From Identification to Enzymatic Activity Profiling: Comprehensive Characterization of BloCAβ, a Probiotic β-Carbonic Anhydrase From Bifidobacterium longum.

The microbiota comprises communities of microorganisms that establish complex relationships with the host and play key roles in physiology, … s. Herein, we report the first biochemical characterization of β-class carbonic anhydrase (CA) from Bifidobacterium longum (BloCAβ) by inhibition and activation profiles. The enzyme was recombinantly expressed, purified, and kinetically characterized, displaying a catalytic efficiency within the same order of magnitude as, although lower than, that of human (h)CA …

Bifidobacterium adolescentis attenuates brown adipose tissue whitening and improves metabolic markers in high-fat diet-induced obese rats.

PURPOSE: Activation of brown adipose tissue (BAT) has emerged as a promising strategy for combating obesity due to its thermogenic capacity. However, chronic high-fat diet (HFD) exposure impairs BAT function and promotes a "whitening" phenotype characterized by lipid droplet enlargement and reduced thermogenic activity. Bifidobacterium adolescentis (B. adolescentis) has been reported to improve lipid metabolism and energy balance. This study investigated whether B. adolescentis supplementation attenuates HFD-induced BAT whitening, evaluated at transcriptional and histological levels, and exami…

Dietary supplementation with milk-derived Bifidobacterium animalis subsp. lactis relieves colitis-linked reproductive disorders via gut-testis axis.

Rapid urbanization, medication, and modern dietary patterns are the main challenges leading to impaired fertility, with a lack of effective therapies. Emerging evidence suggests that reproductive disorders may be closely associated with intestinal damage or occasionally worsen by the side effects of medications. Therefore, the development of dietary supplements as alternatives is crucial for intestinal-linked reproductive health. Milk and dairy products are essential in dietary nutrition with great potential functional ingredients. Bifidobacterium animalis subsp. lactis NJ241 (NJ241), a promis…

Inhibitory Potential of Bifidobacterium longum FB1-1 Cell-Free Supernatant against Carbapenem-Resistant Klebsiella pneumoniae Drug Resistance Spread.

The widespread dissemination of carbapenem-resistant Klebsiella pneumoniae (CRKP) and its drug resistance transfer poses a global public health threat. While previous studies outlined CRKP's drug resistance mechanism, there is limited research on strategies inhibiting CRKP drug resistance spread. This study investigates the potential of Bifidobacterium longum (B. longum) FB1-1, a probiotic, in curbing the spread of drug resistance among CRKP by evaluating its cell-free supernatant (CFS) for antibacterial activity. Evaluating the inhibitory effect of FB1-1 CFS on CRKP drug resistance spread inv…

Effects of Bifidobacterium-Fermented Milk on Obesity: Improved Lipid Metabolism through Suppression of Lipogenesis and Enhanced Muscle Metabolism.

Obesity is a major global health concern. Studies suggest that the gut microflora may play a role in protecting against obesity. Probiotics, including lactic acid bacteria and Bifidobacterium, have garnered attention for their potential in obesity prevention. However, the effects of Bifidobacterium-fermented products on obesity have not been thoroughly elucidated. Bifidobacterium, which exists in the gut of animals, is known to enhance lipid metabolism. During fermentation, it produces acetic acid, which has been reported to improve glucose tolerance and insulin resistance, and exhibit anti-ob…

BACKGROUND: Genus and species level analysis is the best way to characterize alterations in the human gut microbiota that are associated with obesity, because the clustering of obese and lean microbiotas increases with the taxonomic depth of the analysis. Bifidobacterium genus members have been associated with a lean status, whereas different Lactobacillus species are associated both with a lean and an obese status. OBJECTIVES AND METHODS: We analyzed the fecal concentrations of Bacteroidetes, Firmicutes, Methanobrevibacter smithii, the genus Lactobacillus, five other Lactobacillus species pr…

Targeting intestinal microbiota with Bifidobacterium breve and Hafnia alvei probiotics does not improve response to influenza vaccine in mice.

The intestinal microbiota plays a critical role in shaping host immune responses to pathogens and vaccines. Dextran sulfate sodium-treated mice showed reduced vaccinal response following prime-boost immunization with purified ovalbumin, as confirmed by decreased specific immunoglobulin (Ig)G and IgM, confirming the negative impact of dysbiosis and associated inflammation. Two probiotic strains were selected, Bifidobacterium breve and Hafnia alvei, but none of them showed any enhancement of antibody or T cell responses following prime-boost influenza vaccination in mice. While probiotics remain…

Isolation and Characterization of Bifidobacterium longum A1, an Isolate from Faeces of an Exclusively Breast-fed Infant with Probiotic Potential.

UNLABELLED: Microbial colonization of the newborn (primocolonization) plays a crucial role in the development of digestive, metabolic, and immune systems. Disruptions may impair intestinal barrier function and raise the risk of chronic diseases like inflammatory bowel disease, allergies, and obesity. Recent evidence suggests probiotic supplementation in infants may help counter or prevent these effects. This study isolates and characterizes potential probiotic candidates from the gut microbiota of exclusively breastfed infants. Bifidobacterium and Lacticaseibacillus strains were selectively is…
