Diagnosis and management of solid pseudopapillary tumours of the pancreas.

The finding of a solid or cystic mass in the pancreas is becoming more common secondary to the increasing use of cross-sectional imaging and the improved sensitivity of such studies for the detection of pancreatic abnormalities. Because of the aggressive natural history of pancreatic cancer, this has caused concern that all pancreatic abnormalities may be cancer as well as confusion over proper diagnostic and treatment algorithms. This review provides an overview of the natural history, diagnostic considerations, and treatment recommendations for the less common tumor of the pancreas which can

In Silico Validation of OncoOrigin: An Integrative AI Tool for Primary Cancer Site Prediction with Graphical User Interface to Facilitate Clinical Application.

Cancers of unknown primary (CUPs) represent a significant diagnostic and therapeutic challenge in the field of oncology. Due to the limitations of current diagnostic tools in these cases, novel approaches must be brought forward to improve treatment outcomes for these patients. The objective of this study was to develop a machine-learning-based software for primary cancer site prediction (OncoOrigin), based on genetic data acquired from tumor DNA sequencing. By design, this was an in silico diagnostic study, conducted using data from the cBioPortal database (accessed on 21 September 2024) and

Free-ranging avifauna as a source of generalist parasites for captive birds in zoological settings: An overview of parasite records and potential for cross-transmission.

Captive birds in zoological settings often harbor parasites, but little information is available about the potential for free-ranging avifauna to act as a source of infection. This review summarizes the gastrointestinal parasites found in zoo birds globally and in seven common free-ranging avian species [mallard (Anas platyrhynchos), Eurasian blackbird (Turdus merula), common starling (Sturnus vulgaris), Eurasian jackdaw (Corvus monedula), house sparrow (Passer domesticus), European robin (Erithacus rubecula), and rock dove (Columba livia)] to identify the overlap and discuss the potential for

Essential tremor then and now: How views of the most common tremor diathesis have changed over time.

BACKGROUND: Essential tremor (ET) is the most prevalent tremor diathesis. In this "then" and "now" piece, I detail how views of this disorder have changed over time. METHODS: A PubMed search was conducted on June 15, 2017. The term "essential tremor" was crossed in sequential order with 14 s search terms (e.g., genetics, clinical). RESULTS: The traditional view of ET was that it was monosymptomatic. An emerging view of ET is as a more clinically-complex entity with a range of possible motor and non-motor features. Traditionally, ET was viewed as autosomal dominant with complete penetrance by

The Spectrum of Movement Disorders in Children with Genetic Developmental and Epileptic Encephalopathies: A Cross-Sectional Observational Study.

BACKGROUND: Movement disorders (MDs) often remain underexplored in genetic developmental and epileptic encephalopathies (DEEs), largely due to limited awareness, phenotypic heterogeneity and the considerable burden of drug-resistant epilepsy. OBJECTIVES: This study aimed to characterize the etiological profile of genetic DEEs with coexisting MDs, delineate the spectrum of MDs and explore associations between movement disorder phenomenology and underlying genetic variants. METHODS: A single-center cross-sectional observational study was conducted over 1 year (Jan 2024-Jan 2025) at a tertiary

Genetic mutation patterns in Indonesian children with primary steroid-resistant nephrotic syndrome.

BACKGROUND: Approximately 10-30% of pediatric cases of steroid-resistant nephrotic syndrome (SRNS) have pathogenic monogenic variants, particularly in primary SRNS. Identifying these variants aids in predicting outcomes, guiding treatment, and enabling genetic counseling. In Indonesia, data on genetic mutation patterns in pediatric SRNS are still limited. METHODS: A cross-sectional study was conducted using whole-exome sequencing (WES) to detect variants of genetic origin in children diagnosed with primary SRNS at Indonesia's National Referral Hospital, Cipto Mangunkusumo. We assessed the rel

Urinary metabolic age from high-resolution nuclear magnetic resonance spectroscopy reveals longitudinal aging patterns.

Biological age captures inter-individual heterogeneity in aging process arising from genetic and environmental influences. Metabolites, as the end-products of metabolism, integrate these factors and are therefore well suited for biological age estimation. Urinary metabolomics, in particular, provides a non-invasive and information-rich matrix for assessing systemic metabolic states. We applied different machine learning techniques to develop a biological age score from high-resolution 1H nuclear magnetic resonance metabolites measured in urine samples from a large population-based cohort. The

Genetic predisposition to hand grip strength predicts cognitive decline.

Hand grip strength (HGS) is a predictor of cognitive decline in aging. To understand the direct and indirect mechanisms underlying this association, we performed integrative analyses of genetic, cognitive, and autopsy data. A polygenic risk score for HGS (PRSHGS) was calculated in two independent studies of aging (total n = 25,227). Cross-sectional and longitudinal modeling, including mediation with physical activity, assessed the effects of PRSHGS on cognitive performance and postmortem neuropathology. In both cohorts, higher PRSHGS predicted better global cognitive performance (p = 7.77 × 10

Functional deficiency of fibroblasts heterozygous for Bloom syndrome as specific manifestation of the primary defect.

The effect on the rate of sister chromatid exchanges (SCEs) in Bloom syndrome fibroblasts by cocultivation with Fanconi anemia and xeroderma pigmentosum fibroblasts and with Bloom syndrome heterozygotes was studied. Cells of Fanconi anemia and xeroderma origin reduced the rate of SCEs in Bloom cells by about 45%-50%, just as control cells do. In contrast, heterozygous Bloom cells reduced the rate of SCEs by only 16%-28%. In absolute figures, Fanconi cells reduced the mean rate of SCE in Bloom cells from 55.7 +/- 5.50- to 27.7 +/- 6.44, xeroderma cells to 30.5 +/- 5.73, and control cells to 28.

