Contriving multiepitope subunit vaccine by exploiting structural and nonstructural viral proteins to prevent Epstein-Barr virus-associated malignancy.

Cancer is one of the common lifestyle diseases and is considered to be the leading cause of death worldwide. Epstein-Barr virus (EBV)-infected individuals remain asymptomatic; but under certain stress conditions, EBV may lead to the development of cancers such as Burkitt's and Hodgkin's lymphoma and nasopharyngeal carcinoma. EBV-associated cancers result in a large number of deaths in Asian and African population, and no effective cure has still been developed. We, therefore, tried to devise a subunit vaccine with the help of immunoinformatic approaches that can be used for the prevention of E

Designing a Novel di-epitope Diphtheria Vaccine: A Rational Structural Immunoinformatics Approach.

BACKGROUND: The design of an epitope-based vaccine against diphtheria toxin (DTx) originated from the idea that many strong binder epitopes may be structurally located in the depth of DTx. Subsequently, many ineffective antibodies may be produced by the presentation of those epitopes to T and B lymphocytes. The other critical issue is the population coverage of a vaccine that has been neglected in traditional vaccines. OBJECTIVE: Given the issues above, our study aimed to design a peptide-based diphtheria vaccine, considering the issues of unwanted epitopes and population coverage. METHODS:

Immunoinformatics and structural aided approach to develop multi-epitope based subunit vaccine against Mycobacterium tuberculosis.

Tuberculosis is a highly contagious disease caused by Mycobacterium tuberculosis (Mtb), which is one of the prominent reasons for the death of millions worldwide. The bacterium has a substantially higher mortality rate than other bacterial diseases, and the rapid rise of drug-resistant strains only makes the situation more concerning. Currently, the only licensed vaccine BCG (Bacillus Calmette-Guérin) is ineffective in preventing adult pulmonary tuberculosis prophylaxis and latent tuberculosis re-activation. Therefore, there is a pressing need to find novel and safe vaccines that provide robus

Molecular Bases of Protein Antigenicity and Determinants of Immunogenicity, Anergy, and Mitogenicity.

BACKGROUND: The immune system is able to recognize substances that originate from inside or outside the body and are potentially harmful. Foreign substances that bind to immune system components exhibit antigenicity and are defined as antigens. The antigens exhibiting immunogenicity can induce innate or adaptive immune responses and give rise to humoral or cell-mediated immunity. The antigens exhibiting mitogenicity can cross-link cell membrane receptors on B and T lymphocytes leading to cell proliferation. All antigens vary greatly in physicochemical features such as biochemical nature, struc

The humoral immune response against an HLA class I allodeterminant correlates with the HLA-DR phenotype of the responder.

BACKGROUND: The genetic basis for control of alloantibody responses against foreign HLA histocompatibility antigens has never been delineated. The most likely postulate would be that HLA class II alloantigens of the host regulate the response through their ability to present processed HLA allopeptide fragments for the cognate interaction between CD4+ T lymphocytes and B lymphocytes that leads to IgG antibody synthesis. METHODS: We have analyzed our allosensitized transplant patient population with regard to humoral responsiveness to a serologically defined public HLA class I epitope, Bw4. Pep

Conversion of tyrosine to the inflammation-associated analog 3'-nitrotyrosine at either TCR- or MHC-contact positions can profoundly affect recognition of the MHC class I-restricted epitope of lymphocytic choriomeningitis virus glycoprotein 33 by CD8 T cells.

Immunohistochemical detection of increased levels of protein-associated nitrotyrosine has become widely used as a surrogate marker of in situ inflammation. However, the potential consequences of protein-associated nitrotyrosine formation in terms of cellular immune recognition has received surprisingly little attention. Using a well-defined I-E(K)-restricted epitope of pigeon cytochrome c, we previously demonstrated that conversion of a single tyrosine residue to nitrotyrosine can have a profound effect on recognition by CD4 T cells. In this study, we used the MHC class I-restricted epitope of

Autoantibodies to cryptic epitopes elicited by infection with lactate dehydrogenase-elevating virus.

