Smoking and DNA methylation: Correlation of methylation with smoking behavior and association with diseases and fetus development following prenatal exposure.

Among epigenetic mechanisms, DNA methylation has been widely studied with respect to many environmental factors. Smoking is a common factor which affects both global and gene-specific DNA methylation. It is supported that smoking directly affects DNA methylation, and these effects contribute to the development and progression of various diseases, such as cancer, lung and cardiovascular diseases and male infertility. In addition, prenatal smoking influences the normal development of the fetus via DNA methylation changes. The DNA methylation profile and its smoking-induced alterations helps to d

The Chromatin Configuration Promotes Constitutional Chromosomal Rearrangements in Post-Meiotic Sperm.

The complex nuclear packaging of sperm DNA, known as sperm chromatin organization, is essential for protecting the paternal genome during fertilization. Recent studies suggest that changes in the post-meiotic chromatin structure, a specific arrangement that forms after meiosis is completed, may be associated with an increased risk of constitutional chromosomal rearrangements (CCRs) in offspring. This review examines the potential mechanisms by which abnormal post-meiotic organization may impair DNA stability in spermatogonial cells, thereby leading to CCR formation. We discuss the interplay be

Toxicity of Sulfamethoxazole in Model Organism Drosophila melanogaster.

Sulfamethoxazole, a sulfonamide group antibiotic, has been used to prevent bacterial infections for clinical and agricultural purposes. Due to its wide use, sulfamethoxazole has been detected in waste streams. In terms of aquatic toxicity, negative impact of sulfamethoxazole on non-target model organisms has been demonstrated. Furthermore, the drug was also detected in edible crops due to its high mobility in the soil up to 1 mg/L. Therefore, toxicity of sulfamethoxazole at different trophic levels is important task to be investigated. In this manner, Drosophila melanogaster is valuable model

Aspergillus tubingensis and Aspergillus niger as the dominant black Aspergillus, use of simple PCR-RFLP for preliminary differentiation.

This work aimed to identify the species distribution of common clinical and environmental isolates of black Aspergilli based on simple restriction fragment length polymorphism (RFLP) analysis of the β-tubulin gene. A total of 149 clinical and environmental strains of black Aspergilli were collected and subjected to preliminary morphological examination. Total genomic DNAs were extracted, and PCR was performed to amplify part of the β-tubulin gene. At first, 52 randomly selected samples were species-delineated by sequence analysis. In order to distinguish the most common species, PCR amplicons

Development of a miniaturized DNA microarray for identification of 66 virulence genes of Legionella pneumophila.

INTRODUCTION: For the last five years, Legionella sp. infections and legionnaire's disease in Poland have been receiving a lot of attention, because of the new regulations concerning microbiological quality of drinking water. This was the inspiration to search for and develop a new assay to identify many virulence genes of Legionella pneumophila to better understand their distribution in environmental and clinical strains. The method might be an invaluable help in infection risk assessment and in epidemiological investigations. MATERIAL/METHODS: The microarray is based on Array Tube technolog

Development and application of eDNA-based tools for the conservation of white-clawed crayfish.

eDNA-based methods represent non-invasive and cost-effective approaches for species monitoring and their application as a conservation tool has rapidly increased within the last decade. Currently, they are primarily used to determine the presence/absence of invasive, endangered or commercially important species, but they also hold potential to contribute to an improved understanding of the ecological interactions that drive species distributions. However, this next step of eDNA-based applications requires a thorough method development. We developed an eDNA assay for the white-clawed crayfish (

Correlations between oxidative DNA damage and formation of hepatic tumours in two flatfish species from contaminated environments.

Many species in aquatic environments face increased exposure to oncogenic pollution due to anthropogenic environmental change which can lead to higher cancer prevalence. The mechanistic relationship connecting environmental pollution and cancer is multi-factorial and poorly understood, and the specific mechanisms are so far still uncharacterized. One potential mediator between pollutant exposure and cancer is oxidative damage to DNA. We conducted a study in the field with two flatfish species, European flounder (Platichthys flesus L.) and common dab (Limanda limanda L.) with overlapping distri

In Vitro Effects of Titanium Dioxide Nanoparticles (TiO2NPs) on Cadmium Chloride (CdCl2) Genotoxicity in Human Sperm Cells.

The environmental release of titanium dioxide nanoparticles (TiO2NPs) associated with their intensive use has been reported to have a genotoxic effect on male fertility. TiO2NP is able to bind and transport environmental pollutants, such as cadmium (Cd), modifying their availability and/or toxicity. The aim of this work is to assess the in vitro effect of TiO2NPs and cadmium interaction in human sperm cells. Semen parameters, apoptotic cells, sperm DNA fragmentation, genomic stability and oxidative stress were investigated after sperm incubation in cadmium alone and in combination with TiO2NPs

Environment, diet and CpG island methylation: epigenetic signals in gastrointestinal neoplasia.

The epithelial surfaces of the mammalian alimentary tract are characterised by very high rates of cell proliferation and DNA synthesis, and in humans they are highly susceptible to cancer. The role of somatic mutations as drivers of carcinogenesis in the alimentary tract is well established, but the importance of gene silencing by epigenetic mechanisms is increasingly recognised. Methylation of CpG islands is an important component of the epigenetic code that regulates gene expression during development and normal cellular differentiation, and a number of genes are well known to become abnorma

Change in diversity patterns of fish by cascade dams: comprehensive dataset of eDNA and traditional evidence from the Jinsha River.

