Mechanically reinforced hydrogel vehicle delivering angiogenic factor for beta cell therapy.

Type 1 diabetes mellitus (T1DM) is a chronic disease affecting millions worldwide. Insulin therapy is currently the golden standard for treating T1DM; however, it does not restore the normal glycaemic balance entirely, which increases the risk of secondary complications. Beta-cell therapy may be a possible way of curing T1DM and has already shown promising results in the clinic. However, low retention rates, poor cell survival, and limited therapeutic potential are ongoing challenges, thus increasing the need for better cell encapsulation devices. This study aimed to develop a mechanically rei

Hypoparathyroidism: State of the Art on Cell and Tissue Therapies.

Hypoparathyroidism is an endocrine disorder characterized by low serum calcium levels, high serum phosphorus levels, and by inappropriate or absent secretion of the parathyroid hormone (PTH). The most common therapeutic strategy to treat this condition is hormone replacement therapy with calcium and vitamin D but, unfortunately, in the long term this treatment may not be sufficient to compensate for the loss of endocrine function. Glandular autotransplantation is considered the most effective technique in place of replacement therapy. Although it leads to excellent results in most cases, autot

Microencapsulation of cells and molecular therapy of type 1 diabetes mellitus: The actual state and future perspectives between promise and progress.

The history of microencapsulation of live cells started with an idea of Thomas MS Chang in 1964, thereafter applied to isolated pancreatic islets by Anthony M Sun in 1980. The original aim was to provide isolated cells with an immune-protective shield, to prevent physical contact between the transplanted cells and the host's immune system, with retention of the microcapsules' biocompatibility and physical-chemical properties over time. In particular, this revolutionary approach essentially applied to islet grafts, in diabetic recipients who are not immunosuppressed, at a preclinical (rodents)

Germline cysts require Ecdysone receptor for proper timing of encapsulation in the Drosophila ovary.

Oocytes are packaged into ovarian follicles, each containing a maturing germ cell surrounded by a layer of somatic cells. This conserved arrangement is essential for proper oocyte development and reproductive success. Continuous coordination and bi-directional signaling from somatic cells to germ cells are necessary for proper oocyte packaging. In Drosophila, although germ cell packaging has been presumed to be controlled by somatic follicle cells enveloping passive germ cells, recent studies suggest that germ cells themselves produce motor forces that drive somatic encapsulation. Here, in sup

Alginate-gelatine hydrogel microspheres protect NK cell proliferation and cytotoxicity under hypoxic conditions.

AIMS: This study aimed to encapsulate natural killer (NK) cells in a hydrogel to sustain their function within the hypoxic tumour microenvironments. METHODS: An alginate-gelatine hydrogel was generated via electrospray technology. Hydrogel biocompatibility was assessed through cell counting kit-8 and Live/Dead assays to ascertain cell. Moreover, we analysed lactate dehydrogenase assays to evaluate the cytotoxicity against tumours and utilised RT-qPCR to analyse cytokine gene level. RESULTS: Alginate and gelatine formed hydrogels with diameters ranging from 489.2 ± 23.0 μm, and the encapsulat

Microfluidic Generation of Thin-Shelled Polyethylene Glycol-Tyramine Microgels for Non-Invasive Delivery of Immunoprotected β-Cells.

Transplantation of microencapsulated pancreatic cells is emerging as a promising therapy to replenish β-cell mass lost from auto-immune nature of type I diabetes mellitus (T1DM). This strategy intends to use micrometer-sized microgels to provide immunoprotection to transplanted cells to avoid chronic application of immunosuppression. Clinical application of encapsulation has remained elusive due to often limited production throughputs and body's immunological reactions to implanted materials. This article presents a high-throughput fabrication of monodisperse, non-immunogenic, non-degradable,

Efficacy of Salmonella Bacteriophage S1 Delivered and Released by Alginate Beads in a Chicken Model of Infection.

