Bacillus velezensis Enhances Rice Resistance to Brown Spot by Integrating Antifungal and Growth Promotion Functions.

Brown spot, caused by the seedborne fungus Bipolaris oryzae, remains a major constraint in rice production. Here, we used in vitro and in vivo assays to evaluate the biocontrol potential of three Bacillus strains (Ba. cereus OQ725688.1, Ba. velezensis OP938696.1, and Ba. subtilis OP937353.1) against Bi. oryzae in two rice cultivars ("Rubelita" and "Predileta"). Ba. cereus showed the highest in vitro mycelial inhibition (≈95%), whereas Ba. velezensis was the most effective under greenhouse conditions, reducing disease severity by up to 60% and increasing seedling vigor by 51% compared with infe

Exploring the lichenization continuum through the marine tripartite symbiosis of Collemopsidium pelvetiae.

PREMISE: Symbioses between lichen-forming fungi and brown algae (phaeophytes) are extremely rare. We investigated the interactions between the marine fungus Collemopsidium pelvetiae and its two photosynthetic partners, the brown alga Pelvetia canaliculata and a cyanobacterial symbiont to address questions on symbiosis biology, lichenization, and the diversity of fungal-photosynthetic associations in marine environments. METHODS: We combined light microscopy, fluorescence microscope in structural illumination microscopy mode (SIM), and transmission electron microscopy (TEM) to characterize tha

Botryolides F and G from the Fungus Bartalinia robillardoides with Anthelmintic Activity.

The chemical investigation of the fungusBartalinia robillardoides CBS 122686 led to the isolation of two new decarestrictines bearing an unusual three-carbon unit appended to the polyketide moiety and named Botryolides F and G. The compounds were structurally characterized, and their absolute configurations were determined through a combination of spectroscopic methods, single-crystal X-ray diffraction, and electronic circular dichroism (ECD) analyses. Botryolide G was assessed for anthelmintic activity and exhibited promising efficacy at a concentration of 100 μg/mL, with no significant cytot

First occurrence of Corynespora cassiicola infecting chia plant in Bangladesh and sensitivity of the pathogen to selected fungicides.

Chia (Salvia hispanica L.) is an emerging crop in Bangladesh valued for its medicinal properties and economic significance. In March 2024, target spot-like symptoms were observed in an experimental chia field (24.75° N, 90.50° E) at Bangladesh Agricultural University in Mymensingh, Bangladesh, with disease incidence ranged from 23% to 47% across approximately 0.25 ha. Initially appearing as brick-red spots, these symptoms developed into target-shaped concentric rings, affecting leaves, stems, and inflorescences. A total of 24 fungal isolates were recovered from infected tissue; two representat

Hybrid genome assembly of the cannabis powdery mildew agent Golovinomyces ambrosiae uncovers important resources for deciphering virulence factors.

BACKGROUND: Cannabis powdery mildew, caused by the fungal pathogen Golovinomyces ambrosiae, poses a significant threat to licensed producers as its presence on marketable products can compromise product innocuity. While there is a focus in research for resistance genes within the Cannabis sativa germplasm, there is a lack of genetic information regarding the infecting agent, preventing validation of their effectiveness against different populations as the plant-pathogen interaction most likely follows a gene-for-gene relationship. In this paper, we assembled the first G. ambrosiae genome, prov

A multifunctional Streptomyces murinus bioinoculant integrates immune priming and microbiome restructuring for sustainable control of Sclerotinia minor in Brassica rapa.

BACKGROUND: Harnessing beneficial soil microbes provides an eco-friendly alternative to pesticides for sustainable crop protection. Members of the genus Streptomyces combine broad antagonistic activity with plant immune priming and growth promotion, yet their ecological roles and field reliability remain insufficiently understood. We investigated the soil actinobacterium Streptomyces murinus JS029 in the Brassica rapa-Sclerotinia minor pathosystem to elucidate its mechanisms of pathogen suppression, plant immune activation and rhizosphere microbiome modulation, and to develop a field-deployabl

Joint Modeling of Wheat Head Blast and Water-Deficit Risks for Climate-Informed Sowing Calendar Decisions for Rainfed Wheat in the Brazilian Cerrado.

Wheat blast, caused by the Pyricularia oryzae Triticum lineage, is a major constraint to wheat expansion in the Brazilian Cerrado. Delayed sowing is often recommended for disease avoidance; however, late sowing can increase the risk of water deficit. This study quantifies the joint risks from head blast and water deficit in the Brazilian Cerrado to inform sowing-date recommendations. Using daily weather data from a 62-year period (1961 to 2023), crop phenology was simulated based on an accumulated growing degree day approach. Heading and maturity dates were estimated for six sowing dates defin

First molecular confirmation of Lasiodiplodia theobromae causing grapevine trunk disease in southern Egypt.

Grapevine trunk diseases constitute a significant phytopathological concern in Egyptian viticulture, with ongoing debates regarding their origin and transmission dynamics. These complexities are attributed to the heterogeneous manifestation of symptoms and the involvement of multiple wood-associated pathogens, both suspected and confirmed. This study investigates the mycological aspects of grapevine trunk diseases, focusing on Lasiodiplodia theobromae as a causal agent. The pathogen was associated with vascular cankers, dark brown trunk discoloration, pycnidia formation on necrotic tissues, an

Evaluation of Durum and Hard Red Spring Wheat Panels for Sensitivity to Necrotrophic Effectors Produced by Parastagonospora nodorum.