The large-scale construction of cascade dams worldwide has profoundly altered river hydrological regimes and habitats, producing significant threats to fish diversity. This study integrated environmental DNA (eDNA) and traditional fishing data to construct a fish dataset from the middle and upper reaches of the Jinsha River (2003-2024). It was used to systematically reveal the spatiotemporal patterns of fish diversity and analyze key environmental drivers. Additionally, the Isolation Forest algorithm was employed to remove 29 outliers from the historical data. Distance-based redundancy analysi

Cadmium sub-lethal effects on Daphnia magna - A new take on an old subject.

Rising production and use of electronic products leads to higher cadmium usage, increasing the risk of exposure to this dangerous metal. Despite being a classic environmental contaminant, cadmium's full toxicity spectrum and mechanisms of action towards aquatic biota are still in need of further clarification. This work contributes to such clarification by comprehensively characterizing cadmium mechanisms of toxic action resulting from a seven-day exposure to a sub-lethal concentration (EC20; 4.5 µg L-1) on the freshwater species Daphnia magna. A holistic approach starting at molecular level r

Maternal vitamin C regulates reprogramming of DNA methylation and germline development.

Development is often assumed to be hardwired in the genome, but several lines of evidence indicate that it is susceptible to environmental modulation with potential long-term consequences, including in mammals1,2. The embryonic germline is of particular interest because of the potential for intergenerational epigenetic effects. The mammalian germline undergoes extensive DNA demethylation3-7 that occurs in large part by passive dilution of methylation over successive cell divisions, accompanied by active DNA demethylation by TET enzymes3,8-10. TET activity has been shown to be modulated by nutr

Bridging Plant and Human Radiation Response and DNA Repair through an In Silico Approach.

The mechanisms of response to radiation exposure are conserved in plants and animals. The DNA damage response (DDR) pathways are the predominant molecular pathways activated upon exposure to radiation, both in plants and animals. The conserved features of DDR in plants and animals might facilitate interdisciplinary studies that cross traditional boundaries between animal and plant biology in order to expand the collection of biomarkers currently used for radiation exposure monitoring (REM) in environmental and biomedical settings. Genes implicated in trans-kingdom conserved DDR networks often

Epidemiology of ultraviolet-DNA repair capacity and human cancer.

The following conclusions are derived from an epidemiological study. Reduced repair of ultraviolet (UV)-induced DNA damage contributes directly to basal cell carcinoma (BCC) in individuals with prior sunlight overexposure. A family history of BCC is a predictor of low DNA repair. Repair of UV-damaged DNA declines at a fixed rate of approximately 1% per annum in noncancerous controls. The DNA repair differences between young BCC cases and their controls disappear as they age. Hence, BCC, in terms of DNA repair, is a premature aging disease. The persistence of photochemical damage because of red

Real-time malaria detection in the Amazon rainforest via drone-collected eDNA and portable qPCR.

Zoonotic malaria risk at human-wildlife-environment interfaces requires surveillance that integrates signals from reservoirs, vectors and the environment. We coupled a drone-based environmental DNA (eDNA) canopy swabbing approach with portable quantitative PCR (qPCR) to detect Plasmodium DNA in situ during a 24-h field exercise in the Amazon rainforest. Drone-lowered sterile swabs into the canopy, which were then extracted and subjected to a multiplex pan-Plasmodium assay targeting five human-infecting Plasmodium species (limit of detection 0.2 parasites μL-1). Of 12 samples (10 canopy swabs,

Environmental chromium and cadmium co-exposure induces GADD45A-mediated rDNA instability in human cohorts and cell models.

Exposure to heavy metals such as hexavalent chromium [Cr(VI)] and cadmium (Cd) poses a major environmental and occupational health concern due to their widespread presence and genotoxic potential. While the toxicological profiles of individual metals are well-characterized, their combined effects on genome stability remain poorly understood. Ribosomal DNA (rDNA), a repetitive and transcriptionally active genomic region essential for protein synthesis, is increasingly recognized as a sensitive target of environmental stress and genomic instability. Here, we investigated electroplating workers s

Ocular manifestations in the inherited DNA repair disorders.

Deoxyribonucleic acid (DNA) repair is a fundamental process designed to keep the integrity of genomic DNA that is continuously challenged by intrinsic or environmental induced alterations. Numerous genes involved in DNA repair have been cloned and are involved in different DNA repair pathways: base excision repair, nucleotide excision repair, mismatch repair, DNA recombination. Inherited conditions due to mutations in DNA repair genes include mainly: xeroderma pigmentosum, Cockayne syndrome, Trichothiodystrophy, Bloom syndrome, Rothmund-Thomson syndrome, and Werner syndrome. Identification and molecular characterization of a Bacillus subtilis IS13 strain involved in the biodegradation of 4,5,6-trichloroguaiacol.

4,5,6-Trichloroguaiacol (4,5,6-TCG) is a recalcitrant organochlorine compound produced during pulp bleaching and a potential environmental hazard in paper mill effluents. We report here the identification by biochemical tests and molecular biological analysis, using 16S ribotyping, of a 4,5,6-TCG-degrading bacterium, identified as a strain of Bacillus subtilis that is most closely related according to the phylogenetic analysis to B. subtilis strain Lactipan (alignment score 99%). Biodegradation of 4,5,6-TCG by this organism in a mineral salts medium was shown to occur only when the inoculum wa

Detection of azole-resistant Aspergillus fumigatus in the environment from air, plant debris, compost, and soil.

Aspergillus fumigatus is a ubiquitous fungus, a saprophyte of plants, and an opportunistic pathogen of humans. Azole fungicides are used in agriculture to control plant pathogens, and azoles are also used as a first line of treatment for aspergillosis. The continued exposure of A. fumigatus to azoles in the environment has likely led to azole resistance in the clinic where infections result in high levels of mortality. Pan-azole resistance in environmental isolates is most often associated with tandem-repeat (TR) mutations containing 34 or 46 nucleotides in the cyp51A gene. Because the rapid d