Modern bacteriophage encapsulation methods based on polymers such as alginate have been developed recently for their use in phage therapy for veterinary purposes. In birds, it has been proven that using this delivery system allows the release of the bacteriophage in the small intestine, the site of infection by Salmonella spp. This work designed an approach for phage therapy using encapsulation by ionotropic gelation of the lytic bacteriophage S1 for Salmonella enterica in 2% w/v alginate beads using 2% w/v calcium chloride as crosslinking agent. This formulation resulted in beads with an

Development of poly(ethylene glycol) hydrogels for salivary gland tissue engineering applications.

More than 40,000 patients are diagnosed with head and neck cancers annually in the United States with the vast majority receiving radiation therapy. Salivary glands are irreparably damaged by radiation therapy resulting in xerostomia, which severely affects patient quality of life. Cell-based therapies have shown some promise in mouse models of radiation-induced xerostomia, but they suffer from insufficient and inconsistent gland regeneration and accompanying secretory function. To aid in the development of regenerative therapies, poly(ethylene glycol) hydrogels were investigated for the encap

Kinetics of granulocyte phagocytosis: rate limited by cytoplasmic viscosity and constrained by cell size.

Micromanipulation of yeast particles and blood granulocytes has been used to study the kinetics of single phagocytosis events. The ingestion process was quantitated by observation of sequential adhesion and encapsulation times. Both adherence and encapsulation times were found to increase greatly as the temperature was reduced below 37 degrees C; calcium in solution facilitated adhesion of the particle to the phagocyte but not encapsulation; both adhesion and encapsulation processes required a minimum level of plasma components (presumably complement). The general nature of these observations

Directed Molecular Assembly and Crosslinking for Conformal Coating of Mesenchymal Stem Cells Using a Bispecific DNA-Enzyme Conjugate.

Current cell encapsulation methods face problems with large encapsulation volumes. In this study, we present a modular cell-surface conformal coating strategy using a bispecific DNA-enzyme conjugate to direct molecular assembly and crosslinking. The bispecific conjugate exhibited sustained retention on the cell membrane with a half-life on the order of hours. The conjugate catalyzed the formation of a 10 to 20 μm thick fibrin layer upon addition of fibrinogen, allowing the adsorption and deposition of alginate and poly-L-lysine crosslink the coating. The platform was compatible with spheroids

An immunomodulatory encapsulation system to deliver human iPSC-derived dopaminergic neuron progenitors for Parkinson's disease treatment.

Parkinson's disease is a neurodegenerative condition associated with the progressive loss of dopaminergic neurons. This leads to neurological impairments with heightening severity and is globally increasing in prevalence due to population ageing. Cell transplantation has demonstrated significant promise in altering the disease course in the clinic, and stem cell-derived grafts are being investigated. Current clinical protocols involve systemic immunosuppression to prevent graft rejection, which could potentially be avoided by encapsulating the therapeutic cells in a locally immunosuppressive b

Microencapsulation system for scalable differentiation of peripheral motor neurons from human induced pluripotent stem cells.

Stem cell-derived neural cells hold great potential for treating neurological disorders, but clinical translation is limited by the need for scalable, consistent, and functionally robust production systems. To address these challenges, we developed a microfluidic alginate encapsulation chip (MAEC) system for the high-throughput production of mature peripheral motor neurons from human induced pluripotent stem cells. Alginate was selected for its biocompatibility, low immunogenicity, and calcium-triggered gelation, enabling precise size control. Encapsulation conditions were optimized to produce

A bioactive cell sheet interface regulates scaffold-host interactions for cartilage regeneration.

Cartilage tissue engineering remains challenged by uncontrolled scaffold-host interactions after implantation, particularly host tissue infiltration that can disrupt cartilage formation and compromise scaffold integrity. Although biomaterial-based encapsulation strategies have been developed to reduce host interference, excessive restriction of scaffold-host interface may also limit the exchange of nutrients, oxygen, and metabolic wastes. Therefore, an effective encapsulation strategy must protect the developing tissue from excessive host infiltration while maintaining a permissive environment

A Scalable Droplet Microfluidic Platform That Overcomes Large-Cell Constraints for Long-Term Plant Cell Culture and Embryogenesis Analysis.

