An Enhanced Long-Term Wet Adhesion Strategy of Spatial Control the Emergence of Dual Covalent Cross-Linking for Sutureless Cornea Transplant.

Corneal transplantation regeneration requires bioadhesives to perform long-term and stable adhesion functions in a wet environment. However, many current studies focus on the instantaneous or short-term adhesion persistence of bioadhesives, and ignore the evaluation of their long-term wet adhesion behaviors which is urgent for keratoplasty repair process. In view of this situation, a dual covalent cross-linking hydrogel (ASO) bioadhesive is developed. The ASO bioadhesive comprised acrylated gelatin(G-AA), thiolated gelatin(G-SH), and oxidized dextran (OD). Introduction of thiol chemistry made

Cross-Linked Zwitterionic Surface Modifications for Biocompatible Blood-Contacting Medical Devices.

Blood-contacting medical devices, such as vascular catheters, heart valves, membrane oxygenators, or dialysis membranes, enable the treatment and long-term care of countless people suffering from cardiovascular, respiratory, and renal diseases. These devices are largely constructed from polymeric materials, and despite advancements in material biocompatibility, remain susceptible to adverse biological processes such as thrombosis, inflammation, and calcification, introducing risks to patient safety and constraining long-term device performance. This article builds on previous research that int

Clickable Multitargeting Cross-Linkers Based on SuFEx for In Vivo Cross-Linking Mass Spectrometry.

Cross-linking mass spectrometry (XL-MS) is a powerful technique to study protein structures and protein-protein interactions. The coverage of amino acids determines the depth of the XL-MS analysis and accuracy of the structural resolution. In this work, we designed and synthesized a pair of novel trifunctional cross-linkers, succinimidyl-propargyl-aryl sulfonyl fluoride (SPSF), for multitargeting cross-linking in vivo. The highly reactive succinimide ester reacts rapidly with Lys residues first; then, the less reactive sulfonyl fluoride reacts with multiple nucleophilic amino acid side chains

Optimized microfluidic blow spinning process for single-step manufacturing of crosslinked chitosan nanofilaments using a quality-by-design approach.

Nanofibrous patches composed of chitosan (CS) present promising solutions for wound healing by supporting cell growth and enabling controlled drug release. This study introduces a new single-step microfluidic blow-spinning process that blends CS with polyethylene oxide (PEO) and crosslinks it with tripolyphosphate (TPP) using a series of laminar flow chips integrated into a blow-spinning apparatus. The microfluidic mixing between PEO and CS solutions at different concentrations was simulated with computational fluid dynamics (CFD). This manufacturing process was optimized through a Design of E

Polymerization-Induced Self-Assembly Providing PEG-Gels with Dynamic Micelle-Crosslinked Hierarchical Structures and Overall Improvement of Their Comprehensive Performances.

Polymer gels are fascinating soft materials and have become excellent candidates for wearable electronics, biomedicine, sensors, etc. Synthetic gels usually suffer from poor mechanical properties, and integrating good mechanical properties, adhesiveness, stability, and self-healing performances in one gel is more difficult. Herein, polymerization-induced self-assembly (PISA) providing PEG-gels with an overall improvement in their comprehensive performances is reported. PISA synthesis is carried out in PEG (solvent) to efficiently produce various nanoparticles, which are used as the nanofillers

Measurement of Covalent Bond Formation in Light-Curing Hydrogels Predicts Physical Stability under Flow.

Photo-crosslinking hydrogels are promising for tissue engineering and regenerative medicine, but challenges in reaction monitoring often leave their optimization subject to trial and error. The stability of crosslinked gels under fluid flow, as in the case of a microfluidic device, is particularly challenging to predict, both because of obstacles inherent to solid-state macromolecular analysis that prevent accurate chemical monitoring and because stability is dependent on size of the patterned features. To solve both problems, we obtained 1H NMR spectra of cured hydrogels which were enzymatica

Efficacy and safety of microbial transglutaminase-induced scleral stiffening invivo.

