Recent advances in defining the molecular mechanisms of cell cycle control in eukaryotes provide a basis for better understanding the hormonal control of cell proliferation in normal and neoplastic breast epithelium. It is now clear that a number of critical steps in cell cycle progression are controlled by families of serine/threonine kinases, the cdks. These kinases are activated by interactions with various cyclin gene products which form the regulatory subunits of the kinase complexes. Several families of cyclins control cell cycle progression in G1 phase, cyclins C, D and E, or in S, G2 a…

Germ-line mutations in the von Hippel-Lindau (VHL) tumor suppressor disease are associated with a high risk of retinal and cerebellar hemangioblastomas, renal cell carcinoma (RCC), and, in some cases, pheochromocytoma (PHE). In addition, somatic mutation or epigenetic inactivation of the VHL gene occurs in most clear cell RCCs. VHL protein (pVHL) has a critical role in regulating proteasomal degradation of the HIF transcription factor, and VHL inactivation results in overexpression of many hypoxia-inducible mRNAs including vascular endothelial growth factor (VEGF). To identify novel pVHL targe

Protein kinase C encodes a family of enzymes implicated in cellular differentiation, growth control and tumor promotion. However, very little is known with respect to the molecular mechanisms that link protein kinase C to cell cycle control. Here we report that ectopic expression of PKC eta in NIH3T3 fibroblasts blocks the normal phosphorylation of the Rb protein in quiescent cultures restimulated to enter the cell cycle; PKC eta activates a cellular program that includes increased expression of cyclins E (but not cyclin D), as well as the induced expression of the cyclin-dependent kinase inhi…

Cell cycle transitions are driven by cyclins in complex with cyclin-dependent kinases (CDK). Here, we demonstrate that cyclin B3, an evolutionarily divergent B-type cyclin previously thought to function exclusively in female meiosis, is expressed in mitotic cells and interacts with both CDK1 and CDK2. We show that cyclin B3-CDK1/2 is essential for timely mitotic progression by ensuring proper chromosome alignment and stable kinetochore-microtubule interactions, while its overexpression accelerates mitosis and improves chromosome alignment kinetics. Through phospho-mass spectrometry, we identif…

UNLABELLED: Accumulation of D cyclins during the G1-phase of the cell cycle is a key regulatory step that derepresses the transcriptional program required for DNA replication, facilitating S-phase entry. Among the three D cyclins, cyclin D3 predominates in hematopoietic cells. Accordingly, cyclin D3 mutations are implicated in lymphoid malignancies. In this study, we provided evidence demonstrating a critical role for cyclin D3 in myelopoiesis and further demonstrated the stage-specific degradation of cyclin D3 dictates normal myelopoiesis. In a knock-in mouse model expressing a degradation-re…

Cyclin K is overexpressed in multiple tumors and plays critical roles in tumorigenesis, making it a promising therapeutic target. Although the antitumor potential of Cyclin K-targeting molecular glues, their clinical application is limited by insufficient efficacy and tumor selectivity. Herein, we developed BNNC, a photomolecular glue that releases Cyclin K molecular glue (R)-CR8 and phototherapeutic agent BSS-Et in hypoxic tumor microenvironments, thereby enhancing tumor selectivity. Network pharmacology and Western blotting analysis demonstrated that the combination of (R)-CR8 and BSS-Et syn

KEY POINTS: Loss of podocyte-associated cyclin G-associated kinase resulted in mistrafficking of lysosomal hydrolases. Loss of podocyte-associated cyclin G-associated kinase induced immune modulation.
BACKGROUND: Given the postmitotic nature of podocytes, adapting to both physiologic and pathologic stress is crucial to prevent podocyte loss. An important component of maintaining cellular homeostasis are lysosomes, which are membrane-bound organelles responsible for degradation and recycling of damaged organelles and other macromolecules. Lysosome impairment has been shown to cause cellular an

