The Non-Coding Landscape of Cutaneous Malignant Melanoma: A Possible Route to Efficient Targeted Therapy.

Considered to be highly lethal if not diagnosed in early stages, cutaneous malignant melanoma is among the most aggressive and treatment-resistant human cancers, and its incidence continues to rise, largely due to ultraviolet radiation exposure, which is the main carcinogenic factor. Over the years, researchers have started to unveil the molecular mechanisms by which malignant melanoma can be triggered and sustained, in order to establish specific, reliable biomarkers that could aid the prognosis and diagnosis of this fatal disease, and serve as targets for development of novel efficient thera

Reversible methylation of m6Am in the 5' cap controls mRNA stability.

Internal bases in mRNA can be subjected to modifications that influence the fate of mRNA in cells. One of the most prevalent modified bases is found at the 5' end of mRNA, at the first encoded nucleotide adjacent to the 7-methylguanosine cap. Here we show that this nucleotide, N6,2'-O-dimethyladenosine (m6Am), is a reversible modification that influences cellular mRNA fate. Using a transcriptome-wide map of m6Am we find that m6Am-initiated transcripts are markedly more stable than mRNAs that begin with other nucleotides. We show that the enhanced stability of m6Am-initiated transcripts is due

MiREDiBase: A Comprehensive Resource for miRNA Editing Events.

MicroRNA (miRNA) editing introduces a new layer of complexity in post-transcriptional gene regulation, with potential implications for miRNA biogenesis, target specificity, and function. However, the lack of a dedicated repository to comprehensively catalog miRNA editing events has hindered advances in the field. MiREDiBase addresses this gap by offering a meticulously curated database focused exclusively on validated, high-confidence putative miRNA editing events, with emphasis on A-to-I (adenosine-to-inosine) and C-to-U (cytidine-to-uridine) editing. This resource provides extensive annotati

RNA methylation in urological cancers: regulatory logic, biological functions, and clinical relevance.

Urological malignancies, including prostate, bladder, and renal cancers, remain a major clinical challenge because of tumor heterogeneity, disease progression, and therapeutic resistance. In this context, RNA methylation has emerged as an important post-transcriptional regulatory layer in cancer biology. Importantly, the biological consequences of RNA methylation are not dictated by chemical modifications alone, but by coordinated networks of regulatory proteins that install, remove, and interpret these marks, thereby shaping RNA fate and gene expression programs. Among currently studied RNA m

RNA Modifications as Molecular Regulators of Alveolar Epithelial Injury and Aberrant Repair in Pulmonary Fibrosis.

Pulmonary fibrosis is a progressive interstitial lung disease characterized by persistent alveolar epithelial injury, aberrant repair, and excessive extracellular matrix deposition. Increasing evidence indicates that disease progression is closely associated with alveolar type II (AT2) cell dysfunction, impaired AT2-to-AT1 differentiation, and the persistence of transitional epithelial populations, including KRT8+ intermediate populations. Because the formation and resolution of these transitional epithelial populations require dynamic regulation of stress-responsive transcripts and differenti

Multidimensional regulatory mechanisms and translational potential of epigenetic networks in the rheumatoid arthritis disease course.

Rheumatoid arthritis (RA) is an autoimmune disease characterized by chronic synovitis that may progress to irreversible joint destruction and disability, thereby substantially impairing quality of life. RA results from complex interactions among genetic predisposition, environmental exposures, and immune dysregulation; however, current therapies are not curative, and many patients continue to experience pain, morning stiffness, and recurrent inflammation. In recent years, epigenetic mechanisms have emerged as key modulators of RA heterogeneity and disease persistence. Reversible regulatory lay

Crosstalk between m6A modification and metabolic reprogramming in colorectal cancer: from the immune microenvironment to clinical applications.

N6-methyladenosine (m6A) modification is the most common epigenetic alteration in eukaryotic mRNA, significantly impacting metabolic reprogramming and the tumor microenvironment (TME) of colorectal cancer (CRC) by dynamically regulating RNA metabolic processes. Recent studies indicate that m6A modification interacts with CRC metabolic reprogramming, fostering an immunosuppressive TME. Specifically, m6A modification promotes CRC progression by regulating glucose, lipid, and amino acid metabolism while inhibiting the activity of antitumor immune cells (such as T cells, natural killer cells, and

Targeting the METTL5-MGST1 axis for cancer therapy: molecular mechanisms and therapeutic implications.

The epitranscriptomic regulation of cancer represents one of the most rapidly expanding frontiers in oncology. Among the newly characterized epigenetic axes, the Methyltransferase-like protein 5-Microsomal Glutathione S-Transferase 1 (METTL5-MGST1) signaling axis has emerged as a mechanistically compelling circuit implicated in tumor progression, ferroptosis resistance, immune evasion, and treatment-refractory behavior, most directly established in hepatocellular carcinoma and increasingly recognized, though through varying and not always MGST1-dependent mechanisms, across other cancer types.

Uncovering a role for METTL13 in malignant transformation of human hematopoietic stem cells and in the progression of pediatric leukemia.

Post-transcriptional RNA modifications, such as N6-methyladenosine (m6A) methylation and adenosine to inosine (A-to-I) editing, are critical regulators of hematopoietic stem cell (HSC) self-renewal and differentiation, yet their precise contributions to malignant transformation are not fully elucidated. In this study, we uncovered the epitranscriptomic landscape caused by knockdown of genes from the methyltransferase (METTL)-family in hematopoietic stem and progenitor cells (HSPCs). We identified both converging and distinct effects of METTL3 and METTL14, known members of the m6A writer comple

Harnessing a single molecule for dual bioorthogonal regulation of RNA function and m6A methylation.

