Importance of Virulence Factors for the Persistence of Oral Bacteria in the Inflamed Gingival Crevice and in the Pathogenesis of Periodontal Disease.

Periodontitis is a chronic inflammation that develops due to a destructive tissue response to prolonged inflammation and a disturbed homeostasis (dysbiosis) in the interplay between the microorganisms of the dental biofilm and the host. The infectious nature of the microbes associated with periodontitis is unclear, as is the role of specific bacterial species and virulence factors that interfere with the host defense and tissue repair. This review highlights the impact of classical virulence factors, such as exotoxins, endotoxins, fimbriae and capsule, but also aims to emphasize the often-negl

Beyond immune escape: a variant surface glycoprotein causes suramin resistance in Trypanosoma brucei.

Suramin is one of the first drugs developed in a medicinal chemistry program (Bayer, 1916), and it is still the treatment of choice for the hemolymphatic stage of African sleeping sickness caused by Trypanosoma brucei rhodesiense. Cellular uptake of suramin occurs by endocytosis, and reverse genetic studies with T. b. brucei have linked downregulation of the endocytic pathway to suramin resistance. Here we show that forward selection for suramin resistance in T. brucei spp. cultures is fast, highly reproducible and linked to antigenic variation. Bloodstream-form trypanosomes are covered by a d

Histologic and Ultrastructural Findings in Dogs With Chronic Respiratory Disease Suspected of Ciliary Dyskinesia.

Mucociliary clearance is a main defense mechanism of the respiratory tract, which can be inherently impaired in primary ciliary dyskinesia (PCD) or reversibly altered in secondary ciliary dyskinesia (SCD). Limited diagnostic test availability likely leads to misdiagnosis or underdiagnosis of PCD in animals. This study evaluated the light and transmission electron microscopy (TEM) changes in the respiratory mucosa of 15 dogs with chronic respiratory disease suspected of PCD. Necropsy was performed in 1 case and 2 dogs were used as negative controls. PCD was confirmed in 1 dog, which presented w

Antimicrobial peptides: old molecules with new ideas.

Almost 90 years have passed since Alexander Fleming discovered the antimicrobial activity of lysozyme, the first natural antibiotic isolated from our body. Since then, various types of molecules with antibiotic activity have been isolated from animals, insects, plants, and bacteria, and their use has revolutionized clinical medicine. So far, more than 1,200 types of peptides with antimicrobial activity have been isolated from various cells and tissues, and it appears that all living organisms use these antimicrobial peptides (AMPs) in their host defense. In the past decade, innate AMPs produce

Chlorogenic acid mitigates acute respiratory distress syndrome via inhibition of the PI3K/AKT signalling pathway: an integrated analysis of bioinformatics and validation experiments.

OBJECTIVE: High mortality rates are linked to acute respiratory distress syndrome (ARDS), a prevalent type of respiratory failure. Amid the COVID-19 outbreak in particular, a viable defensive method is provided by traditional Chinese medicine (TCM). This study investigated whether chlorogenic acid (CGA), a primary component of honeysuckle, could protect against ARDS. METHODS: We employed network pharmacology to explore the honeysuckle and ARDS component-target-disease network, and enrichment function analysis to uncover the potential mechanisms of honeysuckle in treating ARDS. LPS-induced ARD

The bite of the honeybee: 2-heptanone secreted from honeybee mandibles during a bite acts as a local anaesthetic in insects and mammals.

Honeybees secrete 2-heptanone (2-H) from their mandibular glands when they bite. Researchers have identified several possible functions: 2-H could act as an alarm pheromone to recruit guards and soldiers, it could act as a chemical marker, or it could have some other function. The actual role of 2-H in honeybee behaviour remains unresolved. In this study, we show that 2-H acts as an anaesthetic in small arthropods, such as wax moth larva (WML) and Varroa mites, which are paralysed after a honeybee bite. We demonstrated that honeybee mandibles can penetrate the cuticle of WML, introducing less

Glycoaminoglycan (GAG) deficiency in protective barrier as an underlying, primary cause of ulcerative colitis, Crohn's disease interstitial cystitis and possibly Reiter's syndrome.

Ulcerative colitis, Crohn's disease and interstitial cystitis share many common features, the most important of which is a defect in the glycoaminoglycan (GAG) defensive barrier. This defect allows penetration of toxins causing localized inflammatory response, followed by fibrosis and distant pathological changes, together with a myriad of biochemical and immunological changes. The latter has caused confusion as to etiology of the aforementioned disorders. This hypothesis is somewhat supported by the fact that agents such as glucosamine and pentosan polysulphate (Elmiron) that replace the GAG

Metabolomics reveals McF6'H1-mediated regulation of phenylpropanoid/flavonoid metabolism and enhanced antifungal activity in transgenic Morinda citrifolia.

