Altered excitatory and inhibitory inputs to striatal medium-sized spiny neurons and cortical pyramidal neurons in the Q175 mouse model of Huntington's disease.

The Q175 knockin mouse model of Huntington's disease (HD) carries a CAG trinucleotide expansion of the human mutant huntingtin allele in its native mouse genomic context and recapitulates the genotype more closely than transgenic models. In this study we examined the progression of changes in intrinsic membrane properties and excitatory and inhibitory synaptic transmission, using whole cell patch-clamp recordings of medium-sized spiny neurons (MSNs) in the dorsolateral striatum and cortical pyramidal neurons (CPNs) in layers 2/3 of the primary motor cortex in brain slices from heterozygous (Q1

In Vivo Imaging of the Central and Peripheral Effects of Sleep Deprivation and Suprachiasmatic Nuclei Lesion on PERIOD-2 Protein in Mice.

STUDY OBJECTIVES: That sleep deprivation increases the brain expression of various clock genes has been well documented. Based on these and other findings we hypothesized that clock genes not only underlie circadian rhythm generation but are also implicated in sleep homeostasis. However, long time lags have been reported between the changes in the clock gene messenger RNA levels and their encoded proteins. It is therefore crucial to establish whether also protein levels increase within the time frame known to activate a homeostatic sleep response. We report on the central and peripheral effect

Altered sensitivity of cerebellar granule cells to glutamate receptor overactivation in the Cln3(Δex7/8)-knock-in mouse model of juvenile neuronal ceroid lipofuscinosis.

The juvenile onset form of neuronal ceroid lipofuscinoses (JNCL) is a recessively inherited lysosomal storage disorder characterized by progressive neurodegeneration. JNCL results from mutations in the CLN3 gene that encodes a lysosomal membrane protein with unknown function. Utilizing a Cln3-knock-out mouse model of JNCL that was created on the 129S6/SvEv genetic background, we have previously demonstrated that CLN3-deficient cerebellar granule cells (CGCs) have a selectively increased sensitivity to AMPA-type glutamate receptor-mediated toxicity. Our recent findings that CGCs from 129S6/SvEv

Complete activation of thyroid hormone receptor β by T3 is essential for normal cochlear function and morphology in mice.

BACKGROUND/AIMS: Thyroid hormones (THs) regulate many developmental processes, including the developmental onset of cochlear differentiation and function. TH action is mediated mostly by triiodothyronine (T3) bound to thyroid hormone nuclear receptors (TRs). At positive regulated genes and in the absence of THs, nuclear co-repressors are bound to TRs and decrease basal transcription rate. Ligand (T(3)) binding results in the dissociation of co-repressors and the recruitment of co-activators to the complex, which results in full transcriptional activation. METHODS: We measured cochlear functio

Lung growth and development.

The organogenesis of lung involves several complex mechanisms, including interactions between cells originating from two germ layers--endoderm and mesoderm. Regulation of lung branching morphogenesis with reference to its architecture, growth pattern, differentiation, interactions between epithelium and mesenchyme and / or endothelium, as well as genes regulating these processes have been addressed by the pulmonary biologists through careful molecular biology and genetic experimental approaches. The mammalian lung develops by outpouching from the foregut endoderm as two lung buds into the surr

Glial cytokine modulation improves sleep and circadian disruption in female SAA knock-in mice of Alzheimer's-related pathology.

INTRODUCTION: Sleep and circadian disturbances are early Alzheimer's disease (AD) features, yet mechanisms linking amyloid pathology, neuroinflammation, and sex differences remain unclear. METHODS: We longitudinally assessed sleep, circadian rhythms, and cognition in female and male hAPPSAA knock-in and control mice from 2 to 19 months using piezoelectric monitoring. Aged mice (15 months) received MW151, a glial cytokine inhibitor (2.5 mg/kg, every other day, 6 weeks). RESULTS: Only females exhibited midlife reductions in light-phase sleep, increased rhythm fragmentation, and reduced rhythm

Insulin resistance alters cortical inhibitory neurons and microglia to exacerbate Alzheimer's knock-in mouse phenotypes.