The purpose of this study was to investigate the efficacy and safety of microbial transglutaminases (mTGases) during scleral collagen cross-linking (CXL) in vivo. Sixteen New Zealand white albino rabbits were treated with sub-Tenon's injections of 2 ml of 1 U/ml mTGases in the right eye and 2 ml of phosphate buffer saline (PBS) in the left eye. The rabbits were killed 2 weeks after the injection, and all eyeballs, including some scleral strips, were processed. The elastic modulus was measured with a biomaterials tester. Histopathological analysis and transmission electron microscopy (TEM) were

Dialdehyde xanthan gum and curcumin synergistically crosslinked bioprosthetic valve leaflets with anti-thrombotic, anti-inflammatory and anti-calcification properties.

Currently commercial glutaraldehyde (GA)-crosslinked bioprosthetic valve leaflets (BVLs) suffer from thromboembolic complications, calcification, and limited durability, which are the major stumbling block to wider clinical application of BVLs. Thus, developing new-style BVLs will be an urgent need to enhance the durability of BVLs and alleviate thromboembolic complications. In this study, a quick and effective collaborative strategy of the double crosslinking agents (oxidized polysaccharide and natural active crosslinking agent) was reported to realize enhanced mechanical, and structural stab

Comparison of morphological changes of corneal collagen fibers treated with collagen crosslinking agents using second harmonic generation images.

Corneal cross-linking (CXL) is a common surgical procedure used to modify corneal biomechanics and stabilize keratoconus progression which is still under discussion. Its side effects, which are mostly related to anatomical unpredictability and stromal exposure, are the reason for the search for new CXL agents. In this work we have quantitatively evaluated the porcine corneal stroma architecture treated with collagen crosslinking agents such as riboflavin solutions and açai extract, using second harmonic generation microscopy. Aimed at evaluating the morphological changes in the corneal stroma

Stretchable hydrogels with low hysteresis and anti-fatigue fracture based on polyprotein cross-linkers.

Hydrogel-based devices are widely used as flexible electronics, biosensors, soft robots, and intelligent human-machine interfaces. In these applications, high stretchability, low hysteresis, and anti-fatigue fracture are essential but can be rarely met in the same hydrogels simultaneously. Here, we demonstrate a hydrogel design using tandem-repeat proteins as the cross-linkers and random coiled polymers as the percolating network. Such a design allows the polyprotein cross-linkers only to experience considerable forces at the fracture zone and unfold to prevent crack propagation. Thus, we are

Accurate Measurement of Formaldehyde-Induced DNA-Protein Cross-Links by High-Resolution Orbitrap Mass Spectrometry.

Genomic instability caused by DNA-protein cross-link (DPCs)-induced DNA damage is implicated in disease pathogenesis, aging, and cancer development. The covalent linkages between DNA and protein are induced by chemical reactions catalyzed by the endogenous metabolic intermediates and exogenous agents, such as aldehydes, chemotherapeutic agents, and ionizing radiation. Formaldehyde has been classified as a genotoxic carcinogen. In addition, endogenous formaldehyde-induced DPCs may increase the risks of bone marrow toxicity and leukemia. There is a need to develop an effective detection method f

Instrumented indentation for determination of mechanical properties of human cornea after ultraviolet-A crosslinking.

UVA crosslinking is used for treatment of corneal diseases such as keratoconus in order to stabilize the corneal tissue by crosslinking of the collagen fibers. It has been shown that the crosslinking treatment leads to a stiffening of the central corneal tissue. However, knowledge of lateral extent of the corneal stiffening as well as a systematic study of the mechanical response of human cornea is still missing. In our study we measured the stiffness (elastic modulus) of the anterior surface of healthy and crosslinked human corneas by instrumented indentation using a spherical indenter. The r

Influence of Aspartate Moieties on the Self-Healing Behavior of Histidine-Rich Supramolecular Polymers.

Aspartate incorporated into the protein structure of mussel byssal threads is believed to play an important role, besides the reversible histidine-zinc interactions, in the self-healing behavior of mussel byssal threads. Therefore, copolymers containing both aspartate and histidine moieties are synthesized in order to investigate the influence of aspartate on the complexation of zinc(II) as well as on the self-healing behavior and the mechanical properties of the resulting supramolecular networks. For this purpose, isothermal titration calorimetry measurements of a model aspartate compound as