Phenylpropanoids and flavonoids are major classes of bioactive metabolites that contribute to plant defense and functional value in noni (Morinda citrifolia). Although feruloyl-CoA 6'-hydroxylase 1 (F6'H1) is recognized as a key enzyme in coumarin biosynthesis, its broader role in secondary metabolic regulation and antifungal responses in perennial medicinal plants remains unclear. In this study, we investigated the regulatory function of McF6'H1 by transgenic analysis, HPLC-MS/MS-based metabolomics, and extract-based antifungal assays in noni. Based on an established genetic transformation sy

Progress of Traditional Chinese Medicine in Targeting Ferroptosis to Combat Cerebral Ischemia-Reperfusion Injury.

Cerebral ischemia-reperfusion injury (CIRI) often results in significant morbidity and mortality. The effectiveness of current reperfusion therapies is hindered by substantial secondary damage, which highlights the need for novel molecular targets and neuroprotective strategies. Ferroptosis, a form of programmed cell death driven by iron dysregulation and lipid peroxidation, has emerged as a key player in CIRI pathogenesis. Traditional Chinese medicine (TCM) has demonstrated considerable potential in managing IS, and increasing evidence suggests its neuroprotective effects are primarily mediat

Research Letter: Retrograde Amnesia and Posttraumatic Amnesia in Service Members and Veterans With Remote History of TBI.

OBJECTIVE: The recently updated American Congress of Rehabilitation Medicine diagnostic criteria for mild traumatic brain injury (mTBI) removed retrograde amnesia (RA) as a main criterion for mTBI, recommending it be included as a substitute criterion only when posttraumatic amnesia (PTA) cannot be reliably assessed. This study aimed to investigate the evidence base for this recommendation. SETTING: Military treatment facility. PARTICIPANTS: A total of 752 US military service members/veterans (mean age = 36.1 years, SD = 9.4 years) with a history of TBI prospectively enrolled in the Defense

A Review on Herbal Drugs Used in the Treatment of Peptic Ulcer.

BACKGROUND: An ulcer is a condition characterized by inflammation, irritation, or erosion in the mucosal lining of the stomach or duodenum. Hence, peptic ulcer is the ulcer of both the stomach and the duodenum. 10% of the world's population is affected by chronic peptic ulcers. The formation of peptic ulcers depends on gastric juice pH and the decrease in mucosal defenses. Nonsteroidal antiinflammatory drugs (NSAIDs) and Helicobacter pylori (H. pylori) infection are the two significant factors disrupting mucosal resistance to injury. Indian herbal plants are exceptional for their ethnic, ethno

Single-walled carbon nanotube conjugated cytochrome c as exogenous nano catalytic medicine to combat intracellular oxidative stress.

Mitochondrial dysfunction has been reported to be one of the main causes of many diseases including cancer, type2 diabetes, neurodegenerative disorders, cardiac ischemia, sepsis, muscular dystrophy, etc. Under in vitro conditions, Cytochrome C (Cyt C) maintains mitochondrial homeostasis and stimulates apoptosis, along with being a key participant in the life-supporting function of ATP synthesis. Hence, the medicinal importance of Cyt C as catalytic defense is immensely important in various mitochondrial disorders. Here, we have developed a nanomaterial via electrostatically conjugating oxidize

Withania somnifera in Neurological Disorders: Ethnopharmacological Evidence, Mechanism of Action and its Progress in Delivery Systems.

BACKGROUND: The underlying cause of major neurodegenerative disorders remains a healthcare mystery. The thoroughly investigated causes include oxidative stress, inflammation, environmental factor, mitochondrial dysfunction, and irregular neuronal protein aggregation. Withania somnifera has been used for more than 2500 years as a useful medicinal plant to improve disease defense, prevent aging, rejuvenate the body in a vulnerable situation, and generate a feeling of mental well-being. However, a persuasive paper emphasizing its neuroprotective nature is missing. OBJECTIVE: In the current revie

Ecological risks of microplastics in soils of major Angelica sinensis production areas and biological responses of roots to polyethylene microplastics.

Farmland microplastic pollution has become an emerging environmental issue in agricultural ecosystems; however, systematic studies on soil microplastic contamination in the main production areas of Angelica sinensis in China and its potential effects on medicinal roots remain limited. This study analyzed the composition, ecological risks, and accumulation and biological effects of microplastics in A. sinensis roots through field investigation combined with controlled experiments. The results indicate that farmland soils in the main production areas of Gansu Province contain relatively high lev