BACKGROUND: Metabolic dysfunction contributes to the risk and progression of Alzheimer's disease (AD), yet the cellular mechanisms linking impaired insulin signaling and systemic metabolic stress to brain dysfunction remain incompletely defined. METHODS: We examined the impact of chronic high-fat, high-sugar (HFHS)-induced insulin resistance on metabolic parameters, spatial learning and memory, and in vivo glial activation and neuropathology in Alzheimer's disease knock-in mice expressing human mutant APP and wild-type (WT) tau. Single-nucleus RNA sequencing was performed to resolve cell-type

Impaired spatial coding and neuronal hyperactivity in the medial entorhinal cortex of aged APP knock-in mice.

Advanced amyloid beta (Aβ) pathology is associated with aberrant neuronal network activity and cognitive impairment in preclinical Alzheimer's disease (AD) models. Here, we assess Aβ pathology's impact on spatial information processing in the medial entorhinal cortex (MEC) of 18-month AppNL-G-F/NL-G-F knock-in (APP KI) mice during exploration of open field arenas. Spatial information scores are decreased in APP KI MEC neurons versus age-matched controls. Border cell firing preferences are unstable across sessions and grid cell spatial periodicity is disrupted. Ratemap stability analysis using

Altered Craniofacial and Bone Morphology in Hyperthyroid Knock-In Mice Carrying TSHR D633H and M453T Mutations.

BACKGROUND: Constitutively active mutations (CAMs) in the thyrotropin receptor (TSHR) are the major cause of nonautoimmune hyperthyroidism. TSHR is a key regulator of thyroid hormone synthesis, which is essential for skeletal formation, bone turnover, and craniofacial development. In addition to its role in thyroid hormone regulation, thyrotropin and its receptor have been proposed to independently influence bone formation and resorption. However, the specific effects of constitutively active TSHR mutations on cranial and skeletal development have not been investigated in mouse models. METHOD

Macrophage recruitment is a hallmark of non-syndromic myxomatous mitral valve disease.

AIMS: Myxomatous mitral valve disease (MMVD) is a progressive cardiac disease characterized by extracellular matrix remodelling, leaflet thickening, and proteoglycan accumulation. Despite its significant contribution to cardiovascular morbidity and mortality, the underlying pathophysiological mechanisms involved in its development and progression remain unclear. Recent investigations have revealed a genetic component to the disease, such as mutations in the FLNA gene, and suggested potential involvement of immune cells in the pathogenesis of MMVD. In this study, we investigated the role of mac

Epm2bP71A and Epm2bD148N knock-in mouse models of Lafora disease exhibit distinct and pronounced neurological alterations.

Lafora disease is a rare, autosomal recessive neurodegenerative disorder characterized by the progressive accumulation of abnormal, insoluble, and hyperphosphorylated forms of glycogen, known as Lafora bodies, in the brain and other tissues. The disease typically manifests during early adolescence with myoclonus, seizures, and rapidly progressive cognitive decline, ultimately leading to severe neurological deterioration and death within a decade of onset. Mutations in the EPM2A or EPM2B genes, which encode the proteins laforin and malin-key regulators of glycogen metabolism-are the underlying

Targeting the cGAS-STING pathway mitigates Huntington disease pathogenesis in a knock-in mouse model.

Cyclic GMP-AMP synthase (cGAS) and its downstream effector, stimulator of interferon genes (STING), form a key cytosolic DNA-sensing pathway that drives innate immune activation and proinflammatory signaling. We previously showed that cGAS is upregulated in Huntington disease (HD) cellular models, where it regulates autophagy and inflammation; however, its in vivo role remained unclear. Here, we genetically ablated cGAS in Q175DN knock-in HD mice and performed longitudinal behavioral assessments from 2 to 14 mo of age. cGAS deletion significantly improved HD-associated motor deficits, includin

Preventing MALT1-mediated CYLD cleavage induces intestinal dysbiosis and reduces EAE severity.

