Methyl Jasmonate Was Involved in Hydrogen Sulfide-Alleviated Cadmium Stress in Cucumber Plants Through ROS Homeostasis and Chlorophyll Metabolism.

Cadmium (Cd), as one of the most toxic nonessential elements, severely prohibits plant growth and development. Hydrogen sulfide (H2S) and methyl jasmonate (MeJA) play essential roles in plant response to abiotic stress. However, the potential mechanism of H2S and MeJA in alleviating Cd stress in plants remains unclear. In the current study, the importance and crosstalk of H2S and MeJA in the Cd tolerance of cucumber seedlings have been investigated. Our results revealed that Cd stress obviously prohibited the growth of cucumber seedlings. Optimal concentrations of H2S donor sodium hydrosulfide

A B-Box (BBX) Transcription Factor from Cucumber, CsCOL9 Positively Regulates Resistance of Host Plant to Bemisia tabaci.

B-box (BBX) transcription factors play crucial roles in plant growth, development, and defense responses to biotic and abiotic stresses. In this study, we cloned a BBX transcription factor gene, CsCOL9I, from cucumber and analyzed its role in the plant's defense against the feeding of Bemisia tabaci. CsCOL9 is expressed throughout all developmental stages in cucumber, with the highest expression in the leaves. CsCOL9 is induced by B. tabaci feeding, salicylic acid (SA), methyl jasmonate (MeJA), and hydrogen peroxide (H2O2). Cucumber plants with CsCOL9 silence (TRV2-CsCOL9) and overexpression (

Expression and functional analysis of the transcription factor-encoding Gene CsERF004 in cucumber during Pseudoperonospora cubensis and Corynespora cassiicola infection.

BACKGROUND: Cucumber downy mildew, caused by P. cubensis, is an important leaf disease that can severely affect cucumber production. In recent years, cucumber target spot, caused by C. cassiicola, has been reported in both Asia and Europe and is now considered as a major disease disrupting cucumber production. Single-disease-resistant cucumber varieties have been unable to satisfy production needs. To explore the molecular mechanisms of cucumber resistance to these two diseases, cucumber cultivars D9320 (resistant to downy mildew and target spot) and D0401 (susceptible to downy mildew and targ

Synthetic ultrashort cationic lipopeptides induce systemic plant defense responses against bacterial and fungal pathogens.

A new family of synthetic, membrane-active, ultrashort lipopeptides composed of only four amino acids linked to fatty acids was tested for the ability to induce systemic resistance and defense responses in plants. We found that two peptides wherein the third residue is a d-enantiomer (italic), C16-KKKK and C16-KLLK, can induce medium alkalinization of tobacco suspension-cultured cells and expression of defense-related genes in cucumber and Arabidopsis seedlings. Moreover, these compounds can prime systemic induction of antimicrobial compounds in cucumber leaves similarly to the plant-beneficia

Spent mushroom substrate extracts as natural-substance biostimulants: Effects on growth, chlorophyll and root architecture of cucumber.

Spent mushroom substrate (SMS) is an abundant agro-industrial by-product rich in transformed organic matter, nutrients and potentially bioactive compounds, which makes it a promising raw material for biostimulant production. However, the plant-growth-promoting potential of chemically obtained alkaline SMS extracts, particularly under different application routes, remains insufficiently investigated. This study tested whether an alkaline extract of spent button mushroom substrate stimulates the growth and physiology of cucumber (Cucumis sativus L.). SMS was extracted with 10% potassium hydroxid…

Biochar enhances cucumber production by modulating rhizosphere microbiota and soil metabolites under continuous cropping systems.

Biochar, a soil amendment with diverse regulatory functions, has been widely applied to enhance soil conditions. However, its underlying mechanism for alleviating continuous cropping obstacles, from the perspective of rhizosphere microbe-metabolite-plant coupling, remains to be further elucidated. Using cucumber (Cucumis sativus L.) as the model crop, this study explored the rhizosphere-mediated effects of biochar application under continuous cropping conditions via the analytical methods of metagenomics and metabolomics. Six biochar application rates (0, 5, 10, 20, 30, and 40 t ha-¹) were tes…

The protective role of chloroplast NADH dehydrogenase-like complex (NDH) against PSI photoinhibition under chilling stress.

Chilling stress induces photosystem I (PSI) photoinhibition in various plants, severely impairing their growth. However, the mechanisms suppressing chilling-induced PSI photoinhibition remain unclear. To identify factors preventing PSI photoinhibition, we compared two cucumber cultivars with different susceptibilities to PSI photoinhibition and chilling stress tolerance. Chilling stress caused uncoupling of thylakoid membrane in both cultivars, as previously reported. However, in the chilling-sensitive cultivar, electron efflux from ferredoxin (Fd) was more restricted under chilling stress, re

Virulence assessment of Listeria monocytogenes grown in different foods using a Galleria mellonella model.

Various produce including cantaloupe, caramel-coated apples, and packaged salads, have been recognized in recent years as vehicles for listeriosis, a human foodborne disease caused by intracellular pathogen Listeria monocytogenes. Our knowledge regarding the role of these foods in L. monocytogenes virulence, however, is limited. Understanding their role in modulating L. monocytogenes virulence can be useful in risk assessments and for developing control measures. In this study, we employed the Galleria mellonella larvae model to evaluate virulence potential of fifteen clinical, environmental a

Hydrogen Sulfide Improves the Cold Stress Resistance through the CsARF5-CsDREB3 Module in Cucumber.

As an important gas signaling molecule, hydrogen sulfide (H2S) plays a crucial role in regulating cold tolerance. H2S cooperates with phytohormones such as abscisic acid, ethylene, and salicylic acid to regulate the plant stress response. However, the synergistic regulation of H2S and auxin in the plant response to cold stress has not been reported. This study showed that sodium hydrosulfide (NaHS, an H2S donor) treatment enhanced the cold stress tolerance of cucumber seedlings and increased the level of auxin. CsARF5, a cucumber auxin response factor (ARF) gene, was isolated, and its role in

Survival and growth of Listeria monocytogenes on whole cantaloupes is dependent on site of contamination and storage temperature.

Whole cantaloupes (Cucumis melo L.), marketed as 'Rocky Ford', were implicated in a large multi-state outbreak of listeriosis in the United States in 2011; however, survival and growth of Listeria monocytogenes on whole cantaloupes remains relatively unexplored. The research presented here evaluated three different storage temperatures, two sites of contamination of cantaloupes, and two cantaloupe varieties to determine their effect on the survival of L. monocytogenes. 'Athena' and 'Rocky Ford' cantaloupe cultivars were grown in soil and harvested, and individual melons subsequently received a…

Induction of defense responses in cucumber plants by using the cell-free filtrate of the plant growth-promoting fungus Penicillium simplicissimum GP17-2.

Penicillium simplicissimum GP17-2 is a plant growth-promoting fungus (PGPF) and an inducer of systemic defense responses. The mechanisms underlying the effect of GP17-2 on the reduction of cucumber leaf damage caused by the anthracnose pathogen Colletotrichum orbiculare were investigated. Cucumber leaves treated with the culture filtrate (CF) of GP17-2 exhibited a clear systemic resistance against subsequent infection with C. orbiculare. The number and size of lesions caused by the disease were reduced in CF-treated plants, in comparison with that in the control plants. The results showed that