DNA repair defects and chromosome instability disorders.

Xeroderma pigmentosum (XP), Fanconi anaemia (FA), ataxia telangiectasia (AT) and Bloom disease (BS) are four rare autosomal recessive disorders in which there is defective DNA repair and/or chromosome instability and proneness to malignancy. Between 80 and 90% of patients with XP have a defect, demonstrable at cell level, of excision of DNA lesions induced by ultraviolet rays, while the remainder have a cellular error of post-replication repair. XP cells are also deficient in repairing DNA damage caused by a variety of chemical mutagens. There are at least five different complementation groups

Bayesian methods for fitting mixture models that characterize branching tree processes: An application to development of resistant TB strains.

For pathogens that must be treated with combinations of antibiotics and acquire resistance through genetic mutation, knowledge of the order in which drug-resistance mutations occur may be important for determining treatment policies. Diagnostic specimens collected from patients are often available; this makes it possible to determine the presence of individual drug resistance-conferring mutations and combinations of these mutations. In most cases, these specimens are only available from a patient at a single point in time; it is very rare to have access to multiple specimens from a single pati

Evolution of resistance to Rhynchosporium secalis (Oud.) Davis in barley composite cross II.

Changes in resistance to scald disease which occurred in barley composite Cross II over 45 generations were analyzed genetically. This population, which was synthesized in 1929 by pooling equal numbers of f1 seeds from 378 pair wise crosses among 28 barley varieties, has subsequently been grown at Davis, California under standard agricultural conditions without conscious selection. Progenies derived from self-pollinated seeds from random plants taken from four generations (F8, F13, F23, and F45) were tested against four different races of scald (40, 61, 72, and 74), and rated as resistant, sus

The Association of Wnt Signaling Pathway Gene Variants, Blood Lipoproteins and Cognitive Function in Elderly People.

Wnt signaling is believed to play an important role in the nervous system. However, few studies have examined the association between gene variants of the Wnt signaling pathway and mild cognitive impairment (MCI). Additionally, the potential modulation of this association by blood lipoproteins remains poorly understood. We aimed to investigate these associations in the present analysis. The cross-sectional study comprised 459 participants from 17 villages in Jimo District, Qingdao, Shandong Province. A total of 46 single nucleotide polymorphisms (SNPs) in nine Wnt signaling pathway genes were

"Balanced" karyotypes in six abnormal offspring of balanced reciprocal translocation normal carrier parents.

Among 6800 consecutive blood samples studied for clinical cytogenetic diagnosis, we identified 30 families in which one parent of the proband had a balanced reciprocal autosomal translocation (excluding Robertsonian rearrangements). Twenty-eight of the 30 families had a malformed and/or mentally retarded proband: 19 with an unbalanced derived chromosome, 3 with abnormalities involving chromosomes other than those in the translocation, 5 with a "balanced" reciprocal translocation, and 1 with a normal karyotype. We hypothesize that the latter 6 affected probands with "balanced" karyotypes could

Nonhomologous chromatid exchange in hereditary and sporadic renal cell carcinomas.

For the development of renal cell carcinomas, it has been suggested that a germ-line or somatic mutation occurs on one of the homologous chromosomes 3p, and subsequently the other 3p segment is lost. We have examined the karyotype and/or the allelic combination on chromosomes 3 and 5 by restriction fragment length polymorphism analysis in normal kidney and tumor samples from 28 renal cell carcinomas that developed in two patients with von Hippel-Lindau disease; we then compared the results to those of sporadic tumors. An unbalanced translocation between chromosome 3p and 5q or other chromosome

Whole genome resequencing reveals diagnostic markers for investigating global migration and hybridization between minke whale species.

BACKGROUND: In the marine environment, where there are few absolute physical barriers, contemporary contact between previously isolated species can occur across great distances, and in some cases, may be inter-oceanic. An example of this can be seen in the minke whale species complex. Antarctic minke whales are genetically and morphologically distinct from the common minke found in the north Atlantic and Pacific oceans, and the two species are estimated to have been isolated from each other for 5 million years or more. Recent atypical migrations from the southern to the northern hemisphere hav

Development of sub-tropically adapted diverse provitamin-A rich maize inbreds through marker-assisted pedigree selection, their characterization and utilization in hybrid breeding.

Malnutrition has emerged as one of the major health problems worldwide. Traditional yellow maize has low provitamin-A (proA) content and its genetic base in proA biofortification breeding program of subtropics is extremely narrow. To diversify the proA rich germplasm, 10 elite low proA inbreds were crossed with a proA rich donor (HP702-22) having mutant crtRB1 gene. The F2 populations derived from these crosses were genotyped using InDel marker specific to crtRB1. Severe marker segregation distortion was observed. Seventeen crtRB1 inbreds developed through marker-assisted pedigree breeding and

Influence of ATG SNPs on hip fracture patients' functional status.

One of the key factors influencing the prognosis of older patients with hip fracture is their previous functional status. Autophagy has been related to bone disease and to osteoporosis onset. Some studies have analyzed the relationship of certain genetic determinants with the risk of suffering a fragility fracture but evidence is lacking regarding the influence of genetic factors on the functional status of hip fracture patients. We aimed to evaluate the relationship between autophagy genes single nucleotide polymorphisms and the functional status in older hip fracture patients. A prospective