The paracaspase MALT1 is essential for lymphocyte activation and also plays roles in non-immune cells and cancer. Its protease activity regulates immune signaling by cleaving specific substrates, making it a promising therapeutic target. However, broad inhibition of MALT1 protease activity causes multiorgan inflammation in mice, highlighting the need to understand the effects of individual substrate cleavage. We generated CYLD(R321A) knock-in mice expressing a MALT1-resistant form of the deubiquitinase CYLD. These mice are healthy, with normal lymphocyte development and preserved immune signal

Targeting pericytes in the retina: characterization of the inducible NG2-CreERT2 knock-in mouse model.

Pericytes (PCs) are mural cells embedded in a common basement membrane with endothelial cells (ECs) on capillaries. They are key players in the regulation of stability and function of blood vessels and are an essential component of the blood-brain and blood-retinal barrier. In addition, PCs are reported to be involved in wound healing and tissue regeneration and represent a promising target for the modulation of scarring processes. Due to their heterogeneous expression pattern, a single specific molecular marker for PCs remains elusive. Inducible reporter mouse models represent a frequently us

Alpha-Synuclein Phosphomimetic Y39E and S129D Knock-In Mice Show Cytosolic Alpha-Synuclein Localization without Developing Neurodegeneration or Motor Deficits.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by motor and nonmotor symptoms. Its pathological hallmarks include the accumulation of misfolded alpha-synuclein (α-Syn) in Lewy bodies and Lewy neurites. Phosphorylation of α-Syn is a prominent feature of these inclusions, but its role in disease pathogenesis remains unclear. To identify the role of α-Syn phosphorylation in synucleinopathy, we generated two Snca knock-in (KI) mouse models carrying phosphomimetic mutations at SncaY39 or SncaS129 (SncaY39E or SncaS129D ) which manipulated epitopes phosphorylated

In vivo antibody diversification targeting a conserved coronavirus epitope.

To explore the use of human B cell receptor (BCR) knock-in mice for broadening antibody responses, we diversified CR3022, a monoclonal antibody (mAb) originally identified in a 2003 severe acute respiratory syndrome coronavirus (SARS-CoV) convalescent patient. This mAb targets a conserved epitope on the coronavirus receptor-binding domain (RBD). We took advantage of high- and low-affinity CR3022 BCR knock-in mice and immunized them with SARS-CoV-2 Wuhan RBD trimers to expand the breadth of these antibodies toward this virus. The resulting antibodies retained the ability to neutralize SARS-CoV

Isoflurane activates the type 1 ryanodine receptor to induce anesthesia in mice.

Inhaled anesthetics were first introduced into clinical use in the 1840s. Molecular and transgenic animal studies indicate that inhaled anesthetics act through several ion channels, including γ-aminobutyric acid type A receptors (GABAARs) and two-pore domain K+ (K2P) channels, but other targets may mediate anesthetic effects. Mutations in the type 1 ryanodine receptor (RyR1), which is a calcium release channel on the endoplasmic reticulum membrane, are relevant to malignant hyperthermia, a condition that can be induced by inhaled anesthetics. However, it was previously uncertain whether inhale

Selective striatal pathological changes in a novel human HTT exon 1 knock-in mouse model of Huntington's disease.

Huntington's disease (HD) is an autosomal-dominant neurodegenerative disorder characterized by progressive motor deficits, cognitive decline, and psychiatric disturbances caused by expanded CAG repeats in the huntingtin gene (HTT). Despite the development of various animal models, achieving a comprehensive model that closely replicates the biological mechanisms in order to test therapeutic modalities remains a challenge. Here, we describe a novel human HTT exon 1 knock-in (HEKI-150Q) mouse model that incorporates a 3.465-kb human HTT sequence with 150 polyglutamine and successfully mimics key

Characterization of Ucp1-iCre knockin mice reveals the recombination activity in male germ cells.

Ucp1 promoter-driven Cre transgenic mice are useful in the manipulation of gene expression specifically in thermogenic adipose tissues. However, the wildly used Ucp1-Cre line was generated by random insertion into the genome and showed ectopic activity in some tissues beyond adipose tissues. Here, we characterized a knockin mouse line Ucp1-iCre generated by targeting IRES-Cre cassette immediately downstream the stop codon of the Ucp1 gene. The Cre insertion had little to no effect on uncoupling protein 1 (UCP1) levels in brown adipose tissue. Ucp1-iCre mice of both genders exhibited normal the