Microspore-derived embryogenesis (MDE) is essential for doubled haploid (DH) plant breeding, but conventional bulk culture systems are limited by poor scalability and difficulty for automation. Adapting droplet microfluidics for plant cell culture is promising but challenging due to the large size and fragility of plant cells involved in embryogenesis, such as maize microspores (60-85 µm) and their expanding embryo-like structures (200-500 µm). Here, we present a droplet microfluidic platform engineered for large plant cells, integrating a tubing-based loading strategy that minimizes sedimenta

Biocompatible Multilayered Encapsulation for Organic Light-Emitting Diodes.

Organic light-emitting diodes (OLEDs) have tremendous potential in biotechnology, but their vulnerability to oxygen and moisture presents a significant challenge in encapsulation. In this study, we developed a multilayer thin-film encapsulation consisting of dual inorganic layers and Parylene-C, offering excellent protection and biocompatibility. This encapsulation enhances the suitability of OLEDs for flexible substrates and biological applications. The multilayer structure, composed of Al2O3/SiOxNy/Parylene-C, was fabricated entirely below 100 °C to ensure compatibility with temperature-sens

Alginate-based biofilm-assisted encapsulation of a Co-culture of Bifidobacterium longum DSM20219 and Bacillus subtilis SOM8 enhances resistance to gastrointestinal stress.

Oral probiotic formulations are increasingly explored for supporting gastrointestinal health, restoring gut microbiome balance, and providing non-invasive alternatives to injections or transplants. However, delivering oxygen-sensitive probiotics orally remains challenging because gastric acidity, bile salts, digestive enzymes, and aerobic storage can substantially reduce bacterial viability. Sodium alginate is widely used for probiotic encapsulation because it forms mild, cell-compatible hydrogel beads, but alginate-only matrices often provide limited protection for highly sensitive anaerobes

A Spike-Based mRNA Vaccine Encapsulated in Phospholipid 1,2-Dioleoyl-sn-Glycero-3-PhosphoEthanolamine Containing Lipid Nanoparticles Induced Potent B- and T-Cell Responses Associated with Protection Against SARS-CoV-2 Infection and COVID-19-like Symptoms in Hamsters.

BACKGROUND: Nucleoside-modified mRNA encapsulated in lipid nanoparticles (LNPs) have emerged as a promising vaccine strategy, especially for COVID-19. While the LNPs protect mRNA from degradation and efficiently deliver the mRNA to antigen-presenting cells, the effect of lipid composition on the immunogenicity and protective efficacy of mRNA/LNP vaccines is not well characterized. Studies on using the mRNA/LNP platform for vaccines have largely focused on the nucleic acid cargo with less attention paid to the LNP vehicle. Whether the composition and biophysical properties of LNPs impact vaccin

Microencapsulation of Liver Spheroids with Poly(Vinyl Alcohol) Enhances Function Compared with Alginate.

Background and Aims: Cell therapy approaches to treating chronic liver disease provide only transient improvements, mainly due to loss of hepatocytes after infusion. Microencapsulation in alginate has been shown to protect transplanted cells from physical stress and rejection, but the poor biocompatibility of alginate can lead to graft failure. This study aimed to evaluate a biocompatible poly(vinyl alcohol) (PVA)-based microcapsule against standard alginate for improved transplantation outcome of liver spheroids. Materials and Methods: Human hepatocyte spheroids were microencapsulated in algi

Exploring the effects of cannabidiol encapsulation in liposomes on their physicochemical properties and biocompatibility.

Cannabidiol (CBD) is recognized for its therapeutic properties in various conditions. However, CBD's limited water solubility and sensitivity to environmental stresses hinder its efficacy and bioavailability. Encapsulation in drug delivery systems, particularly liposomes, offers a promising solution. This study aims to prepare CBD-containing liposomes using commercially used lipids distearoyl phosphatidylcholine (DSPC) and dipalmitoyl phosphatidylcholine (DPPC), and 1,2 distearoyl-sn-glycero-3 phosphoethanolamine-N-[carbonyl-amino(polyethylene glycol)-4300] (ammonium salt) (DSPE-PEG) and to pe
