Intracellular ultrastructural characterization of Azadinium and Amphidoma (Amphidomataceae, Dinophyceae), including toxigenic and non-toxigenic strains.

The marine toxigenic dinoflagellate family Amphidomataceae, comprising Amphidoma and Azadinium, has been primarily characterized by thecal morphology examined by SEM. To further clarify the detailed morphology, the intracellular ultrastructure of five Azadinium species, encompassing two toxigenic species (Azadinium poporum and Az. spinosum) and three non-toxigenic species (Az. inconspicuum, Az. dalianense, and Az. zhuanum), was examined using TEM. The ultrastructure of two Amphidoma species was also examined for comparison. All species exhibited typical dinoflagellate organelles, i.e., nucleus

Rapid podocyte loss in murine models triggers the formation of podocyte-parietal epithelial cell intercellular bridges.

Podocytes are highly specialized, terminally differentiated visceral epithelial cells that are critical for the maintenance of the glomerular filtration barrier. In subtypes of glomerulonephritis and focal segmental glomerulosclerosis (FSGS), injured podocytes trigger the activation and proliferation of neighboring parietal epithelial cells (PECs) which line Bowman's capsule. Mechanisms by which injured podocytes trigger the activation of PECs remain poorly understood. In three independent murine models of proliferative glomerulopathy, we observed that rapid podocyte loss triggered the formati

Endogenous spartin, mutated in hereditary spastic paraplegia, has a complex subcellular localization suggesting diverse roles in neurons.

Mutation of spartin (SPG20) underlies a complicated form of hereditary spastic paraplegia, a disorder principally defined by the degeneration of upper motor neurons. Using a polyclonal antibody against spartin to gain insight into the function of the endogenous molecule, we show that the endogenous molecule is present in two main isoforms of 85 kDa and 100 kDa, and 75 kDa and 85 kDa in human and murine, respectively, with restricted subcellular localization. Immunohistochemical studies on human and mouse embryo sections and in vitro cell studies indicate that spartin is likely to possess both

[Ultrastructural study of experimental cerebrocortical necrosis in calves (author's transl)].

Cerebrocortical necrosis (CCN) was experimentally induced in three calves with the thiamine antagonist Amprolium. The calves were followed clinically. At the time of development of typical clinical signs of CCN the calves were killed and necropsied and a complete histopathologic examination was performed. The light microscopic lesions of the cerebral cortex were those of middle laminar necrosis and deep laminar edema. The necrotic zone consisted of fine vesicles and differed only slightly from the edema zone. In the former some neurons were morphologically normal, others showed signs of injury

Cisplatin induces calcium ion accumulation and hearing loss by causing functional alterations in calcium channels and exocytosis.

In recent years, molecular biology and biochemistry have been a focus of studies on the ototoxic side effects of cisplatin. In this paper, the application of cisplatin for 4 h and 72 h was studied from the perspective of electrophysiological function. Patch clamp experiments and immunofluorescence staining were performed on inner hair cells of the cochlea. The patch-clamp results showed that the calcium current amplitude decreased significantly at 4 h and 72 h after cisplatin treatment, the reversal potential was negatively polarized, and the activation time decreased. We suspected that intrac

MicroRNA-451 regulates stemness of side population cells via PI3K/Akt/mTOR signaling pathway in multiple myeloma.

OBJECTIVES: To describe the ultrastructure and relationship to nerves of the myofibroblast in the human bladder lamina propria, and discuss its possible role in bladder function, including sensing stretch, as the response of the bladder to stretch has been thoroughly investigated by afferent nerve recordings, but specialized stretch sensing organs have yet to be identified. MATERIALS AND METHODS: Flexible cystoscopic bladder biopsies were obtained from patients with detrusor hyper-reflexia and from controls. Systematic electron micrographs were obtained throughout the lamina propria, and the

Coupling of calcium transport with ATP hydrolysis in scallop sarcoplasmic reticulum.

Previously, we showed that incubation of the scallop sarcoplasmic reticulum (SR) with EGTA at above 37 degrees C resulted in the uncoupling of ATP hydrolysis with Ca2+ transport [Nagata et al. (1996) J. Biochem. 119, 1100-1105]. We have extended this study by comparing the kinetic behavior of Ca2+ release and binding to the uncoupled SR with that of intact scallop or rabbit SR. The change in the Ca2+ concentration in the reaction medium, as determined as the absorption of APIII, was followed using a stopped flow system. Intact scallop SR was preincubated with Ca2+ in the presence of a Ca2+ ion

The ultrastructural characteristics of the spermatozoa stored in the cauda epididymidis in Chinese soft-shelled turtle Pelodiscus sinensis during the breeding season.