Lactate dehydrogenase-elevating virus (LDV) produces a permanent infection in mice with a B-lymphocyte polyclonal activation leading to hypergammaglobulinaemia. Since LDV specifically suppressed antibodies to native epitopes in CBA/Ht, but not BALB/c, mice immunized against a protein antigen, we explored the relationship between such a change in antibody specificity and the expression of autoantibodies under the influence of LDV. Again in CBA/Ht, but not BALB/c, mice we observed another effect of LDV: the sera from infected CBA/Ht mice were found by enzyme-linked immunosorbant assay to contain

Pilot Studies Testing Novel Minimized Pan-Coronavirus (CoV) Vaccines in Feline Immunodeficiency Virus-Infected Cats With or Without Feline CoV Serotype-1 (FCoV1) Coinfection and in Specific-Pathogen-Free Cats Against Pathogenic FCoV2.

Background: The minimized pan-coronavirus (CoV) vaccine-1 developed by our laboratory contained pDNA sequences of feline coronavirus serotype-1 (FCoV1) and SARS-CoV2 (SCoV2) spike B-cell epitopes plus FCoV/SCoV2-conserved, CoV-specific polymerase cytotoxic T-lymphocyte (CTL) epitopes formulated in lipid nanoparticle (LNP). Only FCoV2 infects feline cell lines needed for developing native challenge inoculum that causes feline infectious peritonitis (FIP). Hence, Pilot Study 1 evaluated the therapeutic efficacy and safety of the pan-CoV vaccine-1 in feline immunodeficiency virus (FIV)-infected c

Bioinformatics designing of an mRNA vaccine for Mokola virus (MOKV) using immunoinformatics as a secure strategy for successful vaccine development.

The Mokola Virus belongs to the family Rhabdoviridae and is genotype 3 of the Lyssavirus genera. A small number of cases of animal and human encephalomyelitis, mainly scattered over sub-Saharan Africa, have been linked to the Mokola Virus (MOKV). Currently there is no vaccine to protect against MOKV infection in people or animals. It has been proven that rabies vaccination does not confer immunity against MOKV infection, even though MOKV and the rabies virus are related. Using immunoinformatics approaches, this study designed an mRNA vaccine that can protect against all the five glycoproteins

Therapeutic efficacy of the multi-epitope vaccine CTB-UE against Helicobacter pylori infection in a Mongolian gerbil model and its microRNA-155-associated immuno-protective mechanism.

Vaccination is an effective means of preventing infectious diseases, including those caused by Helicobacter pylori. In this study, we constructed a novel multi-epitope vaccine, CTB-UE, composed of the cholera toxin B subunit and tandem copies of the B and Th cell epitopes from the H. pylori urease A and B subunits. We evaluated the therapeutic efficacy of the multi-epitope vaccine CTB-UE against H. pylori infection in a Mongolian gerbil model and studied its immuno-protective mechanisms. The experimental results indicated that urease activity, H. pylori colonisation density, the levels of IL-8

Molecular characterization and immunoinformatics-based design of a multi-epitope vaccine against Staphylococcus nepalensis.

Staphylococcus nepalensis has recently been identified in clinical settings, raising questions regarding its potential role as an emerging opportunistic pathogen. However, no vaccine-based preventive strategy has yet been proposed against this bacterium. The objective of this study was to characterize a clinical isolate using PCR-based molecular identification and to design a rational multi-epitope vaccine candidate against S. nepalensis through an integrated immunoinformatics-driven reverse vaccinology approach. Species-level identification was performed by PCR amplification and sequencing of

Protection Against Helicobacter pylori Infection in BALB/c Mouse Model by Oral Administration of Multivalent Epitope-Based Vaccine of Cholera Toxin B Subunit-HUUC.

Vaccination is an increasingly important alternative approach to control Helicobacter pylori infection, since H. pylori resistance to previously efficacious antibiotic regimens is increased, and H. pylori eradication treatment for upper gastrointestinal diseases is becoming less successful. Fortunately, an efficient oral monovalent H. pylori vaccine has been developed. However, compared with monovalent vaccines, multivalent vaccines have the potential to induce more effective and comprehensive protection against H. pylori infection. In this study, we designed and produced a multivalent epitope

A human monoclonal antibody, produced following in vitro immunization, recognizing an epitope shared by HLA-A2 subtypes and HLA-A28.

