Plastid Lipase 3 (PLIP3) is a chloroplast phospholipase A that cleaves linolenic acid, a polyunsaturated fatty acid, from the sn-1 position of the chloroplast membrane lipid monogalactosyldiacylglycerol. Linolenic acid is subsequently converted by enzymes in the chloroplast and the peroxisome to jasmonic acid (JA) requiring transport between the organelles. Overexpression of a cDNA encoding PLIP3 resulted in stunted plant growth with altered leaf morphology caused by accumulation of JA and other oxylipin metabolites redirecting the metabolism from growth to defense. We conducted a genetic supp

BACKGROUND: Insect vectors are responsible for transmitting the majority of plant viruses. While the interplays between insect vectors and viruses have been extensively investigated, how other organisms associated with the shared host plants of the insect and virus impact virus transmission dynamics, and the corresponding viral countermeasures, remain largely unknown.
RESULTS: We explored the modulation of whitefly-mediated transmission of geminiviruses by two non-vector herbivores and the role of viral C2 proteins. The infestation of cotton bollworm induced the accumulation of jasmonic acid

Diseases caused by pathogenic microorganisms impair plant growth, leading to reduced crop yields. While the molecular mechanisms of plant disease response are well understood in annual herbaceous species, they remain largely unknown in perennial woody plants. Here, we found that PtoPHL3, a key transcription factor in poplar's phosphorus starvation response, showed significant expression changes after treatments with salicylic acid and methyl jasmonate, and inoculation of Dothiorella gregaria Sacc (D. gregaria) which causes poplar canker disease. Overexpressing PtoPHL3 conferred increased resis

BACKGROUND: Modern agriculture is based on the application of synthetic agrochemicals to control multiple abiotic and biotic stresses. Nevertheless, owing to their negative effect on ecosystems and human health, their application is increasingly restricted. The use of plant growth-promoting bacteria (PGPB) as well as biological control agents (BCAs) represents a sustainable alternative to chemical pesticides and fertilizers. Bacillus velezensis 83 (Bv83) has been described as a BCA against several fungal phytopathogens.
RESULTS: In this work, we further study the molecular mechanisms behind t

Mechanical wounding triggers rapid transcriptional and hormonal reprogramming in plants, primarily driven by jasmonate (JA) signalling. While the role of JA, ethylene, and salicylic acid in wound responses is well characterised, the contribution of strigolactones (SLs) remains largely unexplored. Here, for the first time, it was shown that SLs modulate wound-induced transcriptional dynamics in Arabidopsis thaliana. Using transcriptome profiling of wild-type (Columbia-0) and the SL biosynthesis mutant more axillary growth3 (max3), a discrete cohort of genes whose wound induction is SL-dependent

Plants perceive signals associated with herbivore eggs and in response initiate ovicidal defence. However, which phytohormone pathways regulate this defence and which defensive compounds dominate it remains largely unknown. Here, we found that the hatching rate of eggs of white-backed planthopper (WBPH) Sogatella furcifera was significantly lower on a japonica rice variety P81 than an indica rice variety NB44. When infested by WBPH, P81 plants showed higher jasmonic acid (JA) and abscisic acid (ABA) responses than did NB44 plants; moreover, P81 plants produced the ovicidal compound benzyl benz

Helicobacter pylori is a Gram-negative bacteria responsible for gastrointestinal disorders, including chronic gastritis and potentially life-threatening conditions like gastric cancer. To manage these adverse outcomes, inhibiting the urease enzyme emerges as a promising strategy. A concise set of cyclopentyl-bearing N-acylthioureas 4a-j was synthesized, characterized and assessed for their ability to inhibit urease enzyme. All the tested compounds exhibited urease inhibitory activities, displaying superior enzyme inhibition when compared to the standard, thiourea (IC50 values 23.00 ± 0.03 μM).

Alfalfa is recognized as a high-quality forage crop exhibiting exceptional saline-alkaline tolerance. However, in China's Songnen Plain region, soda saline-alkaline soils dominated by NaHCO3 constitute a critical limiting factor for local ecosystems and alfalfa production. Consequently, enhancing alfalfa adaptation to saline-alkaline stress is considered essential for vegetation restoration in degraded lands and sustainable development of animal husbandry. This study employed physiological, transcriptomic, and metabolomic techniques to investigate the complex response mechanisms of alfalfa cul

Exploring host plant resistance and elevating plant defense mechanisms through the application of exogenous elicitors stands as a promising strategy for integrated pest management. The fall armyworm, a pernicious menace to grain crops in tropical and subtropical regions, stands as a formidable threat due to its capacity for devastation and a wide-ranging spectrum of host plants. There is no literature regarding artificially induced resistance in maize against fall armyworm (Spodoptera frugiperda) by exogenous application of phytohormones. The present investigation was performed to evaluate the

Trichoderma spp. can enhance plant resistance against a wide range of biotic stressors. However, the fundamental mechanisms by which Trichoderma enhances plant resistance against Meloidogyne incognita, known as root-knot nematodes (RKNs), are still unclear. Here, we identified a strain of Trichoderma asperellum (T141) that could effectively suppress RKN infestation in tomato (Solanum lycopersicum L.). Nematode infestation led to an increase in the concentrations of reactive oxygen species (ROS) and malondialdehyde (MDA) in roots but pre-inoculation with T141 significantly decreased oxidative s

