Chromosome analysis of multipronuclear human oocytes after in vitro fertilization.

Multipronuclear human eggs are frequent after in vitro fertilization. Their chromosome analysis can provide useful information. Before cleavage it can confirm the suspected polyploidy. Among the cleaved multipronuclear eggs it provides an estimation of the incidence of the possible return to diploidy. Ninety-four multipronuclear eggs were fixed at the first, second, or third cleavage according to the air-drying method of Tarkowski with or without colchicine exposure: 60 were successfully analysed. Twelve were stopped before cleavage (six without colchicine treatment and six with colchicine tre

Chromosome aberrations induced in human lymphocytes by 16.5 MeV protons.

In a track segment irradiation experiment with 16.5 MeV protons the induction of dicentrics was studied in human T-lymphocytes. The dose-response relationship was linear quadratic with estimated parameters alpha = (0.44 +/- 0.07) x 10(-1) Gy-1 and beta = (1.95 +/- 0.30) x 10(-2) Gy-2. With respect to X-rays a limiting RBE of 1.1-1.2 exists. The present findings are compared with data from other laboratories obtained with 4.9 and 8.7 MeV protons. It was found that the data do not fit theoretical predictions on a proportional relationship between alpha values and LET. Using the microdosimetric q

Recurrent chromosome aberrations in human lung squamous cell carcinomas.

Cytogenetic study of seven cases of previously untreated lung squamous cell carcinomas (SQC) is reported. Chromosome numbers vary from 38 to 538, with a majority of hypotriploid karyotypes with complex rearrangements. The numbers of recurrent imbalances were evaluated in considering the average number of chromosomes or chromosome segments in each analyzed metaphase and for each case. In decreasing order of frequency, deficiencies for 3p, 5q, 8p, Y, 5p, 10p, 13, and, to a lesser degree, for 8q, 9, 10q, 11pter, 14, 15, and 21 were observed; the excesses principally involve 1q, 3q, and 7q. In thr

Human gene for torsion dystonia located on chromosome 9q32-q34.

Torsion dystonia is a movement disorder of unknown etiology characterized by loss of control of voluntary movements appearing as sustained muscle contractions and/or abnormal postures. Dystonic movements can be caused by lesions in the basal ganglia, drugs, or gene defects. Several hereditary forms have been described, most of which have autosomal dominant transmission with variable expressivity. In the Ashkenazi Jewish population the defective gene frequency is about 1/10,000. Here, linkage analysis using polymorphic DNA and protein markers has been used to locate a gene responsible for susce

Linking of the human immunoglobulin VK and JKCK regions by chromosomal walking.

The linking of the human VK and JKCK gene regions (abbreviations in ref. 1) by chromosomal walking is reported. Hybridization experiments with the DNA of a somatic cell hybrid containing the region between JKCK and the telomer show that none of the major VK gene clusters is located downstream of CK. The distance between the VK and JK genes was found to be 23 kb. The JK proximal VK gene is the B3 gene which is the only representative of subgroup IV in the genome. This gene and the neighbouring B2 gene (accompanying paper) are arranged in opposite orientation to JKCK and can therefore rearrange

Location of genes involved in invasion and metastasis on human chromosome 7.

We have obtained evidence for the existence of genes controlling invasion and metastasis by somatic cell fusion studies. Noninvasive, nonmetastatic mouse BW5147 T-lymphoma cells were fused with invasive human T-cells. The human cells were either activated normal peripheral blood lymphocytes or leukemic T-lymphoblasts. Both fusions resulted in highly invasive human-mouse T-cell hybrids which metastasized in nude mice. Thus the genes derived from either malignant or nonmalignant but inherently invasive cells enable the T-cell hybridomas to metastasize. By continued in vitro selection for invasiv

Pericentric inversion in human chromosome 1 and the risk for male sterility.

A pericentric inversion in chromosome 1 of a severely oligospermic human male is reported. Pachytene analysis in microspread preparations shows an absence of full loop formation in the inversion bivalent and only the rare occurrence of a partial loop. The majority of cells exhibit extensive asynapsis across the inverted segment, or a normal looking synaptonemal complex indicative of heterologous pairing along the length of the inversion. Crossing over is reduced in the No 1 bivalent with only a rare chiasma being seen in the inverted region at metaphase I. Males heterozygous for a pericentric

[Chromosome aberrations produced in human lymphocytes by in vivo and in vitro irradiation].

