Effects of salicylate on organ-of-Corti vibrations in the second turn of the gerbil cochlea.

Sound-evoked responses of the organ of Corti (OoC) in the living cochlea are enhanced by active processes driven by outer hair cells (OHCs). To assess how suppression of OHC function affects intracochlear mechanics, we applied salicylate-a blocker of OHC electromotility-to the round window of anesthetized Mongolian gerbils and measured both distortion product otoacoustic emissions (DPOAEs) and sound-evoked vibrations of the OoC in the second cochlear turn using optical coherence tomography (OCT). Salicylate reduced DPOAE amplitudes in a time- and frequency-dependent manner and decreased intrac

Auditory Neural Activity in Congenitally Deaf Mice Induced by Infrared Neural Stimulation.

To determine whether responses during infrared neural stimulation (INS) result from the direct interaction with spiral ganglion neurons (SGNs), we tested three genetically modified deaf mouse models: Atoh1-cre; Atoh1 f/f (Atoh1 conditional knockout, CKO), Atoh1-cre; Atoh1 f/kiNeurog1 (Neurog1 knockin, KI), and the Vglut3 knockout (Vglut3 -/-) mice. All animals were exposed to tone bursts and clicks up to 107 dB (re 20 µPa) and to INS, delivered with a 200 µm optical fiber. The wavelength (λ) was 1860 nm, the radiant energy (Q) 0-800 µJ/pulse, and the pulse width (PW) 100-500 µs. No auditory re

Level-dependent changes in detection of temporal gaps in noise markers by adults with normal and impaired hearing.

Compression in the basilar-membrane input-output response flattens the temporal envelope of a fluctuating signal when more gain is applied to lower level than higher level temporal components. As a result, level-dependent changes in gap detection for signals with different depths of envelope fluctuation and for subjects with normal and impaired hearing may reveal effects of compression. To test these assumptions, gap detection with and without a broadband noise was measured with 1, 000-Hz-wide (flatter) and 50-Hz-wide (fluctuating) noise markers as a function of marker level. As marker level i

Masker phase effects in normal-hearing and hearing-impaired listeners: evidence for peripheral compression at low signal frequencies.

The presence of cochlear-based compression at low frequencies was investigated by measuring phase effects in harmonic maskers. In normal-hearing listeners, the amount of masking produced depends strongly on the phase relationships between the individual masker components. This effect is thought to be determined primarily by properties of the cochlea, including the phase dispersion and compressive input-output function of the basilar membrane. Thresholds for signals of 250 and 1000 Hz were measured in harmonic maskers with fundamental frequencies of 12.5 and 100 Hz as a function of the masker p

Exploration of cochlear function by otoacoustic emissions: relationship to pure-tone audiometry.

The amplitudes, growth functions and detection thresholds of evoked otoacoustic emissions (EOE) were measured in 44 normally hearing subjects and 138 patients with two categories of cochlear dysfunctions: (a) acoustic trauma; and (b) presbycusis. Separate sets of experiments were also performed: (a) detection of stimulus frequency emissions; and (b) click EOE. EOE properties were studied around 2 kHz, 1 kHz and 0.75 kHz (+/- 0.1 kHz). A partial correlation and multivariate analysis was carried out to investigate the relationships between EOE properties and puretone auditory thresholds (from 0.

Effects of Combined Gentamicin and Furosemide Treatment on Cochlear Macrophages.

Combining aminoglycosides and loop diuretics often serves as an effective ototoxic approach to deafen experimental animals. The treatment results in rapid hair cell loss with extended macrophage presence in the cochlea, creating a sterile inflammatory environment. Although the early recruitment of macrophages is typically neuroprotective, the delay in the resolution of macrophage activity can be a complication if the damaged cochlea is used as a model to study subsequent therapeutic strategies. Here, we applied a high dose combination of systemic gentamicin and furosemide in C57 BL/6 and CBA/C

Stimulus-frequency otoacoustic emissions and middle-ear pressure gains in a finite-element mouse model.

For evoked otoacoustic emissions (OAEs), the stimulus and emission signals traverse the middle ear (ME) in forward and reverse directions, respectively. In this study, a fully coupled three-dimensional finite-element model of the mouse ear canal (EC), ME, and cochlea was used to calculate ME pressure gains, impedances, and reflectances at the EC-entrance and stapes-footplate-cochlear-fluid interfaces. The cochlear model incorporates a series of interdigitated Y-shaped structures sandwiched between the basilar membrane and reticular lamina, each comprised of a Deiters' cell, its phalangeal-proc

Complementary metrics of human auditory nerve function derived from compound action potentials.

