm viability. The experiments showed that over a short term period of six days the viability of the worms did not decline significantly. The use of males isolated by dissection of whole nodules for

Resistance of dogs to reinfection with Ancylostoma ceylanicum following anthelmintic therapy.

A model of human hookworm infection has been developed which shows that dogs with chronic hookworm infection are considerably resistant to reinfection one month after the termination of the primary infection with anthelmintics. Challenge and control dogs were infected with 1,800 larvae and the infection was followed for six weeks. When compared with control dogs, faecal egg excretion and intestinal adult worm burdens in challenge dogs were reduced by 85% and 77%, respectively. Infection had no significant effect on haemoglobin concentrations, total white cell counts, platelet levels or spontan…

Evaluation of selected anthelmintic compounds for activity against Opisthorchis viverrini.

In vitro and in vivo experiments were employed in the screening of potential anthelmintic agents against Opisthorchis viverrini infection in hamsters. A few selected groups of compounds tested included those that are commercially available as well as those that are still being tested by various pharmaceutical firms. The compounds tested in the present study were praziquantel, amoscanate, albendazole, flubendazole, metrifonate, metronidazole and benzodiazepine derivatives. Results from both in vitro and in vivo experiments showed that at the dosages employed, praziquantel was the only one that

Albendazole, an effective single dose, broad spectrum anthelmintic drug.

Albendazole, a new anthelmintic drug was evaluated in Malaysia in 91 patients, with single or mixed infections of Ascaris, Trichuris, and hookworm. Albendazole was administered as a single dose of 400 mg, 600 mg, or 800 mg. The cure rate for Ascaris at all three doses was 100% at days 14 and 21 post-treatment; for hookworm it was 98.8%, 100% and 98%, respectively, at day 14 and 68.8%, 100% and 84%, respectively, at day 21; for Trichuris it was 31.2%, 57.1% and 42.3%, respectively, at day 14 and 27.3%, 60.9% and 48.0%, respectively, at day 21. The egg reduction rate at day 21 was 100% at all th

Comparative trial on the therapeutic effectiveness of the new anthelmintic drug: ciclobendazole.

In a double-blind study in Cameroon the vermicidal effect of Ciclobendazole, a new Benzimidazole derivative, was evaluated and compared to Mebendazole. Ciclobendazole and Mebendazole were equally effective in the treatment of Ascaris and Hookworm infestations. An increase in the dosis of Ciclobendazole from 600 mg to 1200 mg did not lead to an improvement in the effectiveness. When treating trichuriasis significantly better results were achieved with Mebendazole (p = 0.01). Both drugs were tolerated equally well. Side effects, such as vomiting and diarrhoea, only occurred in a small percentage

Anthelmintic activity of pyrantel tartrate against Ascaridia galli in fowls.

At a dose of 15 mg/kg body weight, pyrantel tartrate was 18-51, 99-63, 100 and 100 per cent effective in chickens treated at 10, 20, 30 and 40 days respectively after infection with Ascaridia galli. Similarly, 25 mg/kg was 14-44, 100, 100 and 99-63 per cent effective.

Evaluation of Antimicrobial and Anthelmintic Potential of Some Mushrooms from Pakistan.

The present study is an attempt to evaluate the antimicrobial and anthelmintic potential of eight mushrooms collected from Pakistan. Eight mushrooms, namely Amanita ahmadii, Amanita cinnamomescens, Amanita flavipes, Amanita griseoannulata, Amanita pactosquamata, Amanita sclerisquobrunnea, Amanita citrinoannulatoides, and Limacella subtropicana, were used in the present study. The dried mushroom samples were ground to a fine powder form using pestle and mortar. Crude extracts of dried mushrooms were prepared by the process of maceration in non-polar (petroleum ether and chloroform) and polar (e

Evaluation of the Efficacy of Anthelmintic Drugs Against Trichinella spiralis Larvae.

Background: Albendazole, mebendazole, and ivermectin are effective against adult Trichinella spiralis but show limited efficacy against encapsulated muscle stage larvae. This limitation highlights the need for improved experimental approaches to evaluate anthelmintic activity at this stage and to identify alternative therapeutic candidates. Methods: Seven antiparasitic drugs, albendazole (ABZ), miltefosine (MLT), ivermectin (IVM), tribendimidine (TBD), praziquantel (PZQ), artesunate (ART), and mefloquine (MEQ), were evaluated for in vitro activity against T. spiralis muscle larvae. Larval viab

Early-stage anthelmintic discovery: Integrating phenotype, target and mechanism.

Parasitic helminth infections impose major burdens on human and animal health, and increasing anthelmintic resistance reinforces the need for new drugs. Early-stage discovery remains constrained by the scarcity of novel, potent, selective and chemically tractable starting points, the limited predictive value of some experimental models, mechanistic uncertainty and insufficient capacity to sustain promising series through medicinal chemistry, safety and exposure studies. Discovery can begin through complementary routes, including target-agnostic whole-organism screening, screening of known-phar

Anthelmintic stewardship: An essential step towards sustainable dairy farming.

Helminth infections in grazing dairy cattle can lead to asymptomatic infections, subclinical- or clinical disease. Whether the course of infection leads to disease depends on various factors, leading to a broad variety of prevalence rates and severity of disease between farms, between groups of cattle in a farm and between individual animals. Production losses related to helminth infections, the difficulty in diagnosing helminth disease and the wide availability and high efficacy of anthelmintics have led to overreliance on these products. Combined with management and other factors, this resul…

Evaluating orange oil and lespedeza hay as an alternative anthelmintic for goats.

Identifying alternative practices to control gastrointestinal parasites and overcome resistance to chemical anthelmintics is critical to maintain the productive and economic viability of the small ruminant sector. Here, we evaluated in vivo potential effects of orange oil and sericea lespedeza (Lespedeza cuneata [Dumont] G. Don) supplementation on suppressing gastrointestinal nematodes (GIN) and improving performance of goats. Twenty-four bucks were used in a randomized complete block design and were assigned to12 pens after deworming. Experimental duration was 6 wk (1 wk adaptation and 5 wk o…

Anthelmintic Resistance in Livestock Farming: Challenges and Perceptions of Farmers and Veterinarians.

Anthelmintic resistance in livestock is a growing concern worldwide, with significant implications for animal health and agricultural productivity. This study explores the perceptions of veterinarians and farmers in Bosnia and Herzegovina regarding the factors contributing to anthelmintic resistance in Haemonchus contortus nematodes. Data were collected through structured questionnaires completed by 106 veterinarians and 188 farmers in 2022 and 2023. The analysis focused on self-reported therapeutic practices, farm management and environmental variables. Logistic regression, including Firth's