The production of chromosome aberrations in vivo has been studied in lymphocytes from a patient undergoing a wholebody treatment with gamma-radiation up to a cumulative dose of 1.4 Gy. These results were compared with the observations performed on whole blood samples irradiated in vitro with doses from 0.05 up to 2 Gy of gamma-rays. The frequency of chromosome aberrations, particularly the dicentrics, was found to be similar in vivo and in vitro. The yield of dicentrics could be best related to the dose by using a linear-quadratic model in both cases, the ratio of the coefficients a/b being of

Chromosome Y-specific DNA in related human XX males.

Human 'XX males' are sterile males whose chromosomes seem to be those of a normal female. About 1 in 20,000 males has a 46, XX karyotype, and most cases are sporadic, that is, they are without familial clustering. It has long been argued that maleness in XX males may result from the undetected presence of a small, testis-determining fragment of the Y chromosome, and there is strong evidence for this in sporadically occurring XX males. Indeed, the genomes of three of four sporadic XX males tested were found to contain certain Y-specific DNA sequences. A pedigree in which three XX males occur ha

Partial purification and characterization of DNA from the human X chromosome.

Human X chromosome DNA was partially purified from a mouse-human hybrid cell line containing a single human chromosome, the X. Enrichment of such DNA was accomplished by two sequential reassociations of radiolabeled hybrid cell DNA with large excesses of mouse DNA. Unreassociated hybrid cell DNA was used as a probe for human X chromosome sequences. The human-specific fraction of probe DNA CONTAINED THREE COMPONENTS. Two of these reassociated to human DNAs at rates proportional to the number of X chromosomes present. These two components were thus localized to the X chromosome. One of these X-s

Chromosome aberrations in human lymphocytes exposed to tritiated water in vitro.

The induction of chromosome aberrations in human peripheral blood lymphocytes by tritiated water or 180 kV X-rays in vitro was studied. Lymphocytes were exposed to various concentrations of HTO for 2 h or for 53 h. Chromosome and chromatid type aberrations were scored during the first mitotic division after stimulation with phytohaemagglutinin. For the analysis of the dose-response relationship the data were fitted by the method of least-squares to different models. After acute exposure to tritium beta-rays and X-rays, the dicentrics + centric rings and terminal + interstitial deletions gave t

Structural variability of human chromosome 9 in relation to its evolution.

Human chromosome 9 shows a high susceptibility for structural rearrangements, particularly pericentric inversions, which often are transmitted. Three types of pericentric inversions can be observed on No. 9: 1) Type I, showing the total constitutive heterochromatin in the short arm. 2) Type II with part of the C heterochromatin on the short arm, the rest located on the long arm proximal to the centromere. 3) Type III: a subtelocentric chromosome with part of the C heterochromatin in the very short arm and the rest located interstitially on the long arm. With these inversions as well as with ot

Chromosomal damage in human leukocytes induced by lysergic acid diethylamide.

Addition of lysergic acid diethylamide to cultured human leukocytes resulted in a marked increase of chromosomal abnormalities. The distribution of chromosome breaks deviated significantly from random, with an accumulation of aberrations in chromosome No. 1. Cytogenetic investigation of a patient extensively treated with this drug over a 4-year period for paranoid schizophrenia showed a similar increase in chromosomal damage.

Assembly of Multiple Full-Size Genes or Genomic DNA Fragments on Human Artificial Chromosomes Using the Iterative Integration System.

Human artificial chromosomes (HACs) are gene delivery vectors that have been used for decades for gene functional studies. HACs have several advantages over viral-based gene transfer systems, including stable episomal maintenance in a single copy in the cell and the ability to carry up to megabase-sized genomic DNA segments. We have previously developed the alphoidtetO -HAC, which has a single gene acceptor loxP site that allows insertion of an individual gene of interest using Chinese hamster ovary (CHO) hybrid cells. The HAC, along with a DNA segment of interest, can then be transferred from
