Is family history related to preventive health behaviors and medical management in breast cancer patients?

INTRODUCTION: Women diagnosed with breast cancer who also have a family history of the disease are at increased risk of developing additional primary breast or ovarian cancers. We investigated whether a relationship exists between family history and health behaviors in a cross-sectional study of breast cancer survivors. METHODS: Participants in the Women's Healthy Eating and Living (WHEL) Study (a randomized trial designed to test the effect of a plant-based diet on breast cancer recurrence) completed baseline questionnaires about their family history and health behaviors. Medical records and

Relation of childhood socioeconomic status and family environment to adult metabolic functioning in the CARDIA study.

OBJECTIVE: Low SES and a conflict-ridden, neglectful, or harsh family environment in childhood have been linked to a high rate of physical health disorders in adulthood. The objective of the present investigation was to evaluate a model of the pathways that may help to explain these links and to relate them to metabolic functioning (MF) in the Coronary Artery Risk Development In Young Adults (CARDIA) dataset. METHODS: Participants (n = 3225) in the year 15 assessment of CARDIA, age 33 to 45 years, completed measures of childhood socioeconomic status (SES), risky early family environment (RF),

Inheritance of persistent foramen ovale and atrial septal defects and the relation to familial migraine with aura.

OBJECTIVE: To determine whether smaller atrial shunts (large persistent foramen ovale (PFO) and small atrial septal defect (ASD)) are inherited and whether this has a role in the inheritance of migraine with aura. METHODS: Contrast echocardiography was used to detect atrial shunts in 71 relatives of 20 probands with a significantly sized atrial shunt (large PFO or ASD). Four families with three generations, 14 families with two generations, and two sibships were studied. The contrast echocardiograms were performed blind to history of migraine. A consultant neurologist, who was blinded to card

Familial bilateral medial parietooccipital band heterotopia not related to DCX or LIS1 gene defects.

A father and his daughter displayed strictly similar focal brain dysplasia at MR examination, characterized by regional medial posterior laminar sub-cortical grey matter heterotopia. To our knowledge, no family presenting such anomalies has yet been described. LIS1 and DCX gene defects were excluded. Collecting patients with such inherited dysplasia should improve our knowledge of the genetic basis of cortical malformations.

Violence and hostility among families of Vietnam veterans with combat-related posttraumatic stress disorder.

The current study provides a portrait of emotional-behavioral functioning within a small sample of Vietnam veterans with combat-related posttraumatic stress disorder (PTSD), their partners, and older adolescent and adult children. Veterans, their partners and children reported moderate-low to moderate-high levels of violent behavior. In addition, partner and veteran hostility scores were elevated relative to gender and age matched norms. Partners also reported heightened levels of psychological maltreatment by veterans. Veterans' combat exposure was positively correlated with hostility and vio

Reptilian chemistry: characterization of a family of dianeackerone-related steroidal esters from a crocodile secretion.

The African dwarf crocodile, Osteolaemus tetraspis (Crocodilidae, Reptilia), possesses a pair of skin glands, the paracloacal glands, the secretion of which is thought to be used to mark nest sites or attract mates. Ten aromatic steroidal esters were isolated from this secretion and characterized on the basis of NMR spectroscopic investigations, electrospray ionization-MS analyses, and chemical degradation. These esters, which account for more than 90% of the paracloacal glandular secretion, are derived from either cholesterol or cholestanol, esterified with a C-20 or C-22 acid closely related

DNA-based prenatal diagnosis of a Korean family with tyrosinase-related oculocutaneous albinism (OCA1).

Tyrosinase-related oculocutaneous albinism (OCA1), an autosomal recessive inborn error of pigmentation, is caused by the deficiency of tyrosinase. We had previously identified two different mutations of the TYR gene in a four year old Korean male with mild OCA; a P310insC frameshift in exon 2 and an IVS2-7t-->a,-10-11deltt splice junction mutation in exon 3. Here we report a prenatal diagnostic study of a subsequent fetus of the above family that was at 25% risk of OCA1. SSCP/heteroduplex screening, restriction enzyme digestion, and allele-specific oligonucleotide hybridization analyses of DNA

Pathobiology of human familial hypercholesterolaemia and a related animal model, the Watanabe heritable hyperlipidaemic rabbit.

Defective expression of low density lipoprotein (LDL) receptors is the basic genetic defect in human familial hypercholesterolaemia (FH) and its animal counterpart, the Watanabe heritable hyperlipidaemic (WHHL) rabbit. The pathologic manifestations of human FH were evaluated based on the study of material from six subjects with homozygous FH and a review of the literature. This analysis indicated that homozygous FH is characterized by accelerated atherosclerosis and prominent lipid accumulation in macrophages and other stromal cells of the aortic and mitral valves, skin, tendon, and, variably,

The molecular basis of hereditary 1,25-dihydroxyvitamin D3 resistant rickets in seven related families.

Hereditary 1,25-dihydroxyvitamin D3 [1,25(OH)2D3] resistant rickets (HVDRR) is an autosomal recessive disease caused by target organ resistance to the action of 1,25(OH)2D3, the active form of the hormone. The defect in target cells is heterogenous and commonly appears to be a mutation in the gene encoding the vitamin D receptor (VDR). We have studied cultured skin fibroblasts and Epstein-Barr virus transformed lymphoblasts of seven family branches of an extended kindred having eight children affected with HVDRR. We have previously shown that cells from three affected children in this group co

[Genetic aspects of hypertrophic cardiomyopathy (familial studies and relation of HLA to obstructive hypertrophic cardiomyopathy)].