Plant amino acid transporters regulate not only long-distance transport and reallocation of nitrogen (N) from source to sink organs, but also the amount of amino acids in leaves hijacked by invading pathogens. However, the function of amino acid transporters in plant defense responses to pathogen infection remains unknown. In this study, we found that the rice amino acid transporter gene OsLHT1 was expressed in leaves and up-regulated by maturation, N starvation, and inoculation of the blast fungus Magnaporthe oryzae. Knock out of OsLHT1 resulted in development stage- and N supply-dependent pr

AIM: Currently available medicines have little to offer in terms of supporting the regeneration of injured hepatic cells. Previous experimental studies have shown that resveratrol and metformin, less specific activators of AMP-activated protein kinase (AMPK) and sirtuin 1 (SIRT1), can effectively attenuate acute liver injury. The aim of this experimental study was to elucidate whether modulation of AMPK and SIRT1 activity can modify drug/paracetamol (APAP)-induced hepatocyte damage in vitro.
METHODS: Primary rat hepatocytes were pretreated with mutual combinations of specific synthetic activa

BACKGROUND: Peramivir offers a single-dose intravenous (IV) treatment option for influenza (vs 5-day oral dosing for oseltamivir). We sought to compare outcomes of emergency department (ED) patients at high risk for influenza complications treated with IV peramivir vs oral oseltamivir.
METHODS: During the 2015-16 and 2016-17 influenza seasons, adult patients in two US EDs were randomized to either oral oseltamivir or IV peramivir treatment group. Eligibility included positive molecular influenza test; met CDC criteria for antiviral treatment; able to provide informed consent and agree to foll

Phenolic acids and tanshinones are active principles in Salvia miltiorrhiza Bunge administered for cardiovascular and cerebrovascular diseases. Jasmonic acid (JA) promotes secondary metabolite accumulation, but the regulatory mechanism is unknown in S. miltiorrhiza. We identified and characterized the JA-responsive gene SmMYB97. Multiple sequence alignment and phylogenetic tree analyses showed that SmMYB97 was clustered with AtMYB11, AtMYB12, and ZmP1 in the subgroup S7 regulating flavonol biosynthesis. SmMYB97 was highly expressed in S. miltiorrhiza leaves and induced by methyl jasmonate (MeJ

Molecular mechanisms associated with biochar-elicited suppression of soilborne plant diseases and improved plant performance are not well understood. A stem base inoculation approach was used to explore the ability of biochar to induce systemic resistance in tomato plants against crown rot caused by a soilborne pathogen, Fusarium oxysporum f. sp. radicis lycopersici. RNA-seq transcriptome profiling of tomato, and experiments with jasmonic and salycilic acid deficient tomato mutants, were performed to elucidate the in planta molecular mechanisms involved in induced resistance. In this paper, we investigated the relationship between hydrogen sulfide (H2S) and mitogen-activated protein kinase kinase (MEK1/2) in jasmonic acid (JA)-regulated the redox state of ascorbate in the leaves of Arabidopsis thaliana. The results showed that JA significantly enhanced the phosphorylation level of MEK1/2, the production of endogenous H2S and the ratio of reduced ascorbate (AsA) to dehydroascorbate (DHA) (AsA/DHA) in wild type of A. thaliana (WT). However, there were no obvious effects of JA on above indicators in H2S synthetic mutant of A. thaliana (MT). H2S scavenger hypotaurine (

Evidence that anti-inflammatory and other biological effects of curcumin may at least in part be mediated by its metabolites underscores the importance of identifying novel transformation products. Spontaneous degradation of curcumin in buffer pH 7.5 results mainly in dioxygenated products with a characteristic cyclopentadione ring composed of carbons 2 through 6 of the former heptadienedione chain. When analyzing degradation reactions of 4'- O-methylcurcumin, a product was identified missing one of the terminal carbons of the heptadienedione moiety while containing a cyclopentadione ring and

Aphids induce many transcriptional perturbations in their host plants, but the signalling cascades responsible and the effects on plant resistance are largely unknown. Through a genome-wide association (GWA) mapping study in Arabidopsis thaliana, we identified WRKY22 as a candidate gene associated with feeding behaviour of the green peach aphid, Myzus persicae The transcription factor WRKY22 is known to be involved in pathogen-triggered immunity, and WRKY22 gene expression has been shown to be induced by aphids. Assessment of aphid population development and feeding behaviour on knockout mutan

The known members of plant methyl esterase (MES) family catalyze the hydrolysis of a C-O ester linkage of methyl esters of several phytohormones including indole-3-acetic acid, salicylic acid and jasmonic acid. The genome of grapevine (Vitis vinifera) was found to contain 15 MES genes, designated VvMES1-15. In this report, VvMES5 was selected for molecular, biochemical and structural studies. VvMES5 is most similar to tomato methyl jasmonate esterase. E. coli-expressed recombinant VvMES5 displayed methyl jasmonate (MeJA) esterase activity, it was renamed VvMJE1. Under steady-state conditions,

Between September 2013 and July 2014, 2,482 influenza 2009 pandemic A(H1N1) [A(H1N1)pdm09] viruses were screened in Japan for the H275Y substitution in their neuraminidase (NA) protein, which confers cross-resistance to oseltamivir and peramivir. We found that a large cluster of the H275Y mutant virus was present prior to the main influenza season in Sapporo /: Hokkaido, with the detection rate for this mutant virus reaching 29% in this area. Phylogenetic analysis suggested the clonal expansion of a single mutant virus in Sapporo /: Hokkaido. To understand the reason for this large cluster, we
