MT1H Inhibited Colon Cancer Development via Interacting With DUSP1 and MAPK Signaling Pathway Inactivation.

We focused on examining the impact of metallothionein 1H (MT1H) on colon cancer (CC) proliferation, migration, and invasion, as well as elucidating its underlying molecular mechanisms. Cell proliferation was assessed using the Cell Counting Kit-8 (CCK-8) assay. The invasive and migratory capacities of the cells were evaluated through transwell and wound healing assays. We found that both MT1H were lowly expressed in human CC tissues by immunohistochemistry (IHC). In HCT116 and SW480 cells and a xenograft mouse model, we found that overexpression of MT1H could inhibit CC cell proliferation, mig

Recurring MLH1 deleterious mutations in unrelated Chinese Lynch syndrome families in Singapore.

Lynch syndrome is an autosomal dominant disorder due to mutations in DNA mismatch repair genes, causing young onset colorectal cancer and extra-colonic cancers such as endometrial and stomach cancers. We genotyped MLH1 and MSH2 in patients suspected to have Lynch syndrome and correlated patient clinical characteristics and family history with deleterious mutations. Eighty-five unrelated patients participated. Comprehensive sequencing was performed for MLH1 and MSH2, including all coding regions, splice-site junctions and promoter regions. Of the 29 different mutations found, 11 were deleteriou

DUSP8 ameliorates podocyte mitochondrial dysfunction via PRAS40 dephosphorylation in diabetic kidney disease.

Mitochondrial dysfunction is central to podocyte injury in diabetic kidney disease (DKD). Here, we observed consistent downregulation of dual-specificity phosphatase 8 (DUSP8) in glomeruli from patients with DKD, db/db mice, STZ/HFD-induced DKD mice, and high glucose-treated podocytes. Glomerular DUSP8 levels were positively correlated with the estimated glomerular filtration rate and negatively correlated with the urinary albumin-to-creatinine ratio and serum creatinine level. Functional studies demonstrated that DUSP8 alleviated high glucose-induced mitochondrial fission, reactive oxygen spe

Dual Allosteric Targeting of SHP2: Development of Bivalent Inhibitors with Enhanced Potency for Cancer Treatment.

SHP2 is an attractive anticancer target due to its pivotal role in oncogenic signaling. Although combination therapies involving SHP2 inhibition are promising, none have yet advanced to clinical approval, highlighting the need for innovative SHP2-targeted approaches. Herein, we developed a series of bivalent allosteric inhibitors by linking two distinct ligands targeting the tunnel and latch sites. SDUY127 demonstrated the most potent enzymatic inhibition (IC50 = 16 nM), representing a 3-fold improvement over the parent compounds. We developed new site-specific biotinylated probes and AlphaScr

Modulation of functional properties of laforin phosphatase by alternative splicing reveals a novel mechanism for the EPM2A gene in Lafora progressive myoclonus epilepsy.

The EPM2A gene, encoding the dual-phosphatase laforin, is mutated in a fatal form of progressive myoclonus epilepsy known as Lafora disease (LD). The EPM2A gene, by differential splicing of its transcripts, is known to encode two laforin isoforms having distinct carboxyl termini; a major isoform localized in the cytoplasm (laf331), and a minor isoform that is targeted to the nucleus as well (laf317). We show here that the two laforin isoforms interact with each other and form homo and heterodimers. The homodimer of laf331 display robust phosphatase activity, whereas the laf317 homodimer and th

Role for pituitary adenylate cyclase activating polypeptide in cystitis-induced plasticity of micturition reflexes.

Pituitary adenylate cyclase activating polypeptide (PACAP) peptides are expressed and regulated in sensory afferents of the micturition pathway. Although these studies have implicated PACAP in bladder control, the physiological significance of these observations has not been firmly established. To clarify these issues, the roles of PACAP and PACAP signaling in micturition and cystitis were examined in receptor characterization and physiological assays. PACAP receptors were identified in various tissues of the micturition pathway, including bladder detrusor smooth muscle and urothelium. Bladder

DUSP9-mediated inhibition of IRS1/PI3K/AKT pathway contributes to insulin resistance and metabolic dysfunction in gestational diabetes mellitus.

BACKGROUND: Gestational diabetes mellitus (GDM) is a common pregnancy complication associated with adverse maternal and fetal outcomes. Recent studies suggest a role for dual-specificity phosphatase 9 (DUSP9) in insulin resistance and metabolic dysregulation, though its specific contribution to GDM remains unclear. This study aims to investigate the function of DUSP9 in GDM pathophysiology and its underlying molecular mechanisms. METHODS: We analyzed DUSP9 expression in umbilical cord blood and placental tissues from GDM patients (n = 16) and healthy controls (n = 14) using RT-qPCR and wester

Genomics of T-Cell Lymphomas With CD30/CD15 Co-Expression: Comparison With Anaplastic Large-Cell Lymphoma, ALK-Negative.

