Clinical, Molecular, and Computational Analysis in Patients With a Novel Double Mutation and a New Synonymous Variant in MeCP2: Report of the First Missense Mutation Within the AT-hook1 Cluster in Rett Syndrome.

Rett syndrome is an X-linked neurodevelopmental disorder, primarily caused by MECP2 mutations. In this study, clinical, molecular and bioinformatics analyses were performed in Rett patients to understand the relationship between MECP2 mutation type and the clinical severity. Two double MeCP2 mutations were detected: a novel one (p.G185 V in cis with p.R255X) in P1 and a known one (p.P179 S in cis with p.R255X) in P2. Besides, a novel synonymous mutation (c.807C>T; p.G269G), which could affect mRNA splicing, was identified in P3. The results from clinical severity analysis have shown that P1 wa

Computer-aided molecular modeling study of enantioseparation of iodiconazole and structurally related triadimenol analogues by capillary electrophoresis: chiral recognition mechanism and mathematical model for predicting chiral separation.

Chiral separation of iodiconazole, a new antifungal drug, and 12 new structurally related triadimenol analogues had been developed by capillary electrophoresis (CE) using hydroxypropyl-γ-cyclodextrin (HP-γ-CD) as the chiral selector. The effect of structural features of analytes on Δt and R(s) was studied under the optimum separation conditions. Using molecular docking technique and binding energy calculations, the inclusion process between HP-γ-CD and enantiomers was investigated and chiral recognition mechanisms were discussed. The results suggest that hydrogen bonding between fluorine at po

Markovian chemicals "in silico" design (MARCH-INSIDE), a promising approach for computer aided molecular design II: experimental and theoretical assessment of a novel method for virtual screening of fasciolicides.

A novel method for in silico selection of fluckicidal drugs is introduced. Two QSARs that permit us to discriminate between fasciolicide and non-fasciolicide drugs (the first) and to outline some conclusions about the possible mechanism of action of a chemical (the second) are performed. The first model correctly classified 93.85% of compounds in the training series and 89.5% of the compounds in the predicting one. This model correctly classified 87.7, 93.8, 92.2 and 93.9% of compounds in leave- n-out cross validation procedures when n takes values from 2 to until 6. The model seems to be stab

A multistage computational pipeline integrating ligand-based and structure-based screening with ADMET evaluation, molecular dynamic simulation and free energy calculations for the discovery of potential TNIK inhibitors.

Dysregulation of metabolic signalling pathways, such as Wnt/β-catenin, is a hallmark of various cancers including colorectal cancer, lung squamous cell carcinoma, papillary thyroid carcinoma, multiple myeloma etc. TRAF2 and NCK-interacting kinase (TNIK), a serine/threonine kinase, plays a critical role in this pathway by phosphorylating TCF4, thereby promoting transcription of oncogenic genes linked to cell proliferation, stemness, and therapeutic resistance. Owing to its central role across multiple tumor types, TNIK is a promising target for small-molecule drug development. This study utiliz

Large-scale photonic network with squeezed vacuum states for molecular vibronic spectroscopy.

Although molecular vibronic spectra generation is pivotal for chemical analysis, tackling such exponentially complex tasks on classical computers remains inefficient. Quantum simulation, though theoretically promising, faces technological challenges in experimentally extracting vibronic spectra for molecules with multiple modes. Here, we propose a nontrivial algorithm to generate the vibronic spectra using states with zero displacements (squeezed vacuum states) coupled to a linear optical network, offering ease of experimental implementation. We also fabricate an integrated quantum photonic mi

Chromatin-associated condensates as an inspiration for the system architecture of future DNA computers.

The genome stores and processes approximately 1.5 gigabytes of encoded information. In this article, we propose that the eukaryotic genome and its adaptable three-dimensional packing in the form of chromatin offer a valuable template for the system architecture of DNA-based digital computers. We examine embryonic and stem cells, which exhibit distinct chromatin-associated condensates enriched in transcription machinery. These dynamic biomolecular condensates facilitate the spatial association of genes, genomic control elements, and molecular machinery responsible for reading the genomic code.

Pilot feasibility study investigating 68 Ga-NODAGA-RGD PET-computed tomography molecular angiogenesis imaging in patients with peripheral arterial diseases: can it be of potential clinical value?

INTRODUCTION: Peripheral arterial disease (PAD) is a common vascular condition caused mainly by atherosclerosis that can cause ischemia and increase cardiovascular risk. Noninvasive imaging, such as Doppler ultrasonography, provides hemodynamic information but lack molecular insight. Integrin αvβ3 expression reflects angiogenesis in ischemic tissue and radiolabeled RGD peptides can be used to visualize this process noninvasively. This study prospectively assessed the diagnostic performance of 68 Ga-NODAGA-RGD PET-computed tomography (CT) in patients with suspected or proven PAD and correlate t

Computational prediction of potential aggravating mechanisms of polyethylene terephthalate microplastics in diabetic foot ulcers: An integrated in silico approach combining network toxicology, bioinformatics, machine learning, and molecular dynamics simulations.

The increasing incidence of diabetic foot ulcer (DFU) and growing recognition of environmental pollutants have highlighted polyethylene terephthalate microplastics (PET-MP) as a potential metabolic disease trigger. However, the molecular mechanisms linking PET-MP to DFU remain unclear. This study employed integrated network toxicology and bioinformatics to decipher these mechanisms. PET-MP toxicity targets were screened using SwissTargetPrediction and ChEMBL, and DFU-related differentially expressed genes were obtained from GSE199939 and GSE134431. Deciphering early molecular responses to aristolactam I associated with hepatocellular carcinoma: Computational prediction of a core gene signature and identification of transcription-translation uncoupling under acute aristolactam I exposure.

Aristolochic acid I (AAI) is a potent hepatocarcinogen mainly activated to aristolactam I (ALI). The molecular mechanisms driving ALI‑associated hepatocellular carcinoma (HCC), particularly post‑transcriptional events linking acute liver injury to malignant transformation, remain poorly defined. Here, we integrated pharmacological ALI-target prediction, HCC transcriptomic datasets, weighted gene co-expression network analysis (WGCNA), and SHAP‑based interpretable machine learning to establish a ten-gene signature for ALI-related HCC. Signature dysregulation was validated in a chronic AAI‑carbo

Computational identification of promising genetic markers associated with molecular mechanisms of reduced rice resistance to Rhizoctonia solani under excess nitrogen fertilization using gene network reconstruction and analysis methods.

Although nitrogen fertilizers increase rice yield, their excess can impair plant resistance to diseases, particularly sheath blight caused by Rhizoctonia solani. This pathogen can destroy up to 50 % of the crop, but the mechanisms underlying reduced resistance under excess nitrogen remain poorly understood. This study aims to identify potential marker genes to enhance rice resistance to R. solani under excess nitrogen conditions. A comprehensive bioinformatics approach was applied, including differential gene expression analysis, gene network reconstruction, biological process overrepresentati
