Myocardial Substrate Oxidation and Tricarboxylic Acid Cycle Intermediates During Hypothermic Machine Perfusion.

BACKGROUND: The optimal substrate for hypothermic machine perfusion preservation of donor hearts is unknown. Fatty acids, acetate, and ketones are preferred substrates of the heart during normothermic perfusion, but cannot replete the tricarboxylic acid (TCA) cycle directly. Propionate, an anaplerotic substrate, can replenish TCA cycle intermediates and may affect cardiac metabolism. The purpose of this study was to determine myocardial substrate preferences during hypothermic machine perfusion and to assess if an anaplerotic substrate was required to maintain the TCA cycle intermediate pool i

Temporal proteome and lipidome profiles reveal hepatitis C virus-associated reprogramming of hepatocellular metabolism and bioenergetics.

Proteomic and lipidomic profiling was performed over a time course of acute hepatitis C virus (HCV) infection in cultured Huh-7.5 cells to gain new insights into the intracellular processes influenced by this virus. Our proteomic data suggest that HCV induces early perturbations in glycolysis, the pentose phosphate pathway, and the citric acid cycle, which favor host biosynthetic activities supporting viral replication and propagation. This is followed by a compensatory shift in metabolism aimed at maintaining energy homeostasis and cell viability during elevated viral replication and increasi…

Case Report: Mitochondrial insufficiency underlies cholinergic failure in post-vaccination long COVID: a candidate treatment protocol.

An 80-year-old male with severe long COVID developed acute deterioration following COVID-19 vaccination despite ongoing cholinergic therapy (Mestinon 90 mg daily). A comprehensive mitochondrial support protocol targeting NAD+ repletion, electron transport chain function, and oxidative stress was initiated, with all components delivered in fresh-squeezed orange juice. Clinical response was dramatic within 48 h, with sustained functional recovery over 6 weeks. Challenge-rechallenge testing demonstrated rapid symptom recurrence and resolution with three key observations: missed doses, wrong compo

Multi-omics profiling uncovers metabolic regulation in cumulus cells during oocyte maturation.

Bidirectional signalling between the oocyte and surrounding cumulus cells is essential for coordinating follicular development and acquiring oocyte competence. The metabolic networks governing bovine cumulus cell function, however, remain poorly characterized. To address this gap, we performed integrated metabolomic and proteomic analyses of cumulus cells isolated from cumulus-oocyte complexes at two critical maturation stages: (1) meiotically arrested germinal vesicle stage (0 h) and (2) metaphase II stage (24 h). This temporal profiling reveals dynamic metabolic reprogramming events supporti

Copper homeostasis and cuproptosis in tumor biology: mechanistic insights and clinical perspectives.

Copper is considered a necessary trace element that plays crucial roles in energy production in mitochondria, redox regulation, and metabolism. Copper homeostasis disturbance may promote tumor initiation and progression and therapy resistance. Cuproptosis is a regulated form of cell death when mitochondrial copper interacts with the proteins from the citric acid cycle causing their oligomerization and aggregation, loss of Fe-S proteins, proteotoxic stress, and cell death. Unlike apoptosis, ferroptosis, and necroptosis, cuproptosis can be promoted in cells keeping functioning lipoylation of pro…

An easily overlooked cause of pulmonary arterial hypertension-thiamine deficiency.

Thiamine deficiency (TD), also known as vitamin B1 deficiency, is an often overlooked potential cause of pulmonary arterial hypertension (PAH). It may contribute to the pathological process of PAH through the deficiency of related coenzymes, thereby inhibiting the citric acid cycle. TD can manifest in various clinical forms, including dry beriberi and Shoshin beriberi, the latter characterized by high-output heart failure and lactic acidosis, which can lead to fatal outcomes if not promptly treated. This article reviews the physiological functions of thiamine, the pathophysiological mechanisms…

Late-onset Alzheimer's disease is associated with inherent changes in bioenergetics profiles.

Body-wide changes in bioenergetics, i.e., energy metabolism, occur in normal aging and disturbed bioenergetics may be an important contributing mechanism underlying late-onset Alzheimer's disease (LOAD). We investigated the bioenergetic profiles of fibroblasts from LOAD patients and healthy controls, as a function of age and disease. LOAD cells exhibited an impaired mitochondrial metabolic potential and an abnormal redox potential, associated with reduced nicotinamide adenine dinucleotide metabolism and altered citric acid cycle activity, but not with disease-specific changes in mitochondrial …

Molecular analysis and prenatal diagnosis of human fumarase deficiency.

Fumarase deficiency is a rare autosomal recessive disorder of the citric acid cycle causing severe neurological impairment. The cDNA for both the rat and human enzymes has been cloned previously and shown to encode a coding region of 1.46 kb. To scan for mutations in fumarase-deficient patients we amplified the coding region of fumarase from fibroblast/lymphoblast cDNA employing the oligonucleotide primers designed from the published human and rat cDNA sequence. We then directly sequenced the polymerase chain reaction product. In seven unrelated patients, we detected four missense mutations (A…

In Search of Drug Targets for Mycobacterium tuberculosis.

Mycobacterium tuberculosis is the etiological agent for tuberculosis in humans. The studies related to survival of this pathogen in the human host and development of drugs against reveal that the organism uses a complex physiology to adapt to the host environment. Many studies were targeted to key enzymes that allow this pathogen to either survive or remain latent within the host. Most of the models, which address the survival of pathogen, have evaluated limited dissolved oxygen and prevailing stress conditions. Hence, the truncated citric acid cycle, with the glyoxylate shunt was suggested as…

Pyruvate carboxylase is critical for biofilm formation in Mycobacterium tuberculosis.

Mycobacterium tuberculosis (Mtb) forms biofilms. Biofilm formation is critical in Mtb's virulence. The metabolic pathways governing mycobacterial biofilm formation remain largely unexplored. This study evaluated how different carbon sources influence Mtb biofilm formation. Fermentable substrates-glucose, glycerol, and pyruvate-significantly promoted thick, mature biofilm formation compared to non-fermentable alternatives. In this study, we analyzed the role of pyruvate carboxylase (Pca), an anaplerotic enzyme that converts pyruvate to oxaloacetate, in biofilm formation, as it regulates carbon

Copper Oxide Nanoparticles Impair Mouse Preimplantation Embryonic Development through Disruption of Mitophagy-Mediated Metabolism.

Copper oxide nanoparticles (CuONPs) have been widely applied, posing potential risks to human health. Although the toxicity of CuONPs on the liver and spleen has been reported, their effects on reproductive health remain unexplored. In this study, we investigate the effects of CuONPs on embryonic development and their potential mechanisms. Our results demonstrate that CuONPs exposure impairs mouse preimplantation embryonic development, particularly affecting the morula-to-blastocyst transition. Additionally, CuONPs were found to reduce the pluripotency of the inner cell mass (ICM) and mouse em

Ketogenic β-hydroxybutyrate regulates β-hydroxybutyrylation of TCA cycle-associated enzymes and attenuates disease-associated pathologies in Alzheimer's mice.

Lysine β-hydroxybutyrylation (Kbhb) is a post-translational modification that has recently been found to regulate protein functions. However, whether and how protein Kbhb modification participates in Alzheimer's disease (AD) remains unknown. Herein, we carried out 4D label-free β-hydroxybutylation quantitative proteomics using brain samples of 8-month-old and 2-month-old APP/PS1 AD model mice and wild-type (WT) controls. We identified a series of tricarboxylic acid (TCA) cycle-associated enzymes including citrate synthase (CS) and succinate-CoA ligase subunit alpha (SUCLG1), whose Kbhb modific