The proper categorization of mature T-cell neoplasms with coexpression of CD30/CD15 is unresolved. Prior studies suggested an overlap with ALK-negative anaplastic large cell lymphoma (ALCL). We evaluated the morphologic, immunophenotypic, and molecular features of 28 T-cell lymphomas coexpressing CD30/CD15, and performed a comparison with 8 ALK/CD15-negative ALCL and published data. Clinical information was retrieved from the submitting physician. Immunohistochemistry, TRG and IG gene rearrangement, DNA and RNA targeted next-generation sequencing, and fluorescence in situ hybridization for DUS

DOCK1 insufficiency disrupts trophoblast function and pregnancy outcomes via DUSP4-ERK pathway.

Abnormal trophoblast function is associated with diseases such as recurrent spontaneous abortion, pre-eclampsia, and preterm birth, and endangers maternal and fetal health. However, the underlying regulatory mechanisms remain unclear. In this study, we found DOCK1 expression is decreased in the placental villi of patients with recurrent spontaneous abortion, and that its expression determined the invasive properties of extravillous trophoblasts (EVTs), highlighting a previously unknown role of DOCK1 in regulating EVT function. Furthermore, DOCK1 deficiency disturbed the ubiquitinated degradati

Increased endoplasmic reticulum stress and decreased proteasomal function in lafora disease models lacking the phosphatase laforin.

BACKGROUND: Lafora progressive myoclonus epilepsy (Lafora disease; LD) is a fatal autosomal recessive neurodegenerative disorder caused by loss-of-function mutations in either the EPM2A gene, encoding the dual specificity phosphatase laforin, or the EPM2B gene, encoding the E3-ubiquitin ligase malin. Previously, we and others have shown that both proteins form a functional complex that regulates glycogen synthesis by a novel mechanism involving ubiquitination and proteasomal degradation of at least two proteins, glycogen synthase and R5/PTG. Since laforin and malin localized at the endoplasmic

The E3 ligase tripartite motif 7 drives the progression of non-alcoholic fatty liver disease by targeting DUSP10 degradation in male mice.

Non-alcoholic fatty liver disease (NAFLD) and its more severe form, nonalcoholic steatohepatitis (NASH), have emerged as a burgeoning global epidemic and impose an enormous socioeconomic burden. However, the lack of effective pharmacotherapies is due to incomplete understanding of the molecular mechanisms of NASH. In the present study, we observe that E3 ligase TRIM7 expression is significantly increased in both liver tissues and hepatocytes from NAFLD models. In vivo gain- and loss-of-function experiments reveal that hepatic-specific TRIM7 deletion significantly alleviates hepatic steatosis,

Role of Lipin-1 in Macrophage-Mediated Atherosclerosis: Is It Atherogenic or Atheroprotective?

Macrophages are central regulators of atherosclerosis, governing lipid accumulation, inflammatory signaling, and plaque stability. Lipin-1 is a multifunctional lipid-metabolic regulator that integrates cellular metabolism with inflammatory responses through its dual roles as a phosphatidic acid phosphatase enzyme and a transcriptional coregulator. However, its role in macrophage-driven atherosclerosis remains controversial. This review critically evaluates the domain-specific functions of lipin-1 and their impact on disease progression. Accumulating evidence indicates that lipin-1 exerts diver

In Situ Construction of a Reticulated Peptide Scaffold via Responsive Self-Assembly and Proximal Covalent Labeling for Enhanced Tumor Imaging and Therapy.

Precise imaging and specific retention of therapeutic agents at solid tumors were particularly desirable yet hindered by limited specificity and fast diffusion. Herein, we established a dual-trigger-mediated synergistic strategy to form a reticulated peptide network in situ by functional peptide (CL-YPFG) through responsive self-assembly and covalent labeling for prolonged imaging and boosted accumulation of drugs. Specifically, the CL-YPFG underwent morphology transformation from nanoparticles to nanofibers under alkaline phosphatase and further turned into reticulated peptide structures in t

CD73/adenosine axis exerts cardioprotection against hypobaric hypoxia-induced metabolic shift and myocarditis in a sex-dependent manner.