In vitro immunization and subsequent immortalization of peripheral blood cells of a multiparous woman has resulted in the production of a stable human mouse heterohybridoma, 5C2A2, secreting an HLA-A2/A28-specific human monoclonal antibody. Although possibly exposed to HLA-A2 by transfusions, the cell donor showed no HLA-A2-specific serum antibodies. The present protocol for in vitro immunization includes the elimination of suppressor cells from the responder cell population, the presence of irradiated allogeneic lymphocytes as a source of antigen, as well as stimuli--recombinant interleukin-2

Construction, expression, and characterization of a multi-epitope protein based on envelope glycoproteins B, C, and D of Varicellovirus bovinealpha1/5 in Escherichia coli.

Varicellovirus bovinealpha1 (BoAHV-1) and 5 (BoAHV-5) are important pathogens associated with respiratory, reproductive, and neurological disorders in cattle, remaining globally relevant since their first reports in the 1950s and 1960s. Envelope glycoproteins B, C, and D play critical roles in viral attachment and fusion, making them key targets for immune responses and promising candidates for vaccine development. This study aimed to design a multi-epitope protein using immunoinformatic approaches, incorporating highly conserved, antigenic B- and T- cell epitopes with strong predicted MHC-bin

Next-generation multi-epitope subunit vaccine design: A computational approach utilizing two stable proteins to combat Human Metapneumovirus (HMPV).

Human Metapneumovirus (HMPV) has become a major contributor to acute respiratory infections, particularly in young children, the elderly, and individuals with weakened immune systems. It leads to considerable hospitalizations, morbidity, and treatment costs. Given its clinical impact, the development of novel vaccines against HMPV is essential. This study focused on employing immunoinformatics techniques to produce a multi-epitope vaccine. Multi-epitope vaccines may provide broader protection than live attenuated vaccines by targeting multiple viral strains. Using the NCBI database, consensus

Design of multi-epitope chimeric vaccine against Monkeypox virus and SARS-CoV-2: A vaccinomics perspective.

The current era we experience is full with pandemic infectious agents that no longer threatens the major local source but the whole globe. Almost the most emerging infectious agents are severe acute respiratory syndrome coronavirus-2 (SARS CoV-2), followed by monkeypox virus (MPXV). Since no approved antiviral drugs nor licensed active vaccines are yet available, we aimed to utilize immunoinformatics approach to design chimeric vaccine against the two mentioned viruses. This is the first study to deal with design divalent vaccine against SARS-CoV-2 and MPXV. ORF8, E and M proteins from Omicron

Reverse vaccinology-based design of a universal multiepitope vaccine against chikungunya virus: Phylogenetic and immunoinformatics approaches.

Chikungunya virus (CHIKV) infection is a re-emerging arboviral disease in tropical and subtropical regions. In addition to acute febrile syndrome, CHIKV infection may lead to chronic articular manifestations that significantly affect a long-term quality of life. This study aimed to design a universal vaccine candidate covering all circulating genotypes of CHIKV based on conserved multiepitope platform. We employed a large scale phylogenetic and immunoinformatic approach to identify conserved regions of the open reading frames (ORF2) region encoding viral structural proteins. This study ultimat

In silico identification of Leishmania GP63 protein epitopes to generate a new vaccine antigen against leishmaniasis.

BACKGROUND: The surface of Leishmania spp. presents glycoprotein 63 (GP63), a metalloprotease that acts as one of the parasite's major antigens. A vaccine against leishmaniasis has not yet been developed and stationary phase promastigotes have utmost importance in transmitting Leishmania spp. from phlebotomine sand fly to humans or reservoirs. Therefore, this study aimed to analyze GP63 protein in three different Leishmania spp. to determine new vaccine candidate antigen against leishmaniasis using sequencing data of locally detected Leishmania strains and in silico approaches.