Declines in auditory nerve (AN) function contribute to suprathreshold auditory processing and communication deficits in individuals with normal hearing, hearing loss, hyperacusis, and tinnitus. Procedures to characterize AN loss or dysfunction in humans are limited. We report several novel complementary metrics using the compound action potential (CAP), a direct measure of summated AN activity. Together, these metrics may be used to characterize AN function noninvasively in humans. We examined how these metrics change with stimulus intensity and interpreted these changes within a framework of

A dual filter model describing single-fiber responses to clicks in the normal and noise-damaged cochlea.

This paper presents a composite model of the normal and noise-damaged guinea pig cochlea. The model incorporates a phenomenologically defined cochlear filter, and physiological descriptions of inner hair cell transduction, synaptic adaptation, and spike generation. The latter three model sections were taken from recent literature. The paper first deals with validation and evaluation of the model and adaptation of the relevant parameters to the guinea pig. Then the model is applied to explore to what extent changes in the cochlear filter can be held responsible for abnormal responses to clicks

Acoustic stimulation increases phosphoinositide breakdown in the guinea pig cochlea.

The effect of sound stimulation on phosphoinositide turnover was investigated in the inner ear of the guinea pig. The perilymphatic spaces of the cochlea were perfused with [(32)P]orthophosphate ((32)P(i)) and labeled lipids analyzed in the tissues of the lateral wall of the cochlea (stria vascularis and spiral ligament) and the basilar membrane (organ of Corti). Consistent with previous results, polyphosphoinositides rapidly incorporated (32)P(i) comprising 70-80% of labeled lipids in all tissues analyzed: phosphatidylinositol 4,5-bisphosphate (PtdInsP(2)) was 66% of total [(32)P]phospholipid

[The influence of glucocorticoids on the view of chicken's inner ear damage after exposure to wide-band-noise].

The aim of the study was to assess the influence of glucocorticoids on the view of hair cell regeneration process being in the chicken's inner ear (basilar papilla - BP) after exposure to wide-band noise at the level 120 dB (A) for 48 hours. We found that glucocorticoids given during and/or after exposure to the noise have a cytoprotective activity to the hair cells, they limitate the extensiveness and decrease the dynamics of hair cells injury. We observed that new "young" hair cells reappeared at the sensory epithelium on the 7th day after the end of exposure. Regenerated hair cells have imm

[Evoked acoustic oto-emissions in cochlear deafness].

In order to give a contribution to the genesis of the EOAEs, we have recorded echos in 24 subjects with unilateral sudden deafness (I. group); 20 ears with Menière disease (II. group); 22 ears with progressive sensorineural hearing loss (III. group) and 10 normally hearing young subjects as control group. The results have shown that the EOAEs were present in 100% (I. group), 84.6% (II. group) and 86.3% (III. group) in response to the tone-burst; while the echos were present only in the 57.1% (I. group), 38.4% (II. group) and 45.4% (III. group) in response to the click, although the audiometric

Influence of the endothelium and alpha-adrenoreceptor antagonists on responses to noradrenaline in the rabbit basilar artery.

1. Noradrenaline (10(-6)-10(-2) M) produced slow, concentration-dependent depolarization of smooth muscle cells in the rabbit basilar artery, which preceded the onset of contraction by around 8 s (n = 18). 2. With concentrations greater than 10(-4) M, noradrenaline produced action potentials and fast rhythmic depolarizations superimposed on the slow depolarization. Each fast event was followed by a clear increase in the rate of smooth muscle contraction. The selective alpha 1-adrenoreceptor agonist phenylephrine produced very similar membrane and contractile responses. 3. Action potentials wer

Hypoxia-Stabilized HIF1α Restricts Hair Cell Reprogramming by Suppressing Wnt Signaling in the Mammalian Cochlea.

Sensorineural hearing loss, caused by irreversible hair cell (HC) loss, remains incurable due to the inability of the mammalian cochlea to regenerate HCs spontaneously. Although the combinatorial modulation of Notch and Wnt signaling robustly reprograms cochlear supporting cells to HCs in vitro, these strategies show limited success in vivo, which suggests that the native cochlear microenvironment imposes inhibitory cues. Here, we identify hypoxia-inducible factor 1α (HIF1α) as a critical barrier to HC reprogramming in vivo. We found that HIF1α accumulates in the basilar membrane of the intact

Revealing intracochlear microtrauma during cochlear implantation using optical coherence tomography and electrochemical sensing.

Cochlear implants (CIs) are a highly effective treatment for severe to profound hearing loss, and earlier implantation is associated with improved auditory outcomes. As implantation is now extended to patients with residual hearing, preserving cochlear structures during surgery is a key priority. However, implant insertion trauma remains a risk and can compromise residual hearing. Currently, intraoperative methods capable of detecting such trauma in situ are lacking. We introduce a novel approach for in situ detection of intracochlear trauma using a gerbil model. Specifically, we use optical c