Septoria nodorum blotch (SNB) is an important disease of both durum and hard red spring wheat (HRSW) worldwide. The disease is caused by the necrotrophic fungal pathogen Parastagonospora nodorum when compatible gene-for-gene interactions occur between pathogen-produced necrotrophic effectors (NEs) and corresponding host sensitivity genes. To date, nine sensitivity gene-NE interactions have been identified, but there is little information available regarding their overall frequency in durum and HRSW. Here, we infiltrated a global HRSW panel (HRSWP) and the Global Durum Panel (GDP) with P. nodor

Siderophore Activity of Partially Acetylated Fusarinines from the Sponge-Derived Fungus Pseudogymnoascus verrucosus.

Pseudogymnoascus is a fungal genus prevalent in Antarctica whose metabolism, including the types of siderophores it synthesizes, remains largely unexplored. In this work, we investigated the siderophores produced by Pseudogymnoascus verrucosus FAE27, a strain isolated from the Antarctic marine sponge Hymeniacidon sp., grown under iron-limiting conditions. Two new hydroxamate-type siderophores, N,N'-diacetyl-fusarinine B (7) and N-acetyl-fusarinine A (8), are described here as the first naturally occurring partially acetylated fusarinines to date, along with the known fusarinine (1) and five of

Paleoproteomics reveals chromoblastomycosis as possible cause of King of France Louis XIV's death (1715).

The paleoproteomic study carried out on a sample of the heart of Louis XIV offers us new and original conclusions allowing us to better understand the cause and circumstances of the death of the famous king of France in 1715. Combined micro-CT scans and paleoproteomics confirm the human and cardiac nature of the sample by the identification of 50 order-specific peptides and 15 family-specific peptides, and numerous Homo sapiens species proteins. It raises the hypothesis of a fungal infection by Cyphellophora europea, a fungus known to cause skin infections, worsened in immunocompromised patien

A single-cell atlas of Drosophila trachea reveals glycosylation-mediated Notch signaling in cell fate specification.

The Drosophila tracheal system is a favorable model for investigating the program of tubular morphogenesis. This system is established in the embryo by post-mitotic cells, but also undergoes remodeling by adult stem cells. Here, we provide a comprehensive cell atlas of Drosophila trachea using the single-cell RNA-sequencing (scRNA-seq) technique. The atlas documents transcriptional profiles of tracheoblasts within the Drosophila airway, delineating 9 major subtypes. Further evidence gained from in silico as well as genetic investigations highlight a set of transcription factors characterized b

Biocontrol activity and action mechanism of Pseudomonas aurantiaca ST-TJ4 against Verticillium dahliae, the causal agent of Acer truncatum wilt.

Acer truncatum wilt caused by Verticillium dahliae is a severe soilborne disease that poses a threat to the cultivation of this plant in China. The present study explored the biocontrol efficiency and underlying antagonistic mechanism of Pseudomonas aurantiaca ST-TJ4 against V. dahliae. In vitro, strain ST-TJ4 exhibited excellent inhibitory effects on V. dahliae, causing mycelial deformation. This strain significantly suppressed the production of V. dahliae conidia and microsclerotia. Moreover, the application of ST-TJ4 reduced the incidence of Verticillium wilt in A. truncatum saplings in bot

Low frequency of resistance to thiophanate-methyl in Monilinia fructicola populations from southeastern United States peach orchards.

Methyl benzimidazole carbamate (MBC) fungicides were once widely used for brown rot (Monilinia fructicola) control of peach (Prunus persica (L.) Batsch) in the southeastern US, but their use was substantially reduced and often eliminated due to widespread resistance. In this study, 233 M. fructicola isolates were collected from major peach production areas in Alabama, Georgia, and South Carolina, and sensitivity to thiophanate-methyl was examined. Isolates were also collected from one organic and two experimental peach orchards. A discriminatory dose of 1 μg/ml was used to distinguish sensitiv

Miltefosine Against Scedosporium and Lomentospora Species: Antifungal Activity and Its Effects on Fungal Cells.

Scedosporium and Lomentospora species are filamentous fungi responsible for a wide range of infections in humans and are frequently associated with cystic fibrosis and immunocompromising conditions. Because they are usually resistant to many antifungal drugs available in clinical settings, studies of alternative targets in fungal cells and therapeutic approaches are necessary. In the present work, we evaluated the in vitro antifungal activity of miltefosine against Scedosporium and Lomentospora species and how this phospholipid analogue affects the fungal cell. Miltefosine inhibited different

Coupled Mn(II) and Cr(III) Oxidation Mediated by Ascomycete Fungi.

Manganese (Mn) oxides are considered as the primary oxidant of trivalent chromium [Cr(III)] in the environment. Microbial activities are responsible for the majority of Mn oxide formation in nature, thus likely influencing Cr(III) oxidation. Previous studies have been limited to Cr(III) oxidation by bacterial Mn oxides. Herein, we report coupled Mn(II) and Cr(III) oxidation in the presence of three Mn(II)-oxidizing Ascomycete fungi. In contrast to the previously reported inhibitory effect of Cr(III) on bacterial Mn(II) oxidation, varying effects of Cr(III) on fungal Mn(II) oxidation were obser