This study presents a multifunctional RNA regulation strategy that enables RNA molecules to undergo both bioorthogonal ligation and cleavage reactions within the same system. Using guide RNA (gRNA) as an example, we demonstrate on-demand inactivation via click chemistry and subsequent reactivation by light exposure. Applied to CRISPR-mediated, site-specific RNA methylation, this technology enables continuous, multistep programmable control, overcoming the one-way limitation of traditional methods. Results show that gRNA can stably tolerate both azide and photodegradable groups, facilitating ef

DNA polymorphisms and N6-methyladenosine RNA methylation influencing microRNA expression and regulatory activity in plants.

Plant subspecies that have colonized distinct natural habitats must evolve different phenotypic and physiological features to ensure survival. However, the underlying molecular mechanisms remain poorly understood. In this study, we selected seven Arabidopsis accessions and six rice varieties with significantly different natural habitat parameters to investigate the molecular mechanisms behind the differential expression and activity of microRNAs (miRNAs) at the subspecies level. Compared to Col-0, 24–35 differentially expressed miRNA precursors and 36–110 differentially expressed mature miRNAs

The role and therapeutic prospects of intercellular communication and RNA m6A modification in radiation-induced liver injury.

Radiation-induced liver injury (RILI) is a common and severe complication of radiotherapy for abdominal and thoracic malignancies. Ionizing radiation triggers widespread alterations of the N6-methyladenosine (m6A) modification landscape of hepatic RNA. Dysregulated m6A methylation disrupts hepatocyte homeostasis, immune cell activation and intercellular communication, and thus may contribute to the progression of RILI from acute inflammation to chronic hepatic fibrosis. Although intercellular crosstalk has been reported in RILI-related studies, its crosstalk with m6A modification remains poorl

Codon Usage and mRNA Stability are Translational Determinants of Cellular Response to Canonical Ferroptosis Inducers.

Ferroptosis is a non-apoptotic cell death mechanism characterized by the generation of lipid peroxides. While many effectors in the ferroptosis pathway have been mapped, its epitranscriptional regulation is not yet fully understood. Ferroptosis can be induced via system xCT inhibition (Class I) or GPX4 inhibition (Class II). Previous works have revealed important differences in cellular response to different ferroptosis inducers. Importantly, blocking mRNA transcription or translation appears to protect cells against Class I ferroptosis inducing agents but not Class II. In this work, we examin

MsMTA regulates growth and metabolism in alfalfa (Medicago sativa) through m6A-mediated stabilization of metabolic transcripts.

N6-methyladenosine (m6A) is a widespread RNA modification in eukaryotes, yet its contribution to growth and metabolic regulation in perennial leguminous forage crops remains poorly understood. Here, we identify MsMTA, a nuclear-localized m6A methyltransferase in alfalfa (Medicago sativa), as a key regulator of the global m6A landscape, plant architecture, and forage nutritional composition. Suppression of MsMTA expression reduced overall m6A levels and caused a pronounced dwarf phenotype. At the metabolic level, MsMTA knockdown lines showed a strong increase in soluble sugar content, accompani

Crosstalk between gut microbiota and RNA N6-methyladenosine modification in diabetic retinopathy.

Diabetic retinopathy (DR) is a leading cause of blindness in working-age adults, with a pathogenesis that extends far beyond chronic hyperglycemia. This narrative review synthesizes current evidence to construct a comprehensive model of DR that integrates metabolic memory, epigenetic regulation, and systemic factors such as the gut-retina axis. Specifically, this review focuses on the bidirectional crosstalk between gut microbiota dysbiosis and host RNA N6-methyladenosine (m6A) modification as the central mechanism linking these factors. We examine how gut microbiota dysbiosis contributes to t

Targeting Bromodomain-Containing Protein 9 in Human Uterine Fibroid Cells.

Bromodomain (BRD)-containing proteins are evolutionarily conserved protein-protein interaction modules involved in many biological processes. BRDs selectively recognize and bind to acetylated lysine residues, particularly in histones, and thereby have a crucial role in the regulation of gene expression. BRD protein dysfunction has been linked to many diseases, including tumorigenesis. Previously, we reported the critical role of BRD-containing protein 9 (BRD9) in the pathogenesis of UFs. The present study aimed to extend our previous finding and further understand the role of the BRD9 in UFs.

Integrative analyses of the RNA modification machinery reveal tissue- and cancer-specific signatures.

BACKGROUND: RNA modifications play central roles in cellular fate and differentiation. However, the machinery responsible for placing, removing, and recognizing more than 170 RNA modifications remains largely uncharacterized and poorly annotated, and we currently lack integrative studies that identify which RNA modification-related proteins (RMPs) may be dysregulated in each cancer type. RESULTS: Here, we perform a comprehensive annotation and evolutionary analysis of human RMPs, as well as an integrative analysis of their expression patterns across 32 tissues, 10 species, and 13,358 paired t

Biphasic Liquid Microjunction Extraction for Profiling Neuronal RNA Modifications by Liquid Chromatography-Tandem Mass Spectrometry.

RNA modifications are emerging as critical players in the spatiotemporal regulation of gene expression. Although liquid chromatography-tandem mass spectrometry (LC-MS/MS) enables the simultaneous quantification of numerous enzymatically modified RNAs in a biological sample, conventional RNA extraction and enzymatic digestion protocols that are employed prior to analysis have precluded the application of this technique for small-volume samples. In this study, a biphasic liquid microjunction (LMJ) extraction system using coaxial capillaries that direct and aspirate extraction solvents onto a ∼35