BACKGROUND: Clinical and experimental studies have shown that the myocardial inflammatory response during pathological events varies between males and females. However, the cellular and molecular mechanisms of these sex differences remain elusive. CD73/adenosine axis has been linked to anti-inflammatory responses, but its sex-specific cardioprotective role is unclear. The present study aimed to investigate whether the CD73/adenosine axis elicits sex-dependent cardioprotection during metabolic changes and myocarditis induced by hypobaric hypoxia. METHODS: For 7 days, male and female mice recei

Lafora progressive myoclonus epilepsy: recent insights into cell degeneration.

Lafora disease (LD) is a fatal autosomal recessive form of progressive myoclonus epilepsy. Patients manifest myoclonus and tonic-clonic seizures, visual hallucinations, intellectual, and progressive neurologic deterioration beginning in adolescence. The two genes known to be involved in Lafora disease are EPM2A and NHLRC1 (EPM2B). The EPM2A gene encodes laforin, a dual-specificity protein phosphatase, and the NHLRC1 gene encodes malin, an E3-ubiquitin ligase. The two proteins interact with each other and, as a complex, are thought to regulate glycogen synthesis. It may also be considered as a

Signal-dependent repression of DUSP5 by class I HDACs controls nuclear ERK activity and cardiomyocyte hypertrophy.

Cardiac hypertrophy is a strong predictor of morbidity and mortality in patients with heart failure. Small molecule histone deacetylase (HDAC) inhibitors have been shown to suppress cardiac hypertrophy through mechanisms that remain poorly understood. We report that class I HDACs function as signal-dependent repressors of cardiac hypertrophy via inhibition of the gene encoding dual-specificity phosphatase 5 (DUSP5) DUSP5, a nuclear phosphatase that negatively regulates prohypertrophic signaling by ERK1/2. Inhibition of DUSP5 by class I HDACs requires activity of the ERK kinase, mitogen-activat

Malin decreases glycogen accumulation by promoting the degradation of protein targeting to glycogen (PTG).

Lafora disease (LD) is an autosomal recessive neurodegenerative disease that results in progressive myoclonus epilepsy and death. LD is caused by mutations in either the E3 ubiquitin ligase malin or the dual specificity phosphatase laforin. A hallmark of LD is the accumulation of insoluble glycogen in the cytoplasm of cells from most tissues. Glycogen metabolism is regulated by phosphorylation of key metabolic enzymes. One regulator of this phosphorylation is protein targeting to glycogen (PTG/R5), a scaffold protein that binds both glycogen and many of the enzymes involved in glycogen synthes

Aldosterone and angiotensin II synergistically induce mitogenic response in vascular smooth muscle cells.

Interaction between aldosterone (Aldo) and angiotensin II (Ang II) in the cardiovascular system has been highlighted; however, its detailed signaling mechanism is poorly understood. Here, we examined the cross-talk of growth-promoting signaling between Aldo and Ang II in vascular smooth muscle cells (VSMC). Treatment with a lower dose of Aldo (10(-12) mol/L) and with a lower dose of Ang II (10(-10) mol/L) significantly enhanced DNA synthesis, whereas Aldo or Ang II alone at these doses did not affect VSMC proliferation. This effect of a combination of Aldo and Ang II was markedly inhibited by

Design and synthesis of 3,4-seco-lupane triterpene-tryptamine derivatives and revealing their anti-bladder cancer mechanisms by combining TCGA and transcriptomic approaches.

Bladder cancer is the most common malignant tumor of the urinary tract. In this study, 90 lupane triterpene derivatives, previously synthesized in the laboratory, were systematically evaluated for their potential effects against bladder cancer by cytotoxicity screening against five urinary tumor cell lines. Bioinformatics and molecular dynamics methods were used to investigate the mechanism of action of compound 27 in depth. Most of the derivatives effectively inhibited tumor cell growth, and structure-activity relationship analysis revealed that introducing an indole moiety significantly enha

Co-Stimulation with TWEAK and TGF-β1 Induces Steroid-Insensitive TSLP and CCL5 Production in BEAS-2B Human Bronchial Epithelial Cells.

Steroid-resistant asthma is a common cause of refractory asthma. Type 2 inflammation is the main inflammatory response in asthma, and the mechanism underlying the steroid-resistance of type 2 inflammation has not been completely elucidated. Tumor-necrosis-factor-like apoptosis-inducing factor (TWEAK) and transforming growth factor (TGF)-β1 are involved in epithelial-mesenchymal transition (EMT) and the production of thymic stromal lymphopoietin (TSLP) and C-C motif chemokine ligand 5 (CCL5). We herein hypothesize that the combined exposure to TWEAK and TGF-β1 may result in the development of s