The purpose of the paper was to study the families of 31 patients with hypertrophic cardiomyopathy (HCMP) to establish the hereditary nature of disease as well as to study the peculiarities of distribution of HLA-A, B and DR antigens in 44 patients (Russians) with obstructive HCMP using a method of histocompatibility typing. Echocardiographic investigation of 105 relatives of 31 patients with various types of HCMP revealed 32 patients with the same pathology among 84 persons in 20 families suggesting the familial pattern of disease. Antigen markers of predisposition to obstructive HCMP were es

[Constitutional protein C deficiency in 57 patients from 22 non-related families].

A congenital deficiency in protein C (physiological inhibitor of coagulation) was identified in 57 patients: the deficiency was quantitative (type I) in 20 families, qualitative (type II) in two families. The transmission was autosomal dominant in 21 families but was suspected to be recessive in one family: the 18 years old homozygous propositus has a severe deficiency (protein C = 16 p. 100): both parents are heterozygous (consanguinity was present) and 5 other family members with heterozygous deficiency are asymptomatic. In the 49 patients (25 women, 24 men) belonging to the 21 other familie

Inherited erythrocyte pyruvate kinase deficiency: Studies on 15 members of two related families.

The case of a haemolytic non-spherocytic anaemia with pyruvate kinase (PK) deficiency is reported. The investigation concerns two families with a low level of PK. In the propositus and in the members of his family we have also examined the behaviour of some enzymes and the concentration of red cell metabolites. We confirm the heterogeneity of the manifestation and agree that anaemias due to PK deficiency are complex forms in which the enzyme defect is only one of the symptoms.

Co-Designing a Family e-Hub: A Systemic, Relational, Digital Mental Health Resource.

Access to family and systemic therapies remains limited. Digital mental health interventions offer potential to extend access; however, most are designed for individual users, presenting challenges when engaging multiple members of a family. This study used a participatory co-design approach to inform the development of the Family e-Hub, a systemic, self-guided digital resource grounded in family-specific design principles. Drawing on the perspectives of consumers with lived experience of family therapy and family therapists, the study aimed to co-design recommendations for Family e-Hub conten

The impact of family-related psychosocial factors on suicidal ideation among adolescents: a systematic review.

BACKGROUND: This systematic literature review investigates the impact of family-related psychosocial factors on suicidal ideation. Adolescents experiencing family-related adversities are more likely to experience suicidal ideation. Given that adolescents are a high-risk population, understanding the nature of the impact of family-related factors is important. It would help determine and differentiate the family-specific risk and protective factors and aid in designing targeted suicide prevention strategies. METHOD: The review followed Preferred Reporting Items for Systematic Reviews and Meta-

Feasibility study exploring sleep impacts among families in the aftermath of firearm-related trauma.

OBJECTIVE: The ripple effects of gun violence are far reaching, particularly for children impacted by this trauma. Many of the available sources of mental health support in the wake of gun violence do not address disrupted sleep, which commonly develops as a consequence of loss. METHOD: We recruited adults for focus groups who are caregivers to a child or young adult impacted by loss due to gun violence from the Victim Support Services department of a police department in the US Southeast. The children to whom the adult provides care were required to be 13 years of age or younger. The aim of

Expanding mutational and phenotypic spectrum of CST3-related leukoencephalopathy: a novel family and literature review.

CST3-related leukoencephalopathy is a recently recognized adult-onset neurodegenerative disorder caused by pathogenic CST3 variants with C-terminal truncations, frameshifts, or deletions. Owing to the limited number of reported cases, its clinical and radiological spectrum remains incompletely defined. Here, we report a Chinese family harboring a novel heterozygous nonsense variant of CST3 (c.327C > A; p.Cys109*), which represents the most 5’ pathogenic mutation identified to date. The proband, a 62-year-old woman, presented with recurrent encephalopathic episodes, progressive cognitive declin

Phenotypic Variations in a Large Family with Dominant Optic Atrophy Related to a Novel OPA1 Deletion.

PURPOSE: Dominant optic atrophy (DOA) is a rare disease characterized by the chronic loss of retinal ganglion cells that transduce the visual information from the retina to the brain. Dominant optic atrophy shows interfamilial and intrafamilial phenotypic variations and a restricted 35% molecular diagnosis, with half presenting a pathogenic variant in optic atrophy 1 (OPA1), encoding a large mitochondrial GTPase. Here, we describe the largest ever identified DOA family, with 64 nonsyndromic individuals harboring a novel 10-kb OPA1 deletion. DESIGN: Retrospective, longitudinal cohort study of

MMP13-related Metaphyseal anadysplasia type 1 presenting with rickets-like manifestations in a family.

BACKGROUND: Metaphyseal anadysplasia 1, which includes Spondyloepimetaphyseal dysplasia Missouri type, is a rare autosomal dominant skeletal dysplasia characterized by short stature, mild limb deformities, and transient metaphyseal irregularities that typically resolve with age. The condition is caused by heterozygous missense variants in the MMP13 gene, encoding matrix metalloproteinase 13, a key enzyme in endochondral ossification and extracellular matrix remodeling. Pathogenic variants in MMP13 are exceedingly rare, with only a few families reported. CASE PRESENTATION: We report two siblin

Phenotypic and Molecular Features of a Large ODDD Family: Expanding the Spectrum of CX43-Related Disorder.

We present a family of five siblings who came to our attention with a clinical and radiological diagnosis of familial hypomyelinating leukodystrophy. Despite brain white matter abnormalities being present in all siblings, the clinical phenotype was variable: the three brothers presented with a clear-cut late-onset spastic paraplegia, whereas the two sisters displayed only mild pyramidal signs. Molecular analysis revealed a single relevant variant shared by all affected siblings, namely the likely pathogenic variant c.659C>T (p.Ser220Phe) in the GJA1 gene. Variants in this gene are generally as