The ultrastructure of spermatozoa in cauda epididymidis of soft-shelled turtle, P. sinensis during breeding season was investigated by light microscopy (LM) and electron microscopy (TEM and SEM). The mature spermatozoa appeared elongated and filiform. In general, the turtle spermatozoon contains a characteristic head, midpiece and tail, similar in morphology to that of birds, amphibians and other reptiles. However, several features are unique. These include (1) three intranuclear tubules containing dense core extend from the subacrosomal cone through the rostral nucleus and deep into the nucle

Induction of odontoblast-like cell differentiation in dog dental pulps after in vivo implantation of dentine matrix components.

The effects of dentine extracellular matrix components on dental mesenchymal cells were studied by light and transmission electron microscopy after their implantation at central sites of mechanically exposed pulps in dog molar teeth. The implants were Millipore filters that had been soaked with solutions containing 30 or 300 micrograms/ml of an EDTA-soluble fraction of rabbit incisor dentine. Control filters were soaked with dog albumin or phosphate buffered saline. Columnar, polarized cells were consistently seen after 8 days in close proximity to the filters coated with both concentrations o

Identification of the C-terminal region in Amelogenesis Imperfecta causative protein WDR72 required for Golgi localization.

Amelogenesis Imperfecta (AI) represents a group of hereditary conditions that manifest tooth enamel defects. Several causative mutations in the WDR72 gene have been identified and patients with WDR72 mutations have brown (or orange-brown) discolored enamel, rough enamel surface, early loss of enamel after tooth eruption, and severe attrition. Although the molecular function of WDR72 is not yet fully understood, a recent study suggested that WDR72 could be a facilitator of endocytic vesicle trafficking, which appears inconsistent with the previously reported cytoplasmic localization of WDR72. Salt Gradient-Induced Phoresis of Vesicles and Enhanced Membrane Fusion in a Crowded Milieu.

Phoresis of biocolloidal objects in response to chemical gradients is a matter of interest among diverse scientific disciplines owing to their importance in the spatiotemporal orchestration of biochemical processes. Although there are reports of soft matter transport/phoresis in the gradient of ions or salts in the aqueous system, their phoretic behavior in the presence of macromolecular crowder is largely unexplored. Notably, cellular cytoplasm is illustrated as a crowded milieu and thereby understanding biomolecular phoresis in the presence of polymeric macromolecules would endorse phoretic

Ionic strength and calcium regulate membrane interactions of myelin basic protein and the cytoplasmic domain of myelin protein zero.

The formation of a mature myelin sheath in the vertebrate nervous system requires specific protein-membrane interactions. Several myelin-specific proteins are involved in stacking lipid membranes into multilayered structures around axons, and misregulation of these processes may contribute to chronic demyelinating diseases. Two key proteins in myelin membrane binding and stacking are the myelin basic protein (MBP) and protein zero (P0). Other factors, including Ca2+, are important for the regulation of myelination. We studied the effects of ionic strength and Ca2+ on the membrane interactions

Ultrastructural morphometric observations on the extramural aortico-pulmonary bodies of the domestic fowl.

The extramural aortico-pulmonary bodies of the domestic fowl were analysed in electron micrographs by point-counting morphometry. The Type I cell constituted about 34% of the total volume of the parenchyma, whereas the supporting cells (Type II cells, pericytes and Schwann cells) constituted about 26%. The blood capillaries occupied the lowest volume density of about 5%. The mitochondria and the dense-cored vesicles occupied about the same volume (8%) of the Type I cell cytoplasm. The results obtained in the present study are compared with other similar studies on the aortic and carotid bodies

Bullous esophageal lesions due to cotrimoxazole: an immune-mediated process?