BACKGROUND: Alterations in key cell cycle regulators are strongly linked to tumorigenesis. Therefore, we hypothesized that polymorphisms of genes encoded cyclin-dependent kinase 4 and 6 (CDK4 and CDK6), cyclin D1 (CCND1), CDK inhibitors p16INK4a and p15INK4b, as well as the retinoblastoma protein (RB), could modulate prostate cancer risk and influence the corresponding mRNA levels.
METHODS AND RESULTS: We evaluated CDK4 rs2069502, CDK6 rs2285332, CCND1 rs9344, p16INK4a rs11515, p15INK4b rs3217986, and RB rs3092904 polymorphisms using TaqMan® SNP Assays in a cohort comprising 532 prostate canc

The cyclin-dependent kinase (CDK)/retinoblastoma protein (RB)/early region 2 binding factor (E2F) axis forms the core transcriptional machinery driving cell cycle progression. Alterations in RB1 or other pathway members occur in many cancers, resulting in heightened oncogenic E2F activity. The activity of E2F is regulated by RxL-mediated binding to the hydrophobic patch (HP) of Cyclin A; blocking this interaction results in the hyperactivation of E2F and synthetic lethality in E2F-driven tumors. While mechanistically differentiated and potentially more selective than blocking CDK activity (e.g

BACKGROUND/OBJECTIVE: Bone remodeling is a critical process for skeletal health, and its dysregulation can lead to diseases like osteoporosis. While several Cyclin-Dependent Kinases (CDKs) are known to regulate bone metabolism, the function of CDK10 remains uncharacterized in this context. Human mutations in CDK10 cause the Al Kaissi syndrome, which involves severe skeletal defects, suggesting a role for CDK10 in bone. The objective of this study was to investigate the functional role of CDK10 in bone remodeling in mice in vivo.
METHODS: The in vitro role of CDK10 was examined in the MC3T3-E1

Hirschsprung disease (HD) is a disorder caused by the failed migration of neural crest cells, resulting in abnormal innervation of the colon. Histologic hallmarks include the absence of ganglion cells and the presence of hypertrophic nerve fibers. At present, an immunostain for calretinin is the most used ancillary study. The supportive staining pattern for HD is the absence of immunoreactive ganglion cells and mucosal nerve fibers (neurites). While studying a patient with mantle cell lymphoma involving the colon, we observed a similar immunoreactive pattern for ganglion cells and neurites wit

Urothelial carcinoma (UC) is the most common malignancy of urinary bladder. It is the 9th leading cause of death worldwide and second most common genitourinary malignancy among male. Incidence is increasing in developing countries like Bangladesh. About 80% of patients are found between 50 to 80 years of age. It is 3-4 times more common in male than in female. Determination of therapeutic strategy and prediction of progression of urothelial carcinoma is a major clinical challenge. Treatment of urothelial carcinoma still now mostly depends on pathological stages. Amplification or genomic altera

BACKGROUND: F-box proteins are essential components of the E3 ubiquitin ligases which are involved in the regulation of almost all life activities such as cell cycle, proliferation, and apoptosis, which have become an important research and drug target. However, there are few studies on F-box and leucine-rich repeat protein 16 (FBXL16) in endometrial carcinoma.
METHODS: Clinical samples were collected for determining the correlation between FBXL16 and endometrial carcinoma. Cells were screened and established with Ishikawa cells which proved the fundamental role of FBXL16 in regulating cell p

A 68-year-old male presents with generalized lymphadenopathy and fever of short duration. Axillary lymph node excision was performed and was sent for histopathological evaluation. Microscopic evaluation of the submitted lymph node revealed diffuse proliferation of intermediate-sized atypical lymphoid cells with round nuclei, irregular membranes, finely dispersed chromatin, and inconspicuous nucleoli. Mitotic figures were frequently seen. Immunohistochemical evaluation revealed diffuse expression of CD20, CD5, CD10, B-cell lymphoma 2 (Bcl2), and B-cell lymphoma 6 (Bcl6). Atypical lymphoid cells

OBJECTIVES: Thymic epithelial tumors are the most common adult mediastinal tumors; however, their growth mechanism remains relatively unknown. Among the JAK/STAT pathway-related proteins, which control various intracellular events, STAT3 is deeply involved in cell proliferation. Constitutive activation of STAT3 and the resulting overexpression of cyclin D1 have been confirmed in various tumors, but have not been thoroughly investigated in thymic epithelial tumors. In this study, we immunohistochemically examined STAT3 activation, cyclin D1 expression, and JAK3 activation in thymic epithelial t