A 78-yr-old man experienced a generalized bullous eruption of the skin (a Stevens-Johnson variant of erythema multiforme) with simultaneous involvement of the esophagus due to co-trimoxazole. Immunologic tests revealed specific antibodies of the immunoglobulin G class but not of the immunoglobulin E class against sulfamethoxazole, and in particular against trimethoprim. Lymphocyte transformation tests demonstrated sensitized lymphocytes against trimethoprim but not sulfamethoxazole. The esophageal mucosa showed intraepithelial vesicle formation with diffuse cytoplasmic deposits of immunoglobul

Fine structural survey of the intermediate subnucleus of the nucleus tractus solitarii and its glossopharyngeal afferent terminals.

The intermediate subnucleus of the nucleus tractus solitarii (imNTS) receives somatosensory inputs from the soft palate and pharynx, and projects onto the nucleus ambiguus, thus serving as a relay nucleus for swallowing. The ultrastructure and synaptology of the rat imNTS, and its glossopharyngeal afferent terminals, have been examined with cholera toxin-conjugated horseradish peroxidase (CT-HRP) as an anterograde tracer. The imNTS contained oval or ellipsoid-shaped, small to medium-sized neurons (18.2 x 11.4 microm) with little cytoplasm, few cell organelles and an irregularly shaped nucleus.

Iron transport pathways in the human malaria parasite Plasmodium falciparum revealed by RNA-sequencing.

Host iron deficiency is protective against severe malaria as the human malaria parasite Plasmodium falciparum depends on bioavailable iron f … y required for ferrous iron transport in P. falciparum. Of these, we localized PfVIT and PfZIPCO to cytoplasmic vesicles, PfMRS3 to the mitochondrion, and the novel putative iron transporter PfE140 to the plasma membrane for the first time in P. falciparum. PfNRAMP/PfDMT1 and PfCRT were previously reported to efflux Fe2+ from the digestive vacuole. Our data support a …

Evidence that the wzxE gene of Escherichia coli K-12 encodes a protein involved in the transbilayer movement of a trisaccharide-lipid intermediate in the assembly of enterobacterial common antigen.

The assembly of many bacterial cell surface polysaccharides requires the transbilayer movement of polyisoprenoid-linked saccharide intermediates across the cytoplasmic membrane. It is generally believed that transverse diffusion of glycolipid intermediates is mediated by integral membrane proteins called translocases or "flippases." The bacterial genes proposed to encode these translocases have been collectively designated wzx genes. The wzxE gene of Escherichia coli K-12 has been implicated in the transbilayer movement of Fuc4NAc-ManNAcA-GlcNAc-P-P-undecaprenol (lipid III), the donor of the t

PLB-985 Neutrophil-Like Cells as a Model To Study Aspergillus fumigatus Pathogenesis.

Fungal infections remain a major global concern. Emerging fungal pathogens and increasing rates of resistance mean that additional research efforts and resources must be allocated to advancing our understanding of fungal pathogenesis and developing new therapeutic interventions. Neutrophilic granulocytes are a major cell type involved in protection against the important fungal pathogen Aspergillus fumigatus, where they employ numerous defense mechanisms, including production of antimicrobial extracellular vesicles. A major drawback to work with neutrophils is the lack of a suitable cell line s

Increased GLUT1 expression and localization to Golgi apparatus of acinar cells in the parotid gland of Goto-Kakizaki diabetic rats.

OBJECTIVE: Patients with diabetes are known to have high salivary glucose levels. But the mechanisms are still unclear. We hypothesized that the topological changes of glucose transporters affect the salivary glucose level. METHODS: We used adult Goto-Kakizaki (GK) rats, an animal model of advanced diabetes, and Wistar rats as a control, with or without glucose load. The sections of salivary glands from the animals were processed for standard histological, immunohistochemical, and immunofluorescent staining. RESULTS: Parotid acinar cells of GK rats appeared like mucous filled with low-eosin-

A PARK9 iPSC-Derived Dopaminergic Neuron Model Enables Drug Screening Targeting Autophagy-Lysosome Pathway Dysfunction in Parkinson's Disease.

Parkinson's disease (PD) is a neurodegenerative disease characterized by dopaminergic neuronal degeneration in the substantia nigra, in which lysosomal dysfunction and impaired autophagy-lysosome pathway activity are increasingly recognized as important pathogenic mechanisms. However, disease-modifying therapies targeting this pathway remain unavailable. Here, we generated induced pluripotent stem cells (iPSCs) from a PARK9 patient carrying an ATP13A2 mutation and established mutation-corrected isogenic control iPSCs. PARK9 iPSC-derived neurons recapitulated lysosomal dysfunction-associated ce
